Description
生減弱形態を用いたワクチン接種は不運にも非中和抗体の豊富な生成を誘導するのみである。この時間間隔の間、例えば、限られた量のインターフェロン(IFN)−γ(分泌細胞のみが生成される。したがって、PRRSVは本質的に、常に豊富な体液性(抗体に基づく)免疫、および、多様で限定的だが潜在的に防御的なTヘルパー(Th)1様のIFN−γ反応によって区別される、不均衡免疫反応を刺激すると考えられる。適応免疫の不均衡進行に最も関与していると思われるPRRSV感染の1つの特徴は、十分な自然免疫反応の欠如である。大抵、ウイルス感染細胞はI型インターフェロン「IFN」(IF
N−αおよびIFN−βを含む)を分泌し、隣接する細胞を感染から保護する。さらに、放出されたI型IFNは未感作T細胞のサブセットと相互作用し、ウイルス特有II型IFN(IFN−γ)分泌細胞への転換を促進する。対照的に、PRRSV露出に対するブタのIFN−α反応はほぼ実在しない。そのようなIFN−α生成の病原体による非効果的な刺激は、IFN−αがIFN−γ遺伝子発現を増加させるため、宿主の適応免疫反応の本質において大きな影響を有すると期待されるだろう。したがって、元のサイトカインは優性経路を制御し,適応免疫の発達、すなわちT細胞媒介IFN−γ反応および最大抗ウイルス免疫防御を促進する。

P129−PK継代17感染性CDNAクローンから得られたウイルスは、IFN−アルファ誘導を抑制する能力を減衰させる

は、モノクローナル抗体が商業的に入手可能な状態で調製されたサンドイッチELISAを使用し、(反ブタIFN−α mAb K9およびF17)IFN−αの存在のために測定された。簡潔に、Immulon IIプレート(Dynatech Inc.)は、4℃で一晩の培養、その後5%の牛胎児血清で補われるRPMI媒体でのブロッキングによって、反ブタIFN−α mAb F17(PBL Laboratories)で被覆された。1時間後、媒体が廃棄され、50マイクロリットルの脱離液が複製の測定ウェルに加えられ、検査された。1時間の培養後に、測定ウェルは4回洗浄され、それからビオチン標識、反ブタIFN−α mAb K9(PBL Laboratories)、HRP抱合型ストレプトアビジン(Zymed Laboratories)、およびTMB基質(KPL)で順番に培養された。光学密度はELISAプレートリーダーで決定された。

ZMAC−1細胞系で調製されたP129ウイルスのストックはウイルスが適合させられたPK−9またはMARC−145細胞で得られたものよりも10倍高い力価に達した。これらのウイルスがPBMCによってIFN−α分泌を刺激する能力検査は、ただ1つを例外として(P3412クローンCを隔離する)、IFN−αのわずか少量(<50pg)が検査されたPRRSウイルスストックのいずれかに対するそれらの曝露に反応したこれらの細胞によって分泌されたことを明らかにした。ブタコロナウイルス、伝染性胃腸炎ウイルス(TGEv)へのそれらの曝露の結果、同じ細胞によって生成されたIFN−α(17,540pg)の豊富な分泌との比較によって無視できる。

。50kbより長い遺伝子のカスタム合成(PRRSVゲノムは約15.5kb)は、多数の業者からの商用サービスとして入手可能であり、GenScript、DNA2.0、およびBio Basic Inc.を含むが、これに限られない。合成のウイルスゲノムは、ゲノムに隣接する5’のPacIおよび3’のSpeI制限酵素部位を使用し、感染性クローンpCMV−S−P129のウイルスゲノムを取り替えることによって、方向的にpCMV−Sベクターにクローン化される。合成ゲノムを切断するために、24ヌクレオチド拡張(5’−GCAGAGCTCGTTAATTAAACCGTC−genome−3’,下線PacI部位を含む)が合成ゲノムの5’末端に組み込まれ、83ヌクレオチド拡張(5’−genome−AAAAAAAAAAAAAAAAAAAAATGCATATTTAAATCCCAAGCCGAATTCCAGCACACTGGCGGCCGTTACTAGTGAGCGGCCGC−3’,下線SpeI部位を含む)が合成ゲノムの3’末端に組み込まれる。PacIおよびSpeIでプラスミドおよび合成ゲノムを切断した後に、当業者によく知られた標準のクローン技術を使用することで、適切な断片を精製し、DNA連結酵素を使用して接合し、スクリーニングおよび繁殖のために大腸菌に形質転換する。

Claims (9)
- 離乳前のブタにおいて、ブタ生殖および呼吸症候群(PRRS)ウイルスによる感染に対する保護免疫反応を誘発する方法であって、該方法は、修飾生PRRSウイルスおよび家畜への使用に適切なキャリアを含むワクチンを前記離乳前のブタに投与することを含み、
前記ワクチンは前記離乳前のブタに2週齢以下の時点で投与され;
前記ワクチンを前記ブタが1日齢以下の時点で投与しても安全かつ免疫保護性であり;
前記修飾生PRRSウイルスは、外来のブタCD163受容体タンパク質を発現するブタ腎細胞株中で毒性PRRSウイルスを複数回継代し、培養からウイルスを回収することにより調製され;および
前記保護免疫反応は、PRRSに感染したワクチン投与されていないブタと比較して、PRRS感染中の肺病変の減少をもたらす、前記方法。 - 前記保護免疫反応が最長6ヵ月まで持続する、請求項1の方法。
- 前記ワクチンが、免疫保護を誘導する豚マイコプラズマ(Mycoplasma hyopneumoniae)抗原、免疫保護を誘導する豚サーコウイルス2型抗原、またはそれらの組み合わせをさらに含む、請求項1の方法。
- ブタにおいて、ブタ生殖および呼吸症候群(PRRS)ウイルスによる感染に対する保護免疫反応を誘発する方法であって、該方法は、
(a)修飾生PRRSウイルス、
(b)前記修飾PRRSウイルスをコードする感染性RNA分子、
(c)前記感染性RNA分子をコードするDNA配列を含む単離されたポリヌクレオチド分子、
(d)前記感染性RNA分子を含むウイルス性ベクター、または
(e)前記単離されたポリヌクレオチド分子を含むウイルス性ベクター;
および家畜への使用に適切なキャリア
を含有するワクチンを、離乳前に、前記ブタに投与することを含み;
前記ワクチンは前記ブタが2週齢以下のときに投与され;
前記修飾PRRSウイルスは、外来のブタCD163受容体を発現するブタ腎細胞上で、複数回、毒性PRRSウイルスを継代することにより調製され;
前記修飾PRRSウイルスは、野生型PRRSウイルスまたは修飾ウイルスの由来元である未修飾のPRRSウイルスと比べて低減された毒性を前記ブタにおいて示し;
前記投与は、野生型PRRSウイルスによる感染に対するブタにおける保護免疫反応を誘発し、該保護免疫反応は、PRRSに感染したワクチン投与されていないブタと比較して、PRRS感染中の肺病変の減少をもたらす、前記方法。 - 前記ワクチンが修飾された生の欧州PRRSウイルスを含む、請求項1または4の方法。
- 前記ワクチンが修飾された生の北米PRRSウイルスを含む、請求項1または4の方法。
- 免疫の保護が最長6ヵ月まで持続する、請求項4の方法。
- ワクチン接種が生後1日目に行われたとき、前記ワクチンが最長6ヵ月まで持続する免疫をもたらす、請求項4の方法。
- 前記ワクチンが前記離乳前のブタに1週齢以下の時点で投与される、請求項1の方法。
