結合部位を変化または損傷させないので、チオエーテル
結合を介して磁性粒子に結合した抗体の特異性及び反応
性は、完全に保持されることが見出された。抗体が、上
記のように、純粋に吸着によりまたはそのアミ7基の反
応のどちらかにより磁気粒子に集まり、その結果コンジ
ュゲートされた抗体の特異性と反応性を損うことがある
という前記の従来の結合方法と比較した、本発明の特別
の利点がここKある。
磁気粒子へ化学的に結合し、そしてその結果、本発明の
磁気抗体コンジュゲートが例えば骨頭除去に用いられる
場合に、粒子から脱着しないという利点を有している。

溶媒中で、反応させ、一般式■ の化合物を得、それを最後に1タンパク質を変成させな
いような適当な水性塩含有溶媒、例えば生理食塩水また
はリン酸111食塩水中で、メルカプト基を担持するタ
ンパク質Pと反応させて式Iの化合物を得る。

換基2は、スクシンイミジルオキシの他に、ハロゲン、

ジング(ルケ社双)による除去により優られた結果を、
第1図中に例として示す。これには、BMA031(抗
−T−細胞受容体、(−リングベルケ社製)、BMA
O41またはBMAO81による各場合における未結合
の細胞集団を標識することによって測定された除去効率
が含まれる。FITC−標識第二抗体(前記参照)によ
るインキュベート後、各場合における蛍光強度を線形増
重を用いる細胞蛍光間接撮影機を用いて評価した。

@胞の増殖の検出 単核細胞を実施例7と同様にしてヒトの血痕から単離し
、次いで実施例7と同様にして未結合の磁気粒子(Bi
oMag M4100 、 Bebak )の懸濁液と
ともにインキュベートした。この磁気粒子をその後、永
久磁石を用いて除去した。残りのMNCをco2インキ
ュベーター中で、37℃において、64時間、ポークラ
イードマイトゲン(PWM、 Giboo ;1 :
3000 jl終希釈)または1000g/−植物凝集
素(PHA、 Welloome )の存在下で、無血
清培養基(Iaaove、−ニーリングmodifio
a、−tion)申告場合においてj X j O’
MNC/ウェルの濃度で、96−ウェル平底タイタープ
レート(Nuno )においてインキュベートした。4
8時間の培養時間後w、 14cmチミジン(Ams
rsham)2.7 X 10’ B(1を各培養基へ
加えた。さらに14詩間の椿養後に、細胞を細胞採集機
(Innotea )を用いて、ガラス1誠維フイルタ
ーデイスク上テの吸引により濾過した。フィルターの放
射能を、シンチレーション液の添加後にβ計数器(Pa
o−kard製)で測定した。
またはPWMによるMNCの刺激は、未結合ノtji気
粒子(BioMag M4100 、5ebakR)
Kヨル除去によって損なわれないことを明らかにしてい
る。

Claims (1)
- 【特許請求の範囲】 1)次の式 I ▲数式、化学式、表等があります▼ I (式中、Mはアミノ酸を担持する分散性の磁気的反応物
質又は粒子であり、Pは1またはそれ以上のメルカプト
基を担持するタンパク質であり、Xは化学的方法によつ
て2つのリガントに結合する有機化学構造である) の磁気タンパク質コンジュゲート。 2)式中Xが、場合によりその場合における適当な方法
により置換し得る脂肪族、芳香族、脂環式、脂環式−脂
肪族または芳香族−脂肪族スペーサーである請求項1に
記載の磁気タンパク質コンジュゲート。 3)Xが−(CH_2)_n−(式中、n=1〜8であ
り、好ましくはn=1〜5であり、特に好ましくはn=
2または5である)である請求項2に記載の磁気タンパ
ク質コンジュゲート。 4)Xが、フェニレンであり、2つのリガントがオルト
、メタまたはパラ位に存在し得る請求項2に記載の磁気
タンパク質コンジュゲート。 5)Xがフェニレン−(CH_2)_n−(式中n=1
〜5であり、好ましくはn=3である)であり、フェニ
レン基がスクシンイミジル基に結合する請求項2に記載
の磁気タンパク質コンジュゲート。 6)Xが−CHR−(式中Rがアミノ酸側鎖を示し、好
ましくはアミノ酸アラニン、セリン、スレオニンまたは
メチオニンの側鎖を示し、特に好ましくはアラニンの側
鎖を示す)である請求項2に記載の磁気タンパク質コン
ジュゲート。 7)Xがメチレンシクロヘキシルであり、メチレン基が
スクシンイミジル基に結合し、シクロヘキシル環に結合
したカルボニル基については、シクロヘキシル環の4位
に好適に結合する請求項2に記載の磁気タンパク質コン
ジュゲート。 8)Xが請求項4の定義と同様であるが、そのフェニレ
ン環はメチル、ヒドロキシル、メトキシ、アセトキシ、
ニトロもしくはシアノ基によるか、または塩素もしくは
臭素原子により置換されている請求項2に記載の磁気タ
ンパク質コンジュゲート。 9)タンパク質Pのメルカプト基が、自然状態で存在す
るか、ジスルフィド結合の還元によつて生ずるか、また
は化学反応により導入される請求項1に記載の磁気タン
パク質コンジュゲート。 10)Pが、免疫グロブリンまたは免疫グロブリン残基
で、好ましくはモノクローナル抗体、Fab、Fab′
もしくはF(ab′)_2フラグメント、抗原または酵
素、ホルモン、レクチンも しくは成長因子の残基である請求項1に 記載の磁気タンパク質コンジュゲート。 11)Pが、IgGまたはIgMクラスのモノクローナ
ル抗体を示す請求項10に記載の磁気タンパク質コンジ
ュゲート。 12)Pが水性塩溶液または体液中に溶解された状態で
存在する抗原に対抗するモノクローナル抗体を示す請求
項10に記載の磁気タンパク質コンジュゲート。 13)Pが、細胞、特に骨髄もしくはリンパ系または末
梢血液の細胞、特にBリンパ球、Tリンパ球、それらの
前駆細胞または腫瘍細胞、特に骨髄の腫瘍細胞上に発現
する抗原に対抗するモノクローナル抗体を示す請求項1
0に記載の磁気タンパク質コンジュゲート。 14)Pが細菌、マイコプラズマもしくは原生動物上ま
たはウィルス上に発現される抗原に対抗するモノクロー
ナル抗体を示す請求項13に記載の磁気タンパク質コン
ジュゲート。 15)Pが抗原を示す請求項1に記載の磁気タンパク質
コンジュゲート。 16)Mが、金属酸化物のコアとアミノ基を担持する包
被を有する分散性の粒子であり、上記金属酸化物コアが
、一群の常磁性物質を含有することができる請求項1に
記載の磁気タンパク質コンジュゲート。 17)粒子の径が、約0.1μ〜約100μであるが、
好ましくは約0.1μ〜約1.5μである請求項16に
記載の磁気タンパク質コンジュゲート。 18)Xが化学的または酵素的方法によつて分解され得
る請求項1に記載の磁気タンパク質コンジュゲート。 19)アミノ基を担持する磁気粒子Mを、アミノ基と反
応し式II ▲数式、化学式、表等があります▼II (式中、Zは反応性脱離基である) を有するマレイミドスペーサーと反応せしめ、アミノ結
合の生成を伴つて、式III ▲数式、化学式、表等があります▼III の化合物を得、それが最後にメルカプト基を担持するタ
ンパク質Pと反応して式 I の化合物を得ることからな
る式 I ▲数式、化学式、表等があります▼ I の磁気タンパク質コンジュゲートの製造方法。 20)水性塩溶液または体液から、溶解した抗原、抗体
、受容体、基質、補因子、炭水化物決定基を除去する方
法であつて、その溶液を式 I ▲数式、化学式、表等が
あります▼ I の適当な磁気タンパク質コンジュゲートとともにインキ
ュベートし、除去されるべき成分を特異的に吸着させた
後に、磁気的方法により磁気タンパク質コンジュゲート
を除去し、好適には、特異的に吸着した成分を再び磁気
タンパク質コンジュゲートから溶離することからなる方
法。 21)水性塩溶液または体液から細胞を除去する方法で
あつて、懸濁液を式 I ▲数式、化学式、表等があります▼ I の適当な磁気タンパク質コンジュゲートとともにインキ
ュベートし、除去されるべき成分を特異的に吸着した後
に、磁気的方法により磁気タンパク質コンジュゲートを
除去し、好的には、特異的に吸着した細胞または粒子を
磁気タンパク質コンジュゲートから再び脱着することか
らなる方法。 22)請求項1に記載の磁気タンパク質コンジュゲート
を使用して水性塩溶液または体液から細胞、可溶性抗原
、受容体、基質、補因子または炭水化物決定基を特異的
に除去する方法、または診断法の枠内で又は診断の補助
に使用する方法。 26)好適には骨髄除去またはHLA類型化のための請
求項13または14に定義された細胞を除去するための
請求項22に記載の磁気タンパク質コンジュゲートの使
用方法。
