Description
コラゲナーゼは、特に限定されるものではなく例えば、MMP(Matrix metalloproteinase)1、2、3、9、13、14等のMMPファミリー等が挙げられる。
振動処理の時間は特に限定されるものではないが、例えば10秒〜30分である。
実施例1では、本実施例にかかる培養細胞回収方法と、酵素処理も振動処理も行わない比較例にかかる培養細胞回収方法とを比較する。
実施例2では、酵素処理工程で使用する酵素溶液について検討した。
実施例3では、通常の培養期間の場合において、最も回収効率が優れるトリプシン-EDTA、コラゲナーゼ及びカゼイナーゼの混合液である酵素溶液の混合割合について検討した。
実施例4では、長期の培養期間の場合において、最も回収効率が優れるトリプシン-EDTA、コラゲナーゼ及びカゼイナーゼの混合液である酵素溶液の混合割合について検討した。
Claims (8)
- 培養容器内に封入された3次元多孔性足場にて細胞を培養する培養工程と、
前記培養容器に、トリプシン−EDTAと、コラゲナーゼと、カゼイナーゼと、を有する酵素溶液を添加して、前記培養容器内の培養細胞に酵素処理を行う酵素処理工程と、
水平面内を円運動する台座と前記酵素溶液が添加された培養容器の底面とを面接触させ、酵素処理された培養細胞に対して鉛直方向の振動を与えて振動処理を行い、培養された細胞を前記3次元多孔性足場から剥離する振動処理工程と、を有し、
前記酵素処理工程における酵素の溶液の比率は、トリプシン−EDTA:コラゲナーゼ:カゼイナーゼが0.25〜2.0mg/mL:1〜6mg/mL:3.33〜20mg/mLである、ことを特徴とする、3次元多孔性足場からの培養細胞回収方法。 - 前記培養工程における細胞の培養期間が5日〜14日の場合、前記酵素処理工程における酵素の溶液の比率は、トリプシン−EDTA:コラゲナーゼ:カゼイナーゼが0.25〜1.0mg/mL:1〜2mg/mL:3.33〜6.66mg/mLである、ことを特徴とする請求項1に記載の培養細胞回収方法。
- 前記培養工程における細胞の培養期間が15日〜30日の場合、前記酵素処理工程における酵素の溶液の比率は、トリプシン−EDTA:コラゲナーゼ:カゼイナーゼが1.0〜2.0mg/mL:1〜6mg/mL:6.66〜20mg/mLである、ことを特徴とする請求項1に記載の培養細胞回収方法。
- 前記培養容器は底部の外底面が平坦状であることを特徴とする請求項3又は4に記載の培養細胞回収方法。
- 前記台座の水平面内の円運動は周波数20〜100Hzであることを特徴とする請求項3乃至5の何れか1項に記載の培養細胞回収方法。
- 前記細胞は、ES細胞、iPS細胞又は体性幹細胞である請求項3乃至6の何れか1項に記載の培養細胞回収方法。
- 前記3次元多孔性足場は、平均間隙率80%以上且つ平均ポアサイズ10〜800μmであることを特徴とする請求項3乃至7の何れか1項に記載の培養細胞回収方法。
- 前記3次元多孔性足場の培養面積は800〜900000cm2であることを特徴とする請求項3乃至8の何れか1項に記載の培養細胞回収方法。
