Description
(i)複数の第四級アンモニウム基を含む陰イオン交換不織布基材を含む、第1の濾過媒体と、
(ii)複数のグアニジル基を含む官能化微多孔質膜(functionalized microporous membrane:官能化した微多孔質膜)を含む、第2の濾過媒体と、を含み、
第1の濾過媒体が第2の濾過媒体の上流に配置されている、濾過媒体配列が記載される。
(a)標的生体材料及び夾雑物を含む生体流体を準備することと、
(b)生体流体を、
(i)複数の第四級アンモニウム基を含む陰イオン交換不織布基材を含む、第1の濾過媒体と、
(ii)複数のグアニジル基を含む官能化微多孔質膜を含む、第2の濾過媒体と、を含み、第1の濾過媒体が第2の濾過媒体の上流に配置されている濾過媒体配列と接触させることと、を含む、生体流体の濾過方法が記載される。
「アルキル」は、1〜約12個の炭素原子を有する、直鎖又は分枝鎖の、環状又は非環状の、飽和一価炭化水素、例えば、メチル、エチル、1−プロピル、2−プロピル、ペンチル等を意味する。
「アルキレン」は、1〜約12個の炭素原子を有する直鎖飽和二価の炭化水素、又は3〜約12個の炭素原子を有する分枝鎖飽和二価の炭化水素、例えば、メチレン、エチレン、プロピレン、2−メチルプロピレン、ペンチレン、ヘキシレン等を意味する。
「アルケニル」は、2〜約12個の炭素原子を有する直鎖不飽和一価炭化水素、又は3個〜約12個の炭素原子を有する分枝鎖不飽和炭化水素を意味する。
「アルケノイル」は、カルボニル(−C(=O)−)基を含むアルケニル基を意味する。
「アリール」は、一価の芳香族、例えば、フェニル、ナフチル等を意味する。
「グアニジル」は、グアニジン及びビグアナイドのうちの少なくとも1つから選択される官能基を意味する。
「ヘテロアリーレン」は、芳香族及び複素環である二価の基を指す。すなわち、ヘテロアリーレンは、5又は6員を有する芳香環中に少なくとも1つのヘテロ原子を含む。好適なヘテロ原子は、典型的には、オキシ、チオ、又はアミノである。この基は、連結している、縮合している、又はこれらの組み合わせである1〜5個の環を有し得る。少なくとも1つの環は複素芳香環であり、かつその他の基は、芳香環、非芳香環、複素環、炭素環、又はこれらの組み合わせであり得る。いくつかの実施形態において、ヘテロアリーレンは最大5個、最大4個、最大3個、最大2個、又は1個の環を有する。ヘテロアリーレン基の例としては、これらに限定されるものではないが、トリアジン−ジイル、ピリジン−ジイル、ピリミジン−ジイル、ピリダジン−ジイル等が挙げられる。
「ヒドロカルビル」は、アリール及びアルキルを含む。
「(ヘテロ)ヒドロカルビル」は、ヒドロカルビルアルキル及びアリール基、並びにヘテロヒドロカルビルヘテロアルキル及びヘテロアリール基を含み、後者は、1つ以上のカテナリー(鎖中)ヘテロ原子、例えば、酸素原子又は窒素原子等を含む。ヘテロヒドロカルビルは、任意に、エステル官能基、アミド官能基、尿素官能基、ウレタン官能基、及びカーボネート官能基等の、1つ以上のカテナリー(鎖中)官能基を含有し得る。別途指示がない限り、非ポリマー(ヘテロ)ヒドロカルビル基は、典型的には、1〜60個の炭素原子を含有する。本明細書で用いられるようなヘテロヒドロカルビルのいくつかの例としては、これらに限定されるものではないが、上記の「アルキル」、「ヘテロアルキル」、「アリール」、及び「ヘテロアリール」について記載されたものに加え、メトキシ、エトキシ、プロポキシ、4−ジフェニルアミノブチル、2−(2’−フェノキシエトキシ)エチル、3,6−ジオキサヘプチル、3,6−ジオキサヘキシル−6−フェニルが挙げられる。
「a」、「an」、及び「the」は互換的に用いられ、1つ以上を意味する。
「及び/又は」は、記載された事例の一方又は両方が生じ得ることを示すのに用いられ、例えば、A及び/又はBは、(A及びB)並びに(A又はB)を含む。
α=mf÷ρf×L不織布
式中、mfは試料表面積当たりの繊維質量であり、ρfは繊維密度であり、
L不織布
は不織布の厚さである。ソリディティは、本明細書では不織布基材自体について用い、官能化不織布は対象とならない。不織布基材が2種類以上の繊維の混合物を含む場合、同一のL不織布を用いて繊維のそれぞれの種類について個々のソリディティを求め、これらの個々のソリディティを合計してウェブのソリディティαを得る。
Z−Q−(CH(R5)−CH2−O)m−R6[式中、Zは重合性エチレン性不飽和部分であり、R5はH又はC1〜C4アルキル基であり、QはN(R5)又はOであり、R6はH、C1〜C4アルキル基、アリール基、又はこれらの組み合わせであり、mは2〜100、好ましくは5〜20である]のものである。モノマーの有用なエチレン性不飽和部分Zとしては、以下を挙げることができる。
アミドモノマーは、一般式:
R3は−H又はC1〜C4アルキルであり、X1は−NR3−又は−O−であり、R1はエポキシ官能性又はエーテル官能性の(ヘテロ)ヒドロカルビル基である]のものが挙げられる。より詳細には、エーテル官能基は、低級アルキレンオキシアルキル基である。好ましくは、R1基は、オキシラン(エポキシ)基を有する2〜30個の炭素原子の直鎖、分枝鎖、環状又は多環式炭化水素に基づいている。より好ましくは、R8基は、グリシジルメタクリレート(GMA)のように、3〜10個の炭素を含む。このようなグラフトに関する更なる情報は、米国特許出願公開第2015/0099413号(Berrigan et al.)に見出すことができる。
[式中、R1はH又はC1〜C4アルキルであり、R2は(ヘテロ)ヒドロカルビル基であり、任意に、エステル、アミド、ウレタン又は尿素、好ましくは1〜20個の炭素原子を有する二価のアルキレンを含有し、各R3は独立してH又はヒドロカルビル、好ましくはC1〜C12アルキルであり、R4はH、C1〜C12アルキル又は−N(R3)2であり、R5はH又はヒドロカルビル、好ましくはC1〜C12アルキル又はアリールであり、X1は−O−又は−NR3−であり、oは0又は1であり、nは1又は2である]のリガンド官能性モノマー単位から誘導されるグアニジル基によって処理される。
[式中、R1はH又はC1〜C4アルキルであり、R2は(ヘテロ)ヒドロカルビル基であり、任意に、エステル、アミド、ウレタン又は尿素、好ましくは、1〜20個の炭素原子を有する二価のアルキレンを含有し、各R3は独立してH又はヒドロカルビル、好ましくはC1〜C12アルキルであり、R4はH、C1〜C12アルキル又は−N(R3)2であり、X1は−O−又は−NR3−である]の化合物との縮合により調製することができる。他のリガンドモノマーは、カルボニル含有モノマー、例えば、アクロレイン、ビニルメチルケトン、ジアセトンアクリルアミド又はアセトアセトキシエチルメタクリレート等の、任意に還元剤の存在下で、式Va又はVbの化合物との縮合により調製することができる。
(i)複数の第四級アンモニウム基を含む陰イオン交換不織布基材を含む、第1の濾過媒体と、
(ii)複数のグアニジル基を含む官能化微多孔質膜を含む第2の濾過媒体と、を含み、
第1の濾過媒体が第2の濾過媒体の上流に配置されている、濾過媒体配列。
(a)80〜98重量%のアミノアルキル(メタ)アクリロイルモノマーと、
(b)2〜20重量%のポリ(アルキレンオキシド)モノマーと、
(c)0〜10重量%の第2の親水性モノマーと、が共重合したモノマー単位を含む、実施形態1〜5のいずれか1つに記載の濾過媒体配列。
(a)10〜50重量%の第四級アンモニウム含有リガンドモノマーと、
(b)10〜80重量%のアミドモノマーと、
(c)10〜40重量%のオキシモノマーと、
(d)0〜30重量%のポリ(アルキレンオキシド)モノマーと、が共重合したモノマー単位を含む、実施形態1〜6のいずれか1つに記載の濾過媒体配列。
R1はH又はC1〜C4アルキルであり、
R2は(ヘテロ)ヒドロカルビル基であり、
各R3は独立してH又はヒドロカルビルであり、
R4はH、C1〜C12アルキル又は−N(R3)2であり、
R5はH又はヒドロカルビルであり、
X1は−O−又は−NR3−であり、
oは0又は1であり、かつ
nは1又は2である]のうちの少なくとも1つを含む、実施形態1〜9のいずれか1つに記載の濾過媒体配列。
プライマー層が、エチレン性不飽和重合性基を有する架橋ポリアミンポリマーを含み、エチレン性不飽和重合性基を有する架橋ポリアミンポリマーが、
(a)ポリアミンポリマーと、
(b)ポリアミンポリマーのための多官能性架橋剤と、
(c)アミン反応性官能基及びエチレン性不飽和重合性基を有するモノマーと、の反応生成物である、実施形態10に記載の濾過媒体配列。
流体入口と、
流体出口と、
流体入口及び流体出口を流体連通する濾過媒体と、を含み、
濾過媒体が、
(i)複数の第四級アンモニウム基を含む陰イオン交換不織布基材を含む、第1の濾過媒体と、
(ii)複数のグアニジル基を含む官能化微多孔質膜を含む、第2の濾過媒体と、を含み、かつ
第1の濾過媒体が第2の濾過媒体、複数の濾過媒体の上流に配置されている、フィルタ装置。
分離要素と、
縁部シールと、を更に含み、
分離要素が、
流体入口と流体連通している中心コアと、
第1の側面と、
第2の側面と、を含み、
濾材が、
分離要素の第1の側面上に配置され、外周縁部及び内周縁部を有する第1の媒体ディスクと、
分離要素の第2の側面上に配置され、外周縁部及び内周縁部を有する第2の媒体ディスクと、を更に含み、
第1及び第2の媒体ディスクの外周縁部が縁部シールにより連通されており、第1及び第2の媒体ディスクの内周縁部が中心コアに連通されており、第1及び第2の媒体ディスクの上流側が第1の濾過媒体を含む、実施形態19に記載のフィルタ装置。
(a)標的生体分子及び夾雑物を含む生体流体を準備することと、
(b)生体流体を、実施形態1〜22のいずれか1つに記載の濾過媒体配列に接触させて濾過流体を得ることと、を含む、方法。
[実施例]
供試流体のpH値及び伝導度値を、Beckman Coulter pHI 570 pH及び電気化学計(Beckman Coulter,Inc.(Brea,California)より入手可能)によって特性分析した。
Claims (10)
- (iii)複数の第四級アンモニウム基を含む陰イオン交換不織布基材を含む、第1の濾過媒体と、
(iv)複数のグアニジル基を含む官能化微多孔質膜を含む、第2の濾過媒体と、を含み、
前記第1の濾過媒体が前記第2の濾過媒体の上流に配置されている、濾過媒体配列。 - 前記第1の濾過媒体が、前記第1の濾過媒体1グラム当たり少なくとも0.1mmolの第四級アンモニウム基を含む、請求項1に記載の濾過媒体配列。
- 前記第2の濾過媒体が、前記第2の濾過媒体1グラム当たり少なくとも0.01mmolのグアニジル基を含む、請求項1又は2に記載の濾過媒体配列。
- 前記第1の濾過媒体が、不織布基材の表面にグラフトされたポリマーを含み、前記ポリマーが、
(a)80〜98重量%のアミノアルキル(メタ)アクリロイルモノマーと、
(b)2〜20重量%のポリ(アルキレンオキシド)モノマーと、
(c)0〜10重量%のアミノアルキル(メタ)アクリロイルモノマー及びポリ(アルキレンオキシド)モノマーではない親水性モノマーと、が共重合したモノマー単位を含む、請求項1〜3のいずれか一項に記載の濾過媒体配列。 - 前記第1の濾過媒体が、不織布基材の表面にグラフトされたポリマーを含み、前記ポリマーが、
(a)10〜50重量%の第四級アンモニウム含有リガンドモノマーと、
(b)10〜80重量%のアミドモノマーと、
(c)10〜40重量%のオキシモノマーと、
(d)0〜30重量%のポリ(アルキレンオキシド)モノマーと、が共重合したモノマー単位を含む、請求項1〜4のいずれか一項に記載の濾過媒体配列。 - 前記フリーラジカルグラフト化グアニジル官能性(メタ)アクリロイルモノマーが、微多孔質膜上に配置されたプライマー層にグラフトされ、
前記プライマー層が、エチレン性不飽和重合性基を有する架橋ポリアミンポリマーを含み、
エチレン性不飽和重合性基を有する前記架橋ポリアミンポリマーが、
(a)ポリアミンポリマーと、
(b)前記ポリアミンポリマーのための多官能性架橋剤と、
(c)アミン反応性官能基及びエチレン性不飽和重合性基を有するモノマーと、の反応生成物である、請求項6に記載の濾過媒体配列。 - 前記第1の濾過媒体と前記第2の濾過媒体との間に配置された非官能化濾過媒体を更に含む、請求項1〜7のいずれか一項に記載の濾過媒体配列。
- 流体入口と、
流体出口と、
前記流体入口及び前記流体出口を流体連通する濾過媒体と、を含み、
前記濾過媒体が、
(i)複数の第四級アンモニウム基を含む陰イオン交換不織布基材を含む、第1の濾過媒体と、
(ii)複数のグアニジル基を含む官能化微多孔質膜を含む、第2の濾過媒体と、を含み、かつ
前記第1の濾過媒体が前記第2の濾過媒体の上流に配置されている、フィルタ装置。 - 生体流体の濾過方法であって、前記方法が、
(a)標的生体分子及び夾雑物を含む前記生体流体を準備することと、
(b)前記生体流体を、請求項1〜8のいずれか一項に記載の濾過媒体配列に接触させて濾過流体を得ることと、を含む、方法。
