Description
(1)標的細胞、例えば癌細胞に向かう改変(表面処理)免疫細胞のホーミングを増強し、それにより、環境、例えば腫瘍環境中の免疫細胞の密度を上昇させ、そして、
(2)腫瘍増殖を阻止、および/または癌細胞を死滅させ得ると考えられている。これらの2つの事象は、癌細胞死を相乗的に引き起こし、免疫系を活性化することができ、後者は、転移性癌細胞および/または循環腫瘍細胞を死滅させることができる。本発明は、本明細書に記載する標的化部分-薬物複合体(またはADC)、例えばT-DM1を特徴とするものに限定されない。現在市販されているものおよび開発中のものを含む任意の適当な抗体-薬物コンジュゲートまたは標的化部分-薬物複合体を、本明細書に記載の方法および組成物を用いて、免疫細胞(例えば、任意の免疫細胞)の表面上に取り込み得る。
特に断らない限り、本明細書で使用する全ての技術的および科学的用語は、開示する発明が属する当技術分野の当業者が共通して理解するのと同じ意味を有する。単数用語「a」、「an」、および「the」は、文脈上明らかにそうでない限り、複数形の参照対象を含む。同様に、語「または」は、文脈上明らかにそうでない限り、「および」を含むと意図される。用語「含む(comprising)」は、提示された定義されている要素に加えて、他の要素も存在し得ることを意味する。「含む」の使用は、限定ではなく包含を示す。言い換えれば、用語「含む」は、「主に含むが、それだけでなくてもよい」ことを意味する。さらに、語「含む」の変異形、例えば、「含む(comprise)」および「含む(comprises)」は、同様に同じ意味を有する。一観点において、本明細書に記載する組成物、方法およびその個々の構成要素に関連し、本発明に必須として本明細書に記載する技術は、特定されていない要素を、必須か否かに関わらず、なお受け入れる(「含む」)。
[0026]抗体および抗体断片:本明細書の用語「抗体」は、最も広義の意味で使用され、種々の抗体構造を包含し、モノクローナル抗体、ポリクローナル抗体、多重特異性抗体(例えば二重特異性抗体)および抗体断片が挙げられるが、これらに限定されない。「抗体」は、少なくとも軽鎖および重鎖イムノグロブリン可変領域を含み、抗原のエピトープに特異的に結合するペプチド(例えばポリペプチド)を指していてもよい。抗体は、コンジュゲートされていてもよい。抗体は、検出可能な標識、例えば酵素、ハプテン、またはフルオロフォアにより標識されていてもよい。「抗体断片」は、インタクトな抗体が結合する抗原に結合する、インタクトな抗体の一部を含む、インタクトな抗体とは異なる分子を指す。抗体断片の例としては、Fv、Fab、Fab’、Fab’-SH、F(ab’)2;ダイアボディ;線状抗体;一本鎖抗体分子(例えばscFv);および抗体断片から形成される多重特異性抗体が挙げられるが、これらに限定されない。
本発明は、標的癌化学免疫療法の開発のために、化学療法と免疫療法とを組み合わせる組成物を特徴とする。本明細書の組成物は、標的化部分-薬物複合体(例えば抗体-薬物コンジュゲート(ADC))を含み、これは、リン脂質-ポリエチレングリコール(PEG)リンカーを介して免疫細胞に結合されている。例えば、標的化部分-薬物複合体(例えばADC)は、リン脂質-PEGリンカーを免疫細胞の脂質二重層中に取り込むことにより、免疫細胞に結合していてもよい。
本発明はまた、本発明の組成物を調製する方法、例えば、標的化部分-薬物複合体を調製する方法、標的化部分-薬物複合体を細胞、例えば免疫細胞に連結させる方法等を特徴とする。一例として、DMPE-PEG-ADC(例えば、1,2-ジミリストイル-sn-グリセロ-3-ホスホエタノールアミン(DMPE)を抗体-薬物コンジュゲート(ADC)に連結するポリエチレングリコール(PEG))は、使用準備済の製剤として調製し得る。免疫細胞、例えば事前に増殖させた免疫細胞を、免疫細胞とDMPE-PEG-ADCを共に一定期間(例えば任意の適当な期間、例えば15分間)混合することによって化学免疫療法の標的形態に変換させ得る。ADCを備えた免疫細胞は、ADCによる標的抗原の認識を介して標的腫瘍部位へ向かって遊走し得る。標的腫瘍組織において、ADCは、標的癌細胞のアポトーシスを誘導し、近接して存在する免疫細胞が、ダメージ関連分子パターン(DAMP)を発現する瀕死の癌細胞を破壊する。
[0044]特定の実施形態において、リン脂質-PEGは、PEGの末端上に反応性官能基を保持し、これは、標的化部分-薬物複合体とのカップリングに使用し得る。例えば、反応性官能基がNHSである場合、中間体は反応性アミノ基を有する標的化部分とカップリングして、アミドを形成し得る。反応性官能基がアミンである場合、中間体は反応性イソチオシアネートとカップリングして、チオカルバメートを形成し得る。反応性官能基がアジドである場合、中間体は、反応性ニトリルまたは反応性アルキン基を有する標的化部分とカップリングして、テトラゾールまたはトリアゾールを形成し得る。反応性官能基がマレイミドである場合、中間体は、反応性チオール基を有する標的化部分とカップリングして、チオールの硫黄と得られたスクシンイミドの炭素との間に結合を形成し得る。
T-DM1は、哺乳類細胞におけるトラスツズマブ(TZ)の発現およびそれに次ぐDM1のコンジュゲートにより生成した。調製したT-DM1は次いで、DMPE-PEG-NHSを結合させることにより疎水化し、これによりDMPE-PEG-T-DM1が産生された。合成したT-DM1は、市販品であるKadcylaと比較して同等の細胞毒性を示した。種々の量のDMPE-PEG-T-DM1によるNK細胞の表面処理は、NK細胞の生存率(図1Aを参照のこと)、NK細胞の増殖活性のいずれにも影響を与えない。5×105の免疫細胞の信頼性のある改変には、100μgのDMPE-PEG-T-DM1が必要であり、これにより、1×105のSE-NK/T-DM1細胞の細胞膜上に埋込まれた約2.1μgのT-DM1が得られた。T-DM1は、完全増殖培地中で48時間を超えて、SE-NK/T-DM1細胞の表面上で検出され(図1Bを参照のこと)、重要なNK細胞特異的マーカーの2つである、CD56および2B4は、SE-NK/T-DM1細胞の表面上で利用可能であった。これらの結果は、T-DM1が内在化することなくNK細胞表面上に埋込まれていること、および、ADCによるNK細胞の表面処理は、NK細胞受容体のアクセス可能性に干渉しないことを実証しており、このことは、NK細胞の固有の細胞溶解活性が表面処理時に保持されていることを示唆している(図1Cを参照のこと)。本試験で使用する同種異系NK細胞である、NK-92細胞は、抗体内在化および抗体依存性細胞毒性(ADCC)を開始することができるCD16、CD32およびCD64 IgG受容体を欠損しているため、T-DM1による表面処理は、NK細胞の代謝および生存率に対する効果が最小であると考えられる。癌細胞において、T-DM1内在化は、HER2受容体介在性エンドサイトーシスを介して生じる。NK細胞は、その膜上にHER2を発現しないため、SE-NK/T-DM1細胞の表面上に埋込まれたDMPE-PEG-T-DM1は内在化せず、無視できる程度の細胞毒性を示した。さらに、DMEPとT-DM1の間のPEGスペーサーは、内在化の物理的な障壁を提供する。試験により、PEGスペーサーが長くなると、生体分子の内在化を阻害するだけでなく、立体障害の増加により、膜挿入効率が低下することが報告されている。長いPEGスペーサーの存在によりもたらされたDMPE-PEG-T-DM1の上述の膜挿入効率は、約10%であった。
