Description
前記メタ表面は、金属の相補的積層構造またはナノロッド構造であってもよい。
前記蛍光増強は、可視光領域または近赤外光領域における波長を有する光の強度の増強であってもよい。
前記第1の基板と前記第2の基板とにより形成されるマイクロ流路の高さは、10μm以上100μm以下の範囲であってもよい。
前記第1の基板と前記第2の基板とにより形成されるマイクロ流路の高さは、15μm以上50μm以下の範囲であってもよい。
前記メタ表面は、前記金属の相補的積層構造であり、前記金属の相補的積層構造は、基材と、前記基材の表面に位置するスラブ材と、少なくとも前記スラブ材上に位置する金属材料とを含み、前記基材は、少なくとも前記スラブ材と接する表面層を備え、前記スラブ材は、前記表面層の屈折率よりも高い屈折率を有する材料からなると共に、その表面から前記基材の前記表面層に達する、周期的に配列した複数の穴を有し、前記金属材料は、前記スラブ材の表面、および、前記複数の穴の底面を形成する前記基材の表面層上にそれぞれ位置し、これにより上記課題を解決する。
前記金属材料は、ドルーデ金属に近似できる複素誘電率を有する物質であってもよい。
前記ドルーデ金属に近似できる複素誘電率を有する物質は、金(Au)、銀(Ag)、銅(Cu)、アルミニウム(Al)、白金(Pt)、チタン(Ti)、ニッケル(Ni)、および、これらの合金からなる群から選択されてもよい。
前記複数の穴は、2以上の異なる直径を有する円孔、もしくは2以上の異なる一辺の長さを有する角柱状孔であり、かつ/または、2以上の異なる周期で配列されていてもよい。
前記複数の穴の周期は、300nm以上1000nm以下の範囲であってもよい。
前記複数の穴の直径または一辺の長さは、100nm以上500nm以下の範囲であってもよい。
前記スラブ材の厚さは、100nm以上2μm以下の範囲であってもよい。
前記メタ表面は、前記ナノロッド構造であり、前記ナノロッド構造は、基材と、前記基材の表面に周期的に起立する複数のナノロッドとを備え、前記基材は、前記複数のナノロッドの屈折率よりも小さい屈折率を有する材料からなってもよい。
前記基材は、1以上1.6以下の屈折率を有する材料からなり、前記複数のナノロッドは、2以上5以下の屈折率を有する半導体または誘電体からなってもよい。
前記半導体または誘導体は、シリコン、ゲルマニウム、窒化ガリウム、および、二酸化チタンからなる群から選択されてもよい。
前記複数のナノロッドの周期は、300nm以上1000nm以下の範囲であってもよい。
前記複数のナノロッドの直径または一辺の長さは、100nm以上500nm以下の範囲であってもよい。
前記複数のナノロッドの高さは、100nm以上2μm以下の範囲であってもよい。
前記複数のナノロッドは、2以上の異なる直径を有する円柱、もしくは2以上の異なる一辺の長さを有する角柱であり、かつ/または、2以上の異なる周期で配列されていてもよい。
図2は、本発明の蛍光検出用生体分子検査チップを用いた検出原理を示す模式図である。
ステップS610:標的生体分子を捕捉するための捕捉分子を含有する溶液を、図1に示す検査チップ100のマイクロ流路130に流し、メタ表面110に固定する。メタ表面110は間隙を有するため、捕捉分子を含有する溶液をマイクロ流路130に流すだけで、捕捉分子は容易に固定される。このような捕捉分子は、標的生体分子に応じて適宜選択される。捕捉分子を含有する溶液を流す前に、流路内を液体で満たすためにリン酸生理食塩水(PBS)などのバッファーなどを流してもよい。
メタ表面として、シリコンナノロッド構造および金の相補的積層構造をそれぞれ作製した。
図8は、金の相補的積層構造であるメタ表面を示すSEM像である。
捕捉分子には、プロテインAG-シスまたはシス-ストレプトアビジン(Click Biosystems、PRO1001またはPRO1005)を用いた。捕捉抗体には抗アビジン抗体(アブカム、ab53494)を用いた。
標的生体分子にはイミュノグロブリンG(IgG、アブカム、ab206202)、p53抗体(Novus Biologicals、NB200-171)、マウスモノクローナルp53抗体(アブカム、ab16465)、CEA(アブカム、ab742)、および、蛍光分子(AF555、Alexa Fluor555)標識済みp53抗体(アブカム、ab210717)、蛍光分子(AF555、Alexa Fluor555)標識済みヤギ抗マウスIgG抗体(アブカム、ab150114)、蛍光分子(AF555、Alexa Fluor555)標識済みヤギ抗ウサギIgG抗体(アブカム、ab150078)、または、蛍光分子(DL555、DyLight555)標識済みIgG(Novus Biologicals、NBP1-76055)を用いた。
また、ビオチン標識キット(和光純薬347-90891)、蛍光分子HiLyte555(HL555)標識キット(和光純薬348-90141)、蛍光分子インドシアニングリーン(ICG)標識キット(和光純薬345-91431)を使用し、必要に応じて、標的生体分子に蛍光標識、ビオチン標識を付した。
実施例1では、周期400nm、直径320nmである円柱状シリコンナノロッド構造のメタ表面110(図1)の第1の基板120(図1)に、マイクロ流路(流路高さD30μm)を有するポリジメチルシロキサン(PDMS)製の第2の基板140(図1)を自己吸着により貼り合わせ、検査チップを構成した。検査チップの外観を図9(A)に、これを配置した流路装置の外観を図9(B)に、それぞれ示す。なお、PDMSは、可視光の平均透過率が60%以上であることが知られている。
実施例2では、周期300nm、直径236nmである円柱状シリコンナノロッド構造のメタ表面を用いた以外は実施例1と同様にして検査チップおよび流路装置を構成した。得られた検査チップの外観は図9(A)と同様であり、流路装置の外観は図9(B)と同様であった。
実施例3では、周期410.5/710nm、穴の直径302nmである金の相補的積層構造のメタ表面110(図1)を有する第1の基板120(図1)に、マイクロ流路(流路高さ30μm)を有するPDMS製の第2の基板140(図1)を自己吸着により貼り合わせ、検査チップを構成した。この検査チップの外観を図10に示す。
実施例4では、直径316nmである円柱状シリコンナノロッド構造のメタ表面を有する第1の基板を用いた以外は、実施例1と同様にして検査チップを構成した。検査チップの外観は、図9(A)と同様であった。標的生体分子として、AF555で蛍光標識された腫瘍マーカーであるp53抗体を用いた以外は、実施例1と同様の手順で、標的生体分子の検出を行った。標的生体分子濃度と蛍光強度との関係を図17に示す。
実施例5では、穴の直径が291nmである金の相補的積層構造のメタ表面を有する第1の基板を用いた以外は、実施例3と同様にして検査チップを構成した。検査チップの外観は図10と同様であった。捕捉分子としてシス-ストレプトアビジンを用いると共に、標的生体分子として、HL555で蛍光標識したp53抗体をさらにビオチン標識して用いた以外は、実施例1と同様の手順で標的生体分子の検出を行った。標的生体分子濃度と蛍光強度との関係を図18に示す。
実施例6では、実施例5で作製した検査チップおよび流路装置を用いて、捕捉抗体の有無による標的生体分子の検出効率を比較した。
実施例7では、周期400nm、直径302nmである円柱状シリコンナノロッド構造のメタ表面を有する第1の基板を用いると共に、PDMS製の第2の基板に形成されたマイクロ流路の流路高さDを15μmとした以外は、実施例1と同様にして検査チップおよび流路装置を構成した。検査チップの外観は、図9(A)と同様であり、流路装置の外観は図9(B)と同様であった。得られた流路装置を用い、実施例1と同様の手順で標的生体分子の検出を行った。標的生体分子として、AF555で蛍光標識されたIgG分子をさらにビオチン標識して用いた。ここで、IgG濃度は5ng/ml、0.33ng/mlおよび0.022ng/mlとした。
比較例1では、周期400nm、直径302nmである円柱状シリコンナノロッド構造のメタ表面を用いた以外は、実施例1と同様にして、検査チップおよび流路装置を構成し、標的生体分子の検出を行った。標的生体分子の濃度は、50ng/mL、5ng/mLおよび0.5ng/mLとした。蛍光検出の測定時に、PDMS製の第2の基板を除去してメタ表面を有する基板(第1基板)単独について測定し、マイクロ共振器を通して検出する実施例との比較を行った。標的生体分子の蛍光検出は、実施例1と同様の手順で行った。濃度が50ng/mLの標的生体分子についての蛍光スペクトルを図13に示す。蛍光スペクトルから得られた標的生体分子濃度と蛍光強度との関係を図19に示す。
比較例2では、穴の直径が250nmである金の相補的積層構造のメタ表面を有する第1の基板のみを単独で用いた。捕捉分子として、プロテインAのPBS希釈液500ng/mLをメタ表面に滴下し、表面への固定のため、室温・湿潤環境で1時間保持したのち、PBSでリンスした。次いで、標的生体分子として、ICG標識したIgG分子のPBS希釈液2μL(濃度90ng/mL、9000ng/mLおよび450000ng/mL)を、別々に準備した基板のメタ表面にそれぞれ滴下し、室温・湿潤環境下で2時間インキュベーションした。その後、PBSでリンスして固定されていない浮遊分子などを除去し、乾燥窒素をフローしてメタ表面を乾燥させた。
実施例8では、穴の直径が315nmである金の相補的積層構造のメタ表面を有する第1の基板を用いた以外は、実施例3と同様にして検査チップおよび流路装置を構成した。検査チップの外観は図10と同様であった。
図13は、比較例1の検査チップを用いた発光スペクトルを示す図である。
図15は、実施例2による蛍光測定から得た標準曲線を示す図である。
図16は、実施例3による蛍光測定から得た標準曲線を示す図である。
図17は、実施例4による蛍光測定から得た標準曲線を示す図である。
図18は、実施例5による蛍光測定から得た標準曲線を示す図である。
図19は、比較例1による蛍光測定から得た標準曲線を示す図である。
図20は、比較例2による蛍光測定から得た標準曲線を示す図である。
実施例9では、実施例3と同じ検査チップおよび流路装置を使用し、再現性、ならびに、バッファー(緩衝液)の影響について調べた。
実験1)実施例1と同様に捕捉分子を流路装置に流した後、NSバッファー(Abcam社製、ab193972)で希釈した標的生体分子を、平均流量レート7μL/分で30分間、流した。
実験2)標的生体分子を、90%NSバッファーおよび10%ヒト血清で希釈した以外は、実験1と同様に行った。
実験3)事前に捕捉分子を流路装置に流さなかった以外は実験1と同様に行った。
実験4)実験1の測定の翌日に、再現性を確かめるため、同じ設計により作製したメタ表面を用いて、実験1と同様の実験を行った。
実験5)実施例1と同様に捕捉分子を流路装置に流した後、100%ヒト血清で希釈した標的生体分子を、平均流量レート4μL/分で50分間、流した。
実験6)バックグラウンド信号レベルを決定するために、捕捉分子および標的生体分子を流すことなく、NSバッファーのみを平均流量レート7μL/分で30分間、流路装置に流した。
実施例10では、実施例3と同じ検査チップおよび流路装置を使用し、マウスモノクローナルp53抗体の間接的な検出を実施した。
図25は、実施例10による蛍光強度の比較を示す図である。
実施例11では、実施例3と同じ検査チップおよび流路装置を使用し、サンドウィッチ検定配置においてCEAの間接的な検出を調べた。CEAは医療診断に使用される腫瘍マーカーである。
図28は、実施例11による蛍光測定から得たCEAの蛍光強度の濃度依存性を示す図である。

Claims (19)
- メタ表面を有する第1の基板と、
前記第1の基板と対向して位置し、マイクロ流路を有する第2の基板と
を備え、
前記メタ表面は、検出されるべき生体分子の固定を効率化する間隙を有すると共に、前記生体分子が発する蛍光の波長範囲を含む領域において蛍光増強を示し、
前記第2の基板は、可視光または近赤外光を透過する材料からなり、
前記第1の基板と前記第2の基板との間で前記蛍光が共振する、蛍光検出用生体分子検査チップ。 - 前記メタ表面は、金属の相補的積層構造またはナノロッド構造である、請求項1に記載の検査チップ。
- 前記蛍光増強は、可視光領域または近赤外光領域における波長を有する光の強度の増強である、請求項1または2に記載の検査チップ。
- 前記第1の基板と前記第2の基板とにより形成されるマイクロ流路の高さは、10μm以上100μm以下の範囲である、請求項1~3のいずれかに記載の検査チップ。
- 前記第1の基板と前記第2の基板とにより形成されるマイクロ流路の高さは、15μm以上50μm以下の範囲である、請求項4に記載の検査チップ。
- 前記メタ表面は、前記金属の相補的積層構造であり、
前記金属の相補的積層構造は、基材と、前記基材の表面に位置するスラブ材と、少なくとも前記スラブ材上に位置する金属材料とを含み、
前記基材は、少なくとも前記スラブ材と接する表面層を備え、
前記スラブ材は、前記表面層の屈折率よりも高い屈折率を有する材料からなると共に、その表面から前記基材の前記表面層に達する、周期的に配列した複数の穴を有し、
前記金属材料は、前記スラブ材の表面、および、前記複数の穴の底面を形成する前記基材の表面層上にそれぞれ位置する、請求項2に記載の検査チップ。 - 前記金属材料は、ドルーデ金属に近似できる複素誘電率を有する物質である、請求項6に記載の検査チップ。
- 前記ドルーデ金属に近似できる複素誘電率を有する物質は、金(Au)、銀(Ag)、銅(Cu)、アルミニウム(Al)、白金(Pt)、チタン(Ti)、ニッケル(Ni)、および、これらの合金からなる群から選択される、請求項7に記載の検査チップ。
- 前記複数の穴は、2以上の異なる直径を有する円孔、もしくは2以上の異なる一辺の長さを有する角柱状孔であり、かつ/または、2以上の異なる周期で配列されている、請求項6~8のいずれかに記載の検査チップ。
- 前記複数の穴の周期は、300nm以上1000nm以下の範囲である、請求項6~9のいずれかに記載の検査チップ。
- 前記複数の穴の直径または一辺の長さは、100nm以上500nm以下の範囲である、請求項6~10のいずれかに記載の検査チップ。
- 前記スラブ材の厚さは、100nm以上2μm以下の範囲である、請求項6~11のいずれかに記載の検査チップ。
- 前記メタ表面は、前記ナノロッド構造であり、
前記ナノロッド構造は、基材と、前記基材の表面に周期的に起立する複数のナノロッドとを備え、
