rAAVウイルス粒子

rAAV粒子は、当技術分野において公知の方法を用いて生産することができる。例えば、米国特許6,566,118号;同第6,989,264号;および同第6,995,006号を参照されたい。本発明を実施する場合、rAAV粒子を生産する宿主細胞としては、哺乳動物細胞、昆虫細胞、植物細胞、微生物細胞および酵母が挙げられる。宿主細胞は、AAV repおよびcap遺伝子が宿主細胞内に安定的に維持されるパッケージング細胞、またはAAVベクターゲノムが安定的に維持されるプロデューサー細胞である場合もある。例示的パッケージングおよびプロデューサー細胞は、293、A549またはヒーラ細胞に由来する。AAVベクターは、当技術分野において公知の標準的技術を用いてを精製され、製剤化される。

MYOC突然変異体が眼、特に、IOPの一因となることがある細胞、例えば線維柱帯網細胞、の機能にどのような影響を及ぼすのかを理解することは、ミオシリン(MYOC)緑内障の病態形成を知る上での手がかりとなる。MYOC機能の理解は、ミオシリン(MYOC)緑内障のための、可能性がある治療戦略を見いだすのにも役立つだろう。本明細書に記載する結果は、MYOC突然変異体が野生型MYOC発現を低減させ、Wntシグナル伝達を遮断することを実証する。さらに、これらの結果は、R−スポンジン3(RSPO3)の発現および/またはMYOCのサイレンシングが、突然変異型MYOCの発現によって遮断されたWntシグナル伝達を回復させることができることを示唆する。

HEK293細胞(Microbix Biosystems Inc.)をDMEM、10%FCSおよび5%CO2で培養した。HEK293T(293T)細胞株をATCCから得、DMEM、10%FCSおよび5%CO2で培養した。
SV40ラージT抗原(SV40 TAg)をAAV2−SV40 T抗原ベクターでの形質導入による不死化に使用した。完全線維芽細胞増殖培地(ScienCell)中で維持した第7継代hTM細胞(ScienCell Research Laboratories、Carlsbad、CA)を10cm細胞培養プレートに播種し、1×105DRPのAAV2−SV40−Tag(「hTM−T」と標示した)またはAAV2−EGFP(陰性対照、「hTM−ENT」と標示した)のどちらか一方を24時間、形質導入した。細胞が集密に達したら、それらを2×15cmプレート(P8)で継代させた。細胞をおおよそ3〜4日ごとに繰り返し継代した。第10継代で、アリコートを採取して細胞数を判定した。hTM−T細胞からの全細胞数は、hTM−ENT細胞からの全細胞2.5×105と比較して、5.2×106であった。
hTM−T細胞と初代hTM細胞の比較は、細胞形態、集団倍加時間、およびプラスミドトランスフェクション効率の著しい差を示した。初代hTM細胞は、長い紡錘形の細胞体で、より大きく、線維芽細胞のように見えたが、不死化hTM−T細胞は、より小さく、立方体状の形状であり、比較的均一なサイズであった。hTM−T細胞株は、増殖速度の増加を示し、集団倍加が初代細胞よりおおよそ3〜4倍速く起こった。加えて、hTM−T細胞は、20細胞継代を超えて増殖し続けたが、初代hTM細胞は、第10継代までに増殖減少を示し、第12継代までに最終的な増殖停止を示した。同様の細胞密度の両方の細胞タイプに対してEGFPプラスミドおよびリポフェクタミンを使用して、トランスフェクション効率を判定した。簡単に言うと、Lipofectamine 2000(Life Technologies)をその製造業者のプロトコルに従って使用して、集密に達していないhTM−TまたはhTM細胞にEGFPプラスミドをトランスフェクトした。hTM−T細胞のほうが細胞培養面のmm2当りの細胞数は多かったが、初代hTM細胞(約5%)と比較してEGFP+ hTM−T細胞のパーセンテージ(約50%)のほうが明確に高かった。
wtMYOC、MYOC突然変異体P370LおよびY437H RSPO3ならびに/またはshRNAを発現するプラスミドを、Lipofectamine 2000(Life Technologies)を使用して293TまたはhTM−T細胞にトランスフェクトした。簡単に言うと、プロテアーゼ阻害剤カクテルを含有するRIPA緩衝液50〜100μLに細胞を溶解した。細胞溶解物10〜13μLをSDS−PAGEによって分析し、その後、iBlot高速転写システム(Life Technologies)を使用して免疫ブロットした。Tris緩衝食塩水、0.05%Tween 20(TBST)、0.2% I−Block(カゼインベースのブロッキング試薬;Life Technologies)を使用してブロットをブロックし、マウス抗ヒトMYOC抗体とともにインキュベートした。その後、ブロットを抗マウスHRP標的抗体(R&D Systems、Minneapolis、MN)とともにインキュベートした。ECL Chemiluminescent Substrate(Thermo Fisher)を使用して免疫反応性バンドを可視化し、BioMax XARフィルム(Carestream Health)を用いてKodak X−Omat 2000 Processorで現像して可視化した。
293TまたはhTM−T細胞をCostar 96ウェルホワイトまたはブラックウォールプレートに2×104細胞/ウェルで播種した。細胞播種の1〜2日後に、Fugene HDトランスフェクション試薬(Promega、Madison、WI)をその製造業者のプロトコルに従って使用してトランスフェクションを行った。
野生型MYOC(wtMYOC)は培養細胞から分泌されるが、5種の異なる突然変異型のMYOCを発現する細胞からはMYOCが殆どまたは全く分泌されない。また培養細胞への正常MYOCと突然変異型MYOCのコトランスフェクションはwtMYOC分泌を抑制することが報告されている(Jacobsonら(2001)Hum.Mol.Genet.10(2):117〜25頁)。MYOC分泌に対する突然変異型MYOC発現の効果を調査するために、野生型MYOC、P370L突然変異型MYOCまたはY437H突然変異型MYOCを発現するプラスミドを293細胞にトランスフェクトした。
MYOCは、ストレスファイバーの形成を刺激するアクチン細胞骨格の機構を修飾するWntシグナル伝達経路の成分、例えば、Frizzled(Fzd)ファミリーのWnt受容体、分泌Frizzled関連タンパク質(sFRP)ファミリーのWntアンタゴニスト、およびWnt阻害因子1(WIF−1)と相互作用すると考えられている(Kwonら(2009)Mol.Cell.Biol.29:2139〜54頁)。ストレスファイバーの形成は、線維柱帯網(TM)の収縮性およびIOP調節に肝要である。しかし、正確にどのようにMYOCがWntシグナル伝達に関連しているのか、およびこの関連性がIOPにどのような影響を及ぼすのかは不明である。細胞生物学実験に基づいて、マトリックス細胞タンパク質としてのミオシリンの役割が提案されている(ReschおよびFautsch、2009;Kochら、2014)。他のグループにより、ミオシリンはオリゴデンドロサイト分化の媒介因子であり、マウスの視神経のミエリン形成に関与することが立証されている(Kwonら、2014)。
前の実施例は、緑内障性MYOC突然変異体P370LおよびY437Hがヒト細胞におけるWntシグナル伝達を遮断するように作用することを実証した。これらのMYOC突然変異体を発現する細胞においてWntシグナル伝達を回復させることができる可能性があるメカニズムを調査するためのさらなる実験に着手した。
AAV粒子が線維柱帯網の細胞に形質導入することができるかどうかを判定するために、EGFPをコードするAAV2ベクターを、(VP1に基づいてナンバリングして)R471Aアミノ酸置換を含むAAV2粒子の野生型AAV2粒子にパッケージングした。AAV2 EGFPおよびAAV2 R471A EGFPでのhTM細胞の処置(上記)によって、ウイルス粒子をin vitroで評価した。図10(左パネル)に示すように、AAV2 R471A EGFPは、野生型AAV2と比較して高いTM細胞形質導入を示した。TM細胞形質導入をin vivoで評価するために、AAV2 EGFPおよびAAV2 R471A EGFPをマウスの眼に注射した。その後、マウスを屠殺し、EGFP発現について分析した。図10(右パネル)に示すように、AAV2 R471A EGFPは、野生型AAV2と比較して高いTM細胞形質導入をin vivoで示した。
上の実施例は、緑内障性MYOC突然変異(例えば、P370LおよびY437H)がWntシグナル伝達を遮断すること、およびWntシグナル伝達のこの阻害をR−スポンジン3(RSPO3)発現またはMYOC shRNAによって反転させることができることを実証した。いかなる特定の理論にも拘束されることを望むものではないが、MYOC突然変異(例えば、P370Lおよび/またはY437H)は、TMにおけるWntシグナル伝達に影響を及ぼすことによって、IOPを変更しPOAGに寄与する可能性があると考えた。次の実験では、AAV2ベクターによって送達されるR−スポンジン3(RSPO3)発現またはMYOC shRNAが、緑内障の症状をこの疾患のマウスモデルにおいて改善することができるかどうかを試験した。

- 哺乳動物のミオシリン(MYOC)緑内障を処置するための、RSPO1、RSPO2、RSPO3、RSPO4またはその機能的変異体をコードするベクターを含む組換えアデノ随伴ウイルス(rAAV)粒子を含む、医薬組成物であって、

ここで:
(i)組換えAAV粒子は、AAV1、AAV2、AAV3、AAV4、AAV5、AAV6、AAV6 ShH10、AAV7、AAV8、AAVrh8、AAVrh8R、AAV9、AAV10、AAVrh10、AAV11、AAV12、ウシAAV、ヤギAAVもしくはマウスAAV血清型カプシド、もしくはクレードA〜FからのAAV血清型カプシドを含む;
(ii)組換えAAV粒子は、チロシンカプシド突然変異体、ヘパリン結合カプシド突然変異体、AAV2R471Aカプシド、AAVAAV2/2−7m8カプシド、AAV DJカプシド、AAV2 N587Aカプシド、AAV2 E548Aカプシド、AAV2 N708Aカプシド、AAV V708Kカプシド、AAV1/AAV2キメラカプシド、もしくはAAV2/HBoV1カプシドを含む; および/または
(iii)組換えAAV粒子は、AAV2のVP1に基づいてナンバリングしてR484、R487、K527、K532、R585もしくはR588位のうちの1カ所またはそれ以上におけるアミノ酸置換を含むAAVカプシドを含む;
前記医薬組成物。 - 請求項1〜10のいずれか1項に記載の医薬組成物であって、
ここで:
(i)ベクターは、AAV1、AAV2、AAV3、AAV4、AAV5、AAV6、AAV7、AAV8、AAVrh8、AAVrh8R、AAV9、AAV10、AAVrh10、AAV11、AAV12、ヤギAAV、ウシAAVまたはマウスAAV血清型逆位末端反復配列(ITR)を含む; および/または
(ii)ベクターは、AAV血清型2 ITRを含む、場合によりここで:
(a)組換えAAV粒子は、同じAAV血清型に由来する1つもしくはそれ以上のITRおよびカプシドを含む;もしくは
(b)組換えAAV粒子は、組換えAAV粒子のカプシドとは異なるAAV血清型に由来する1つもしくはそれ以上のITRを含む;
前記医薬組成物。 - 請求項10に記載の医薬組成物であって、
ここで、組換えAAV粒子は、AAV血清型2カプシドを含み、そして
場合によりここで:
(i)AAV血清型2カプシドは、AAV2 VP1を基準にしてナンバリングしてR471Aアミノ酸置換を含むAAV2カプシドタンパク質を含む; および/または
(ii)ベクターはAAV2 ITRを含む;
