Description
(第1実施形態)
本発明の第1実施形態に係る測定方法は、検出部材の表面に位置しており、第1検出対象物よりも第2検出対象物に高い反応性を有する第1反応物質と第2検出対象物よりも第1検出対象物に高い反応性を有する第2反応物質とを準備する準備工程と、第1検出対象物および第2検出対象物を含む試料を検出部材の表面に供給する供給工程と、供給工程の後、第1反応物質と試料との反応に基づく第1信号値と、第2反応物質と試料との反応に基づく第2信号値とを測定する測定工程と、第1信号値および第2信号値に基づいて、試料における第1検出対象物と第2検出対象物との関係性を算出する算出工程と、を備える。
本実施形態の準備工程は、検出部材の表面に位置しており、第1検出対象物よりも第2検出対象物に高い反応性を有する第1反応物質と、第2検出対象物よりも第1検出対象物に高い反応性を有する第2反応物質とを準備するものである。
(第2実施形態)
図1は、本発明の第2実施形態である測定装置100の機能的構成を示すブロック図である。本発明の第1実施形態に係る測定方法は、測定装置100で実行される。
分子間相互作用解析装置であるBiacore T200(GE ヘルスケア製)を用いた。この装置は、4つのフローセル(Fc1〜4)を有している。
なお、図6(a)に示される白丸、白三角、白菱形、および白四角は、それぞれ図6(b)に示される白丸、白三角、白菱形、および白四角に対応する。
実施例1における(1)〜(7)の操作を、標準試料に代えて血液試料について実施し、アプタマーAおよびアプタマーBについての信号値を得た。得られた信号値と実施例1で作成された検量線とに基づいて、血液試料に含まれる総Hbに対するHbA1cの比率を算出した。
第1反応物質として、RNAアプタマーAに換えてRNAアプタマーC(配列番号4)を用い、第2反応物質として、RNAアプタマーBに換えてRNAアプタマーD(配列番号5)を用いた他は、特に言及しなければ、実施例1および2と同様に、検量線の作成、血液試料の測定を行った。
以下において、実装基板上に、5’末端に6量体のオリゴエチレングリコール(OEG)を介してアミノ基を有するRNAアプタマーCおよびDを固定化し、HbA1cを検出する手順を示す。

Claims (14)
- 検出部材の表面に位置しており、第1検出対象物よりも第2検出対象物に高い反応性を有する第1反応物質と、第2検出対象物よりも第1検出対象物に高い反応性を有する第2反応物質とを準備する準備工程と、
試料を前記検出部材の表面に供給する供給工程と、
一回の前記試料の供給で、前記第1反応物質と前記試料との反応に基づく第1信号値と、前記第2反応物質と前記試料との反応に基づく第2信号値とを測定する測定工程と、
前記第1信号値および前記第2信号値に基づいて、前記試料における総検出対象物に対する前記第1検出対象物の比率を算出する算出工程と、を備える測定方法。 - 前記算出工程は、前記総検出対象物に含まれる前記第1検出対象物と前記第2検出対象物との割合の変化に応じた、前記総検出対象物に対する前記第1検出対象物の比率を算出する、請求項1に記載の測定方法。
- 前記算出工程は、予め作成された検量線と、前記第1信号値および前記第2信号値とに基づいて、前記第1検出対象物の前記比率を算出する、請求項1または2に記載の測定方法。
- 前記検量線は、前記総検出対象物の濃度と、前記総検出対象物に対する前記第1検出対象物の比率とが予め定められた複数の標準試料を用いて測定された、前記第1反応物質と前記標準試料との反応に基づく第1標準信号値と、前記第2反応物質と前記標準試料との反応に基づく第2標準信号値とを用いて作成される、請求項3に記載の測定方法。
- 前記第1反応物質および前記第2反応物質は、タンパク質、ポリペプチド、ペプチド、核酸、およびボロン酸化合物からなる群から選択される、請求項1〜4のいずれか1項に記載の測定方法。
- 前記第1反応物質および前記第2反応物質は、抗体あるいはアプタマーである、請求項1〜5のいずれか1項に記載の測定方法。
- 前記第1反応物質および前記第2反応物質はいずれも、RNAアプタマーである、請求項1〜5のいずれか1項に記載の測定方法。
- 前記検出部材は、表面弾性波素子を有し、
前記第1信号値および前記第2信号値は、前記表面弾性波素子の位相特性の値である、請求項1〜7のいずれか1項に記載の測定方法。 - 前記第1信号値および前記第2信号値は、QCM、SPRおよびFETから選択された方法によって測定された値である、請求項1〜8のいずれか1項に記載の測定方法。
- 前記試料は、血液試料である、請求項1〜9のいずれか1項に記載の測定方法。
- 前記第1検出対象物および前記第2検出対象物は、タンパク性物質である、請求項1〜10のいずれか1項に記載の測定方法。
- 前記タンパク性物質は、疾患関連タンパク性物質である、請求項11に記載の測定方法。
- 第1検出対象物と第2検出対象物とを含む試料を、検出部材の表面に位置しており、前記第1検出対象物よりも前記第2検出対象物に高い反応性を有する第1反応物質と、前記第2検出対象物よりも前記第1検出対象物に高い反応性を有する第2反応物質とに供給する供給部と、
一回の前記試料の供給で、前記第1反応物質と前記試料との反応に基づく第1信号値と、前記第2反応物質と前記試料との反応に基づく第2信号値とを測定する測定部と、
前記第1信号値および前記第2信号値に基づいて、前記試料における総検出対象物に対する前記第1検出対象物の比率を算出する算出部と、を備える装置。 - 前記算出部は、前記総検出対象物に含まれる前記第1検出対象物と前記第2検出対象物との割合の変化に応じた、前記総検出対象物に対する前記第1検出対象物の比率を算出する、請求項13に記載の装置。
