Description
本出願は、2016年3月31日に出願された、米国仮特許出願第62/315,774号に対する優先権を主張するものであり、その開示は、参照によりその全体が本明細書に組み込まれる。
本発明は、癌の治療のための併用療法に関し、特に、ニトロキソリン、または、その類似体またはその薬学的に許容される塩の、少なくとも一つの追加の抗癌化学療法剤または免疫療法剤とその併用療法に関し、併用療法は、相乗的な抗癌効果を示す。
癌の治療のための併用療法は、いずれかの抗癌治療に対する癌発症の機会の可能性を最小限に抑え、そして、より低い用量の個々の抗癌治療の使用を可能にし、毒性を低下させる。薬物−薬物相互作用の性質に応じて、2つの活性成分の併用は、相乗効果、相加効果または拮抗効果をもたらすことができる。
ニトロキソリンは、血管新生の阻害(4)および癌の増殖および浸潤の阻害(5,6)においてもまた、活性であることが最近発見された。
オキシキノリン、クリオキノールおよびヨードキノールのような、NITおよびその類似体は、感染症の治療において共通の臨床用途を共有する。
NIT類似体はまた、抗癌細胞傷害性を有することも報告されている(7)。
しかしながら、本発明者の知識によれば、これらの薬物は、いずれも、他の抗癌剤と組み合わせて調査されておらず、そして、NITまたはその類似体は、他の癌療法剤のいずれかとの薬物−薬物相互作用は報告されていない。
本発明は、ニトロキソリン、その類似体またはその薬学的に許容される塩を、少なくとも1つのさらなる抗癌化学療法剤または免疫療法剤を含む、新規な併用療法を提供することにより、これらのニーズを満たす。
本併用療法は、癌細胞増殖の阻害に対して相乗効果を示す。

細菌の増殖を阻害するその作用機序は未知のままである。
NITが、尿路病原性大腸菌が、NITのサブMICで、尿路上皮細胞および尿カテーテルに付着することを阻害することが見出され、そして、NITが、二価イオンのマグネシウム2+およびマンガン2+をキレート化することによって、細菌の外膜の崩壊を促進すると考えられている(1)。
オキシキノリン(オキシン)は、軽度の静真菌性、静菌性、駆虫性、およびアメーバ活性を有する消毒剤である。オキシンは、マンガン2+およびマグネシウム2+の酵素を本質的に奪うことによって、酵素に直接接触することなく、単離された大腸菌RNAポリメラーゼを阻害することができる(2)。
オキシンはまた、金属キレート剤としてもまた、診断目的のためのラジオ−インジウム用の担体としても使用される。
クリオキノールは、長期にわたり、局所的抗感染性、腸の抗アメーバ性および膣トリコモナシドとして使用されてきた。クリオキノールの経口製剤は、亜急性骨髄−視神経症を引き起こすことが示されており、従って、世界中で禁止されている。クリオキノールは、おそらく、Cu2+とZn2+のキレーターとして作用するその能力に起因して、動物モデルにおけるアルツハイマー病の認知低下を停止させることが判明した(3)。
ヨードキノールは、腸の消毒剤として、特に、抗アメーバ剤として、広く使用されている、別のハロゲン化8−キノリノールである。それはまた、他の感染のために局所的に使用され、そして、CNSおよび眼の損傷を引き起こす可能性がある。その作用メカニズムは未知のままである。
NITはまた、前立腺癌細胞のアポトーシスを誘導することができることも示され(5)、そして、癌細胞に対するその細胞傷害性は、Cu2+と組み合わせて投与された場合により顕著であった(6)。
クリオキノール(Clioquinol)、オキシン(oxine)およびイオドキノール(iodoquinol)などのNIT類似体もまた、抗癌細胞毒性を有することが報告されている(7)。
がんは、他の特徴の中でも、浸潤および転移などの悪性の行動と結びついている、細胞の制御不能な増殖であり、それは、遺伝的感受性と環境因子との間の相互作用によって引き起こされる。
これらの要因は、細胞の増殖速度を制御する遺伝子である、癌遺伝子、および、癌を予防する助けとなる遺伝子である、癌抑制遺伝子に、遺伝的変異の蓄積をもたらし、癌細胞に制御不能な増殖のような悪性の特徴をもたらす。化学療法剤は、化学物質、特に1つ以上の抗癌剤を使用する、癌治療の1つのカテゴリーである。広い意味で、ほとんどの化学療法剤は、有糸分裂(細胞分裂)を損なうことによって作用し、速く分裂する細胞を効果的に標的とする。
これらの薬物は、細胞を損傷させるため、細胞傷害性と呼ばれている。抗癌剤は、DNAの損傷および細胞分裂に関与する細胞機構の阻害を含む、様々な機作によって、有糸分裂を防止する。これらの薬物が癌細胞を殺す理由の一つの理論は、それらがアポトーシスとして知られている、プログラムされた形態の細胞死を誘導することである。

併用療法は、多数の異なる薬物で同時に患者を治療することを含む。
薬剤はそのメカニズムおよび副作用が異なるので、併用療法は、いずれか1つの薬剤に耐性が生じる機会を最小限に抑え、したがって、耐性癌または毒性を低下させるためにより低用量で治療するために使用することができる。しかしながら、薬物−薬物相互作用の予測不可能な性質のため、既知の治療法の組み合わせはまた、有効性の低下または副作用の増加をもたらす可能性がある。
癌免疫療法は、腫瘍を拒絶して、そして、破壊する免疫系を刺激する。
癌は、腫瘍の微小環境を操作し、そして、免疫抑制を推進することにより、
ホストの免疫を逃れることができ、そのため、有効な抗腫瘍免疫応答を取り込むために必要な成分は、癌患者に存在するが、ホストは通常、腫瘍進行を阻止することができないことを発見した(8)。
この直感でわかるものではない(counterintuitive)シナリオを説明するために、過去に、腫瘍の「免疫脱出」に主に焦点を当て、そして、腫瘍が、ホストによって生成された腫瘍特異的免疫応答を回避する能力を発達させるか、または、一緒になってホストの抗癌免疫作用を働かせないことを示唆する、多数の分子および細胞のメカニズムが提案されている。このプロセスは、「腫瘍誘発性免疫抑制」と呼ばれ、近年認識されており、そして、鋭意研究中である。腫瘍は、免疫系の全ての成分を妨害し、抗腫瘍免疫応答の全ての段階に影響を及ぼすことができるようである。
免疫系が、免疫介入に敏感であるそれらの悪性細胞を排除し、そして、免疫耐性変異体の選択と生存を可能にすることによる、「免疫編集(immune editing)」を経由して、これらの抗原性の変化を駆動するかもしれないことが示唆されている。
免疫編集(immune editing)の最終結果は、腫瘍が宿主免疫系から逃れることである。制御性T細胞(Treg)および骨髄球由来サプレッサー細胞(MDSC)は、免疫逃避(immune escape)を実施するために腫瘍によって使用される2つのタイプの細胞である。Treg、MDSCまたは阻害因子の有害作用からの免疫細胞の保護、そして、したがって、そのエフェクター機能の増強は、癌患者の有効な抗腫瘍免疫を回復させることができる。
腫瘍関連抗原(TAA)が自己抗原であるので、Treg細胞の蓄積は、免疫寛容の実施に対する応答である可能性がある。
Treg細胞は、
IL−10およびTGF−β1産生、
免疫抑制性アデノシンへのATPの酵素分解、または
Fas/FasLおよびグランザイム/パーフォリン経路の関与を含む、
様々なメカニズムによるエフェクターT細胞の免疫活性の下方制御に寄与する。
腫瘍は、Tregによって媒介される免疫抑制効果の恩恵を受ける。
アルギナーゼ−1の産生、
L−アルギニン代謝に関与する酵素、ならびに
誘導性一酸化窒素シンターゼ(iNOS)の活性化を含む、いくつかの機序を介して、T細胞応答を抑制することによって腫瘍増殖を促進する。
それらはまた、T細胞分化に必須のアミノ酸である、トリプトファンの異化に関与するインドールアミン−2,2−ジオキシゲナーゼ(IDO)の腫瘍の生成を制御する。
BCGによる尿路上皮および膀胱腫瘍細胞の感染は、抗原提示分子の発現を増加させるBCGの内部移行をもたらす。
これは、IL−8)およびIL−17と共に、Th1サイトカイン(IL−2、腫瘍壊死因子、IL−12、およびIFN−γ)およびTh2サイトカイン(IL−4、IL−5、IL−6、およびIL−10)などのサイトカインの放出を介して免疫応答を誘導する。
この複雑な免疫カスケードは、細胞傷害性Tリンパ球、ナチュラルキラー細胞、好中球およびマクロファージによって媒介される、抗腫瘍活性を誘導する。
同所性(orthotopic)マウス膀胱腫瘍モデルにおける、BCGの別の研究では、低用量療法と比べ、高用量BCG療法に続いて、MDSCの集団は、有意に下方調整された。
同じ研究では、CD4+/Foxp3+ Tregも同じ変化を示した。
BCG処理後、CD4+/Foxp3+ Tregの集団は血液中で減少した。免疫抑制因子に対するこれらの阻害効果は、BCG療法の強い抗腫瘍治療効果を説明することができる(10)。
最も特徴づけられた免疫チェックポイント受容体は、細胞傷害性Tリンパ球関連抗原4(また、CD152としても知られているCTLA−4)、プログラムされた細胞死タンパク質1(また、CD279としても知られるPD−1)および、インドールアミン 2,3−ジオキシゲナーゼ−(IDO)であり、そして、これらの分子を標的とする薬剤は、複数の固形または血液癌の治療のために承認されているか、臨床試験において、または広範に試験されている。
CTLA−4は、CD28よりも、著しく高い親和力および親和力で、CD80およびCD86に結合し、CTLA−4が、そのリガンドに対して、CD28を打ち負かすことを可能にし、T細胞活性化の有効な阻害をもたらす(12)。
拮抗性抗体を用いることによる、CTLA−4活性の遮断は、この機作を妨害し、そして、したがって、T細胞の活性を保存する。
現在、Bristol−Myers Squibbの抗CTLA−4 モノクローナル抗体である、イピィリムマブ(ipilimumab)は、転移性メラノーマ患者について、米国食品医薬品局(FDA)の承認を受けている。
さらに、アストラゼネカの抗CTLA−4モノクローナル抗体である、トレメリムマブ(tremelimumab)は、悪性中皮腫の患者の治療のために、FDAによってオーファンドラッグ指定を受けた。
癌の微小環境における、PD−L1またはPD−L2の過剰発現は、腫瘍内の免疫応答の阻害をもたらす(14)。抗PD−1/PD−L1抗体による、PD−1とPD−L1との間の相互作用の阻害は、T細胞の不活性化を阻害し、したがって、抗腫瘍応答を増強し、腫瘍増殖を遅延させ、そして、腫瘍拒絶を促進し得る(15)。
2つの抗PD−1モノクローナル抗体である、ブリストル・マイヤーズ・スクイブのニボルマブ、およびメルクのペンブロリズマブは、転移性黒色腫および非小細胞肺癌患者について、米国のFDAの承認を受けた。最近、FDAは、転移性腎細胞癌患者の治療としてニボルマブを承認した。
2016年に、ロシュの抗PD−L1モノクローナル抗体である、アテゾリズマブは、転移性尿路上皮癌および非小細胞肺癌の治療薬としてFDAの承認を受けた。
アストラゼネカのドュルバルマブ(durvalumab)およびファイザーのアベルマブ(avelumab)のような他の抗PD−L1 モノクローナル抗体は、後期臨床試験中である。
T細胞膜タンパク質−3(T cell membrane protein−3)(TIM−3)、
LAG3、
Igおよび免疫受容体チロシンベースの阻害モチーフ(ITIM)ドメイン(TIGIT)を有するT細胞免疫受容体(T−cell immunoreceptor with Ig and immunoreceptor tyrosine−based inhibitory motif (ITIM) domains(TIGIT))、

いくつかの実施形態では、追加の癌療法は、手術、放射線療法、化学療法、毒素療法、免疫療法、凍結療法または遺伝子治療を含む。
いくつかの実施形態において、癌は、化学療法抵抗性、免疫療法抵抗性または放射線耐性の癌である。
本発明における治療方法は、任意の動物モデルまたは臨床において、有効量のニトロキソリンまたは類似体もしくは、その薬学的に許容される塩と有効量の免疫療法剤または有効量の化学療法剤との組み合わせとして適用することができる。

本発明の他の実施形態によれば、キットは、有効量のニトロキソリン、または類似体もしくはその薬学的に許容される塩、および有効量の化学療法剤を有する1つまたは複数の医薬組成物を含む。
ニトロキソリン、または類似体または、その薬学的に許容される塩の有効量、および免疫療法剤のまたは化学療法剤の有効量は、一つの医薬組成物に存在させることができる。
それらは、別個の医薬組成物中に存在することもできる。
キットはさらに、有効量のニトロキソリンまたは類似体もしくはその薬学的に許容される塩と、癌を治療するための免疫療法剤または化学療法剤の有効量との組み合わせの使用に関する説明書を含む。
(i)治療される癌またはそれに関連する症状の重篤度を低減または緩和する;
(ii)治療される癌またはそれに付随する症状の持続時間を短縮する;
(iii)治療される癌またはそれに関連する症状の進行の予防;
(iv)治療されるべき癌またはそれに関連する症状の退行を引き起こす;
(v)治療される癌またはそれに関連する症状の発症または発症の予防;
(vi)治療される癌またはそれに関連する症状の再発の予防;
(vii)処置される癌またはそれに付随する症状を有する被験体の入院の減少;
(viii)治療される癌またはそれに関連する症状を有する被験体の入院期間の短縮;
(ix)治療される癌またはそれに関連する症状を有する被験体の生存を増加;
(xi)被験体において、治療される癌またはそれに関連する症状を阻害または軽減すること;および/または
(xii)別の療法の予防的または治療的効果を増強または改善すること。
実施形態1は、ニトロキソリン、または類似体またはその薬学的に許容される塩の有効量、および免疫療法剤の有効量を、それを必要とする被験体に投与することを含む、癌を治療する方法である。ここで、好ましくは、免疫療法剤は、制御性T細胞または骨髄由来のサプレッサー細胞の阻害剤またはモジュレーターであり、ならびにニトロキソリンまたは類似体またはその薬学的に許容される塩と免疫療法剤との組み合わせは、相乗効果をもたらす。
ニトロキソリン、その類似体、またはその薬学的に許容される塩の有効量を含む、第1の医薬組成物を得る工程、
有効量の免疫療法剤または化学療法剤を含む、第2の医薬組成物を得る工程、そして、
キットの中の第1及び第2医薬組成物を組み合わせる工程。
本発明の以下の実施例は、本発明の性質をさらに説明するためのものである。以下の実施例は本発明を限定するものではなく、そして、本発明の範囲は添付の特許請求の範囲によって決定されるべきであることを理解すべきである。
NITおよび1つ以上の化学療法剤との組み合わせによる腫瘍増殖の相乗的阻害
2つのヒト癌細胞株の増殖阻害において、カルボプラチン、パクリタキセル、マイトマイシンC、エピルビシンおよびピラルビシンを含む一群の化学療法剤のグループで、NITを試験した。表2および3に示すように、NITは、ヒト膀胱癌細胞株5637およびヒト肝癌細胞株HepG2の増殖を阻害した。
表10および11に示すように、NITは、それぞれ、カルボプラチンおよびパクリタキセルと組み合わせて、ヒト膀胱癌5637の増殖を阻害し、そして、それぞれ、研究のCI値を計算し、図2および3にプロットした。
したがって、図2および4のCIプロットは、NITおよびカルボプラチンが、相乗的、または強く相乗的、に分類されるように、膀胱癌細胞株5637および肝臓癌細胞株HepG2の増殖を阻害するという結論を支持する。
図3のCIプロットは、NITとパクリタキセルとの組み合わせが、膀胱癌細胞株5637の増殖阻害について、わずかな相乗効果を示したことを示し、そして、図5は、この組み合わせによる、肝臓癌細胞株HepG2の増殖阻害は、薬剤濃度Fa(吸収率)が0.3よりも低い場合、部分的に相乗的である。
表 15および図6に示されるように、薬剤のIC50値より低い濃度でのこの部分的相乗作用はまた、膀胱癌細胞株5637の阻害についてのNITおよびマイトマイシンCの組み合わせについても見出された。
表16および17ならびに図7および8は、NITと、エピルビシンまたはピラルビシンとの組み合わせは、膀胱癌細胞株5637の増殖の阻害について、いくつかの濃度で部分的に相乗的であることを示した。
腫瘍体積は、指示された治療後、Xenogen IVIS200システムで分析した。
図9Aは、各処置群の典型的なIVIS画像を示す。各マウスの腫瘍体積は、1秒あたりの総光子の、関心領域の分析(region−of−interest analysis)によって決定された。
各群、6匹のマウスで分析した。
図9Bに示すように、15ミリグラム/キログラムを、1日2回、経口で、NITを、1ミリグラム/ミリリットルのマイトマイシンC(NIT+MMC)を、7日に1回、膀胱内投与と組み合わせて、投与した群の平均腫瘍体積は、NIT単独またはMMC単独治療群のそれとは有意に相違し(それぞれ、p<0.0001およびp=0.0001)、MMCとNITの併用治療により、腫瘍増殖阻害の著しい増強を示した。
各処理についてのマウス生存を評価するために、カプラン−マイヤー解析(Kaplan−Meier analysis)も行い、図9Cにまとめた。一貫して、NITとMMC群の生存状態は、対照群および単剤治療群とは対照的に、有意に改善した。
膀胱内投与でMMC処理された、生存マウスの腫瘍抑制率と生存割合は、それぞれ、0.56と0.57であった。
経口NITを、MMCと投与した場合、観察された腫瘍抑制率および生存マウスの割合は、有意に増加し、それぞれ、0.71と1に、有意に増加し、NITおよびMMCの組み合わせによる腫瘍阻害の相乗効果を示す、予測されるブリス相加値(Bliss Additive Values)よりも大きかった。
免疫療法剤とNITの組み合わせによる、腫瘍増殖の相乗的阻害
NITは、マウス膀胱および前立腺癌の同所性(orthotopic)異種移植(xenograft)モデルの腫瘍増殖阻害のためのBCGおよび抗PD−1抗体を含む免疫療法剤の群を用いて試験した。
各マウスの腫瘍体積は、1秒あたりの総光子について、関心のある領域解析(region−of−interest analysis)により決定した。各群において8匹のマウスを分析した。図10Bに示されるように、経口投与での30ミリグラム/キログラムのNITは、連続でも、また48時間の休薬を伴っていても、いずれも、腫瘍増殖を阻害し、膀胱内投与されたBCGによって引き起こされるものと比較して、わずかに高い、または類似の活性であった。
経口NIT(連続して、または48時間の休薬で)と共に、BCGを投与された場合、NIT単独またはBCG治療単独と比較して、阻害率は有意に増加した(全ての比較について、p<0.0001)。
カプラン・マイヤー分析もまた、各治療についてのマウスの生存を評価するために行われ、そして、図10Cに纏めた。
BCGと組み合わせて、経口NITで投与(連続的に、または48時間の休薬)された群の生存状態は、単一の薬物治療群と比較して、有意に改善された。
各示された処理群についての典型的なIVIS画像を、図11Aに示す。
各マウスの腫瘍体積は、1秒あたりの総光子の関心領域分析によって決定された。各群、5匹のマウスを分析した。
図11Bに示すように、10ミリグラム/キログラムの腹腔内抗PD−1は、腫瘍増殖を80%の割合で、強力に阻害した。他方、経口投与で、15ミリグラム/キログラムを投与したNITは、31%の阻害割合を示した。経口NITが、抗PD−1と組み合わせられた場合、単味のNITまたは抗PD−1処置と比較して、阻害割合は、有意に増強された(それぞれ、p<0.0001およびp=0.039)。カプラン−マイヤー分析もまた行い、図11Cに纏めた。
NIT+抗PD−1群、または単味の抗PD−1群の生存状態は、単味のNIT群よりも有意に高かった。
各マウスの腫瘍体積は、1秒あたりの総光子の関心領域(region−of−interest)分析によって決定された。各群について、8匹のマウスを分析した。図12Bに示すように、10ミリグラム/キログラムの腹腔内投与の抗PD−1は、堅牢な腫瘍増殖抑止を示した(76%)。他方、経口で、15ミリグラム/キログラムのNITの投与は、腫瘍抑制率52%を示した。NITが、抗PD−1と組み合わせた場合、腫瘍阻害率が、96%まで大幅に向上した(p値は、それぞれ、単味のNITおよび抗PD−1処理と比較して、0.017と0.027であった)。
図12Cは、各群から採取した腫瘍の画像を示す。
各マウスの腫瘍重量は測定され、そして、図12Dにまとめた。腫瘍重量データの分析から、NITおよび抗PD−1の組み合わせは、単味のNITおよび抗にPD−1治療(それぞれ、37%および56%)と比較して、それぞれ、0.0006と0.0111のp値を有する、腫瘍阻止の顕著な増強(74%)を齎すことが明らかになった
NITを、抗PD−1と組み合わせて投与したとき、その実際の観察された腫瘍の体積と重量の阻害率は、それぞれ、0.96と0.74であって、予想されたブリス独立基準(Bliss Independence model)値(それぞれ、0.89および0.72)より大きく、腫瘍の阻害において、相乗効果を示唆した。
実験手順
3.1研究材料
化合物:NIT、カルボプラチン、パクリタキセル、エピルビシンおよびピラルビシンを供給者から購入し、そして、DMSOに溶解し、ストック溶液にして、そして、−20℃で貯蔵した。
使用前に、ストック溶液を異なる濃度の作業溶液に希釈した。作業溶液中のDMSO濃度は1%未満であった。全ての癌細胞株は、ベンダーから購入した。
G418選択の後、MBT−2−LucおよびRM−9−Lucからの増殖培地をルシフェラーゼ活性について試験して、ルシフェラーゼの細胞培地への発現および分泌を確認した。
インビトロアッセイ
MTTアッセイを、試験法として使用した。
適切な量の細胞(2X103/100μL)を、96ウェルのプレートに播種し、そして、CO2インキュベータ中で、37℃で、細胞接着および適応のために、一夜、インキュベートした。
癌細胞もまた、別の96ウェルプレート(プレートT0)に、12ウェル/細胞株で播種し、時間ゼロ(T0)でOD値を測定した。
一晩の順応後、プレートT0に、ウェル当たり、20μLのMTT試薬(最終濃度0.5ミリグラム/ミリリットル)を添加し、そして、37℃で、4時間インキュベートした。
上清培地をピペットで取り出し、そして、ウェル当たり、約150μLのDMSOを添加した。
試験波長として550nmおよび基準波長として630nmを使用して、時間ゼロ(T0)でのOD値を得るために、プレートをプレートリーダーで読み取った。
一晩の適応後、試験化合物およびビヒクルをプレートに加えて、細胞を処理した。
試験化合物を、3倍のウェル中において、6つの濃度で試験した。プレートを37℃で、CO2インキュベーターで、48時間インキュベートした。より長いインキュベーション時間を使用することができる。
1ウェルあたり、20μLのMTT試薬を添加し、そして、37℃で、4時間インキュベートした。その上清培地をピペットで取り出し、そして、ウェル当たり、約150μLのDMSOを添加した。
試験波長として約550nMおよび参照波長として約630nMを使用して、プレートリーダー(TECAN、Infinite M200)上でプレートを読み取った。
細胞生存度(%)は、2つの方法で計算した:
マウス膀胱および前立腺癌モデルにおける、インビボ効果
同所性(orthotopic)膀胱癌腫瘍を確立するために、マウスを、ケタミン/キシラジン溶液、0.1ミリリットル/10g体重(K113;シグマーアルドリッチ ジャパン合同会社、東京、日本)を、腹腔内に(ip)投与することにより麻酔した。
次いで、24ゲージのテフロン(登録商標)静脈内カテーテルを、不活性潤滑剤を使用し、尿道を通して膀胱または前立腺に挿入した。腫瘍の埋め込みのための膀胱を準備するために、短時間の酸暴露と、その後のアルカリ中和した。8μlの1MOI硝酸銀の膀胱内滴下によって、膀胱壁上の化学的病変が促進した。これにより、適切かつ制御された拡散性膀胱壁病変が形成された。
15秒後、PBSの経尿道注入により、内容物を洗い流した。
最初のカテーテルを取り出し、そして、MBT−2−Luc細胞(0.1ミリリットルPBSと混合した5x104の細胞)の膀胱内滴下のために、尿道に、新しい24−ゲージのカテーテルを挿入し、ステッチで、1.5時間保持した。
10日ごとに、生物発光技術(Xenogen IVIS200 システム;Xenogen Corporation、Hopkinton、MA、米国)を用いて、ルシフェリンの腹腔内投与後に腫瘍イメージングを行った。
薬物投与の詳細な説明を表23に示す。
ルシフェラーゼ発現によって決定された腫瘍造影結果によれば、BCG(1X105CFU/100μL)は、1週間に1回、3週間、膀胱内投与した。
NITは、継続的に、または48時間休薬して投与した。
RM−9−Luc細胞のトリプシン処理に続いて、ハンクス平衡塩溶液10μl中の5.0X103個の細胞を、新しい24−ゲージのカテーテルを用いて、前立腺の右背葉部(dorsal lobe)に直接、注射した。
注入された前立腺背葉内の明確に識別可能な小胞(bleb)は、技術的に満足のいく注射のサインと考えた。腹部の創傷をステンレス鋼クリップ(Autoclip;Becton Dickinson Co.、Sparks、MD)で閉じた。
RM−9−Luc細胞注射の一週間後、腫瘍径が5mmに達した時、同所性前立腺癌腫瘍を有するマウスを、無作為に、4群に分けた:コントロール(PBS)、NIT、抗PD−1、およびNITと抗PD−1との組み合わせ。

EXY=EX+EY−(EXEY)、

Claims (12)
- 免疫療法剤の有効量を組み合わせた、癌の治療のための医薬品の製造における、ニトロキソリン、オキシキノリン、クリオキノール、ヨードキノールまたはその薬学的に許容される塩の有効量の使用であって、
ニトロキソリン、オキシキノリン、クリオキノール、ヨードキノールまたはその薬学的に許容される塩と、免疫療法剤の組合せが、相乗効果をもたらし、そして、
a)免疫療法剤が、抗PD−1抗体もしくはその抗原結合断片、または抗PD−L1抗体もしくはその抗原結合断片であり、癌が膀胱癌または前立腺癌であるか、または
b)免疫療法剤が、Bacillus Calmette−Guerin(BCG)治療剤であり、癌が膀胱癌である、使用。 - 免疫療法剤が、抗PD−1抗体またはその抗原結合断片である、請求項1に記載の使用であって、ニトロキソリン、オキシキノリン、クリオキノール、ヨードキノールまたはその薬学的に許容される塩の有効量が、1日当たり、経口投与で、100ミリグラム〜1600ミリグラムであり、そして、抗PD−1抗体またはその抗原結合断片の有効量が、静脈投与で、1〜4週間毎に、1ミリグラム/キログラム〜20ミリグラム/キログラムである使用。

d)化学療法剤が、カルボプラチン、シスプラチン、オキサリプラチンまたはネダプラチンであり、癌が膀胱癌または肝臓癌である、使用。 - 癌が膀胱癌であり、そして、化学療法剤が膀胱内に投与される、請求項5に記載の使用。
- 請求項5または6に記載の使用であって、
化学療法剤が、パクリタキセル、ドセタキセルまたはカバジタキセルであり、
ニトロキソリン、オキシキノリン、クリオキノール、ヨードキノールまたはその薬学的に許容される塩の有効量が、1日当たり、経口投与で、100ミリグラム〜1600ミリグラムであり、そして、パクリタキセル、ドセタキセルまたはカバジタキセルが、膀胱内に、膀胱点滴液中0.5ミリグラム/ミリリットル〜10ミリグラム/ミリリットルの濃度で、投与される使用。 - 請求項5または6に記載の使用であって、
化学療法剤が、マイトマイシンCであり、
ニトロキソリン、オキシキノリン、クリオキノール、ヨードキノールまたはその薬学的に許容される塩の有効量が、1日当たり、経口投与で、100ミリグラム〜1600ミリグラムであり、そして、マイトマイシンCが、膀胱内に、膀胱点滴液中0.1ミリグラム/ミリリットル〜5ミリグラム/ミリリットルの濃度で、投与される、使用。 - 化学療法剤が、エピルビシン、ドキソルビシンまたはピラルビシンである、請求項5または6のいずれか1項に記載の使用であって、

a)免疫療法剤が、抗PD−1抗体もしくはその抗原結合断片、または抗PD−L1抗体もしくはその抗原結合断片であり、癌が膀胱癌または前立腺癌であるか、または
b)免疫療法剤が、Bacillus Calmette−Guerin(BCG)治療剤であり、癌が膀胱癌である、キット。 - それを必要とする被験体の癌を治療するためのキットであって、ニトロキソリン、オキシキノリン、クリオキノール、ヨードキノールまたはその薬学的に許容される塩の有効量、化学療法剤の有効量、および1種以上の薬学的に許容される担体、および、ニトロキソリン、オキシキノリン、クリオキノール、ヨードキノールまたはその薬学的に許容される塩の有効量と、化学療法剤の有効量の組合せを用いることのインストラクションを含み、
a)化学療法剤が、パクリタキセル、ドセタキセル、またはカバジタキセルであり、癌が膀胱癌または肝臓癌であるか、
b)化学療法剤が、マイトマイシンCであり、癌が膀胱癌であるか、
c)化学療法剤が、エピルビシン、ドキソルビシン、またはピラルビシンであり、癌が膀胱癌であるか、または
