Description
1.培養オーランチオキトリウム(Aurantiochytrium)属藻類を培養するための培地の製造方法であって、以下の工程:
(1)細胞抽出物を強酸で処理し、これを加熱する工程;
(2)工程(1)の抽出物を中和する工程;
(3)工程(2)の抽出物を基礎として細胞培養培地を調製する工程;
を含む、当該製造方法。
2.前記細胞培養培地を用いてオーランチオキトリウム属藻類を培養すると、当該藻類が蓄積する全脂肪酸中の奇数脂肪酸含有率が増大する、項目1に記載の製造方法。
3.前記工程(3)の細胞培養培地に、工程(1)及び(2)の処理をしていない0.01〜1重量%の細胞抽出物を添加する、項目1又は2のいずれかに記載の製造方法。
4.前記工程(1)の加熱が、60〜180℃で10〜180分間行われる、項目1〜3のいずれか1項に記載の製造方法。
5.前記工程(1)の強酸処理が、塩酸、硫酸、リン酸、及び硝酸からなる群から選択される酸の添加によりなされる、項目1〜4のいずれか1項に記載の製造方法。
6.前記工程(2)の中和がアルカリの添加によりなされる、項目1〜5のいずれか1項に記載の製造方法。
7.前記工程(1)の強酸処理がプロトン型(H型)強酸性陽イオン交換樹脂の添加であり、前記工程(2)の中和が、当該強酸性陽イオン交換樹脂の除去である、項目1〜4のいずれか1項に記載の製造方法。
8.前記細胞抽出物が、培養細胞に対して十分な主要栄養素を供給することが出来る限りにおいて、原核生物、細菌、真菌、原生動物、無脊椎動物又は脊椎動物から選択される生物種の細胞に由来する、項目1〜7のいずれか1項に記載の製造方法。
9.前記細胞抽出物が魚肉抽出物又は酵母抽出物である、項目8に記載の製造方法。
10.項目1〜9のいずれか1項に記載の製造方法により製造した培地を用いてオーランチオキトリウム属藻類を培養して細胞内にトリグリセリドを蓄積させる工程を含む、オーランチオキトリウム属藻類を培養する方法。
11.前記培養したオーランチオキトリウム属藻類が蓄積する全脂肪酸中の奇数脂肪酸含有率が増大する、項目10に記載の培養方法。
12.前記培養工程を経てオーランチオキトリウム属藻類が蓄積する全脂肪酸中の奇数脂肪酸の含有率が5〜40重量%である、項目10又は11のいずれかに記載の培養方法。
13.奇数脂肪酸を主要成分として含有するトリグリセリドを製造する方法であって、以下の工程:
(1)項目10〜12のいずれか1項に記載の方法を用いて培養したオーランチオキトリウム属藻類を提供する工程;
(2)当該オーランチオキトリウム属藻類からトリグリセリド成分を抽出する工程;及び
(3)抽出したトリグリセリドを精製する工程;
を含む、当該製造方法。
14.項目1〜9のいずれか1項に記載の製造方法を用いて製造された培養オーランチオキトリウム属藻類を培養するための培地。
最終濃度2%(w/v)の魚肉抽出物(マルハニチロ株式会社より提供、鰹節製造残渣のプロテアーゼ分解産物)20g、炭素源としてグルコース20g、および海水塩10gを添加して培養液を調製し、当該培地200mLを、500mLの坂口フラスコに分注した。こうして調製した培地を参照培地としてオーランチオキトリウム属藻類の培養に用いた。
上記のように強酸処理した魚肉抽出物に0.01、0.05、0.1、0.25、0.5、1及び5%(w/v)の上記未処理酵母抽出物を添加し、上記と同様に培養液を調製して、当該培養液中で培養したSA−96株のバイオマス(培養物1Lあたりの藻類乾燥重量(g/L))及び全脂肪酸中の奇数脂肪酸の比率(%)を測定した。
Claims (13)
- 培養オーランチオキトリウム(Aurantiochytrium)属藻類を培養するための培地の製造方法であって、以下の工程:
(1)細胞抽出物を強酸で処理し、これを加熱する工程;
(2)工程(1)の抽出物を中和する工程;
(3)工程(2)の抽出物を基礎として細胞培養培地を調製する工程;
を含み、当該細胞抽出物がオーランチオキトリウム属藻類の細胞抽出物ではない、当該製造方法。 - 前記細胞培養培地を用いてオーランチオキトリウム属藻類を培養すると、当該藻類が蓄積する全脂肪酸中の奇数脂肪酸含有率が増大する、請求項1に記載の製造方法。
- 前記工程(3)の細胞培養培地に、工程(1)及び(2)の処理をしていない0.01〜1重量%の細胞抽出物を添加する、請求項1又は2のいずれかに記載の製造方法。
- 前記工程(1)の加熱が、60〜180℃で10〜180分間行われる、請求項1〜3のいずれか1項に記載の製造方法。
- 前記工程(1)の強酸処理が、塩酸、硫酸、リン酸、及び硝酸からなる群から選択される酸の添加によりなされる、請求項1〜4のいずれか1項に記載の製造方法。
- 前記工程(2)の中和がアルカリの添加によりなされる、請求項1〜5のいずれか1項に記載の製造方法。
- 前記工程(1)の強酸処理がプロトン型(H型)強酸性陽イオン交換樹脂の添加であり、前記工程(2)の中和が、当該強酸性陽イオン交換樹脂の除去である、請求項1〜4のいずれか1項に記載の製造方法。
- 前記細胞抽出物が、培養細胞に対して十分な主要栄養素を供給することが出来る限りにおいて、原核生物、細菌、真菌、原生動物、無脊椎動物又は脊椎動物から選択される生物種の細胞に由来する、請求項1〜7のいずれか1項に記載の製造方法。
- 前記細胞抽出物が魚肉抽出物又は酵母抽出物である、請求項8に記載の製造方法。
- 請求項1〜9のいずれか1項に記載の製造方法により製造した培地を用いてオーランチオキトリウム属藻類を培養して細胞内にトリグリセリドを蓄積させる工程を含む、オーランチオキトリウム属藻類を培養する方法。
- 前記培養したオーランチオキトリウム属藻類が蓄積する全脂肪酸中の奇数脂肪酸含有率が増大する、請求項10に記載の培養方法。
- 前記培養工程を経てオーランチオキトリウム属藻類が蓄積する全脂肪酸中の奇数脂肪酸の含有率が5〜40重量%である、請求項10又は11のいずれかに記載の培養方法。
- 奇数脂肪酸を主要成分として含有するトリグリセリドを製造する方法であって、以下の工程:
(1)請求項10〜12のいずれか1項に記載の方法を用いて培養したオーランチオキトリウム属藻類を提供する工程;
(2)当該オーランチオキトリウム属藻類からトリグリセリド成分を抽出する工程;及び
(3)抽出したトリグリセリドを精製する工程;
を含む、当該製造方法。
