Description
「網膜細胞」という用語は、網膜を構成する任意の細胞型(網膜神経節細胞、アマクリン細胞、水平細胞、双極細胞、光受容体細胞(杆体及び錐体を含む)、ミュラーグリア細胞、アストロサイト(例えば、網膜アストロサイト)、ならびに網膜色素上皮など)を本明細書で指し得る。
本開示は、変異AAVカプシドタンパク質を提供する。本開示の変異AAVカプシドタンパク質は、親AAVカプシドタンパク質の表面露出(例えば、溶媒露出)部分における挿入部位に5つのアミノ酸~20個のアミノ酸の長さの異種ペプチド挿入断片を含み、その結果、変異カプシドタンパク質は、AAVビリオンに存在するとき、対応する親AAVカプシドタンパク質を含むAAVビリオンによる網膜細胞に対する感染力と比較して、網膜細胞に対する感染力を増加させる。他の場合では、網膜細胞は、ミュラー細胞である。他の網膜細胞には、アマクリン細胞、双極細胞、及び水平細胞が含まれる。「約5つのアミノ酸~約20個のアミノ酸の挿入断片」は、本明細書ではペプチド挿入断片(例えば、異種ペプチド挿入断片)とも称される。「対応する親AAVカプシドタンパク質」は、ペプチド挿入断片を含まない同一のAAV血清型のAAVカプシドタンパク質を指す。多くの場合、変異AAVカプシドは、5つのアミノ酸~20個のアミノ酸(例えば、5つ~7つ、7つ~10個、10個~12個、12個~15個、または15個~20個のアミノ酸)の長さの単一の異種ペプチド挿入断片を含む。
上記のように、約5つのアミノ酸~約20個のアミノ酸の長さの異種ペプチドが、AAVカプシドのGHループに挿入される。場合によっては、挿入ペプチドは、5つのアミノ酸~20個のアミノ酸の長さを有する。場合によっては、挿入ペプチドは、7つのアミノ酸~15個のアミノ酸の長さを有する。場合によっては、挿入ペプチドは、9つのアミノ酸~15個のアミノ酸の長さを有する。場合によっては、挿入ペプチドは、9つのアミノ酸~12個のアミノ酸の長さを有する。挿入ペプチドは、5つのアミノ酸、6つのアミノ酸、7つのアミノ酸、8つのアミノ酸、9つのアミノ酸、10個のアミノ酸、11個のアミノ酸、12個のアミノ酸、13個のアミノ酸、14個のアミノ酸、15個のアミノ酸、16個のアミノ酸、17個のアミノ酸、18個のアミノ酸、19個のアミノ酸、または20個のアミノ酸の長さを有する。場合によっては、挿入ペプチドは、7つのアミノ酸の長さを有する。場合によっては、挿入ペプチドは、8つのアミノ酸の長さを有する。場合によっては、挿入ペプチドは、9つのアミノ酸の長さを有する。場合によっては、挿入ペプチドは、10個のアミノ酸の長さを有する。場合によっては、挿入ペプチドは、11個のアミノ酸の長さを有する。場合によっては、挿入ペプチドは、12個のアミノ酸の長さを有する。場合によっては、挿入ペプチドは、13個のアミノ酸の長さを有する。場合によっては、挿入ペプチドは、14個のアミノ酸の長さを有する。場合によっては、挿入ペプチドは、15個のアミノ酸の長さを有する。
対象rAAVビリオンは、遺伝子産物(異種遺伝子産物)をコードするヌクレオチド配列を含む異種核酸を含む。場合によっては、遺伝子産物は、ポリペプチドである。場合によっては、遺伝子産物は、RNAである。遺伝子産物がRNAである場合、場合によっては、RNA遺伝子産物は、ポリペプチドをコードする。場合によっては、本開示のrAAVビリオンは、単一の異種遺伝子産物をコードするヌクレオチド配列を含む単一の異種核酸を含む。場合によっては、本開示のrAAVビリオンは、2つの異種遺伝子産物をコードするヌクレオチド配列を含む単一の異種核酸を含む。場合によっては、本開示のrAAVビリオンは、2つの異種核酸を含み、それぞれが、異種遺伝子産物をコードするヌクレオチド配列を含む。
遺伝子産物が干渉RNA(RNAi)である場合、適切なRNAiには、細胞におけるアポトーシス因子または血管新生因子のレベルを低減するRNAiが含まれる。例えば、RNAiは、細胞におけるアポトーシスを誘導または促進する遺伝子産物のレベルを低減するshRNAまたはsiRNAであり得る。その遺伝子産物がアポトーシスを誘導または促進する遺伝子は、本明細書で「アポトーシス促進遺伝子」と称され、そうした遺伝子の産物(mRNA、タンパク質)は、「アポトーシス促進遺伝子産物」と称される。アポトーシス促進遺伝子産物には、例えば、Bax遺伝子産物、Bid遺伝子産物、Bak遺伝子産物、及びBad遺伝子産物が含まれる。例えば、米国特許第7,846,730号を参照のこと。
遺伝子産物がアプタマーである場合、目的アプタマーの例には、VEGFを標的とするアプタマーが含まれる。例えば、Ng et al.(2006)Nat.Rev.Drug Discovery 5:123、及びLee et al.(2005)Proc.Natl.Acad.Sci.USA 102:18902を参照のこと。例えば、VEGFアプタマーは、5’-cgcaaucagugaaugcuuauacauccg-3’というヌクレオチド配列(配列番号//)を含み得る。血小板由来増殖因子(PDGF)特異的アプタマー(例えば、E10030)もまた使用に適しており、例えば、Ni and Hui(2009)Ophthalmologica 223:401、及びAkiyama et al.(2006)J.Cell Physiol.207:407)を参照のこと。

場合によっては、目的の遺伝子産物は、遺伝子機能を部位特異的にノックダウンする部位特異的エンドヌクレアーゼであり、例えば、エンドヌクレアーゼは、網膜疾患と関連する対立遺伝子をノックアウトする。例えば、野生型であるときに網膜の構造タンパク質であり、及び/または正常な網膜機能を提供する遺伝子の欠陥コピーを優性対立遺伝子がコードする場合、部位特異的エンドヌクレアーゼは、その欠陥対立遺伝子を標的とし、その欠陥対立遺伝子をノックアウトすることができる。場合によっては、部位特異的エンドヌクレアーゼは、RNAガイド型エンドヌクレアーゼである。

場合によっては、目的の遺伝子産物をコードするヌクレオチド配列は、転写制御要素に機能可能なように連結される。例えば、場合によっては、目的の遺伝子産物をコードするヌクレオチド配列は、構成的プロモーターに機能可能なように連結される。他の場合では、目的の遺伝子産物をコードするヌクレオチド配列は、誘導性プロモーターに機能可能なように連結される。場合によっては、目的の遺伝子産物をコードするヌクレオチド配列は、組織特異的または細胞型特異的な調節要素に機能可能なように連結される。例えば、場合によっては、目的の遺伝子産物をコードするヌクレオチド配列は、網膜細胞特異的プロモーターに機能可能なように連結される。例えば、場合によっては、目的の遺伝子産物をコードするヌクレオチド配列は、光受容体特異的調節要素(例えば、光受容体特異的プロモーター)に機能可能なように連結され、例えば、光受容体細胞において機能可能なように連結された遺伝子を選択的に発現させる調節要素に機能可能なように連結される。適切な光受容体特異的調節要素には、例えば、ロドプシンプロモーター、ロドプシンキナーゼプロモーター(Young et al.(2003)Ophthalmol.Vis.Sci.44:4076)、ベータホスホジエステラーゼ遺伝子プロモーター(Nicoud et al.(2007)J.Gene Med.9:1015)、網膜色素変性症遺伝子プロモーター(前出のNicoud et al.(2007))、受容体間レチノイド結合タンパク質(IRBP)遺伝子エンハンサー(前出のNicoud et al.(2007))、IRBP遺伝子プロモーター(Yokoyama et al.(1992)Exp Eye Res.55:225)が含まれる。
本開示は、a)上記の対象rAAVビリオンと、b)医薬的に許容可能な担体、希釈剤、賦形剤、または緩衝剤とを含む医薬組成物を提供する。いくつかの実施形態では、医薬的に許容可能な担体、希釈剤、賦形剤、または緩衝剤は、ヒトにおける使用に適したものである。
本開示は、個体の網膜細胞への遺伝子産物の送達方法を提供し、方法は、上記の対象rAAVビリオンを個体に投与することを含む。遺伝子産物は、上記のポリペプチドもしくは干渉RNA(例えば、shRNA、siRNA、及び同様のもの)、アプタマー、または部位特異的エンドヌクレアーゼ(例えば、RNAガイド型エンドヌクレアーゼ)であり得る。網膜細胞への遺伝子産物の送達によって、網膜疾患が治療され得る。網膜細胞は、光受容体、網膜神経節細胞、ミュラー細胞、双極細胞、アマクリン細胞、水平細胞、または網膜色素上皮細胞であり得る。場合によっては、網膜細胞は、光受容体細胞(例えば、杆体細胞または錐体細胞)である。
本開示は、上記の対象変異アデノ随伴ウイルス(AAV)カプシドタンパク質をコードするヌクレオチド配列を含む単離された核酸を提供し、変異AAVカプシドタンパク質は、対応する親AAVカプシドタンパク質と比較して約5つのアミノ酸~約20個のアミノ酸の挿入断片をGHループまたはループIVに含み、変異カプシドタンパク質は、AAVビリオンに存在するとき、対応する親AAVカプシドタンパク質を含むAAVビリオンによる網膜細胞に対する感染力と比較して、網膜細胞に対する感染力を増加させる。対象の単離された核酸は、AAVベクターであり得、例えば、組換えAAVベクターであり得る。
対象核酸によってコードされる変異AAVカプシドタンパク質は、AAVカプシドのGHループに挿入された約5つのアミノ酸~約20個のアミノ酸の長さの挿入ペプチドを有する。挿入ペプチドは、5つのアミノ酸、6つのアミノ酸、7つのアミノ酸、8つのアミノ酸、9つのアミノ酸、10個のアミノ酸、11個のアミノ酸、12個のアミノ酸、13個のアミノ酸、14個のアミノ酸、15個のアミノ酸、16個のアミノ酸、17個のアミノ酸、18個のアミノ酸、19個のアミノ酸、または20個のアミノ酸の長さを有する。適切な挿入ペプチドは、上記のものである。適切な挿入ペプチドには、上記の式I~Xのいずれか1つのペプチドが含まれる。

大型動物の眼では、強力かつ複雑な障壁によってAAVによる汎網膜感染が阻止されるが、インビボの反復スクリーニング法を使用することで、この障壁を克服することができるカプシド変異体を有するAAVを創出した。イヌは、網膜変性疾患の重要な前臨床モデルであり、その眼のサイズ及び構造はヒトと類似しており、多くの型の網膜疾患がさまざまなイヌの種で天然に生じる。このスクリーニング法を使用することで、イヌの網膜において汎網膜感染を生じさせる能力を有する96個のAAV変異体を同定した。ディープシークエンシングを使用することで、イヌ網膜において、スクリーニングしたAAV変異体のプールに由来するこうした変異体のうちの18個の変異体の能力を定量化した。硝子体内注射の後に網膜細胞におけるウイルスのDNA及びmRNAのレベルに基づいて感染力を定量化した。こうした変異体は、大型動物及びヒトの眼における多種多様な遺伝子送達方針に使用することができるものである。

Claims (20)

a)干渉RNAまたはアプタマーである、
b)ポリペプチド、任意には、神経保護ポリペプチド、抗血管新生ポリペプチド、抗アポトーシスポリペプチド、もしくは網膜細胞の機能を増進するポリペプチド、または、RNAガイド型エンドヌクレアーゼである、または、
c)RNAガイド型エンドヌクレアーゼ及びガイドRNAである、
請求項1~4のいずれか1項に記載のrAAVビリオン。 - 前記異種遺伝子産物は、グリア細胞由来神経栄養因子、線維芽細胞増殖因子2、ニュールツリン、毛様体神経栄養因子、神経成長因子、脳由来神経栄養因子、上皮増殖因子、ロドプシン、X連鎖アポトーシス抑制物質、レチノスキシン、RPE65、網膜色素変性症GTPase相互作用タンパク質-1、ペリフェリン、ペリフェリン-2、ロドプシン、RdCVF、網膜色素変性症GTPase調節因子(RPGR)、またはソニックヘッジホッグである、請求項1~4のいずれか1項に記載のrAAVビリオン。
- 前記異種ペプチドは、約12個~20個のアミノ酸の長さを有する、請求項1~6のいずれか1項に記載のrAAVビリオン。
- a)請求項1~7のいずれか1項に記載のrAAVビリオンと、
b)医薬的に許容可能な賦形剤と
を含む、医薬組成物。 - 個体の網膜細胞へ遺伝子産物を送達する方法に使用のための請求項1~7のいずれか1項に記載の組換えアデノ随伴ウイルス(rAAV)ビリオン。
- 眼疾患を治療する方法に使用のための請求項1~7のいずれか1項に記載の組換えアデノ随伴ウイルス(rAAV)ビリオン。
- 前記眼疾患は、緑内障、網膜色素変性症、黄斑変性、網膜分離症、レーバー先天黒内障、糖尿病性網膜症、色覚異常(achromotopsia)、または色覚異常(color blindness)である、請求項10に記載のrAAVビリオン。
- 対応する親AAVカプシドタンパク質と比較してカプシドタンパク質GHループに異種ペプチドの挿入断片を含むアデノ随伴ウイルス2(AAV2)カプシドタンパク質VP1をコードするヌクレオチド配列を含む単離された核酸であって、
前記異種ペプチドは,アミノ酸配列PDSTTRSと、アミノ酸Pの前のN末端における2アミノ酸長リンカー及びアミノ酸Sの後のC末端における1アミノ酸長リンカーとを含んでおり、
前記カプシドタンパク質は、AAVビリオンに存在するとき、網膜細胞に対する前記AAVビリオンの感染力を増加させる、
単離された核酸。 - 前記挿入断片は、AAV2のVP1のアミノ酸570及びアミノ酸611に対応するアミノ酸の間に存する、請求項12に記載の単離された核酸。
- 前記挿入断片は、AAV2のVP1のアミノ酸587及びアミノ酸588の間に存する、請求項13に記載の単離された核酸。
- 前記異種ペプチドは、約12個~20個のアミノ酸の長さを有する、請求項12~14のいずれか1項に記載の単離された核酸。
- 請求項12~15のいずれか1項に記載の単離された核酸を含む、単離された遺伝子改変宿主細胞。
- アデノ随伴ウイルス2(AAV2)カプシドタンパク質VP1であって、
前記AAV2カプシドタンパク質VP1は、異種ペプチドの挿入断片をGHループに含んでおり、
