Description
[1] 水不溶性担体の表面に、炭化水素基を介してアミノ基、カルボキシル基、リン酸基、ホスフォノ基、ホスフィノ基、チオール基から選択される少なくとも1種が共有結合していることを特徴とする、カルシプロテインパーティクルの吸着材。
[2] 前記炭化水素基が直鎖状炭化水素基である、[1]に記載の吸着材。
[3] 前記炭化水素基に、2つ以上のホスフォノ基が共有結合していることを特徴とする、[1]又は[2]に記載の吸着材。
[4] 前記炭化水素基が直鎖状プロピル基、直鎖状ブチル基、又は直鎖状ヘキシル基であり、その末端の炭素原子に1つの水酸基と2つのホスフォノ基が共有結合している、[1]~[3]のいずれかに記載の吸着材。
[5] 前記炭化水素基が直鎖状ウンデシル基であり、その末端の炭素原子に1つのホスフォノ基が共有結合している、[1]又は[2]に記載の吸着材。
[6] 前記担体が多孔質である、[1]~[5]のいずれかに記載の吸着材。
[7] 体液中に存在するカルシプロテインパーティクルの吸着用である、[1]~[6]のいずれかに記載の吸着材。
[8] [1]~[7]のいずれかに記載の吸着材が液体の入口及び出口を有する容器内に充填されている、カルシプロテインパーティクルの吸着器。
[9] [8]に記載の吸着器と、該吸着器に液体を供給するためのポンプとを備えてなる、透析器または血漿分離器と接続可能な、カルシプロテインパーティクルの吸着システム。
[10] [1]~[7]のいずれかに記載の吸着材とカルシプロテインパーティクルを含有する液体とを接触させる工程を包含する、カルシプロテインパーティクルの吸着方法。
[11] [1]~[7]のいずれかに記載の吸着材とカルシプロテインパーティクルを含有する液体とを接触させる工程を包含する、カルシプロテインパーティクルが除去された液体の製造方法。
多孔質セルロースビーズ(排除限界分子量500万、粒径400~500μm)40mLにアルカリ性水溶液を加えて全容量を80mLにした後、エピクロルヒドリン30mLを加え、40℃で2時間反応させた。反応後ビーズを水で十分洗浄してエポキシ化セルロースビーズを得た。得られたエポキシ化セルロースビーズにパミドロン酸水溶液を加え50℃で5時間以上振盪した。その後、水で十分洗浄してパミドロン酸固定セルロースビーズ(吸着材A)を得た。乾燥させた吸着材Aに硫酸及び硝酸を加えてマイクロウェーブ分解装置で加圧酸分解を行い、ICP-AES法により得られた溶液のP元素の含有量測定を行った。パミドロン酸の固定化量は元素分析の結果から2μmol/mLであった。
実施例1のパミドロン酸水溶液をアレンドロン酸水溶液に代えて、アレンドロン酸固定セルロースビーズ(吸着材B)を得た。アレンドロン酸の固定化量は元素分析の結果から5μmol/mLであった。
実施例1のパミドロン酸水溶液をネリドロン酸水溶液に代えて、ネリドロン酸固定セルロースビーズ(吸着材C)を得た。ネリドロン酸の固定化量は元素分析の結果から6μmol/mLであった。
実施例1で得られたエポキシ化セルロースビーズに11-メルカプトウンデシルホスホン酸を含むエタノール水溶液を加え40℃で24時間振盪した。その後、水で十分洗浄して11-メルカプトウンデシルホスホン酸固定セルロースビーズ(吸着材D)を得た。11-メルカプトウンデシルホスホン酸の固定化量は元素分析の結果から10μmol/mLであった。
実施例1~4の吸着材A、B、C、Dのビーズ30μLにカルシプロテインパーティクル(CPP)を含む血清120μLを加え、室温で1時間振盪した。未処理血清と振盪1時間後の上清中のCPP量は表1のようになった。
CPPを含む測定対象液に近赤外線蛍光プローブ(Osteosense(登録商標))を加えてCPPに結合させた後、ゲル濾過スピンカラムで分画し、CPPを含む高分子量画分の蛍光を近赤外線スキャナーで定量した。

Claims (9)
- 水不溶性担体に、炭化水素基を介して少なくとも2つの電子供与性基が共有結合しており、前記電子供与性基が、窒素原子、硫黄原子、若しくはリン原子に水素原子が結合した基、又は酸性プロトンを有する基であることを特徴とする、リン酸カルシウムとタンパク質との複合体であるカルシプロテインパーティクルの吸着材。
- 前記炭化水素基の最長部分の炭素数が1以上20以下である、請求項1に記載の吸着材。
- 前記電子供与性基が電子供与性基間の炭素原子の数が1~5個となるものを含む、請求項1又は2に記載の吸着材。
- 前記炭化水素基に前記電子供与性基以外の極性基がさらに結合している、請求項1~3のいずれかに記載の吸着材。
- 前記極性基が水酸基である、請求項4に記載の吸着材。
- 前記電子供与性基が電子供与性基と前記水酸基との間の炭素原子の数が1~5個となるものを含む、請求項5に記載の吸着材。
- 前記水不溶性担体の形状が粒状、板状、繊維状、中空糸状、および不織布状のいずれかであることを特徴とする、請求項1~6のいずれかに記載の吸着材。
- 請求項1~7のいずれかに記載の吸着材が、液体の入口及び出口を有する容器内に充填されている、カルシプロテインパーティクルの吸着器。
- 請求項8に記載の吸着器と、前記吸着器に液体を供給するためのポンプとを備えてなる、カルシプロテインパーティクルの吸着システム。
