Description
(1)次式(I):
で示される化合物もしくはその塩又はそれらの溶媒和物。
(2)前記式(I)において、R1がエトキシ基又はトリフルオロメトキシ基である前記(1)に記載の化合物もしくはその塩又はそれらの溶媒和物。
(3)前記(1)又は(2)に記載の化合物もしくはその塩又はそれらの溶媒和物を含有する鎮痛薬。
(4)神経障害性疼痛、がん化学療法に伴う疼痛、糖尿病性神経痛、糖尿病性末梢神経障害性疼痛及び片頭痛からなる群から選択される疾患又は症状を治療及び/又は予防するための前記(3)に記載の鎮痛薬。
(5)次式(II):
で示される化合物もしくはその塩又はそれらの溶媒和物を含有する鎮痛薬。
(6)前記式(II)において、Rがハロゲン原子で置換されたインダゾリル基である前記(5)に記載の鎮痛薬。
(7)前記式(II)において、Rが塩素原子で置換されたインダゾリル基である前記(5)に記載の鎮痛薬。
(8)神経障害性疼痛、がん化学療法に伴う疼痛、糖尿病性神経痛、糖尿病性末梢神経障害性疼痛及び片頭痛からなる群から選択される疾患又は症状を治療及び/又は予防するための前記(5)~(7)のいずれかに記載の鎮痛薬。
(9)次式(IIa):
で示される化合物もしくはその塩又はそれらの溶媒和物。
(10)前記式(IIa)において、R’がハロゲン原子で置換されたインダゾリル基である前記(9)に記載の化合物もしくはその塩又はそれらの溶媒和物。
(11)前記式(IIa)において、R’が塩素原子で置換されたインダゾリル基である前記(9)に記載の化合物もしくはその塩又はそれらの溶媒和物。

すなわち、対応する芳香族アルデヒド化合物、2,4-ジアミノ-6-ヒドロキシピリミジン(別名、2,6-ジアミノピリミジン-4(3H)-オン)及びメルドラム酸を、(i)水中、トリエチルベンジルアンモニウムクロリドの存在下、加熱下反応させるか、(ii)マイクロウェーブ照射下で加熱下反応させるか、(iii) 有機溶媒中で加熱下反応させることにより、目的とする化合物(I)を製造することができる。
すなわち、対応するアミン化合物とイタコン酸を反応させて、γ-ラクタムカルボン酸に変換した後、縮合剤(例えば、カルボジイミド)の存在下、ヒスタミンと反応させることにより、目的とする化合物(II)を製造することができる。

マウスPAC1受容体発現CHO細胞(PAC1/CHO細胞)及びマウスVPAC1受容体発現CHO細胞(VPAC1/CHO細胞)を用い、PACAP刺激により生じるCREB(cAMP-responsive element-binding protein)のリン酸化に対する各化合物の効果を、抗リン酸化CREB(pCREB)抗体を用いたウェスタンブロット解析により検討した。
PACAP(100pmol/5μL)をマウスくも膜下腔に単回投与する(単回i.t. injection)と、下半身を舐めたり噛んだりする自発性疼痛様行動を、投与後5分後程度から生じる(数時間持続する)(Ohnou et al. J. Pharmacol. Sci. 130, 194-203, 2016)。
PACAP(100pmol/5μL)をマウスに単回i.t.投与すると、機械刺激に対する過敏現象(機械的アロディニア)を長期(投与後少なくとも3か月間)に渡って生じた(非特許文献1)。このPACAP誘発長期機械的過敏現象は、PA-8、化合物2j、化合物2o(それぞれ10、100pmol、図5A、B)、あるいはPA-9、化合物3d(それぞれ100pmol、図5C)の同時i.t.投与によりその発症は強く抑制された。前記化合物に代えて溶媒(VEH:0.2%DMSO含有人工脳脊髄液)を使用した例では、PACAP誘発機械的過敏現象を抑制しなかった。
オキサリプラチンは5%グルコース含有生理食塩水に溶解し、2mg/kgの用量で腹腔内投与した。薬物の投与はオキサリプラチン投与後4日目に行った。マウスは雄性ddY(6~12週齢)を使用した。縦軸は後肢足蹠へのアセトンドロップ(10μL)適用後の舐め行動、足振り行動の持続時間である。溶媒(VEH:10%DMSO水溶液)は無効であったが、PA-8、化合物2j及び化合物2o(10mg/kg、単回経口投与)はいずれもオキサリプラチンによる冷的アロディニアを4~5時間にわたって有意に抑制するが、化合物2oはより早い時間帯(投与後1時間目)から有効であった(図6)。
オキサリプラチンは5%グルコース含有生理食塩水に溶解し、2mg/kgの用量で腹腔内投与した。薬物投与(経口投与)は、オキサリプラチン投与前30分(図7A)、あるいはオキサリプラチン投与前30分、投与後2.5時間及び投与後5.5時間後の3回行った(図7B)。マウスは雄性ddY(6~12週齢)を使用した。縦軸は後肢足蹠へのアセトンドロップ(10μL)適用後の舐め行動、足振り行動の持続時間である。溶媒(VEH:10%DMSO水溶液)は無効であったが、PA-8、化合物2j及び化合物2o(10mg/kg)はいずれもオキサリプラチンによる冷的アロディニアの発症を有意に抑制するが、化合物2oの3回投与は、ほぼ完全に発症を抑制した(図7B)。
雄性ddY(6~12週齢)の第4腰髄脊髄神経を絹糸で結紮するspinal nerve ligation(SNL)モデルを使用した。溶媒(VEH:10%DMSO含有生理食塩水)、PA-8、化合物2j及び化合物2oは結紮5~10分前、その後は1日1回7日間腹腔内投与(30mg/kg)した。機械刺激の方法は実施例5と同様であり、結紮後1日目(D1)以降のデータは、VEHあるいは薬物投与後1時間のデータである。化合物2jはVEH群同様、機械的アロディニアの発症を抑制できなかった。一方、PA-8は有意に機械的アロディニアの発症を抑制したが、化合物2oは更に強く機械的アロディニアの発症を抑制し、非損傷側と同等レベルに機械的閾値を保った。また、PA-8と化合物2oは、投与を7日目で打ち切っても少なくともその後7日間はVEH群に比べ有意に機械的閾値を高く保った(図8)。
雄性ddY(6~12週齢)の第4腰髄脊髄神経を絹糸で結紮するspinal nerve ligation(SNL)モデルを使用した。溶媒(VEH:10%DMSO含有生理食塩水)、PA-8、化合物2j及び化合物2o(図9A)、あるいはPA-9及び化合物3d(図9B)は術後14日目にi.t.投与した。機械刺激の方法は実施例5と同様であり、i.t.投与後24時間目まで、経時的にデータを取得した。
ホルマリンをマウス後肢に皮下注射すると、動物は注射された後肢を舐めたり、振り回したりするなどの疼痛様行動を示す。この行動は2相性を示し、第1相(ホルマリン投与後10分くらいまで)はホルマリンの感覚神経に対する直接作用、第2相(ホルマリン投与後10分から60分くらいまで)は第1相の間に脊髄後角細胞で形成される過敏化(中枢性感作とも呼ばれる神経系における一種の可塑的現象)が主要な要因となり、ホルマリン投与部位における炎症反応が重要な寄与をする。

Claims (11)
- 前記式(I)において、R1がエトキシ基又はトリフルオロメトキシ基である請求項1記載の化合物もしくはその塩又はそれらの溶媒和物。
- 前記式(I)において、R1がエトキシ基又はトリフルオロメトキシ基である請求項3記載の鎮痛薬。
- 神経障害性疼痛、がん化学療法に伴う疼痛、糖尿病性神経痛、糖尿病性末梢神経障害性疼痛及び片頭痛からなる群から選択される疾患又は症状を治療及び/又は予防するための請求項3又は4記載の鎮痛薬。
- 前記式(II)において、Rがハロゲン原子で置換されたインダゾリル基である請求項6記載の鎮痛薬。
- 前記式(II)において、Rが塩素原子で置換されたインダゾリル基である請求項6記載の鎮痛薬。
- 神経障害性疼痛、がん化学療法に伴う疼痛、糖尿病性神経痛、糖尿病性末梢神経障害性疼痛及び片頭痛からなる群から選択される疾患又は症状を治療及び/又は予防するための請求項6~8のいずれか1項に記載の鎮痛薬。
- 前記式(IIa)において、R’が塩素原子で置換されたインダゾリル基である請求項10記載の化合物もしくはその塩又はそれらの溶媒和物。
