Description
高純度医療用アテロコラーゲンをNaOAc/HAc溶液(1.2g CH3COONa、27.5ml CH3COOH in 50ml)に3wt%で入れ、pH3.0に維持し、攪拌して完全に溶かした。この溶液をTFF(タンジェンシャルフロー・フィルトレーション(Tangential Flow Filtration))によりPBS溶液でダイアフィルトレーション(dia-filtration)して、PBS溶液の医療用アテロコラーゲン分散液を製造した。
実施例2.抗-ヌクレオリンGROアプタマー-リンカー-ゲムシタビンコンジュゲート(anti-Nucleolin GRO aptamer-linker-Gemcitabine Conjugate)[AS1411-Gemコンジュゲート]の合成
マレイミドカプロイル-(Gly-Phe-Leu-Gly)-ゲムシタビン(Maleimidecarproyl-(Gly-Phe-Leu-Gly)-Gemcitabine)[MC-GFLG-ゲムシタビン(Gemcitabine)]に、HS-C6-tttggtggtggtggttgtggtggtggtgg[HS-C6-T3-AS1411]を反応し、ゲムシタビン(Gemcitabine)-(Gly-Leu-Phe-Gly)-Mal-S-C6-tttggtggtggtggttgtggtggtggtgg[Gemcitabine-(GLFG-MC-S-C6)-T3-AS1411,AS1411-Gemコンジュゲート]を合成した。つまり、RSS-C6-tttggtggtggtggttgtggtggtggtgg[RSS-C6-T3-AS141])をDTTの存在下で約3時間還元反応した後、残っているDTTをセントリコン(centricon)で除去し、SB17緩衝液に交換した。少量のDMSOに溶かしたMal-GPLG-ゲムシタビン(Gemcitabine)を入れて一晩振とうさせた。逆相(Reverse-phase)HPLC(Waters-Xbridge OST C18 10×50mm、65、TEAE/ACN緩衝液)により精製し、ゲムシタビン(Gemcitabine)-(GLFG-MC-S-C6)-T3-AS1411[AS1411-Gemコンジュゲート]を得た。ESI-MSによりゲムシタビン(Gemcitabine)-(GLFG-MC-S-C6)-T3-AS1411[AS1411-Gemコンジュゲート]の分子量を確認した。C334H422F2N117O198P29S[Cal.MW=10211.90 Obs.MW=10211.87]

AS1411にドキソルビシン-HCl(Doxorubicin-HCl)(アドリアマイシン-HCl(Adramycin-HCl))を反応させて、AS1411-Dox付加(AS1411-Dox adduct)を合成した。つまり、0.5mlの緩衝液(20mMリン酸ナトリウム、150mM NaCl、0.5mM EDTA;pH7.0)に20uMのGRO、150uMのドキソルビシン-HCl(Doxorubicin-HCl)、0.37%ホルムアルデヒドを入れて10℃で一晩振とうしながら反応を進行した。反応後、逆相(Reverse-phase)HPLC(Waters-Xbridge OST C18 10×50mm、0.1M TEAE/ACN緩衝液、260nm、490nm)により分離/精製した。分離精製した後、セントリコン(centricon)(ミリポア(Millipore)、MW3000 cut)を用いて濃縮および脱塩を行った。
AS1411-Dox付加(AS1411-Dox adduct)は、HPLCリテンションタイム(retention time)の確認とUV-Visスペクトラ(spectra)、ESI-MSにより確認した(図1および図2)。
コントロールアプタマー(Control aptamer)にドキソルビシン-HCl(Doxorubicin-HCl)(アドリアマイシン-HCl(Adramycin-HCl))を反応してコントロール-Dox付加(control-Dox adduct)を合成した。つまり、0.5mlの緩衝液(20mMリン酸ナトリウム、150mM NaCl、0.5mM EDTA;pH7.0)に20uMのコントロールアプタマー(Control aptamer)、150uMのドキソルビシン-HCl(Doxorubicin-HCl)、0.37%ホルムアルデヒドを入れて10℃で一晩振とうしながら反応を進行した。反応後、逆相(Reverse-phase)HPLC(Waters-Xbridge OST C18 10×50mm、0.1M TEAE/CAN緩衝液、260nm、490nm)により分離/精製した。分離精製した後、セントリコン(centricon)(ミリポア(Millipore)、MW3000 cut)を用いて濃縮および脱塩を行った。
CRO-Dox付加(CRO-Dox adduct)は、HPLCリテンションタイム(retention time)の確認とUV-Visスペクトラ(spectra)、ESI-MSにより確認した。
本発明者は、KR10-1279584およびUS9309515B2で出願したERBB2アプタマー[a6g66agag666gcc6gag6gcc6cgcaagggcg6aacaa、6=NapdU[5-(N-ナフチルカルボキシアミド)-2’-デオキシウリジン(5-(N-Naphthylcarboxyamide)-2´-deoxyuridine)]]からERBB2アプタマー-siRNAコンジュゲートを合成した。細胞内でBcl-2を標的とするsiRNAを末端部分にマレイミド基を有するように合成した。そしてERBB2アプタマーの末端には、チオール基で変形して合成した。チオール化したアプタマーとマレイミド-siRNAを8mM スルフォ-SMPB(Sulfo-SMPB)とともに反応し、ERBB2アプタマーは、10mM TCEPが溶けているPBSに37℃、1時間反応して活性化した。そして、この2つの物質をセントリ-セップ(Centri-Sep)カラムで精製して準備し、2つの物質をよく混ぜた後、37℃で2時間反応した。最後に、結合が完了した2つの物質をPAGE精製し、UV分光光度計で定量した(図3)。
実施例7.アテロコラーゲン-[アプタマー-薬物コンジュゲート]複合体(ゾル-ゲルタイプ)組成物の製造
合成により製造したアプタマー-薬物(drug)コンジュゲートをPBSに入れ、室温でミキサー(mixer)を用いて完全に溶かした。このようにして準備した試薬を高濃度アテロコラーゲン分散液と適正濃度0.5~3.0%で混ぜて、室温でローテーターカフ(rotator cuff)装置を用いて30分間混合液になるようにした。低い温度で液体状態(ゾル(Sol))であるアテロコラーゲン-[アプタマー-薬物コンジュゲート]複合体(ゾル-ゲルタイプ)は、生体内の腫瘍に直接注入時37℃で固形化(ゲル(Gel))され、アプタマー-薬物コンジュゲート(ゾル-ゲルタイプ)を包んでいたアテロコラーゲンが徐々に溶けながら薬物が徐々に放出され、効果的な腫瘍治療剤として使用可能である(図4)。
1)膵臓がん:アテロコラーゲン-[AS1411-Gemコンジュゲート]複合体(Complex)(ゾル-ゲルタイプ)
2)乳がん:アテロコラーゲン-[ERBB2アプタマー-siRNAコンジュゲート]複合体(Complex)(ゾル-ゲルタイプ)
3)肝臓がん:アテロコラーゲン-[AS1411-Dox付加]複合体(Atelocollagen-[AS1411-Dox Adduct]Complex)(ゾル-ゲルタイプ)
先に製造した高濃度コラーゲン分散液と合成されたアプタマー-薬物コンジュゲート(Aptamer-Drug Conjugate)をゾル-ゲルタイプと同様な方法により混合液を作成した。準備された混合液を48ウエル細胞培養プレート(48 well cell culture Plate)に入れて拡散させて0.5mmの均一な膜を形成した後、零下80℃で凍結させた。凍結が完了した試料を-70℃に維持された凍結乾燥機で30時間凍結乾燥し、多孔性膜を製造した(図5)。
生体適合性、生分解性天然高分子であるアテロコラーゲン分散液と合成されたアプタマー-薬物コンジュゲート(Aptamer-Drug Conjugate)を材料として、3次元構造を有するアテロコラーゲン-[アプタマー-薬物コンジュゲート]複合体(スカフォールドタイプ)を製造した。アテロコラーゲンが分解されて出てくるアプタマー-薬物コンジュゲートが、がん細胞に選択的に作用できるように結合した。製造には、3Dプロッター(3D plotter)にSFF(solid free-form fabrication)工程を適用し、製造方法としては、アテロコラーゲン-[アプタマー-薬物コンジュゲート]複合体混合溶液を精密ノズル(直径300μm)が結合されたバレル(barrel)に注入し、3Dプロッター(3D plotter)を用いて、37℃のプレート(Plate)に吐出圧力300±50kPa、フローティング速度5mm/sで固定して、3層(3 layer)の3次元アテロコラーゲン-[アプタマー-薬物コンジュゲート]複合体(スキャナフォールドタイプ)を作製した。
実施例10.アテロコラーゲン-[アプタマー-薬物コンジュゲート]複合体組成物の生体内安定性の実験(ゾル-ゲルタイプ)
Cy3で標識されたAS1411-Gem Conjugate(AS1411-Gemコンジュゲート)[Cy3-AS1411-Gemコンジュゲート((Cy3-AS1411-Gem Conjugate))]およびアテロコラーゲン-[Cy3-AS1411-Gemコンジュゲート]複合体(Atelocollagen-[Cy3-AS1411-Gem Conjugate]Complex)を凍結乾燥してシートに製作した。シートは5mmと8mmの大きさで0.5%、1.5%、3.0%の濃度のアテロコラーゲンを用いて製作した(図8)。
準備されたシートを、マウスの表皮層に移植して分解度を肉眼で確認した。0.5%と1%のシートは、5日内にすべて溶けて吸収されることが観察された。1.5%のシートは、5日経過時は一部残っていて、8日目にすべて溶けた。3.0%のシートは、8日経過後も完全に溶けずに維持されることが観察された(図9)。
アテロコラーゲンを3Dプラッター(3D plotter)を用いて、格子構造の3次元細胞担体として製作した。細胞担体は、5×5×2mm3、10×10×2mm3の大きさに0.5%、1.5%、3.0%の濃度のアテロコラーゲンを用いて製作し、生体内での分解速度を調節するために、1-エチル-(3-3-ジメチルアミノプロピル)カルボジイミドヒドロクロリド(EDC、Sigma Chemical Co.,St.Louis、MO、USA)溶液で24時間交差結合した。または生体適合性物質であるゲニピンを用いて架橋化した。このようにして得られた支持体は、超音波破砕機を使用して蒸留水で洗浄した後、-50℃で再び冷凍乾燥処理を行った(図10a、図10b)。
架橋化したスカフォールドを先に実施した方法と同様の方法で、マウスの表皮層に移植して経過を観察した。移植後8日が経過した時点でもほとんど分解されていないことが確認された。架橋化による分解速度の減少が顕著であることを確認した(図11)。
Cy3-AS1411-Gemコンジュゲート((Cy3-AS1411-Gem Conjugate))をこのスカフォールドに担持して電子顕微鏡で観察した(図12)。
実施例13.アプタマー-薬物コンジュゲート(Aptamer-Drug Conjugate)の膵臓がんに対するin vitro効能
膵臓がん細胞株BxPC3、Miapaca-2、Panc-1、capan-1に対してWST-1アッセイ(assay)を行った。膵臓がん細胞株でAS1411-Gemコンジュゲート(AS1411-Gem Conjugate)細胞阻害(cell inhibition)効能の検証をWST-1アッセイ(assay)によりin vitroで確認した。まず、抗がん剤が結合されていない裸の(naked)形態であるコントロールアプタマーCRO(control aptamer CRO)、および、がん細胞標的化機能を持っているAS1411、そしてそれらにそれぞれ、膵臓がんに効果のある抗がん剤であるゲムシタビンが結合している形態のCRO-Gemコンジュゲート(CRO-Gem Conjugate)およびAS1411-Gemコンジュゲート(AS1411-Gem Conjugate)を準備した(表2)。
膵臓がん細胞株であるBxPC3、Miapaca-2、Panc-1、capan-1(ATCC、RPMI+10%FBS)は、適正細胞濃度を決定するcell testにより定められた細胞数(cell number)1~2×104個/ウエル(well)を96ウエルプレート(96 well Plate)に蒔種(seeding)した後、一日育てた。アプタマー(Aptamer)およびアプタマー-薬物コンジュゲート(Aptamer-Drug Conjugate)PBSに溶けており、0.1uMで最大100uMの濃度で各ウエル(well)にすぐに処理した。処理された細胞(cell)は、5%CO2インキュベーター(incubator)で72時間培養した後、WST-1アッセイ(assay)用試薬溶液(WST-1、Roche)を10ulずつ処理し、ELISAリーダー(reader)機で440nmでの吸光度を測定した(図13)。
膵臓がん細胞株Miapaca-2およびMCF-7に対してWST-1アッセイ(assay)を行った。膵臓がん細胞株でアテロコラーゲン-[AS1411-Gemコンジュゲート]複合体(Atelocollagen-[AS1411-Gem Conjugate]Complex)の細胞阻害(cell inhibition)効能の検証をWST-1アッセイ(assay)によりin vitroで確認した。まず、抗がん剤が結合していない裸の(naked)形態であるコントロールアプタマー(control aptamer)CRO、および、がん細胞標的化機能を持っているAS1411、そしてそれらにそれぞれ、膵臓がんに効果のある抗がん剤であるゲムシタビンが結合している形態のアテロコラーゲン-[AS1411-Gemコンジュゲート]複合体(Atelocollagen-[AS1411-Gem Conjugate]Complex)およびアテロコラーゲン-[CRO-Gem コンジュゲート]複合体(Atelocollagen-[CRO-Gem Conjugate]Complex)を準備した。
膵臓がん細胞株であるMiapaca-2およびMCF-7(ATCC、RPMI+10%FBS)は、適正細胞濃度を決定する細胞試験(cell test)により定められた細胞数(cell number)1~2×104個/ウエル(well)を96ウエルプレート(96 well Plate)に蒔種(seeding)した後、一日育てた。アプタマー(Aptamer)およびアテロコラーゲン-[アプタマー-薬物コンジュゲート]複合体(Atelocollagen-[Aptamer-Drug Conjugate]Complex) PBSに溶けており、0.1uMで最大100uMの濃度で各うえる(well)にすぐに処理した。処理された細胞(cell)は、5%CO2インキュベーター(incubator)で72時間の間インキュベーション(incubation)させた後、WST-1アッセイ(assay)用試薬液(reagent solution)(WST-1、Roche)を10μLずつ処理し、ELISAリーダー(reader)機で440nmでの吸光度を測定した(図14)。
乳がん標的細胞に対するターゲティングを確認するために、siRNAにCy3に変形されたERBB2アプタマー-リンカー-siRNA-Cy3(ERBB2 aptamer-linker-siRNA-Cy3)と、対照群であるsiRNA-Cy3を製作した。ERBB2が過発現されたMCF7細胞ライン(cell line)にこの2つの合成物質を処理して蛍光顕微鏡で確認した。何の処理もしていないものとCy3のみが結合しているsiRNA対照群の場合には、細胞が蛍光を示さないことが確認され、ERBB2アプタマー(ERBB2 aptamer)が結合しているものにおいてのみ蛍光を示すことが確認された。この結果から、アプタマーによる標的化によってsiRNAが標的細胞に正確に伝達されることを確認することができる(図15)。
乳がん細胞株BT-474とMCF-7細胞ライン(cell line)でアテロコラーゲン-[ERBB2アプタマー-siRNAコンジュゲート]複合体(Atelocollagen-[ERBB2 aptamer-siRNA Conjugate]Complex)の細胞阻害(cell inhibition)効能の検証をWST-1アッセイ(assay)により行った。裸の(naked)ERBB2と活性効果のないAtelocollagen-[control aptamer-siRNA Conjugate]Complex、そしてがん細胞標的化機能を持っているERBB2標的アプタマーに、抗がん効果に優れた抗がん剤であるsiRNAが結合しているアテロコラーゲン-[ERBB2アプタマー-siRNAコンジュゲート]複合体(Atelocollagen-[ERBB2 aptamer-siRNA Conjugate]Complex)を乳がん細胞株に処理してWST-1アッセイ(assay)を行った。細胞の細胞生存能力(cell viability)および増殖(proliferation)を比較したところ、ERBB2アプタマー-siRNAコンジュゲート(ERBB2 aptamer-siRNA Conjugate)が対照群であるアテロコラーゲン-[コントロールアプタマー-siRNAコンジュゲート]複合体アプタマー(Atelocollagen-[control aptamer-siRNA Conjugate]Complex aptamer)よりも優れた効能を示すことが確認された(図17)。
前述の実験と同様の方法により肝臓がん細胞株HepG2、Huh-7でアプタマー-薬物コンジュゲート(aptamer-drug conjugate)であるAS1411-Dox付加(AS1411-Dox adduct)と、コントロールアプタマー(control aptamer)のコントロール-Dox付加(control-Dox adduct)をもって、細胞阻害(cell inhibition)効能の検証をWST-1アッセイ(assay)によりin vitroで確認した。コントロール-Dox付加(control-Dox adduct)と比較したところ、AS1411-Dox付加(AS1411-Dox adduct)が肝臓がん細胞株でにおいて優れた効能を示すことが確認され、組成物の活性能力を検証した(図18)。
実施例17と同様の方法で肝臓がん細胞株を準備し、抗がん効果のあるアテロコラーゲン-[AS1411-Dox付加]複合体(Atelocollagen-[AS1411-Dox Adduct]Complex)、標的化機能のないアテロコラーゲン-[CRO-Dox付加]複合体(Atelocollagen-[CRO-Dox Adduct]Complex)、そしてアテロコラーゲン-Dox(Atelocollagen-Dox)を処理してWST-1アッセイ(assay)で効能の検証を行った。予想通り、標的化機能のあるアテロコラーゲン-[AS1411-Dox付加]複合体(Atelocollagen-[AS1411-Dox Adduct]Complex)が対照群であるアテロコラーゲン-[CRO-Dox付加]複合体(Atelocollagen-[CRO-Dox Adduct]Complex)と比較して優れた効能を示すことが観察された。コラーゲンとDox単独投与時には、非選択的に細胞に透過による細胞阻害(cell inhibition)が観察された(図19)。
実施例19.アテロコラーゲン-[AS1411-Gem コンジュゲート]複合体(Atelocollagen-[AS1411-Gem Conjugate]Complex)(ゾル-ゲル型(Sol-Gel type))の膵臓がん動物モデルの効能の検証
動物モデルの作成:6週齢Balb/cヌード雌マウス(nude female mouse)を用いて、腫瘍in vivoモデルを作成した。実験に使用されたマウスの平均体重は、20gであった。ヒトに由来する膵臓細胞株であるcapan-1細胞をATCC社から購入して増殖させた。増殖されたcapan-1細胞を5×107細胞数分の量でマウスに皮下注射して腫瘍を形成した。注射後2~3週間毎日観察して腫瘍の成長を肉眼で確認し、キャリパー(caliper)を使用して腫瘍の大きさを測定した。
複合体の効能の検証:腫瘍が適当な大きさに成長した後、前記表3に示すようなアテロコラーゲン-[AS1411-Gem コンジュゲート]複合体(Atelocollagen-[AS1411-Gem Conjugate]Complex)(ゾル-ゲル型(Sol-Gel type))をマウスの腫瘍部位に注射器で直接注入した(表3)。
動物モデルの作成:6週齢Balb/cヌード雌マウス(nude female mouse)を用いて、腫瘍in vivoモデルを作成した。実験に使用されたマウスの平均体重は、20gであった。ヒトに由来する乳がん細胞株であるBT-474細胞をATCC社から購入して増殖させた。増殖されたBT-474細胞を1×107細胞数分の量でマウスに皮下注射して腫瘍を形成した。注射後2~3週間毎日観察して腫瘍の成長を肉眼で確認し、キャリパー(caliper)を使用して腫瘍の大きさを測定した。
実施例21の方法で腫瘍動物モデルを作成し、実施例7および8の方法で製造したアテロコラーゲン-[ERBB2アプタマー-siRNAコンジュゲート]複合体(Atelocollagen-[ERBB2 aptamer-siRNA Conjugate]Complex)(パッチタイプ)、およびアテロコラーゲン-[ERBB2アプタマー-siRNAコンジュゲート]複合体(Atelocollagen-[ERBB2 aptamer-siRNA Conjugate]Complex)(スカフォールドタイプ)を腫瘍周囲に移植した。
移植されたアテロコラーゲン-[ERBB2アプタマー-siRNAコンジュゲート]複合体(Atelocollagen-[ERBB2 aptamer-siRNA Conjugate]Complex)(パッチタイプ)、およびアテロコラーゲン-[ERBB2アプタマー-siRNAコンジュゲート]複合体(Atelocollagen-[ERBB2 aptamer-siRNA Conjugate]Complex)(スカフォールド型(scaffold type))の抗がん効果を測定するために、一定期間、腫瘍のサイズ変化と移植体の重量変化を観察した。アテロコラーゲン-[ERBB2アプタマー-siRNAコンジュゲート]複合体(Atelocollagen-[ERBB2 aptamer-siRNA Conjugate]Complex)(スカフォールドタイプ)がアテロコラーゲン-[ERBB2アプタマー-siRNAコンジュゲート]複合体(Atelocollagen-[ERBB2 aptamer-siRNA Conjugate]Complex)(パッチタイプ)よりも更に良い抗がん効果を示すことを確認した(図28)。
動物モデルの作成:6週齢Balb/cヌード雌マウス(nude female mouse)を用いて、腫瘍in vivoモデルを作成した。実験に使用されたマウスの平均体重は、20gであった。ヒトに由来する肝臓がん細胞株であるHuh-7細胞をATCC社から購入して増殖させた。増殖されたHuh-7細胞を5×106細胞数分の量でマウスに皮下注射して腫瘍を形成した。注射後2~3週間毎日観察して腫瘍の成長を肉眼で確認し、キャリパー(caliper)を使用して腫瘍の大きさを測定した。
腫瘍が適当な大きさに成長した後、前記表5に示すようなアテロコラーゲン-[AS1411-Dox付加]複合体(Atelocollagen-[AS1411-Dox adduct]Complex)(ゾル-ゲル型(Sol-Gel type)を注射器でマウスの腫瘍に直接注入した。そして、複合体の抗がん効果を測定するために、一定期間、腫瘍のサイズ変化と移植体の重量変化を観察した。対照群の0.5%アテロコラーゲンのみを注入した場合と、活性のないアテロコラーゲン-[CRO-Dox付加]複合体(Atelocollagen-[CRO-Dox adduct]Complex)(ゾル-ゲル型(Sol-Gel type))と比較したところ、アテロコラーゲン-[AS1411-Dox付加]複合体(Atelocollagen-[AS1411-Dox adduct]Complex)(ゾル-ゲル型(Sol-Gel type))が優れたがん抑制効果を示すことが観察された(図29)。
Claims (17)
- 前記アテロコラーゲン分散液は、アテロコラーゲンNaOAc/HAc溶液をTFF(タンジェンシャルフロー・フィルトレーション)によりPBS溶液でダイアフィルトレーションして得られたものである、請求項1に記載の薬物伝達体。
- 前記アテロコラーゲン-[アプタマー-薬物]複合体は、アプタマー-薬物コンジュゲートをPBS分散液の状態でアテロコラーゲンPBS分散液と混合して得られたものである、請求項1に記載の薬物伝達体。
- 前記複合体は、固形化されてアプタマー-薬物コンジュゲートを包んでいるアテロコラーゲンが体温により溶けるゾル-ゲル型である、請求項3に記載の薬物伝達体。
- 前記複合体は、ゾル型の複合体を細胞培養プレートに入れて拡散させて膜を形成し、これを凍結乾燥した多孔性膜状である、請求項3に記載の薬物伝達体。
- 前記複合体は、ゾル型の複合体をノズルが結合されたバレルに注入し、3Dプラッターを用いてプレートに吐出して得られたスカフォールド型である、請求項3に記載の薬物伝達体。
- 前記アプタマー-薬物コンジュゲートは、AS1411とゲムシタビンのコンジュゲートである、請求項1に記載の薬物伝達体。
- 前記アプタマー-薬物コンジュゲートは、ゲムシタビン-(Gly-Leu-Phe-Gly-マレイミドカプロイル-S-(CH2)6)-ttt-AS1411、AS1411-ドキソルビシン付加、またはERBB2-siRNAコンジュゲートである、請求項1に記載の薬物伝達体。
- 請求項1に記載の薬物伝達体を含む抗がん組成物。
- 前記抗がん組成物は、がん手術前の患者に投与して腫瘍の大きさを縮小するためのものである、請求項9に記載の抗がん組成物。
- 前記抗がん組成物は、がん手術後の患者に投与して残存がん細胞を除去するためのものである、請求項9に記載の抗がん組成物。
- 前記がんは、膵臓がん、乳がんまたは肝臓がんである、請求項9に記載の抗がん組成物。
- 前記アプタマー-薬物コンジュゲートは、AS1411とゲムシタビンのコンジュゲートである、請求項9に記載の抗がん組成物 。
- アテロコラーゲンNaOAc/HAc溶液をTFF(タンジェンシャルフロー・フィルトレーション)によりPBS溶液でダイアフィルトレーションして前記アテロコラーゲン分散液を得るステップをさらに含む、請求項14に記載の薬物伝達体の製造方法。
