Description
用語「細胞潜在力」とは本明細書において、その他の細胞種に分化する細胞の能力。細胞が分化できる細胞種が多いほど、その潜在力は大きい。
in vitro培養後にin vivoで単離された多能性幹細胞の細胞潜在力を拡張するために使用され得る因子のカクテル、多能性の化学的拡張剤(CEP)を同定した。CEPは、例えば、isPSCに、in vivoで胚外系統を作り出す能力を付与することによって、未処置の対応する細胞と比較した場合に、isPSCの細胞潜在力を拡張する。CEPは、拡張されていない多能性細胞などの混入細胞種を少なくとも10%、20% 30%、35%、40%、45%、50%、55%、60%、65%、70%、75%、80%、85%、90%、95%または99%含まないciEPSCの単離された集団を提供するために使用され得る。
1.サイトカイン
好ましいサイトカインは、1〜100ng/ml、好ましくは、1〜50、さらにより好ましくは、1〜30ng/mlの濃度範囲で使用される、ヒト白血病抑制因子(LIF)(「L」)、インターロイキン6クラスサイトカインである。IL−6は、ニューロポイエチン類、IL−6、IL−11、IL−27、IL−31、白血病抑制因子、オンコスタチンM、カルジオトロフィン−1、ニューロポイエチンおよびカルジオトロフィン様サイトカイン因子1(新規ニューロトロフィン1およびB細胞刺激因子−3としても知られる)ならびにIL−6の2種のウイルス性類似体を含む構造的に関連するサイトカインの群の創始者メンバーである、プロトタイプの4へリックスバンドルサイトカインである。サイトカインのインターロイキン6ファミリーのこれらのメンバーは、LIFについて開示される同等の濃度で、本明細書において開示される組成物において使用され得る。
多能性を拡張する化学化合物、すなわち、多能性の化学拡張剤(CEP)として、単独の、またはタンパク質と組み合わせた、2,000ダルトン未満、より好ましくは、1,500ダルトン未満、最も好ましくは、1,000ダルトン未満の分子量を有する小分子が挙げられる。小分子は、900ダルトン以下または500ダルトン以下の分子量を有し得る。化学的に誘導される再プログラムにおいて、好ましくは、本明細書において同定された小分子と同一の経路を標的とする、より大きな分子が使用され得る。
PARP1阻害剤は、好ましくは、MiH(ミノサイクリンヒドロクロリド)(「M」)、0.5〜5μMの、より好ましくは、の範囲の濃度で使用される強力なPARP1選択的阻害剤である。例えば、組成物中のMiHの濃度は、0.5、1、1.5、2、2.5、3、3.5、4、4.5、5または5μMであり得る。さらなるPARP1阻害剤として、それだけには限らないが、BSI−201(4−ヨード−3−ニトロベンズアミド)、NAM(ニコチンアミド)およびPJ34(N−(6−オキソ−5,6−ジヒドロ−フェナントリジン−2−イル)−N,N−ジメチルアセトアミド(pPARP1およびPARP2阻害剤))、PARP阻害剤XIV、4−[(1−メチル−1H−ピロール−2−イル)メチレン]−1,3(2H,4H)−イソキノリンジオン(強力なPARP1阻害剤)、ベリパリブ、2−[(2R)−2−メチルピロリジン−2−イル]−1H−ベンズイミダゾール−4−カルボキサミドジヒドロクロリド(強力なPARP−1およびPARP−2阻害剤)、オラパリブ(Olaparidb)(1−(シクロプロピルカルボニル)−4−[5−[(3,4−ジヒドロ−4−オキソ−1−フタラジニル)メチル]−2−フルオロベンゾイル]ピペラジン);6−アミノ−1H−ベンゾ[de]イソキノリン−1およびINH2BP(5−ヨード−6−アミノ−1,2−ベンゾピロン;PARP阻害剤II)が挙げられる。PARP1および/またはPARP2阻害剤を含むその他の既知PARP阻害剤は、市販されており、細胞をciEPSCに再プログラムするのに有効な量で本明細書において開示されるCEP組成物中に含まれ得る。MiHをTHと、DESまたはDIMをNAMと置換する実験などにおいて、例えば、本明細書において例示されるようなin vitroアッセイを使用して、その他のPARP1阻害剤の同等濃度を決定することは、当業者の能力内である。
GSK阻害剤は、好ましくは、GSK3を阻害し、好ましくは、GSK3に対して選択的である。適したGSK阻害剤として、アミノピリミジン、グリコーゲンシンターゼキナーゼ3阻害剤であるCHIR99021(「C」)がある。CEP組成物は、0.1〜5μMの、好ましくは、1から3の間の、さらにより好ましくは、1.5から3μMの間の濃度範囲のCHIR99021を含む。例えば、CEPは、1、1.5、2、2.5、3、3.5、4、4.5または5μMの濃度のCHIR99021を含み得る。当業者ならば、必要な有効量を容易に微調整できるので、これらの数の間に入る濃度が考慮される。
最も好ましいGPCR阻害剤として、1〜5μMの濃度範囲で使用される(S)−(+)−ジメチンデンマレエート(「D」)がある。ジメチンデンマレエートは、サブタイプ選択的mAChR(ムスカリン性アセチルコリン受容体)M2、mAChR M1、mAChR M3およびmAChR M4アンタゴニストならびにヒスタミンH1受容体アンタゴニストであるエナンチオマーである。しかし、その他のGPCR阻害剤は、本明細書において開示されるCEP組成物中に含まれ得、それらとして、それだけには限らないが、エチレンジアミン、例えば、トリペレナミンHCL、(中枢および末梢ヒスタミンH1受容体(receptorss)を競合的に遮断するヒスタミンH1アンタゴニスト)、メピラミンおよびアンタゾリン;ロラチジン(loratidine)またはその代謝産物、デスロラタジン(選択的ヒスタミンH1アンタゴニスト)などの三環系または四環系が挙げられる。その他のものは当技術分野で公知であり、それだけには限らないが、レボセチリジン、フェキソフェナジン、アステミゾール、ケトチフェン、セチリジン、ロラタジン、ルパタジン、ミゾラスチン、アクリバスチン、エバスチン、ビラスチン、ベポタスチン、テルフェナジン、キフェナジン(Quifenadine)、シクリジン、クロルシクリジン、ヒドロキシン(hydroxine)、ペニラミン(peniramine)、クロルフェナミン、トリポリジン(tripolidine)、ジフェンヒドラミン、カルビノキサミン、ブロマジンなどが挙げられる。
拡張された多能性幹細胞は、任意の哺乳類(例えば、ウシ、ヒツジ、ブタ、イヌ、ネコ、ウマ、霊長類)、好ましくは、ヒトなどの哺乳類から得られた多能性細胞、または部分的もしくは完全に分化した細胞を誘導することによって得られる。供給源として、骨髄、線維芽細胞、胎児組織(例えば、胎児肝臓組織)、末梢血、臍帯血、膵臓、皮膚または任意の臓器もしくは組織が挙げられる。
CiEPSCは、(i)形態学的に−マウスESC様形態学および(ii)(a)細胞の、3種の胚性胚葉の組織に分化する能力、(b)1種以上の胚外マーカーの上方制御された発現、(c)1種または複数の多能性のマーカーの上方制御および/または下方制御ならびに(d)in vivoでのTEおよびICM両方への寄与に基づいて機能的に、を含め特性に基づいて拡張された多能性細胞として同定される。ciEPSCは、対応する細胞と比較した場合にin vivoで拡張された細胞潜在力を示す。好ましい実施形態では、ciEPSCは、in vivoで1種以上の胚外系統を作り出す/それに寄与する。in vivoでの1種以上の系統へのciEPSC寄与は、当技術分野で公知の、実施例に記載されるような方法を使用するin vivo移植後に、胚外マーカー(以下に論じられる)のうち1種以上のマーカーの存在を調べることによって決定され得る。
ヒトドナー細胞から得られたciEPSCは、マウス胚性幹(ES)細胞と形態学的に似ている。ヒトEPS細胞は、マウス胚幹細胞と似ているドーム型コロニーを形成する。マウスEPS細胞は、マウス胚幹細胞と形態学的に同一である。
ciEPSCは、当技術分野で公知の方法を使用して、三胚葉の各々、外胚葉、中胚葉および内胚葉に由来する1種以上の細胞/組織に分化する能力を有する。
対応する細胞はそのように寄与できない、in vivoでTEおよびICMの両方へ寄与する能力は、in vivoで胚外系統を作り出す/それに寄与する改善された能力のしるしである。例えば、三胚葉の各々から1種以上の細胞/組織へ分化し得る、ヒト胚性幹(hES)細胞の細胞潜在力は、本明細書において開示されるようなCEPSと接触されたhESに、in vivoでTEおよびICMの両方に寄与する能力を付与することによって改善される/拡張される。in vivoでTEおよびICMに寄与する細胞の能力は、当技術分野で公知の方法を使用して決定され得る。いくつかの実施形態では、TEおよびICMに寄与する能力は、実施例において開示されるように、ciEPSCをマウスE2.5またはE3.5胚に微量注入し、in vivoでE10.5胚に発達させることおよび注入された細胞のTEおよびICMへの寄与を調べることによって決定される。
ciEPSCは、対応する生物から単離された、未処置の対応する細胞(すなわち、本明細書において開示されるようなCEPSと接触されていない対応する細胞)と比較した場合に、TBX3およびGBX2などの多能性の1種または複数のマーカーの上方制御を示す。OCT4、REX1、DPPA3、TBX3およびGBX2を含むいくつかの多能性マーカー遺伝子のmRNA発現は、hEPS細胞において、CEP処理されていないプライム型hES細胞におけるものよりも均一であった。
A.多能性幹細胞における拡張された多能性の誘導
ciEPSCは、誘導/再プログラムされる予定の細胞(本明細書において、ドナー細胞)を、CEPを含有する培養培地と、細胞の化学的に誘導された拡張された多能性幹細胞(ciEPSC)への再プログラミングをもたらすのに十分な時間接触させることによって製造される。
ciEPSCの実質的に精製された集団は、培養供給源から例えば、抽出によって(例えば、密度勾配遠心分離および/またはフローサイトメトリーによる)得られ得る。純度は、任意の適当な方法によって測定され得る。多能性細胞は、例えば、フローサイトメトリー(例えば、FACS解析)によって精製された99%〜100%であり得る。ヒトの誘導された拡張された多能性幹細胞は、例えば、誘導多能性幹細胞上のマーカーまたはマーカーの組合せと結合する分子(例えば、抗体、抗体誘導体、リガンドまたはFc−ペプチド融合分子)を利用し、それによって、分子と結合する細胞を正に選択すること(すなわち、正の選択)によって単離され得る。正の選択方法のその他の例として、所望のおよび望ましくない細胞種の混合集団において、所望の細胞種の成長を優先的に促進する方法が挙げられる。あるいは、所望の細胞種上に存在しないが、望ましくない細胞種上に存在するマーカーと結合する分子を使用することによって、このようなマーカーを含有する望ましくない細胞が、所望の細胞から除去され得る(すなわち、負の選択)。その他の負の選択方法は、所望のおよび望ましくない細胞種の混合集団において、望ましくない細胞種を優先的に死滅させることまたはその成長を阻害することを含む。したがって、負の選択、正の選択またはそれらの組合せを使用することによって、幹細胞の濃縮された集団が作製され得る。
ciEPSCは培養で増殖され、後の回収および使用のために貯蔵され得る。細胞の培養物または幹細胞の混合培養物が確立されると、細胞の集団は、組織形成をともなって、または伴わずに、細胞増殖を促す条件下で細胞密度が示すように新鮮培地へ継代することによってin vitroで有糸分裂的に増殖される。このような培養方法は、例えば、分化を誘導する特定の増殖因子(例えば、IGF、EGF、FGF、VEGFおよび/またはその他の増殖因子)を欠く培養培地中で細胞を継代することを含み得る。培養細胞は、十分な細胞密度が到達されたときに新鮮培地に移され得る。
所望の細胞種または形態学を生じさせ得る幹細胞の容易に入手可能な供給源の同定は、治療的処置、組織工学および研究にとって重要である。幹細胞を入手可能であることは、移植、組織工学、血管新生の調節、脈管形成、臓器再生、ヒト化動物モデル、細胞置換または細胞療法ならびに疾患の予防などにおいて極めて有用となる。このような幹細胞は、遺伝子療法レジメンの一部として対象中に遺伝子を導入するために使用され得る。
幹細胞の培養物は、ひとたび確立されると、新規組織を生成可能な後代細胞、例えば、線維芽細胞を生成するために使用され得る。ciEPSCは、三胚葉のいずれかから細胞、例えば、上皮細胞を含む皮膚および有毛細胞、ケラチノサイト、メラノサイト、脂肪細胞、骨、筋肉および結合組織を形成する細胞、例えば、筋細胞、軟骨細胞、骨細胞、肺胞細胞、実質細胞、例えば、肝細胞、腎細胞、副腎細胞および島細胞、血液細胞、網膜細胞(および耳中の有毛細胞または舌上の味蕾を形成するものなどの知覚に関与するその他の細胞)および神経を含む神経組織に分化するように導入され得る。
誘導多能性幹細胞の治療的使用は、癌、創傷、新生物、傷害、ウイルス感染、糖尿病などに起因する疾患および障害を含む種々の病理学的状態を治療するために、誘導多能性幹細胞、幹細胞集団またはその後代を個体中に移植することを含む。治療は、現在当技術分野で公知の任意の方法に従って、新規組織を生成するための細胞の使用およびこのように生成された組織の使用を伴い得る。細胞は、in vivoで新規組織を生成するように組織損傷の部位に移植、注入またはそうでなければ直接投与され得る。一実施形態では、投与は、遺伝子改変されたciEPSCまたはその後代の投与を含む。
糖尿病(DM)は、膵臓が十分なインスリンを産生しないため、または細胞が産生されるインスリンに応答しないためのいずれかのために対象が高血糖を有する一群の代謝疾患である。
神経変性障害は、疾患の結果としてニューロンの変質、遺伝性の状態または外傷性もしくは虚血性脊髄もしくは脳傷害などの傷害が関与する状態を特徴とする。神経変性状態として、ニューロンの損傷または変質が関与する任意の疾患または障害またはその症状または原因または効果が挙げられる。神経変性状態として、それだけには限らないが、アレキサンダー病、アルパース病、アルツハイマー病、筋萎縮性側索硬化症、毛細血管拡張性運動失調症、カナバン病、コケイン症候群、大脳皮質基底核変性症、クロイツフェルト・ヤコブ病、ハンチントン病、ケネディ病、クラッベ病、レビー小体認知症、マシャド・ジョセフ病、多発性硬化症、パーキンソン病、ペリツェウス・メルツバッハ病、ニーマン−ピック病、原発性側索硬化症、レフサム病、サンドホフ病、シルダー病、スティール・リチャードソン・オルゼウスキー病、脊髄ろうまたは損傷を受けたニューロンと関連する任意のその他の状態を挙げることができる。その他の神経変性状態として、外傷性脊髄損傷、虚血性脊髄損傷、卒中、外傷性脳損傷および遺伝性の状態を挙げることができる、またはそれによって引き起こされ得る。
ciEPSCおよびその後代の治療的使用は、癌と関連する症状を治療および/または回復させるために誘導多能性幹細胞、幹細胞集団またはその後代を個体に移植することを含む。例えば、一実施形態では、ciEPSCは、対象の細胞を死滅させ、低減させ、または損傷を与えた化学療法を受けている癌患者に投与され得る。癌のための通常の幹細胞移植では、すべての癌細胞を破壊しようとして、しばしば、放射線療法とともに極めて高用量の化学療法が使用される。この治療はまた、骨髄中の幹細胞を死滅させる。治療の直後に、破壊されたものと置き換えるために幹細胞が与えられる。
ciEPSCおよびその後代は、当技術分野で公知の方法を使用して組織工学によって作製される構築物を作製するために使用され得る。組織工学によって作製された構築物は、損傷を受けた臓器または組織を修復するまたはそれに取って代わるための人工デバイスとして、を含む種々の目的のために使用され得る。一般に、それらはまた、構築物の細胞によって分泌されるタンパク質またはその他の分子のin vivo 送達システムとして、または薬物送達システムとして役立ち得る。組織工学によって作製された構築物はまた、組織機能のin vitroモデルとして、または種々の治療もしくは医薬品の効果を試験するためのモデルとして用途がある。幹細胞の移植のための最もよく使用される生体材料足場は、Willerth,S.M.およびSakiyama−Elbert,S.E.、Combining stem cells and biomaterial scaffolds for constructing tissues and cell delivery(2008年7月9日)、StemBook編The Stem Cell Research Community、StemBookに概説されている、幹細胞の移植のための最もよく使用される生体材料足場に概説されている。組織工学技術は、組織成長を持続し、促進するために適当な培養基質の選択を頻繁に含む。一般に、これらの基質は、三次元でなくてはならず、対象の組織の所望の形状の足場を形成するように処理可能でなくてはならない。
ciEPSCは、三胚葉のいずれかから細胞、例えば、上皮細胞を含む皮膚および有毛細胞、ケラチノサイト、メラノサイト、脂肪細胞、骨、筋肉および結合組織を形成する細胞、例えば、筋細胞、軟骨細胞、骨細胞、肺胞細胞、実質細胞、例えば、肝細胞、腎細胞、副腎細胞および島細胞(例えば、α細胞、δ細胞、PP細胞およびβ細胞)、血液細胞(例えば、白血球、赤血球、マクロファージおよびリンパ球)、網膜細胞(および耳中の有毛細胞または舌上の味蕾を形成するものなどの知覚に関与するその他の細胞)および神経を含む神経組織に分化するように誘導され得る。
ciEPSCは、in vivoまたはin vitro/ex vivoで脱分化を促進するために、投与、送達または対象、組織もしくは細胞との接触のために製剤化され得る。増殖因子、分化または脱分化を誘導するその他の因子、分泌産物、免疫調節物質、抗炎症剤、退縮因子、神経支配、血管新生を促進する、またはリンパネットワークを増強する生物学的に活性な化合物などのさらなる因子ならびに薬物が組み込まれ得る。
支持構造は、緩い織ったメッシュまたは不織メッシュであり得、これでは、細胞はメッシュ中またはメッシュ上に播種される。構造は、固体構造支持体を含み得る。支持体は、チューブ、例えば、神経軸索の再成長のための神経管であり得る。支持体は、ステントまたは弁であり得る。支持体は、多孔質構造中への細胞の内殖および/または細胞の播種を可能にする多孔質界面を有する膝または臀部またはその一部などの関節人口装具であり得る。多数のその他の種類の指示構造もあり得る。例えば、支持構造は、スポンジ、発泡体、サンゴまたは内部孔を有する生体適合性無機構造または織り合わされたポリマー繊維のメッシュシートから形成され得る。これらの支持構造は、既知方法を使用して調製され得る。
別の実施形態では、細胞は、ヒドロゲルと混合されて、細胞−ヒドロゲル混合物を形成する。ヒドロゲルは、注入もしくはカテーテルによって、またはその他の支持構造の移植の時点で投与され得る。架橋は、投与に先立って、その間に、またはその後に起こり得る。
単離されたciEPSCは、所望の種(ドナー)に由来するciEPSCを同一または異なる種の第2の動物(レシピエント)中に組み込む動物モデルを作り出すために使用され得る。ドナー動物は、ヒト、マウス、ラット、ブタ、ウシ、ヒツジ、ヤギ、ウマ、イヌ、チンパンジー、ゴリラ、オランウータン、サル、マーモセットなどといった哺乳類であり得る。いくつかの好ましい実施形態では、ドナー哺乳類は、ヒトであり、レシピエント哺乳類は、ヒト化動物モデルを提供するために使用される非ヒトである。その他の実施形態では、ドナーおよびレシピエント動物は、大きさが対応している。レシピエントは、ブタ、ラット、マウス、ウシ、ヒツジ、ヤギ、ウマ、イヌ、チンパンジー、ゴリラ、オランウータン、サル、マーモセットおよびボノボなどのヒト以外の任意の動物であり得る。ciEPSCは、ヒトではない哺乳類において臓器再生のために使用され得、ciEPSCは、哺乳類において所望の臓器を生成するために使用され得、これでは、哺乳類は、発生段階におけるその臓器の発達の欠如と関連する異常を有する。
本明細書において開示される拡張された多能性(CEP)の化学誘導物質を含むキットが提供される。CEPは、上記で記載される通りである。これらは、所望の濃度をもたらすための細胞培養培地への添加を容易にする既定の濃度を有する形態であり得る。キットは、ドナー細胞種に基づいて所望の濃度範囲および投与時間を提供する指示書を含み得る。キットはまた、拡張された多能性を誘導するためのドナー細胞の培養のための、CEPと事前混合されている細胞培養培地を含み得る。
小分子ライブラリー
スクリーニングのために使用される小分子ライブラリーは、表1に記載されるように、購入した、または自家作製した。
最初の試みは、ヒトにおいてナイーブ型多能性を支持する新規小分子を同定することに焦点を当てた。マウスにおいてナイーブ型多能性を支持する2iおよびLIF条件(ERK阻害剤PD0325901、GSK3β阻害剤CHIR99021およびヒトLIF(hLIF))に基づいて、プライム型hES細胞(Yeomら、Development、第122巻:881〜894頁(1996年);Tesarら、Nature、第448巻:196〜199頁(2007年)を使用して最初のスクリーニングを実施して、ナイーブ型多能性マーカーOCT4遠位エンハンサー(DE)を活性化し得る化学化合物を同定した(図1Aおよび表1)。
プライム型hESは、フラットなコロニーである。対照的に、ナイーブ型ヒト多能性細胞は、ドーム型コロニーである。したがって、あらゆる得られたヒトEPS細胞は、プライム型ヒト多能性幹細胞から形態学的に区別され得る。
以下のすでに確立されたプライム型hES/hPS細胞系統を使用した(EPS変換のために用いられた細胞株の継代数は、括弧内に示されている):H1(継代30)、0227E(おおよその継代20)、HSF1(おおよその継代50)およびHSF6(おおよその継代60)およびH9(継代40)。hES/hPS細胞株H1(WA01)およびH9(WA09)は、WiCellから入手し、核型解析によって立証した。プライム型hES細胞を、20%ノックアウト血清置換(KSR)(Invitrogen)、1mmグルタミン(Invitrogen)または1% GlutaMAX(Invitrogen、35050)、1%非必須アミノ酸(Invitrogen)、0.1mm β−メルカプトエタノール(Invitrogen)および4〜10ng/ml bFGF(塩基性(basis)線維芽細胞増殖因子)(Peprotech)を補給した従来のhES/hPS細胞培地:DMEM−F12(Invitrogen)中、マイトマイシンC不活性化MEFフィーダー細胞(2×104個/cm2)またはマトリゲルコーティングされたディッシュ上で20% O2、5% CO2条件で維持した。細胞株を、ディスパーゼを使用して5〜7日毎に1:3〜1:5の分割比で継代した。ナイーブ型NHSM−hES細胞を、これまでの報告(Gafniら、Nature、第504巻(7479号):282〜6頁(2013年))に従って培養した。
マウスナイーブ型ES細胞を、10ng/mL hLIF(Peprotech)、3μm CHIR99021(Tocris)および1μm PD0325901(Tocris)を補給した2i培地含有血清不含N2B27培地中、マイトマイシンC不活性化MEFフィーダー細胞またはゼラチンコーティングされたディッシュ上で20% O2、5% CO2条件で維持した。細胞を、0.05% トリプシン−EDTA(Invitrogen)によって2〜4日毎に継代した。
N2B27−LCDM培地の調製
500mlのN2B27培地を、240mlのDMEM/F12(ダルベッコの改変イーグル培地/栄養混合物F−12)(Invitrogen、11320)、240mlのNEUROBASAL(登録商標)培地(基礎細胞培養培地)(Invitrogen、21103−049)、2.5mlのN−2栄養補助剤(Invitrogen、17502048)、5mlのB−27栄養補助剤(Invitrogen; 17504010)、1mmグルタミンまたはGlutaMAX(商標)(Invitrogen)、1%非必須アミノ酸(Invitrogen)、0.1mm β−メルカプトエタノール(Invitrogen)、ペニシリン−ストレプトマイシン(Invitrogen)、(任意選択)5mg/mLのBSA(ウシ血清アルブミン)(Sigma)および小分子阻害剤を以下のように含めることによって作製した。
変換は、通常、プライム型hES/hPS細胞の最後の継代後3日目または4日目に実施した(それらが5〜6日毎に継代された場合には。コロニーは、通常、60〜70%コンフルエンスに到達した。
臨床目的でin vitro受精によって生成された胚盤胞期のヒト胚を、書面によるインフォームド・コンセントおよび承認を得て入手した。プロテアーゼ(Sigma、P8811)によって透明帯を除去した後、全胚をマイトマイシンC不活性化MEFフィーダー細胞上に播種し(4×104個/cm2)、LCDM培地で培養した。胚が播種される少なくとも30分前に、MEF細胞培養培地をFBS−LCDM培地に交換した。
書面によるインフォームド・コンセントおよびClinical Research Ethics Committeeによる承認を得て入手した2〜3ヶ月齢の胚からヒト胚性線維芽細胞を単離し、誘導多能性幹細胞(iPSC)を作製するために使用した。
すべてのマウスの研究は、動物実験委員会(Institutional Animal Care and Use Committee)によって承認された。この研究に使用されるマウスの株は、C57BL/6J−Tg(GOFGFP)11Imeg/Rbrc(OG)、C57BL/6NCrlVr(C57)、ICRおよびJackson Laboratoryから購入したSTOCK Tg(Sox2−cre)1Amc/JとB6.Cg−Gt(ROSA)26Sortm14(CAG−tdTomato)Hze/Jの間のF1ハイブリッドおよびC57BL/6NCrlVr (C57)と129の間のF1ハイブリッドまたはC57BL/6NCrlVr(C57)とDBAの間のF1ハイブリッドを含んでいた。奇形腫形成アッセイのために使用された免疫不全マウスは、商業的に購入した。
胚盤胞から直接誘導されるmEPS細胞のために、OG、C57、ICRまたはSTOCK Tg(Sox2−cre)1Amc/JとB6.Cg−Gt(ROSA)26Sortm14(CAG−tdTomato)Hze/Jマウスの間のF1ハイブリッドまたはC57BL/6NCrlVr(C57)と129の間のF1ハイブリッドの胚盤胞を、LCDM培地とともにMEFフィーダー上に播種した。4日後、成長が観察され、単細胞に解体した。mEPS細胞を2〜3日毎に継代し、さらなる解析のために凍結または使用した。
ヒトおよびマウスの拡張された多能性幹細胞を、20% O2、5% CO2条件で血清不含N2B27−LCDM培地中で培養した。ヒトEPS細胞を未分化状態で維持するために、以下の基準を使用した:a)ヒトEPS細胞をあまりにも低密度にプレーティングすることを避けること、b)適切な量の新たに準備されたMEFフィーダー細胞の使用およびc)ヒトEPS細胞が過成長すること(例えば、90%超のコンフルエンスを達成する;好ましくは、細胞は、90%未満のコンフルエントでなくてはならない)を可能にしないこと。したがって、ヒトおよびマウスEPS細胞を、マイトマイシンC不活性化マウス胚性線維芽細胞(MEF)フィーダー細胞(1cm2あたり3×104個細胞)上で培養し、単細胞トリプシン処理(0.05%トリプシン−EDTA、Invitrogen)によって2〜4日毎に(通常、1:6〜1:10の分割比で)継代した。EPS細胞の継代数は、拡張された多能性状態の獲得後にカウントされた継代の数を示す。N2B27−LCDM培地を新鮮なLCDM培地と毎日交換した。
異種間キメラアッセイが、倫理委員会(Ethics Committee)によって承認された。
hEPS細胞の胚外キメラ化を検出するために、E10.5受胎産物から胎盤および卵黄嚢を単離し、コラゲナーゼIVを使用して消化した。単離された一次細胞を24ウェルプレート中に播種し、さらなる解析の前に、10% FBSおよび10% KSRを補給したノックアウトDMEM中で3〜4日間培養した。
この実験において使用した細胞は、複数細胞微量注入のものと同様である方法で培養し、準備した。細胞懸濁液を注入の前に氷上に20〜30分間置いた。氷上に置いた後に、消化された単細胞を1時間以内に注入のために使用した:言い換えれば、全注入プロセスは、30分超かかってはならない。その後、注入された胚は、37℃で5% CO2を有する加湿インキュベーター中に1〜2時間回復される。細胞を氷上に1時間超置いた場合には、残りの注入のために別のバッチの細胞を消化した。単細胞(Tdtomato標識されたヒト細胞、未標識ヒト細胞、Tdtomato標識EPSおよびナイーブ型マウスES細胞)を、8細胞期ICR2倍体マウス胚中に微量注入し、E5.0までex vivoで発達させた(補給されたKSOM(K(カリウム)補給された単純最適化培地);Summersら、J.Assist Reprod.Denet.、第30巻(8号):995〜999頁*(2013年) 中で60時間)。その他の実験では、キメラ胚盤胞の作製のために、注入された胚を、N2B27−LCDM培地中で最初の4時間培養し(単一hEPS細胞が注入されたキメラ胚の培養には、10μMのY27632(Tocris、1254)が添加されることが推奨された)、次いで、それらをG2 PLUS培地(Vitrolife、10131)に交換した。60時間後、胚を固定し、免疫染色した。
単一Tdtomato標識mEPS細胞を、8細胞マウス胚中に注入し、N2B27−LCDM培地中で4時間培養した。注入された胚を、G2 PLUS培地に移し、さらに56時間培養した。同一キメラ胚の半分を伝統的なmES誘導培地中に播種し、もう一方を、伝統的なTS誘導培地13に播種したことによって、同一キメラ胚を使用して、ESおよびTS細胞株の両方を作製した。同一キメラ胚からES様およびTS様コロニーを同時に誘導した。Tdtomato標識された伝統的なマウスES TT2およびmc2i−1を、このアッセイにおいて対照として別個に使用し、各8細胞胚に、10〜15個の細胞を注入した。
確立されたTS様またはES様細胞株の10〜15個細胞を、8細胞マウス胚中に注入した。in vitroキメラアッセイのために、注入された胚をG2 PLUS培地中で60時間培養した。次いで、それらを固定し、免疫染色した。in vivoキメラアッセイのために、受胎産物を、E13.5発達段階で解体し、蛍光陽性細胞の存在を検出するために免疫蛍光実体顕微鏡を使用して観察した。
細胞を、4%パラホルムアルデヒド中、室温で15分間固定し、0.2% Triton X−100および3%正常ロバ血清(Jackson Immuno Research)を含有するPBSを用いて、室温で45分間ブロッキングした。細胞を一次抗体とともに4℃で一晩インキュベートした。二次抗体(Jackson Immuno Research)を室温で1時間インキュベートした。核をDAPI(Roche)を用いて染色した。抗体の詳細は、以下に提供した。
胚、卵黄嚢および胚盤のキメラ組織を単離し、コラゲナーゼIVを使用して単細胞に消化した。懸濁液を、セルストレーナー(40μm)を通して濾過した。次いで、サンプルをBD LSRFortessa機器で解析した。FlowJoソフトウェア(Ashland)を使用してデータ解析を実施した。
キメラ胎盤組織を単離し、コラゲナーゼIVを使用して消化した。初代Tdtomato+およびTdtomato−胎盤細胞の両方を、FACSを使用して精製した。Trizol(Invitrogen)を使用して精製細胞の全RNAを抽出した。以前に記載されたようにcDNAを調製した31。qPCR鋳型による必要に応じて、増幅されたcDNA産物を10倍希釈した。Bio RAD CFX Connect Real−TimeシステムでKAPA SYBR(登録商標)FAST qPCRキットを使用して定量的PCR解析を実施した。リアルタイムPCRに使用したプライマーは、表2に列挙されている。
キメラ胎盤組織を単離し、コラゲナーゼIVを使用して消化した。キメラ胎盤の1つまたは半分に由来する初代胎盤細胞を、24ウェルプレート中でマトリゲル(商標)コーティングされたフィルター(8μm孔サイズ;BD Biosciences、Franklin Lakes、NJ、USA)上に播種した。手短には、細胞を、血清不含DMEM/F12培地中でTranswellの上側チャンバー上に播種した。Transwellの下部チャンバーは、10% FBSを含有するDMEM/F12培地で満たした。チャンバーを37℃で5% CO2とともに24時間インキュベートした。インキュベーションの最後に、フィルターの上側表面上の細胞を、コットンスワブを使用して除去した。下側表面にフィルターを通して浸潤する細胞を、4%パラホルムアルデヒドを用いて20分間固定し、免疫蛍光によってさらに解析した。免疫蛍光に以下の抗体を使用した:CK8(1:200、sc−52324;Santa Cruz)、CK7(1:40、MA5−11986;Thermo Scientific)およびTdtomato/RFP(1:400、ab62341; Abcam)。
検出されたヒト細胞株のヒトOCT4転写制御を評価するために、OCT4−DEルシフェラーゼプラスミド(Addgene)を、ヌクレオフェクション(4D−Nucleofector(商標)システム、Lonza)によって細胞中にトランスフェクトした。正規化のために、対照ベクターpGL4.74[hRluc/TK](Promega、E6921)を同時トランスフェクトした。空のベクターを用いるトランスフェクションによってベースライン活性を解析した。トランスフェクション後、細胞株を、ウェルあたり5*103個細胞の密度でマトリゲルコーティングされた96ウェルプレート中に播種した。次いで、48時間後、デュアルルシフェラーゼリポーターアッセイ系(Promega、E1960)を使用してルシフェラーゼ活性を検出するために細胞を溶解した。
マウスおよびヒトEPS細胞を、単細胞にトリプシン処理し、ゼラチンコーティングしたプレート上にプレプレーティングすることによってMEFフィーダー細胞から分離し、15%ウシ胎児血清(Gibco)を補給したIMDM(Iscoveの改変ダルベッコ培地)(Gibco)中、超低接着プレート(Corning)で6日間培養した。次いで、EBを集め、同一培地中、マトリゲルコーティングされたプレート上に6日間プレーティングし、固定し、検出した。ヒト細胞のために、抗体は、抗SOX17抗体(AF1924;R&D)、抗FOXA2抗体(ab108422;Abcam)、抗LHX5抗体(sc−130469;Santa Cruz)、抗α−SMA抗体(CBL171;Millipore)、抗CDX2抗体(ab74339、Abcam)および抗GATA6抗体(sc−9055、Santa Cruz)を含む。マウス細胞のために、抗体は、抗FOXA2抗体(ab60721;Abcam)、抗β−IIIチュブリン抗体(sc−80016;Santa Cruz)、抗α−SMA抗体(CBL171;Millipore)および抗CDX2抗体(CDX2−88、AM392;Biogenex)を含む。
ヒトおよびマウスEPS細胞を、注入前にトリプシン処理によって集めた。およそ106個細胞を免疫不全マウス中に皮下注入した。奇形腫は、一般に、2〜6週間以内に発生し、動物は腫瘍の大きさが直径1.5cmを超える前に死亡した。次いで、奇形腫を、パラフィン中に包埋し、ヘマトキシリンおよびエオシン染色のために処理した。
CGH実験のために、ゲノムDNAを抽出し、参照として初期継代の細胞株を使用してImagenesによってCustom SurePrint G3 8x60Kヒト全ゲノムAGI−CGHアレイとハイブリダイズした。
Gバンド染色体解析を、報告されたように(Longoら、Transgenic Res.、第6巻:321〜328頁(1997年))実施した。
0.05%トリプシン−EDTA(Invitrogen)を使用してプレートから細胞を回収し、それらをカウントし、フィーダー細胞を、Y27632を含まない適当な培地中でウェルあたり10,000個細胞の密度でプレシードした24ウェルプレート上にプレーティングした。時間の関数として血球計を使用して細胞数をカウントすることによって成長速度を調べた。成長の対数期(48および72時間の時点)から得たデータを使用して、対数増殖曲線を得た。倍加時間を、式:DT=48*[lg2/(lgNt(4日目の細胞数)−lgNo(2日目の細胞数))]に従って算出した。
RNeasy Miniキット(Qiagen)を使用して、プライム型hES細胞、hEPS細胞およびナイーブ型NSHM−hES細胞、mES細胞およびmEPS細胞から全RNAを単離した。Illumina(登録商標)(NEB、USA)のNEBNext(登録商標)Ultra(商標)RNA Library Prepキットを使用して、RNAシーケンシングライブラリーを構築した。Illumina HiSeq 2000を使用して、断片化されたランダムプライムされた200bp対末端ライブラリーをシーケンシングした。遺伝子発現レベルを、FPKM(100万のマッピングされたリードあたりの1キロベースの転写物の断片)として算出した。その他の実験では、作製されたシーケンシングリードを、TopHatアラインメントソフトウェアツールを使用して、ヒトについてヒトゲノムから構築されたhg19およびマウスについてGRCm38/mm10に対してマッピングした。各遺伝子のリードカウントを算出し、RPKM(100万リードあたりのキロベースあたりのリード)を使用して各遺伝子の発現値を正規化した。
シーケンシングおよびデータ解析.
EZ−Magna ChIP A/Gキット(Millipore)を、製造業者のプロトコールに従って使用してChIPを実施した。抗H3K27me3(抗トリメチル−ヒストンH(Lys27)(07−449、Millipore)および抗H3K4me3(抗ヒストンH3(トリメチルK4)(ab8580、Abcam)))抗体を使用した。精製ChIP DNAを使用して、Illuminaマルチプレックスドシーケンシングライブラリーを調製した。Illumina 1 TruSeq DNAサンプル調製v2キットプロトコールに従って、Illuminaシーケンシングのライブラリーを調製した。次いで、増幅されたライブラリーを、100から500bpの間の断片を捕獲するよう設定された、Sage Science製のPippin Prepシステム中の2%ゲルカセットを使用してサイズ選択した。個別のTruSeq指数を有するライブラリーを等モル比で混合することおよびペアードリード様式で100塩基のためのIllumina HiSeq 2500上のレーンに一緒に流すことによってマルチプレックス化した。Bowtieソフトウェアバージョン2.0を使用して、ヒトリードをヒトリファレンスゲノムhg19(UCSC、2009年2月)に対してアラインした。最大で2つ以下のミスマッチを有するゲノムに対して独特にアラインされたリードのみをさらなる解析のために考えた。ngsplotを使用してすべてのRefSeq遺伝子にわたってクロマチンプロファイルを算出して、TSSの2kb上流からTES の2kb下流の間のリード密度を解析した。ヒトサンプルのプロファイルは、プライム型およびhEPS細胞の平均およびエラーバーを表す。MACSバージョン2.0(ChIP−Seqのモデルベースの解析)ピーク発見アルゴリズムを使用して、バックグラウンドを上回るChIP−Seq濃縮の領域を同定し、BAMPE配列モデルを使用して、ピークを見い出し、FDR<0.05を使用して有意なピークを選択した。最後に、HommerおよびRを使用して、ピークをアノテートし、ピークと関連する遺伝子機能要素および遺伝子および遺伝子オントロジーならびにKEGG(Kyoto Encyclopedia of Genes and Genomics(京都遺伝子ゲノム百科事典))pathwayにおけるピークの分布を解析した。
プロテアーゼ阻害剤カクテル(78443;Thermo Fisher Scientific)およびホスファターゼ阻害剤カクテル(78428;Thermo Fisher Scientific)を補給したRIPA(放射免疫沈降アッセイ)溶解バッファー(P0013B;Beyotime)を使用して、プライム型hES細胞およびhEPS細胞から全細胞タンパク質抽出物を単離した。ブロットを2% BSA(A1470;Sigma−Aldrich)/TBST中、室温で1時間インキュベートし、次いで、5% BSAまたは5%スキムミルクパウダー(P1622;Applygen)/TBST(0.1% Tween−20を有するTris塩基理食塩水バッファー)中、4℃で一晩、以下の抗体とともにインキュベートした:抗p−STAT3(ホスホ−STAT(シグナルトランスデューサーおよび転写のアクチベーター)3(Tyr705)(9145S;Cell Singaling Technology)、抗STAT3(sc−482;Santa Cruz)、抗GP(糖タンパク質)130(3732S;Cell Singaling Technology)、抗GSK−3β(AG751−1;Beyotime)、抗p−GSK−3β(Ser9)(9322S;Cell Singaling Technology)、抗PARP1(sc−7150;Santa Cruz)、抗TBX3(ab89220、Abcam)、抗GBX2(sc−22230;Santa Cruz)および抗β−アクチン(A1978;Sigma))。
培養細胞の全ウェルから得た全RNAを、RNeasy Plus Miniキット(QIAGEN)を使用して単離した。TransScript First−Strand cDNA Synthesis SuperMix(TransGen Biotech)を使用してRNAをcDNAに変換した。ABI Prism 7300 Sequence Detection SystemでPower SYBR(登録商標)Green PCR Master Mix(Applied Biosystems)を使用してPCRを実施した。Δ−ΔCT法を使用してデータを解析した。リアルタイムPCRに使用されたプライマーが表2に列挙されている。
細胞、胚および胚盤の総DNAを、DNeasy Blood & Tissueキット(QIAGEN)を使用して単離した。EasyTaq PCR SuperMix(TRANSGEN BIOTECH)を使用してゲノムPCRを実施した。Q−PCRによってヒト特異的ミトコンドリアDNAエレメントを検出するために、サンプルあたり 70ngの総DNAを使用した。Δ−ΔCT法を使用してデータを解析し、これらは、ヒト−マウス保存ミトコンドリアDNAエレメントの値に対して最初に正規化した。次いで、相対発現値を、注入されていない野生型マウス組織から単離された対照サンプルから作製された値に対してさらに正規化した。ゲノムPCRに使用したプライマーは、表2に列挙されている。
第1に、細胞を0.5%トリプシン−EDTAを用いて1単細胞懸濁液(1% BSA−PBS)中に脱凝集させ、次いで、各細胞を手作業で選び、0.2mlの、低張溶解バッファーを含有するPCRチューブ中に移した。第2に、単細胞cDNAを、以前に記載されたように調製した(Picelliら、Nature Protocols、第9巻:(171〜181頁(2014年)))。qPCR鋳型による必要に応じて、増幅されたcDNA産物を10倍希釈した。Bio RAD CFX Connect Real−TimeシステムでKAPA SYBR(登録商標)FAST qPCRキットを使用して定量的PCR解析を実施した。リアルタイムPCRに使用したプライマーは、表2に列挙されている。
shRNAレンチウイルスベクター(Sigma−Aldrich)を使用してPARP1ノックダウンを達成した。shRNA配列は、表2に列挙されている。それぞれこれらのベクターを用いてhEPS細胞をトランスフェクトし、解析の前に3継代の間培養した。
ガイドRNA配列を、プラスミドpx330(Addgene)中にクローニングした。gRNAを含有するPx330を、ヌクレオフェクション(4D−Nucleofector(商標)システム、Lonza)によって消化された単一mEPS細胞中に同時トランスフェクトした。シングルコロニーを選び、個別に増殖させた。DNeasy Blood & Tissueキット(QIAGEN)を使用してコロニーのゲノムDNAを抽出し、ゲノムPCRによって、これをさらに解析した。Parp1遺伝子座のエキソン1およびエキソン2が欠失したコロニーを同定した。
2種の小分子、(S)−(+)−ジメチンデンマレエート(DiM)およびミノサイクリンヒドロクロリド(MiH)は、この条件下で、マウス胚性幹(ES)細胞に形態学的に似ているドーム型hESコロニー形成を支持するとわかった。これらの小分子をさらに組み合わせ、試験した後、LCDMと名付けた新規処置組合せが確立され、これは、hLIF、CHIR99021、DiMおよびMiHを含有していた(図1A)。この処置組合せ下で、プライム型hES細胞の変換によって(示されていないデータ)、胚盤胞から直接誘導される細胞によって(示されていないデータ)、多能性因子を用いる体細胞再プログラミングによって(示されていないデータ)、マウスES細胞に形態学的に似ている細胞株を作り出すことができる。LCDMによって支持された細胞は、in−vitro分化およびin−vivo奇形腫形成の両方で3種の胚性胚葉に分化する能力を示した(示されていないデータおよび表3)。
Claims (9)
- 単離された多能性幹細胞の細胞潜在力を拡張するための細胞培養培地組成物であって、以下の群の各々
(1)サイトカイン、
(2)グリコーゲンシンターゼキナーゼ(GSK)阻害剤、
(3)Gタンパク質共役受容体(GPCR)阻害剤
(4)ポリ(ADP−リボース)ポリメラーゼ−1(PARP1)阻害剤、および
(5)任意選択で、Rho関連コイルドコイル含有タンパク質キナーゼ(ROCK)阻害剤
に由来する多能性の化学的拡張剤(CEP)を、未処置細胞の拡張された多能性幹細胞への再プログラミングを誘導するのに有効な量で含み、
前記サイトカインが、ヒト白血病抑制因子(Leukemia inhibitory fact)(LIF、「L」)、インターロイキン(IL)−6、IL−11、IL−27、IL−31、白血病抑制因子、オンコスタチンM、カルジオトロフィン−1、ニューロポイエチンおよびカルジオトロフィン様サイトカイン因子1からなる群から選択され、
前記GSK阻害剤が、CHIR99021(「C」)[6−[[2−[[4−(2,4−ジクロロフェニル)−5−(5−メチル−1H−イミダゾール−2−イル)−2−ピリミジニル]アミノ]エチル]アミノ]−3−ピリジンカルボニトリル]、BIO−アセトオキシム、GSK 3I阻害剤XV、SB−216763、CHIR99021トリヒドロクロリド、GSK−3阻害剤IX[((2Z,3E)−6’−ブロモ−3−(ヒドロキシイミノ)−[2,3’−ビインドリニリデン]−2’−オン]、GSK3IX[6−ブロモインジルビン−3’−オキシム]、GSK−3β阻害剤XII[3−[[6−(3−アミノフェニル)−7H−ピロロ[2,3−d]ピリミジン−4−イル]オキシ]フェノール]、GSK−3阻害剤XVI[6−(2−(4−(2,4−ジクロロフェニル)−5−(4−メチル−1H−イミダゾール−2−イル)−ピリミジン−2−イルアミノ)エチル−アミノ)−ニコチノニトリル]、SB−415286[3−[(3−クロロ−4−ヒドロキシフェニル)アミノ]−4−(2−ニトロフェニル)−1H−ピロール−2,5−ジオン]およびビオ[(2’Z,3’E)−6−ブロモインジルビン−3’−オキシム]からなる群から選択され、
前記GPCR阻害剤が、DiM((S)−(+)−ジメチンデンマレエート)(「D」)であり、
前記PARP1阻害剤が、MiH(ミノサイクリンヒドロクロリド)(「M」)、BSI−201(4−ヨード−3−ニトロベンズアミド)、NAM(ニコチンアミド)、PJ34(N−(6−オキソ−5,6−ジヒドロ−フェナントリジン−2−イル)−N,N−ジメチルアセトアミド、PARP阻害剤XIV、4−[(1−メチル−1H−ピロール−2−イル)メチレン]−1,3(2H,4H)−イソキノリンジオン、ベリパリブ、2−[(2R)−2−メチルピロリジン−2−イル]−1H−ベンズイミダゾール−4−カルボキサミドジヒドロクロリド、オラパリブ(Olaparidb)(1−(シクロプロピルカルボニル)−4−[5−[(3,4−ジヒドロ−4−オキソ−1−フタラジニル)メチル]−2−フルオロベンゾイル]ピペラジン)、6−アミノ−1H−ベンゾ[de]イソキノリン−1およびINH2BP(5−ヨード−6−アミノ−1,2−ベンゾピロン)からなる群から選択される、組成物。 - サイトカインが、ヒト白血病抑制因子(Leukemia inhibitory fact)(LIF、「L」)であり、
GSK阻害剤が、[6−[[2−[[4−(2,4−ジクロロフェニル)−5−(5−メチル−1H−イミダゾール−2−イル)−2−ピリミジニル]アミノ]エチル]アミノ]−3−ピリジンカルボニトリル](「C」)であり、
GPCR阻害剤が、DiM((S)−(+)−ジメチンデンマレエート)(「D」)であり、
PARP1阻害剤が、MiH(ミノサイクリンヒドロクロリド)(「M」)である、請求項1に記載の組成物。 - Lが、1〜100ng/mlの濃度範囲にあり、Cが、0.5〜5μMの濃度範囲にあり、Dが、1〜5μMの濃度範囲にあり、Mが、0.5〜5μMの濃度範囲にある、請求項2に記載の組成物。
- ROCK阻害剤が、0.2〜20μMの範囲の濃度で存在する、Y27632[(+)−(R)−トランス−4−(1−アミノエチル)−N−(4−ピリジル)シクロヘキサンカルボキサミド++ +ジヒドロクロリド)]またはフサジル(fusadil)である、請求項1に記載の組成物。
- 細胞培養培地を含み、任意選択で、エンド−IWR1(4−[(3aR,4S,7R,7aS)−1,3,3a,4,7,7a−ヘキサヒドロ−1,3−ジオキソ−4,7−メタノ−2H−イソインドール−2−イル]−N−8−キノリニル−ベンズアミド)またはXAV939(3,5,7,8−テトラヒドロ−2−[4−(トリフルオロメチル)フェニル]−4H−チオピラノ[4,3−d]ピリミジン−4−オンを含む、請求項1から4のいずれか一項に記載の組成物。
- マウス胚幹細胞、ヒト胚幹細胞および誘導多能性幹細胞からなる群から選択されるドナー細胞において拡張された多能性を誘導する方法であって、
ドナー細胞を、請求項1から5のいずれかに記載の組成物とともに、拡張された多能性を誘導するのに有効な期間培養することを含む、方法。 - ドナー細胞が、CEPを含む再プログラミング培地中で9〜20日の範囲の期間培養される、請求項6に記載の方法。
- ドナー細胞が、単細胞または小コロニーとして播種される、請求項7に記載の方法。
- ドナー細胞が、単細胞として播種され、細胞を、LCDMを含む培地における培養の前にROCK阻害剤を含む細胞培養培地中で12〜48時間の範囲の期間培養することをさらに含む、請求項8に記載の方法。
