Description
〔1〕炭素原子数が2〜18のアシル基を有するアシル中性アミノ酸及びその塩からなる群より選ばれる1又は2以上を含有する保湿剤。
〔2〕アシル中性アミノ酸は、アシルロイシン、アシルイソロイシン、アシルバリン、アシルグリシン及びアシルアラニンからなる群より選ばれる1又は2以上である、〔1〕記載の保湿剤。
〔3〕アシル基の炭素原子数は、2〜12である、〔1〕または〔2〕記載の保湿剤。
〔4〕アシル基の炭素原子数は、2〜10である、〔1〕〜〔3〕のいずれか1項記載の保湿剤。
〔5〕アシル基の炭素原子数は、8である、〔1〕〜〔4〕のいずれか1項記載の保湿剤。
〔6〕アシル中性アミノ酸は、アシルバリン、アシルグリシン、及びアシルアラニンからなる群より選ばれる1又は2以上である、〔1〕〜〔5〕のいずれか1項記載の保湿剤。
〔7〕フィラグリン、インボルクリン、トランスグルタミナーゼ1、ケラチン10、及びロリクリンからなる群より選ばれる1又は2以上の保湿関連タンパク質の産生を促進する、〔1〕〜〔6〕のいずれか1項記載の保湿剤。
〔8〕炎症性サイトカインの増殖を抑制する、〔1〕〜〔7〕のいずれか1項記載の保湿剤。
〔9〕〔1〕〜〔8〕のいずれか1項記載の保湿剤を含有する化粧料。
〔10〕〔1〕〜〔8〕のいずれか1項記載の保湿剤を含有する皮膚外用剤。
ヒト正常表皮角化細胞NHEK(NB)(クラボウ社製)をHuMedia−KG2(クラボウ社製)で前培養し、30x104細胞/mLの濃度となるよう調整し、調整後の培養液2mL(すなわち、60x104細胞/well)を6wellプレートに添加して1日培養した。サンプル(オクタノイルバリン(実施例1:C8Val)、オクタノイルアラニン(実施例2:C8Ala)、オクタノイルロイシン(実施例3:C8Leu)、オクタノイルイソロイシン(実施例4:C8Ile)、オクタノイルグリシン(実施例5:C8Gly)、グリシルプロリン(比較例1:Gly−Pro)、及びバリン(比較例2:Val))を各図に示す濃度となるように添加し、48時間反応させた。反応後、細胞より全RNAを抽出し、逆転写を行い、リアルタイムPCRにて標的遺伝子(フィラグリン、インボルクリン、卜ランスグルタミナーゼ(TG)1、ケラチン10、及びロリクリン)の増幅量を測定し(n=2)、コントロール(無添加)の増幅量と比較した。補正遺伝子としてGAPDHを用いた。計算は比較CT法を用い、プライマー及びプロ一ブはTaqMan(登録商標) Gene Expressionを用い、蛍光色素はFAMを用いた。結果を図1〜2に示す。
サンプルとして、ブタノイルバリン(実施例6:C4Val)、ヘキサノイルバリン(実施例7:C6Val)、オクタノイルバリン(実施例8:C8Val)、デカノイルバリン(実施例9:C10Val)、ドデカノイルバリン(実施例10:C12Val)及びアセチルバリン(実施例11:C2Val)を用いたこと、及び、標的遺伝子をフィラグリン、インボルクリン、及び卜ランスグルタミナーゼ(TG)1としたことのほかは、実施例1と同様に行った。結果を図3に示す。
秤量瓶に各化合物を約1g精秤し、65%RHに設定された恒温恒湿器(ナガノ科学機械製作所:LH−30−10)に入れ、7日後の重量増加率[A]を測定した。続いて、35%RHに設定された恒温恒湿器に同じサンプルを入れ、7日後の重量増加率[B]を測定した。[A]に対する[B]の割合が大きいほど、低い湿度になったときにも水分を保持できると考えられるため、この値が高い方がより高い保湿力を有する化合物であると考えられる。検討結果を表1に示す。
ヒト正常表皮角化細胞NHEK(NB)(クラボウ社製)をHuMedia−KG2(クラボウ社製)で前培養し、15x104細胞/mLの濃度となるよう調整し、調整後の培養液2mL(すなわち、30x104細胞/well)を6wellプレートに添加して1日培養した。サンプル(ラウロイルバリン(実施例14:C12Val)、デカノイルバリン(実施例15:C10Val)、オクタノイルバリン(実施例16:C8Val)、ヘキサノイルバリン(実施例17:C6Val)、ブタノイルバリン(実施例18:C4Val)、アセチルバリン(実施例19:C2Val)、ラウロイルアラニン(実施例20:C12Ala)、デカノイルアラニン(実施例21:C10Ala)、オクタノイルアラニン(実施例22:C8Ala)、ヘキサノイルアラニン(実施例23:C6Ala)、ブタノイルアラニン(実施例24:C4Ala))を各図に示す濃度となるように添加し、24時間反応させた。反応後、細胞より全RNAを抽出し、逆転写を行い、リアルタイムPCRにて標的遺伝子(フィラグリン)の増幅量を測定し(n=2)、コントロール(無添加)の増幅量を100%とし、この値と比較した。補正遺伝子としてGAPDHを用いた。計算は比較CT法を用い、プライマー及びプロ一ブはTaqMan(登録商標) Gene Expressionを用い、蛍光色素はFAMを用いた。

ヒト正常表皮角化細胞NHEK(NB)(クラボウ社製)をHuMedia−KG2(クラボウ社製)で前培養し、15x104細胞/mLの濃度となるよう調整し、調整後の培養液2mL(すなわち、30x104細胞/well)を6wellプレートに添加して1日培養した。サンプル(ラウロイルバリン(実施例25:C12Val)、デカノイルバリン(実施例26:C10Val)、オクタノイルバリン(実施例27:C8Val)、ヘキサノイルバリン(実施例28:C6Val)、ブタノイルバリン(実施例29:C4Val)、アセチルバリン(実施例30:C2Val))を各図に示す濃度となるように添加し、24時間反応させた。その後、培地を捨ててNeutral Red溶液2mL/wellに置換し、さらに2時間培養した。Neutral Red溶液はNeutral Red(クラボウ社)をHuMedia−KG2培地/生理食塩水(2:1)混合溶液で1/100に希釈したものを使用した。Neutral Red溶液で染色した細胞は、PBS(−)で一回洗浄した後、1%CaCl2/1%ホルマリン溶液1.5mL/wellで1分固定し、1%酢酸/50%エタノール溶液1.5mL/wellを添加して室温で20分抽出を行い、プレートリーダーで吸光度540/630nmを測定した。

Claims (10)
- 炭素原子数が2〜18のアシル基を有するアシル中性アミノ酸及びその塩からなる群より選ばれる1又は2以上を有効成分として含有する保湿剤であって、アシル中性アミノ酸がアシルロイシン、アシルイソロイシン、アシルバリン、アシルグリシン、及びアシルアラニンからなる群より選ばれる1又は2以上である保湿剤。
- 炭素原子数が2〜18のアシル基を有するアシル中性アミノ酸及びその塩からなる群より選ばれる1又は2以上を含有する保湿剤であって、アシル中性アミノ酸がアシルロイシン、アシルイソロイシン、及びアシルバリンからなる群より選ばれる1又は2以上である保湿剤。
- アシル基の炭素原子数は、2〜12である、請求項1又は2に記載の保湿剤。
- アシル基の炭素原子数は、2〜10である、請求項1〜3のいずれか1項に記載の保湿剤。
- 炭素原子数が2〜10のアシル基を有するアシル中性アミノ酸及びその塩からなる群より選ばれる1又は2以上を有効成分として含有する保湿剤。
- アシル基の炭素原子数は、8である、請求項1〜5のいずれか1項に記載の保湿剤。
- フィラグリン、インボルクリン、トランスグルタミナーゼ1、ケラチン10、及びロリクリンからなる群より選ばれる1又は2以上の保湿関連タンパク質の産生を促進する、請求項1〜6のいずれか1項に記載の保湿剤。
- 炎症性サイトカインの増殖を抑制する、請求項1〜7のいずれか1項に記載の保湿剤。
- 請求項1〜8のいずれか1項に記載の保湿剤を含有する化粧料。
- 請求項1〜8のいずれか1項に記載の保湿剤を含有する皮膚外用剤。
