Description
DBTA組成物のうちの1つは、43塩基のオリゴヌクレオチドセグメントによるタンデムリピートを含むヒトベータ-アクチンに由来するローリングサークル法による増幅産物(RCAact)などの捕捉用DNAポリマー、ならびに以下の構成要素:(1)ヒトベータ-アクチンに由来する20塩基のDNA配列(o20b(+)act)(配列番号4、第1の結合性DNA配列)に共有結合的に付加した抗ヒトCD3モノクローナル抗体(抗CD3、T細胞活性化剤)、および(2)これもまたヒトベータ-アクチンに由来する20塩基のDNA配列(o20b(+)act)(配列番号4、第2の結合性DNA配列)に共有結合的に付加した抗ヒトCD28モノクローナル抗体(抗CD28)を含む。このようなDBTA組成物を、「DBTA[抗CD3-o20b(+)act+抗CD28-o20b(+)act]」と表す。別のDBTA組成物は、ローリングサークル法による増幅産物(RCAact)、ならびに以下の構成要素:(1)ヒトベータ-アクチンに由来する20塩基のDNA配列(o20b(+)act)(配列番号4、第1の結合性DNA配列)に共有結合的に付加した抗CD3、および(2)抗CD28を含む。このようなDBTA組成物を、「DBTA[抗CD3-o20b(+)act+非修飾抗CD28]」と表す。RCAの鋳型およびローリングサークル法による増幅産物であるRCAactを作出するためのプライマーの配列、ならびに対応する第1の結合性DNA配列、および第2の結合性DNA配列を、下記の表1に示す。
第1の結合性DNA配列および第2の結合性DNA配列の、抗CD3および抗CD28抗体(Ab)のそれぞれへの共有結合的付加は、下記で記載される、マレイミド-チオールカップリング戦略を介して進める。以下の、抗CD3-o20b(+)act(抗CD3-DNA)コンジュゲートおよび抗CD28-o20b(+)act(抗CD28-DNA)コンジュゲートの、合成、精製、および特徴付けについての具体的記載は、異なる抗体(Ab)クローンのほか、異なる長さおよび組成の、異なる第1および第2の結合性核酸配列のための、一般的なコンジュゲーション戦略として適合させることができる。
マレイミドにより活性化させた第1の結合性DNA配列および第2の結合性DNA配列の調製
この例では、第1の結合性DNA配列および第2の結合性DNA配列は、同じヌクレオチド配列を有する。工程は、第1の結合性DNA配列について記載されているが、また、第2の結合性DNA配列へも適用される。出発DNA配列(prot-mal-o20b(+)act)(TriLink Biotechnologies、San Diego、CA、USA)は、N末端のC6スペーサーでキャッピングされた、20塩基の第1の結合性DNA配列に続き、保護された三環式のマレイミド部分(prot-mal)を含む。反応性マレイミド活性化誘導体(mal-o20b(+)act)は、prot-mal-o20b(+)actから、逆電子要請型ディールス-アルダー脱保護ステップを介して作出した。マレイミドにより活性化させた第1の結合性DNA配列(mal-o20b(+)act)を、無水トルエン中に懸濁させ(約1mg/mL)、90℃で4時間にわたり加熱した。ベンチトップ型遠心分離を使用して、マレイミドにより活性化させた第1の結合性DNA配列(mal-o20b(+)act)を沈殿させ、溶媒を除去した。沈殿したmal-o20b(+)actを、低温エタノール(3×1mL)で洗浄した。減圧下で、残留有機溶媒をさらに低減したら、洗浄された固体生成物を、100mMのHEPES緩衝液、pH 7.3中に溶解させた。mal-o20b(+)actの最終濃度は、UV-Vis分光法(NanoDrop(商標)Spectrophotometer、Thermo Fisher Scientific、Waltam、MA、USA)を介して決定した。結果として得られる原液(DNA中に0.5~1mM)を、チオール修飾抗体による抗体コンジュゲーションのために、直接使用した。残りのmal-o20b(+)act原液は、反応性を著明に喪失させずに、-20℃で、数カ月間にわたり保管した。
チオール修飾抗体中間体の調製
トラウト試薬(2-イミノチオラン塩酸塩)の10mMの原液を、100mMのHEPES緩衝液、pH 7.3中でまず調製した。次いで、PBS中に1mg/mLの、抗CD3 Ab(OKT3 クローン、eBioscience、Thermo Fisher Scientific、Waltam、MA、USA)または抗CD28 Ab(9.3クローン、GeneTex、USA)溶液を、20倍濃度のpH 8.5ホウ酸緩衝液(Thermo Fisher Scientific、Waltam、MA、USA)および10~15mMのトラウト試薬原液の両方と、8:1:1の容量比で混合した。結果として得られる溶液を、完全に混合し、室温で、0.75~1.25時間にわたりインキュベートした。トラウト混合物の未使用分は、反応性を大きく喪失させずに、-20℃で、数カ月間にわたり保管しうるが、新鮮な溶液が好ましい。抗体を活性化させた後、100mMのHEPES、pH 7.3緩衝液で平衡化させた、従来の脱塩カラム(例えば、NAP-5またはPD-10、GE Healthcare Life Sciences)を使用して、反応混合物を精製した。次いで、回収画分を、UV-Vis分光法(NanoDrop(商標)Spectrophotometer、Thermo Fisher Scientific、Waltam、MA、USA)により、速やかに解析した。この段階で結果として得られるタンパク質の回収率は、抗体について公知の、280nmにおけるモル吸光係数を使用して測定される、抗CD3および抗CD28抗体のいずれについても、>60%であることが典型的であった。
マレイミド活性化DNA配列(第1の結合性DNA配列および第2の結合性DNA配列)の、チオール修飾Ab中間体とのコンジュゲーションであって、Ab-DNAコンジュゲート(抗CD3-o20b(+)actおよび抗CD28-o20b(+)act)を作出するコンジュゲーション
抗体-DNAコンジュゲートを作出するために、ある容量のmal-o20b(+)actを、調製されたばかりの精製チオール活性化抗体のアリコートへと添加して、10~40:1のo20b(+)act:Abの標的モルインプット比を達成した。完全に混合した後、結果として得られる溶液を、25℃で、一晩(16~24時間)にわたりインキュベートした。最終的なコンジュゲートの精製は、飽和塩化アンモニウム溶液を使用する、Abの選択的沈殿を介して達成する。第一に、総反応容量と等容量の飽和塩化アンモニウム溶液を添加し、完全に混合し、氷上に置いた。15分後、試料を、10℃、15,000×gの相対遠心力(rcf)で、10分間にわたり遠心分離した。上清を除去した後、適切な最小容量の、0.1Mリン酸ナトリウム、0.15M NaClによる、pH 7の緩衝液を添加して、最終ペレットを再溶存させた。抗体-DNAコンジュゲート(Ab-DNA)の最終的な回収率および標識化効率は、DNA含量(ε=210,100M-1cm-1)を決定するために、Pierce BCA Protein Assay Kit(Thermo Scientific、Waltam、MA、USA)を、A260の測定(NanoDrop(商標)Spectrophotometer、Thermo Fisher Scientific、Waltam、MA、USA)と組み合わせて使用して決定した。これらの条件下で、1つ~5つの、第1の結合性DNA配列を、抗CD3抗体の各分子に付加することができ、かつ/または1つ~5つの、第2の結合性DNA配列を、抗CD28抗体の各分子に付加することができる。付加は、一般に、>60%の最終コンジュゲート回収率で達成される。代替的に、抗体-DNAコンジュゲートの標識化および収率はまた、参考文献である、Anal Chem. 2014 Apr 15; 86(8): 3869-3875の付録に記載されている方法により測定することもできる。コンジュゲート純度のさらなる確認は、解析的サイズ除外クロマトグラフィー(SEC)を使用し、標準タンパク質サイズの検量線に照らして決定する。典型的な解析的SEC条件は、以下の通り:10μLの試料注射容量、0.5mL/分の流量、30分間にわたる試行であって、100mMのリン酸ナトリウム、100mMの硫酸ナトリウム、0.05%のアジドナトリウムによる、pH 6.7の緩衝液を、TSK Gel 3000SWxLカラム(日本、東京、東ソー株式会社)上で使用する試行である。典型的な解析的SECの溶出時間は、以下の通り:非標識化Ab=16.8~17.0分間、付加を施していないo20b(+)act=20.0分間、Ab-DNA(Ab-o20b(+)act)コンジュゲート混合物=10~15分間である。精製された最終Ab-DNAコンジュゲート(沈殿および再懸濁の後の)は、SEC解析において、結合しなかったDNA中間材料または出発材料の、>95%の除去を示す。
実施例1で調製したAb-DNAコンジュゲートが、それらの、T細胞への特異的な細胞結合能力を保持することを確認するために、フローサイトメトリー(Cytoflex S、Beckman Coulter)を使用して、各コンジュゲートバッチについて、検証研究を実施した。Normal Peripheral Blood(NPB)Pan T Cells(AllCells、USA)を、10mLの温熱完全X-vivo培地(表2を参照されたい)中、37℃で融解させ、次いで、300×gのrcfで、10分間にわたり遠心分離した。次いで、細胞を、10mLの新鮮な完全培地中に再懸濁させ、Nucleocounter(登録商標)NC-200システム上で解析して、細胞のカウントおよび生存率を決定した。濃度を、1mL当たりの細胞1×106個へと調整し、PBSで洗浄した後で、T細胞を、PBS中に10%のNormal Goat Serum(NGS)中、4℃で、15分間にわたりブロッキングした。ブロッキング溶液を除去した後で、細胞を、1%のNGS/PBS溶液中の抗体および抗体-DNAコンジュゲートと共に、4℃で、15分間にわたりインキュベートした。並行する実験において、細胞を、抗CD3-DNA(抗CD3-o20b(+)act)、または抗CD28-DNA(抗CD28-o20b(+)act)と共にインキュベートした。付加を施していない、または非修飾の抗CD3 Abおよび抗CD28 Abと共にインキュベートされた細胞を、陽性対照として使用した。インキュベーション後、細胞を、PBS中で洗浄し、抗CD3 Abおよび抗CD28 Abの両方のマウスアイソタイプに特異的な、フルオロフォア標識化二次抗体(Jackson ImmunoResearch、PA、USA)の、1%NGS/PBS溶液と共にインキュベートした。二次抗体を標識化するための典型的な希釈率は、1mg/mLの原液に対して、1:200である。4℃で15分間にわたるインキュベーションの後で、細胞を、前出と同様に洗浄し、PBS中に再懸濁させ、フローサイトメトリーにより解析して、Ab-DNAコンジュゲートと結合したT細胞の百分率を、非標識化Ab(陽性対照)と結合したT細胞の百分率と比べて決定した。
一本鎖ローリングサークル増幅(ssRCA)の工程であって、ローリングサークル法による増幅産物(例えば、RCAact)を、T細胞を活性化させるための第2の薬剤として作製する工程は、2つのステップ:1)環状の鋳型を作出する、直鎖状DNA鎖のライゲーションと、2)一本鎖のコンカタマーを合成する、ライゲーションされた環状鎖の増幅とを含む。ライゲーションは、製造元のプロトコール(New England Biolabs(登録商標)、MA、USA)に従い、T4 DNA Ligaseを使用して達した。ライゲーションに、2つのオリゴヌクレオチドを用意した。1つのオリゴヌクレオチド(RCAact no-CpG、配列番号3)は、ヒトβ-アクチン遺伝子の、43塩基のヌクレオチド配列を含有し、5’リン酸基を含有した。この43塩基のオリゴヌクレオチドは、ライゲーションの後、環状の鋳型を形成した。第2のオリゴヌクレオチドは、20塩基の長さ(RCAactプライマー)であり、43塩基のオリゴヌクレオチドの両端に対して相補的であった。この第2のオリゴヌクレオチドを使用して、ライゲーション(43塩基のオリゴヌクレオチドの鋳型依存ライゲーション)の前に、環状鎖を形成し、次いで、かつ、その後のssRCA反応において、環状鎖を増幅した(図2を参照されたい)。ライゲーションのために、450ピコモルの、43塩基のオリゴヌクレオチドを、10mMのトリス、pH 8と、50mMの塩化ナトリウムとからなるアニーリング緩衝液120μLの容量中で、300ピコモルの、20塩基のオリゴヌクレオチドと混合した。混合物を、95℃で、2分間にわたり加熱し、次いで、1秒ごとに、温度を0.1℃ずつ下げることにより、+4℃へと冷却した。冷却した後、混合物を、室温へと暖め、このアニーリング反応物96μLを、480μLの最終容量中で、10mMのATPを含有する、10倍濃度のT4 DNA Ligase緩衝液48μL、および24μlのT4 DNA Ligase(400単位/μl)と混合した。ライゲーション反応物を、23℃で、20時間にわたりインキュベートし、次いで、65℃で、20分間にわたりインキュベートして、リガーゼを熱殺菌した。
AllCells(USA、型番PB009-IF)製のヒトPan T Cellsの凍結アリコートを、全ての活性化/増加研究に使用した。Pan T Cellsを、実施例2で記載した通りに、融解させ、加工し、以下の完全X-Vivo培地(Lonza、Basel、Switzerland)へと添加して、表2に示される通り、1mL当たりの細胞1×106個の初期濃度をもたらした。
実施例4で例示した、6ウェルプレートによる研究に加えて、72C VueLife(登録商標)バッグ(CellGenix GmbH、Breisgau、Germany)を使用して、T細胞の活性化および増加の効率についての、大スケールの比較を実施した。実施例4で記載した、同じ比および構成要素を、直線的にスケーリングして、32mLの完全X-Vivo培地中の細胞32×106個(1mL当たりの細胞1×106個)の出発条件に適合させた。培養物は、標準的な細胞培養インキュベーター内の72C VueLifeバッグ(CellGenix GmbH、Breisgau、Germany)内に維持した。4日目に、細胞を回収し、新鮮な培地で洗浄し、カウントし、適切な培地容量中で希釈して、1mL当たりの細胞0.5×106個をもたらし、VueLife 72Cバッグ内に再播種した。6日目に、さらなる増加のために、細胞を希釈し、揺動WAVE bioreactor(商標)プラットフォーム(GE Healthcare、Bio-Sciences)上の、Waveバッグ内の250mL中に播種した。8日目までに、Nucleocounter(登録商標)(ChemoMetec、Allerod、Denmark)を使用して、細胞をカウントし、フローサイトメトリーにより、多様なCD表面マーカーの発現について検討した。下記の表3~5は、Dynabeads(登録商標)試料およびDBTAで活性化させた試料の両方について、同等レベルの、CD25発現、x倍の増加、細胞サイズ、および生存率を描示する。加えて、8日目のフロー解析は、Dynabeads(登録商標)およびDBTAのいずれについても、CDマーカーの全パネルについて、ほぼ同一レベルの表現型の発現を示す。調製物および結果を、下記の表3~5に例示する。
一連のT細胞活性化実験を行って、DBTA系の構成要素(例えば、抗CD3-DNA(抗CD3-o20b(+)act)、抗CD28-DNA(抗CD28-o20b(+)act)、およびRCAact)の全てをプレインキュベートするか、またはあらかじめ会合させ、次いで、あらかじめ会合させた複合体を、T細胞へと添加することの効果を評価した。結果を、in situにおける会合のために、各個別の構成要素を、T細胞へと、個別に添加する、標準的なプロトコールと比較した。これらのあらかじめ会合させた試料について、実施例4で使用した、同じ数量および比のDBTA構成要素を、1.5mLの試験管に、併せて添加し、完全に混合し、室温で、30分間にわたりインキュベートした。次いで、あらかじめ会合させたDBTA混合物を、6ウェル-プレート内の、新鮮なT細胞培養物へと添加した。並行して、かつ、実施例4で行った通り、あらかじめ会合させなかった、標準的なDBTA試料を、T細胞培養物へと添加した。結果を、Dynabeads(登録商標)および細胞だけの対照試料(それぞれ、陽性対照および陰性対照)と比較した。
一連のT細胞活性化実験を行って、抗CD3-DNA:抗CD28-DNAの異なる比の、T細胞の活性化および増加に対する効果を評価した。全ての場合に、RCAact濃度を、67nMに維持する一方で、抗CD3-DNAコンジュゲート:抗CD28-DNAコンジュゲートのモル比1:1を、1:2と対比して検討した。実施例4で概括した、他の全てのプロトコール条件を維持した。
抗CD3-DNA抗体コンジュゲートおよび抗CD28-DNA抗体コンジュゲートを、T細胞培養物へと添加して、30分後における、ローリングサークル法による増幅産物(RCAact)の、別個の第2の添加の前に、Ab-DNA/細胞プレインキュベート試料を作出することの効果を評価する実験を行った。この研究のために使用した、抗CD3-DNA:抗CD28-DNA比は、1:1であった。結果を、全てのDBTA構成要素を、in situにおける会合のために、個別かつ同時に添加する手順と比較した。この実験では、実施例4で概括した、他の全てのプロトコールの詳細および数量に従った。in situにおける会合プロトコールでは、使用した抗CD3-DNA:抗CD28-DNA比は、1:1および1:2であった。
一連のT細胞活性化実験を行って、異なるAb-DNA:RCAactインプット比の、T細胞活性化効率に対する効果を評価した。全ての場合に、抗CD3-DNA(抗CD3-o20b(+)act)コンジュゲートだけを使用し、図8Aおよび図8Bでは、Ab-DNAと表した。各Ab-DNAコンジュゲート試料が、活性化のために、ウェル1つ当たり同じ標準濃度(1μg/mLのインプットまたは6.7nM)を特徴とする一方で、3log範囲(6.7nM、67nM、および670nM)のインプットRCAact産物について探索した。CpGを含有するRCA鋳型およびCpGを含有しないRCA鋳型から作製される、ローリングサークル法による増幅産物(実施例1および表1を参照されたい)について探索した。CpGを含有しないRCA鋳型から作製される、ローリングサークル法による増幅産物を使用して、以下の実験条件:i)比を1:10とするAb-DNA:RCAact(CpGなし、1:10のAb-DNA:RCAにより表される);ii)比を1:1とするAb-DNA:RCAact(CpGなし、1:1のAb-DNA:RCAにより表される);およびiii)比を1:100とするAb-DNA:RCAact(CpGなし、1:100のAb-DNA:RCAにより表される)を使用した。同様に、CpGを含有するRCA鋳型から作製される、ローリングサークル法による増幅産物については、以下の実験条件:i)比を1:10とするAb-DNA:RCAact(CpG、1:10のAb-DNA:RCAにより表される);ii)比を1:1とするAb-DNA:RCAact(CpG、1:1のAb-DNA:RCAにより表される);およびiii)比を1:100とするAb-DNA:RCAact(CpG、1:100のAb-DNA:RCAにより表される)を使用した。いずれの実験セットについても、最適のT細胞活性化(CD25の発現により測定される)は、Ab-DNA:RCAact比を1:10とするときに達成された。活性化効率の低下は、Ab-DNA:RCAの比を増大させたときにも、これを低下させたときにも観察された。しかし、全ての試料は、7日目の実験において、10倍またはこれを超える細胞の増加を裏付けた。実施例4で概括した、他の全てのプロトコールの詳細および数量を維持した。図8Aは、1および4日間にわたる培養の後における、CD25の発現%を示し、図8Bは、4および7日後における、細胞の増加を示す。
本開示についての、前出の全ての例(実施例1~9)および付随する図(図1~8)は、ヒトB-アクチンに由来する配列を有する、ローリングサークル法による増幅産物を伴うDBTA系(実施例1)を用いる。ヒトT細胞の活性化および増加の成功はまた、多様なヌクレオチドの配列および長さを有する捕捉オリゴヌクレオチドを含む、代替的な捕捉核酸ポリマーによっても達成することができる。1つの代替的な例は、メチシリン耐性Staphylococcus aureus(MRSA)ゲノムに由来する、ローリングサークル法による増幅産物を特徴とする。対応する、相補的な第1の結合性DNA配列および第2の結合性DNA配列を、o25b(+)mrsa(配列番号7)と称し、対応するDBTA産物を、DBTA[抗CD3-o25b(+)mrsa+非修飾抗CD28]またはDBTA[抗CD3-o25b(+)mrsa+抗CD28-o25b(+)mrsa]と称する。これらのDBTA構成要素の各々についての、具体的な配列情報を、下記の表6に示す。
MRSAベースのDBTA系(全長RCA産物を作出するのに使用される一般的な条件については、実施例10および実施例3を参照されたい)を使用して、異なるサイズのローリングサークル法による増幅(RCA)産物を使用するヒトT細胞の活性化および増加の効率の比較を行った。これらの実験のために、全長RCA産物を、超音波処理条件下に置き、異なる相対サイズ分布をもたらした。超音波処理されたRCA産物をもたらすために、製造元の推奨インプット条件に従い、Covaris M220 Focused ultrasonicator(商標)(Covaris、Woburn、MA)を用いて、大型のゲノムDNA断片から出発して、1500塩基(b)および150塩基(b)のサイズの断片をもたらした。標的インプットパラメータは、入射ピークパワー、占有率、バースト1つ当たりのサイクル、処理時間、および温度を含む。超音波処理の後で、実施例1の条件に従い、解析的サイズ除外クロマトグラフィーを使用して、産物の相対サイズ分布を確認した。RCA産物について、この方法による絶対サイズおよび分子量の決定は知られていないが、小型断片について予測される、溶出時間増大の一般的な傾向が観察された。こうして、以下の溶出時間:約16Kb(理論的な最大長)の全長RCA産物に対する10.1分間(空隙容量に対応する)、1.5Kbの超音波処理RCA産物に対する10.8分間、0.15Kbの超音波処理RCA産物に対する14.8分間、43塩基の出発RCA鋳型(対照注入)に対する19.2分間、非コンジュゲートo25b(+)mrsa(対照注入)に対する20.0分間、およびヌクレオチドを含む残留低分子に対する23.7分間が観察された。1.5Kbの超音波処理産物はまた、ゲル電気泳動によってもサイズ決定したところ、予測される通り、超音波処理は、単一の長さの産物をもたらさないので、サイズが400~1600塩基の範囲(1.5Kbの標的範囲に近い)の断片を含有することが見いだされた。
抗CD3抗体を付加した第1の結合性DNA配列(抗CD3-o20b(+)act)および抗CD28抗体を付加した第2の結合性DNA配列(抗CD28-o20b(+)act)を合成するための実験条件は、DNA(D):抗体(P)比の変化をもたらすように、抗体-DNAコンジュゲートを調製したことを除き、実施例1で記載した通りであった。これは、抗CD3抗体を付加した第1の結合性DNAおよび抗CD28抗体を付加した第2の結合性DNA配列の、ローリングサークル法による増幅産物とのハイブリダイゼーションのレベルを制御する意図で行った。図11は、2つの異なるD/P比における、DBTA[抗CD3-o20b(+)act+抗CD28-o20b(+)act]の性能を示す。CD25の発現により測定されるT細胞の活性化は、D/P比を2.8~3.0(DBTA[抗CD3-o20b(+)act+抗CD28-o20b(+)act](D/P:2.8~3.0)により表される)としたとき、D/P比を1.5~1.8(DBTA[抗CD3-o20b(+)act+抗CD28-o20b(+)act](D/P:1.5~1.8)により表される)としたときと比較して高度であった。D/P比を大きくしたとき、観察されるT細胞の活性化は高度であった。図11のデータは、8つの個別の実験から取った平均を表す。
抗CD3-o20b(+)actおよび抗CD28-o20b(+)actを合成するための実験条件については、実施例1において記載した。市販のJet PEI(登録商標)(VWR International、Pennsylvania、USA、型番89129-914)およびポリL-リシン(PLL)(Sigma-Aldrich、Missouri、USA、型番P9155)を、この実験のための第2の薬剤として使用した。本研究で使用したN/P比(カチオン性捕捉ポリマーのアミン基のモルの、結合性DNA配列のリン酸基のモルに対する比)は、jet PEI(登録商標)の3のN/P、およびPLLの1のN/Pであった。抗CD3-o20b(+)actコンジュゲートおよび抗CD28-o20b(+)actコンジュゲートを、1μg/mLの濃度で使用した。実施例1で概括した、他の全てのプロトコールの詳細および数量に従った。T細胞の活性化については、実施例4で記載したプロトコールに従った。図12Aおよび図12Bは、抗CD3-o20b(+)act、抗CD28-o20b(+)actおよびカチオン性捕捉ポリマー(PEIまたはPLL)を使用する、T細胞の活性化および増殖を示す。
AllCells製のヒトPan T Cells(型番PB009-IF)の凍結アリコートを、全ての活性化研究および増加研究に使用した。Pan T Cellsを、実施例2で記載した通りに融解させ、加工し、以下の完全X-Vivo培地へと添加して、1mL当たりの細胞1×106個の初期濃度をもたらした。
第1の結合性DNA配列および/または第2の結合性DNA配列を、二次抗体に付加することにより、二次Ab-DNAコンジュゲートを作出する:
異なるマウスIgG亜型をターゲティングする二次抗体(ヤギ抗マウスIgG(H+L)、型番115-005-062、ヤギ抗マウスIgG1、型番115-005-205、ヤギ抗マウスIgG(1+2a+2b+3)、型番115-005-164、およびヤギ抗マウスIgG2a、型番115-005-206)は、Jackson Immunoresearchから得、抗CD3コンジュゲートおよび抗CD28コンジュゲートについて、上記の実施例1で記載した通り、結合性DNA配列である、o20b(+)actへとコンジュゲートさせた。
マウスIgG1亜型をターゲティングする二次抗体(ヤギ抗マウスIgG1、型番115-005-205)は、Jackson Immunoresearchから得、抗CD3抗体および抗CD28抗体の、結合性DNA配列への付加について、上記の実施例1で記載した通り、結合性DNA配列である、o20b(+)actに付加した。
Claims (15)
- T細胞およびNK細胞から選択される免疫細胞を活性化させる方法であって、
a)T細胞およびNK細胞から選択される、用意された免疫細胞の集団を、第1の薬剤および第2の薬剤と接触させるステップ
を含み、
第1の薬剤が、第1の結合性核酸配列に付加した免疫細胞活性化剤を含み、
第2の薬剤が、少なくとも1つの捕捉オリゴマーを含み、
少なくとも1つの捕捉オリゴマーが、第1の結合性核酸配列と会合することが可能であり、
少なくとも1つの捕捉オリゴマーが、捕捉オリゴヌクレオチドであり、および、少なくとも1つの捕捉オリゴヌクレオチドが、第1の結合性核酸配列と相補的である、方法。 - 免疫細胞活性化剤を、第1の結合性核酸配列に、共有結合を介して付加する、請求項1記載の方法。
- 第2の薬剤が、ローリングサークル法による増幅産物である、請求項1または2のいずれか一項記載の方法。
- 免疫細胞が、T細胞であり、免疫細胞活性化剤が、T細胞活性化剤である、請求項1乃至3のいずれか一項記載の方法。
- T細胞活性化剤が、抗CD3抗体である、請求項4記載の方法。
- T細胞の集団を、T細胞共刺激剤と接触させるステップをさらに含む、請求項4または5記載の方法。
- T細胞共刺激剤が、抗CD28抗体、抗4-1BB抗体、またはこれらの組合せである、請求項6に記載の方法。
- 免疫細胞活性化剤を、第1の結合性核酸配列に、中間結合性部分を介して、非共有結合的に付加する、請求項1記載の方法。
- 中間結合性部分が、二次抗体、ビオチン、アビジン、およびこれらの組合せからなる群から選択される、請求項8記載の方法。
- 中間結合性部分を、第1の結合性核酸配列に、共有結合を介して付加する、請求項8記載の方法。
- 中間結合性部分が、非共有結合的相互作用を介して、免疫細胞活性化剤に付加することが可能である、請求項10記載の方法。
- 第1の薬剤を、T細胞およびNK細胞から選択される免疫細胞の集団を含有する培地中、in situで形成する、請求項1乃至11のいずれか一項記載の方法。
- 第2の薬剤が、少なくとも1つの捕捉オリゴヌクレオチドを含む捕捉核酸ポリマーであり、少なくとも1つの捕捉オリゴヌクレオチドが、第1の結合性核酸配列と相補的である、請求項1乃至12のいずれか一項記載の方法。
- 免疫細胞が、ナチュラルキラー細胞であり、免疫細胞活性化剤が、ナチュラルキラー細胞活性化剤である、請求項1乃至3、8乃至13のいずれか一項記載の方法。
- ナチュラルキラー細胞活性化剤が、抗CD335抗体、抗CD244抗体、抗CD2抗体、またはこれらの組合せである、請求項14に記載の方法。
