本発明は、インビトロ診断のための、カートリッジ及び改善された方法を提供することを目的とする。

以下、本発明の好ましい実施形態に従って構成されかつ動作するカートリッジの前部、後部、上部、底部、第1の側面及び第2の側面の平面図、並びに前部及び後部の斜視図をそれぞれ簡略化して示す図である図1A~図1Hと、密閉する前の、開いた位置にある図1のカートリッジを簡略化して示す図である図2を参照する。
Claims (14)
- 第1の操作用容積と第2の操作用容積とを少なくとも含む複数の操作用容積を有する、インビトロ診断用カートリッジの作動方法であって、前記方法は、
前記複数の操作用容積の少なくとも1つから前記複数の操作用容積の他の少なくとも1つに流体溶液を移動させることであって、前記流体溶液を移動させることは、輸送要素を移動させて前記複数の操作用容積の前記少なくともいくつかの内部と連続的に連通させることを含む、前記移動させることと、
前記複数の操作用容積の少なくとも1つを流通することを含み、
前記流体溶液を移動させることが、
前記第1の操作用容積において細胞膜破壊物質の位置を突き止めることと、
前記第1の操作用容積と連通する中空の針の開口端を突き止めることと、
前記細胞膜破壊物質の少なくとも一部を前記中空の針内に吸引することと、
内部に位置する試料を有する前記第2の操作用容積と連通するように前記中空の針の前記開口端を移動させることと、
前記試料と前記細胞膜破壊物質の少なくともいくつかを前記中空の針内に繰り返し吸引して前記試料と前記細胞膜破壊物質を前記中空の針から前記第2の操作用容積内に放出して、これにより、前記試料と前記細胞膜破壊物質を混合すること
を含む、インビトロ診断用カートリッジの作動方法。 - 前記移動させることは、前記複数の操作用容積のそれぞれの間の前記輸送要素を通るように前記流体溶液を操作することも含む、請求項1に記載の方法。
- 前記流体溶液を移動させることは、細胞物質を含む流体溶液を前記カートリッジ内に取り付けられたマイクロ流体力学PCRアレイに移動させることを含む、請求項1または2に記載の方法。
- 前記流体溶液を移動させることは、細胞物質を含む流体溶液を前記マイクロ流体力学PCRアレイから前記カートリッジに関連するセンサアレイに移動させることを含む、請求項3に記載の方法。
- 前記複数の操作用容積のいくつかに細胞物質を供給する前に、その中に物質を注入することも含む、請求項1~4のいずれか1項に記載の方法。
- 前記流体溶液を移動させることは、更に、
細胞溶解液と磁気ビーズを含む第3の操作用容積と連通するように前記中空の針の前記開口端を移動させることと、
前記試料と前記細胞膜破壊物質に係合させるように前記細胞溶解液と磁気ビーズの少なくとも一部を前記中空の針内に吸引することと、
前記試料、前記細胞膜破壊物質の少なくともいくつか、前記細胞溶解液及び磁気ビーズを繰り返し前記中空の針内に吸引して、前記試料、前記細胞膜破壊物質の前記少なくともいくつか、前記細胞溶解液及び前記磁気ビーズを前記中空の針から前記第3の操作用容
積内に放出して、これにより、核酸を前記試料から解放すること
を含む、請求項1に記載の方法。 - 前記流体溶液を移動させることは、更に、
洗浄緩衝液を含む第4の操作用容積と連通するように前記中空の針の前記開口端を移動させることと、
前記洗浄緩衝液の少なくとも一部を前記中空の針内に吸引して、前記磁気ビーズとこれに結合する前記核酸も一緒に係合させることと、
前記洗浄緩衝液と前記磁気ビーズをこれに結合する前記核酸も一緒に前記中空の針内に繰り返し吸引して、これにより、細胞残屑と結合されていない核酸を前記磁気ビーズから洗い落とすこと
を含む、請求項6に記載の方法。 - 前記流体溶液を移動させることは、更に、
溶出緩衝液を含む第5の操作用容積と連通するように、前記中空の針の前記開口端を移動させることと、
前記溶出緩衝液の少なくとも一部を前記中空の針内に吸引して、前記磁気ビーズとこれに結合する前記核酸も一緒に係合させることと、
前記溶出緩衝液と前記磁気ビーズとこれに結合する前記核酸も一緒に前記中空の針内に繰り返し吸引して、これにより、前記核酸を前記磁気ビーズとの係合から解放することを含む、請求項7に記載の方法。 - 前記流体溶液を移動させることは、更に、
内部に並置された少なくとも1つの磁石を有する第6の操作用容積と連通するように、前記中空の針の前記開口端を移動させることと、
前記少なくとも1つの磁石が前記磁気ビーズを引きつけるように、前記溶出緩衝液と前記磁気ビーズとこれから解放された前記核酸も一緒に前記第6の操作用容積内に移動させることと、
前記溶出緩衝液を前記核酸と一緒に前記中空の針内に吸引すること
を含む、請求項8に記載の方法。 - 前記流体溶液を移動させることは、更に、
マイクロ流体力学PCRアレイと連通する第7の操作用容積と連通するように、前記中空の針の前記開口端を移動させることと、
前記マイクロ流体力学PCRアレイが、前記核酸を増幅することによって増幅される核酸を生成するように、前記溶出緩衝液と前記核酸を前記マイクロ流体力学PCRアレイ内に移動させることと、
前記増幅された核酸を前記中空の針内に吸引すること
希釈緩衝液を含む第8の操作用容積と連通するように、前記中空の針の前記開口端を移動させることと、
前記希釈緩衝液を前記中空の針内に吸引して前記増幅された核酸と係合させることと、
前記希釈緩衝液と前記増幅された核酸を前記中空の針内に繰り返し吸引して、これにより、希釈核酸を生成すること
を含む、請求項9に記載の方法。 - 前記流体溶液を移動させることは、更に、
センサアレイと連通する第9の操作用容積と連通するように、前記中空の針の前記開口端を移動させることと、
前記希釈核酸の少なくとも第1の部分を前記センサアレイ内に移動させて、これにより、前記センサアレイを洗浄することと、
その後、前記希釈核酸の第2の部分を移動させて前記センサアレイと動作するように係合させること
を含む、請求項10に記載の方法。 - 前記流体溶液を移動させることは、更に、
濃縮ディスクリミネーターを含む第10の操作用容積と連通するように、前記中空の針の前記開口端を移動させることと、
前記濃縮ディスクリミネーターを前記中空の針内に吸引することと、
ディスクリミネーター用緩衝液を含む第11の操作用容積と連通するように、前記中空の針の前記開口端を移動させることと、
前記ディスクリミネーター用緩衝液を前記中空の針内に吸引して、前記濃縮ディスクリミネーターと係合させることと、
前記ディスクリミネーター用緩衝液と前記濃縮ディスクリミネーターを前記中空の針内に繰り返し吸引して前記ディスクリミネーター用緩衝液と前記濃縮ディスクリミネーターを前記中空の針から前記第11の操作用容積内に放出し、これにより、希釈ディスクリミネーターを生成することと、
前記希釈ディスクリミネーターを前記中空の針内に吸引することと、
前記センサアレイと連通する前記第9の操作用容積と連通するように、前記中空の針の前記開口端を移動させることと、
前記希釈ディスクリミネーターを移動させて前記センサアレイと動作するように係合させること
を含む、請求項11に記載の方法。 - 前記流体溶液を移動させることは、更に、
レポーター再構成用緩衝液を含む第12の操作用容積と連通するように、前記中空の針の前記開口端を移動させることと、
前記レポーター再構成用緩衝液を前記中空の針内に吸引することと、
前記レポーター再構成用緩衝液を前記中空の針内に繰り返し吸引して前記レポーター再構成用緩衝液を前記中空の針から乾燥レポーターを含む第13の操作用容積を経由して前記第12の操作用容積内に放出し、これにより、再構成されたレポーターを生成することと、
前記再構成されたレポーターを前記中空の針内に吸引することと、
前記センサアレイと連通する前記第9の操作用容積と連通するように、前記中空の針の前記開口端を移動させることと、
前記再構成されたレポーターを移動させて前記センサアレイと動作するように係合させること
を含む、請求項12に記載の方法。 - 前記流体溶液を移動させることは、更に、
アレイ洗浄用緩衝液を含む第14の操作用容積と連通するように、前記中空の針の前記開口端を移動させることと、
前記アレイ洗浄用緩衝液を前記中空の針内に吸引することと、
前記センサアレイと連通する前記第9の操作用容積と連通するように、前記中空の針の前記開口端を移動させることと、
前記アレイ洗浄用緩衝液を移動させて前記センサアレイと動作するように係合させること
を含む、請求項13に記載の方法。
