1.1 ヒト肝臓の採取及びカニューレ挿入
肝臓移植に適していない廃棄されたヒト肝臓臓器(DHLO)を移植に対する標準的な手順に従ってヘパリン化した。0.2cm〜1.0cmの小さい血管柄のない肝臓単位(肝臓組織キューブ、LTC)の複数ブロックの細区画によって、及び/又は血管−胆管柄が与えられた単位の区域若しくは亜区域の作製により、DHLOを適切に作製した。全ヒト肝臓又は代替的には左葉(区域2−区域3−区域4±区域1)、右葉(区域5〜区域8)、又は左外側の肝臓(区域2−区域3±区域1)、また同様に肝臓組織キューブ(LTC)を−80℃で少なくとも24時間凍結して細胞溶解を促進した。
最初に、全ヒト肝葉を4℃にてPBS中で一晩解凍した。次に、大静脈又は肝静脈にカニューレ挿入を行い、逆行性灌流系を開始した。
LTCを37℃で1時間〜1.5時間解凍した。LTCの脱細胞化に関するプロトコルを表3〜表5に示す。
生得の肝臓組織及び脱細胞化されたLTC(dLTC)を4サイクルの脱細胞化の後に組織学的に比較した。脱細胞化サイクルは、LTCから細胞及び細胞形質成分を除去したのに対し、コラーゲンを保存することが分かった(図3)。
Claims (14)
- ヒト肝臓スキャフォールドを製造する方法であって、
(i)全肝臓又はその機能的単位を含む健康な又は病的なヒト肝臓組織中の細胞を機械的に傷害することと、
(ii)前記肝臓組織を低張性試薬又は高張性試薬に暴露することによって前記組織中の細胞を浸透圧ストレスに供することと、
(iii)任意に、前記組織をプロテアーゼ及び/又はDNAaseに暴露することと、
(iv)前記組織を洗浄剤に暴露することと、
(v)工程(ii)、工程(iii)及び工程(iv)の各々を任意の順序で1回又は複数回繰り返すことと、
を含み、それによりヒト肝臓スキャフォールドを製造し、
前記ヒト肝臓組織を工程(ii)〜(v)の間に、工程(ii)〜(iv)の2以上の繰返しに亘って最大値まで次第に増加した後に維持する速度で前記ヒト肝臓組織を逆行性に灌流することによって発生させられるフローせん断応力に供する、
方法。 - (i)前記組織を1巡又は複数巡回(round)の凍結及び解凍に供することによって、又は(ii)前記組織をHIFU又は超音波処理に供することによって、前記細胞を機械的に傷害する、請求項1に記載の方法。
- 前記低張性剤が脱イオン水である、及び/又は前記高張性剤が水又は生理食塩水である、請求項1又は2に記載の方法。
- 前記組織を工程(iii)においてプロテアーゼ、任意にトリプシン又はプロナーゼに暴露する、請求項1〜3のいずれか一項に記載の方法。
- 前記洗浄剤がアニオン性洗浄剤、任意にドデシルリン酸ナトリウム(SDS)又はデオキシコール酸ナトリウム(SdC)である、請求項1〜4のいずれか一項に記載の方法。
- 前記洗浄剤がイオン性洗浄剤、任意にポリエチレングリコールp−(1,1,3,3−テトラメチルブチル)−フェニルエーテル(Triton X100(商標))である、請求項1〜5のいずれか一項に記載の方法。
- 初期灌流速度が0.1ml/分/組織グラム〜1.99ml/分/組織グラムである、及び/又は前記標的灌流速度が2ml/分/組織グラム〜20ml/分/組織グラムである、請求項1〜6のいずれか一項に記載の方法。
- 工程(v)が、
a)前記肝臓組織を通る灌流によって次の順序:(ii)、(iii)、(ii)、(iii)、(iv)、[(ii)、(iv)]n、(ii)、(iii)、[(ii)、(iv)]n(ここで、nは1〜25である)で工程(ii)〜工程(iv)を繰り返すこと、又は
b)前記肝臓組織を通る灌流によって工程(ii)及び工程(iv)を1回〜25回繰り返すこと、
を含む、請求項1〜7のいずれか一項に記載の方法。 - 前記肝臓組織を表1に示される灌流レジメンに供する、請求項1〜8のいずれか一項に記載の方法。
- 前記スキャフォールドを脱細胞化の後に滅菌することを含む、請求項1〜9のいずれか一項に記載の方法。
- 前記スキャフォールドを細胞により再増殖させて人工肝臓組織を製造することを含み、任意に、前記細胞が、ヒト初代肝臓細胞及び細胞株肝臓細胞、ヒト初代肝細胞、内皮細胞、ヒトiPSC又は患者特異的iPSC由来細胞、ヒト胚性幹細胞(hESC)、ヒト間葉系幹細胞(hMSC)、ヒト胎生幹細胞、ヒト癌細胞、及びヒト内皮前駆細胞(EPC)である、請求項1〜10のいずれか一項に記載の方法。
- 請求項1〜10のいずれか一項に記載の方法によって製造されたヒト肝臓スキャフォールド、又は請求項11に記載の方法によって製造された人工肝臓組織。
- ヒト肝臓スキャフォールドをそれを必要とする個体に移植することを含む肝疾患又は肝機能障害の治療方法における使用のための、
請求項1〜10のいずれか一項に記載の方法によって製造されたヒト肝臓スキャフォールド。 - ヒト人工肝臓組織をそれを必要とする個体に移植すること、又は
人工ヒト肝臓組織を、それを必要とする個体の血管系に、前記個体からの循環血が前記組織を通過して前記個体へと戻るように、前期組織が個体に肝臓機能を提供するように、体外で接続すること
を含む肝疾患又は肝機能障害の治療方法における使用のための、
請求項11に記載の方法によって製造されたヒト人工肝臓組織。
