Abstract
Description
〔1-1.材料〕
以下に示す材料を被験試料の調製のために準備した。
・大豆抽出物(フェリチン鉄として10質量%含有、鉄として5質量%含有):株式会社アンチエイジング・プロ社製
・イヌリン(アガベ由来)(平均分子量約5000):株式会社アルマテラ社製
・難消化性デキストリン(平均分子量約2000):松谷化学工業株式会社製
・αシクロデキストリン(分子量973):株式会社シクロケム
・アラビアガム(平均分子量約200000):ネキシラ株式会社
以下に示す試験群を各被験試料ごとに設定した。
・試験群1-1:大豆抽出物200mg(鉄として10mg)
・試験群1-2:大豆抽出物200mg(鉄として10mg)とイヌリン1000mg とを混合した
・試験群1-3:大豆抽出物200mg(鉄として10mg)とイヌリン1000mgとを混合し、造粒した
・試験群1-4:大豆抽出物200mg(鉄として10mg)と難消化性デキストリン1000mgとを混合した
・試験群1-5:大豆抽出物200mg(鉄として10mg)とαシクロデキストリン1000mgとを混合した
・試験群1-6:大豆抽出物200mg(鉄として10mg)とアラビアガム 1000mgとを混合した
・試験群1-7:大豆抽出物200mg(鉄として10mg)+アラビアガム 1000mgとを混合し、造粒(流動層造粒)した
鉄剤が苦手な女性16名(20~39歳)に、各試験群に対応するように被験試料を変えながら6日間にわたり朝食前の空腹時に摂取してもらい、その摂取後の胸やけの症状について評価してもらった。評価は、下記に示す胸焼けスコアの基準で1点ごとに10段階で点数付けをしてもらうアンケートを実施することにより行った。
(胸やけスコア)
0点:胸焼けが全くない、10点:胸焼けが非常に強い
(1)大豆抽出物だけを摂取した試験群1-1では、胸やけスコアの平均値が4.5あったのに対して、イヌリン、難消化性デキストリン、αシクロデキストリン、アラビアガムなどの難消化性多糖類を併用すると、胸やけの症状が軽くなることが明らかとなった(試験群1-2~試験群1-7)。
(2)試験群1-6と試験群1-1との比較から、アラビアガムによる胸やけの症状の軽減の効果は限定的であり、造粒加工で加熱された場合のみ(試験群1-7)、効果が示された。
(3)なお、大豆抽出物だけを摂取した際の被験者は、他の鉄剤に比べて胸やけの症状が少ないとの評価であった。また、本試験例全般にわたって鉄剤特有の便秘の症状は認められなかった。
試験例1で使用した大豆抽出物について、フェリチン鉄由来の鉄量と遊離鉄ならびに鉄塩として存在する鉄量の比率を計測するため、その大豆抽出物を、ビタミンCを0.1w/v%含有する水溶液に溶解し、遠心式限外ろ過フィルターユニット(分画分子量:10000)にて高分子層と低分子層を分離し、回収した。高分子層と低分子層とについて、それぞれの鉄含有量をICP-MS(日本食品分析センター)で定量した。また、フィルターユニットにかける前の試料についても同様にして鉄含有量を測定した。
〔3-1.材料〕
以下に示す材料を被験試料の調製のために準備した。
・大豆抽出物(フェリチン鉄として10質量%含有、鉄として5質量%含有):株式会社アンチエイジング・プロ社製
・ビタミンC:東北製薬社製
・ゼイン(トウモロコシたん白):商品名「小林ツェインDP-N」、小林香料株式会社製
・腸溶性ハードカプセル:商品名「DRcaps」、Lonza社製
・豚ゼラチンハードカプセル:商品名「ハードカプセル」、カプスゲル・ジャパン社製
以下に示す試験群を各被験試料ごとに設定した。
・試験群3-1:大豆抽出物200mg(鉄として10mg)をゼインでコーティング(腸溶コーティング錠)
・試験群3-2:大豆抽出物200mg(鉄として10mg)とビタミンC50mgとを混合し、ゼインでコーティング(腸溶コーティング錠)
・試験群3-3:大豆抽出物200mg(鉄として10mg)を腸溶性ハードカプセルに封入(腸溶カプセル)
・試験群3-4:大豆抽出物200mg(鉄として10mg)とビタミンC50mgとを腸溶性ハードカプセルに封入(腸溶カプセル)
・試験群3-5:大豆抽出物200mg(鉄として10mg)を豚ゼラチンハードカプセルに封入(ゼラチンカプセル)
・試験群3-6:大豆抽出物200mg(鉄として10mg)とビタミンC50mgとを豚ゼラチンハードカプセルに封入(ゼラチンカプセル)
鉄不足を実感する女性30名(20~39歳)に対して、月経終了時に採血を行った後、被験試料の摂取を開始し、摂取開始から10日後に再度採血を行って、ヘモグロビン値を摂取前後で比較した。なお、被験食品の摂取は朝食前の空腹時とし、各被験試料につき5名で行った。
(1)試験群3-1~試験群3-6の結果から、大豆抽出物の摂取によりヘモグロビン値が11.3~26.9%の変化率で上昇が認められた。
(2)試験群3-2(変化率16.4%)と試験群3-1(変化率11.3%)との比較や、試験群3-4(変化率26.9%)と試験群3-3(変化率14.4%)との比較や、試験群3-6(変化率13.8%)と試験群3-5(変化率13.3%)との比較から、大豆抽出物に加えてビタミンCを摂取することにより、ヘモグロビン値のより高い上昇が得られることが明らかとなった。
(3)上記(2)のうち、試験群3-2と試験群3-1との比較(変化率の差が5.1%)や試験群3-4と試験群3-3との比較(変化率の差が12.5%)に対し、試験群3-6と試験群3-5との比較(変化率の差が0.5%)から、大豆抽出物に加えてビタミンCを摂取することによりヘモグロビン値のより高い上昇が得られる効果は、腸溶加工したときにより顕著であった。
(4)なお、本試験例全般にわたって、被験者から胸やけなどの症状の訴えは認められなかった。
Claims (4)
- 難消化性多糖類を有効成分とすることを特徴とする豆由来フェリチン鉄供与材に含まれる遊離鉄及び鉄塩による胸やけの抑制剤(但し、有効成分がペクチンである場合を除く)。
- 前記難消化性多糖類として、分子量1万以下のものを用いる、請求項1記載の剤。
- 前記難消化性多糖類として、イヌリン、難消化性デキストリン、及びシクロデキストリンからなる群から選ばれた1種又は2種以上を用いる、請求項1又は2記載の剤。
- 前記難消化性多糖類は、前記豆由来フェリチン鉄供与材に含まれる遊離鉄及び鉄塩の合計量の1質量部に対して10~1000質量部を使用する、請求項1~3のいずれか1項に記載の剤。
