Description
リン脂質型オメガ3の原料としては、クリルオイルやイクラ等の魚卵、水産廃棄物がある。特に、南極オキアミを原料としたクリルオイルが、販売されている。
南極オキアミ由来(クリルオイル)のリン脂質型オメガ3脂肪酸の組成物は、リン脂質濃度が約40%(W/W組成物)であり、高濃度のトリグリセリド(約45%)および遊離脂肪酸(約15%)を含んでいる(特許文献1)。また魚介類、特にその加工過程から出る廃棄物由来の有用リン脂質の製造方法が報告されている(特許文献2)。
しかし、水産物を原料とした場合、海洋汚染による重金属・ヒ素等の問題が懸念される。さらに、季節変動により漁獲高・リン脂質含量が不安定となることが懸念されている。そのため、季節変動を受けにくく、高品質でかつ安定的な生産が可能な代替原料が求められている。
さらに、クリルオイルや魚介類・廃棄物由来の脂質は、リン脂質型EPA・DHAが主成分であるため、リン脂質型αリノレン酸が主成分となる原料がより好ましい。
トリグリセリド型のαリノレン酸を豊富に含むアマニ油から酵素でエステル交換反応し、リン脂質型αリノレン酸を調製しようとする例(非特許文献3)がある。しかし、酵素等を用いるため、煩雑な操作が必要であり、コストも高くなる可能性がある。大豆・卵黄を原料として、一般的にはレシチンがあるが、リノール酸が主要脂肪酸であり、αリノレン酸は10%以下であり、充分な含量ではない。
乳系の原料として、牛乳から精密濾過膜を用いてリン脂質含量を高める方法(特許文献3)があるが、リン脂質型αリノレン酸を高含有化することに着目したものではない。また、酵母を原料としては、代表的なパン酵母またはビール廃酵母からヘキサンにてリノール酸などの油脂を抽出する方法(特許文献4)が知られている。しかし、パン酵母はαリノレン酸を蓄積できないことが知られており(非特許文献4)、さらにヘキサンではリン脂質が抽出されにくい。よって、これまでリン脂質型αリノレン酸を高含有できる組成物は、知られていなかった。
(1) 酵母エキス抽出後の酵母残渣から抽出されたリン脂質型αリノレン酸を含有する機能性食品、
(2) 酵母エキス抽出後の酵母残渣から抽出されたリン脂質型αリノレン酸を含有する医薬品、
(3) 酵母エキス抽出後の酵母残渣から抽出されたリン脂質型αリノレン酸を抽出する製造方法、
であって、この有用リン脂質組成物が、リン脂質含量30%質量%以上を含むことを特徴とする。上記構成によれば、ホスファチジルコリンPCを豊富に含有する組成物が得られる。
(1) 酵母から抽出したリン脂質型αリノレン酸を含む組成物であって、
一般式(1)の化合物を含み、前記リン脂質型αリノレン酸組成物中における一般式(1)の化合物含量が30質量%以上である組成物。
R2は、水素または任意の脂肪酸から選択され、
R1又はR2うち、少なくとも1つは、脂肪酸であり、
Xは、コリン残基(PC)、エタノールアミン残基(PE)、イノシトール残基(PI)、又はセリン残基(PS)から1つが選択される)
(2) 一般式(1)の前記化合物のうちコリン残基を有するホスファチジルコリン(PC)が、50質量%(PC/全リン脂質)以上である(1)に記載の組成物、
(3) R1およびR2は、それぞれ独立したαリノレン酸である、(1)に記載の組成物、
(4) R1が、αリノレン酸である、(1)に記載の組成物、
(5) R2が、αリノレン酸である(1)に記載の組成物、
(6) R1とR2がαリノレン酸である(1)に記載の組成物、
(7) リン脂質型αリノレン酸組成物中の脂肪酸組成であるαリノレン酸が総脂肪酸のうち30%以上を含む、(1)に記載の組成物、
(8) (1)〜(7)のいずれかに記載の脂質型αリノレン酸組成物を含有する機能性食品、
(9) (1)〜(7)のいずれかに記載の脂質型αリノレン酸組成物を含有する医薬品、
を提供するものである。
トルラ酵母の酵母エキスは、調味料として好適であることから大量に生産されており、本発明はその残渣を有効利用できるため、コスト・廃棄物削減の点でも、極めて有利な方法である。
本発明における好ましい原料としては、トルラ酵母と称されるCandida utilisであるが、目的の成分を含有するものであれば特に限定されず、例えば、真菌類としては、キャンディダ属、クルイベロマイセス属、チゴサッカロマイセス属の酵母が挙げられる。酵母の培養方法は、特に限定されない。酵母の培養形式はバッチ培養、あるいは連続培養のいずれかが用いられる。培地には一般的に炭素源として、ブドウ糖、酢酸、エタノール、グリセロール、糖蜜、亜硫酸パルプ廃液等が用いられ、窒素源としては、尿素、アンモニア、硫酸アンモニウム、塩化アンモニウム、硝酸塩などが使用される。リン酸、カリウム、マグネシウム源も過リン酸石灰、リン酸アンモニウム、塩化カリウム、水酸化カリウム、硫酸マグネシウム、塩化マグネシウム等の通常の工業用原料でよく、その他亜鉛、マンガン、鉄イオン等の無機塩を添加する。その他、ビタミン、アミノ酸、核酸関連物質等を添加しても良い。カゼイン、酵母エキス、肉エキス、ペプトン等の有機物を添加しても良い。
抽出方法は、エタノールを加えて十分攪拌後、適当な温度および時間で反応を行う。温度は特に限定はないが、通常20℃〜80℃未満であり、好ましくは50℃〜70℃である。抽出時間は、特に限定はないが、通常は1時間以上、好ましくは3〜9時間である。抽出方法は、攪拌、還流、浸漬、振盪、超音波抽出が挙げられる。
抽出前に酵素処理する場合は、抽出時間の短縮が可能となる。酵素としては、細胞壁溶解酵素であるストレプトマイセス属由来のβグルカナーゼ「デナチームGEL」(ナガセケムテックス社製)、Taloromyces属由来のβグルカナーゼ「Filtrase BRX」(DSMジャパン社製)、天野エンザイム社製「ツニカーゼFN」がある。好ましくはツニカーゼFNである。超臨界抽出する場合は、酵母懸濁液に超臨界状態の二酸化炭素を接触させた後、減圧して大気圧に戻すことにより酵母懸濁液から二酸化炭素を分離する方法である。
(酵母の培養)
キャンディダ・ウチリスCS7529株(FERMP−3340)を予めYPD培地(酵母エキス1%、ポリペプトン2%、グルコース2%)を含む三角フラスコで種母培養し、これを30L容発酵槽に18L培地に1〜2%植菌した。培地組成は、グルコース4%、燐酸一アンモニウム0.3%、硫酸アンモニウム0.161%、塩化カリウム0.137%、硫酸マグネシウム0.08%、硫酸銅1.6ppm、硫酸鉄14ppm、硫酸マンガン16ppm、硫酸亜鉛14ppmを用いた。培養条件は、pH4.0、培養温度30℃、通気量1vvm、撹拌600rpmで行った。pHのコントロールは、アンモニアで行った。16時間菌体培養した後、培養液を回収し、遠心分離により集菌して、180gの湿潤酵母菌体を得た。
(酵母エキスの抽出)
菌体培養後の湿潤酵母菌体を蒸留水に懸濁して遠心分離を繰り返すことで洗浄した。洗浄後は湿潤菌体を蒸留水に再度懸濁する、又は、凍結乾燥、若しくは熱風乾燥させた乾燥菌体を蒸留水に懸濁する。2N NaOHでpH13に調整後、70℃で20分間攪拌し、酵母エキスを抽出した。酵母エキス抽出後の菌体残渣をドラムドライヤーで乾燥し、乾燥物100gを原料として使用した。乾燥物中のホスファチジルコリン含量は2.8%である。
上記原料100gに対して95%エタノールで60℃、9時間抽出した。その抽出液に水をエタノール濃度70%となるように添加し、5℃達温後に1時間保持した後、ろ紙(ADVANTEC社、NO.5C)で濾過し不溶物を取り除いた。さらに、ろ過液をロータリーエバポレーターにて減圧濃縮乾固し、組成物5.5gを得た。乾固物の残留エタノール濃度は2.5%であった。組成物のリン脂質含量は71.7%、ホスファチジルコリン含量48%となった。リン脂質組成物の脂肪酸組成として、αリノレン酸38%、リノール酸29%、オレイン酸15%、パルミトレイン酸4.2%となった。
上記原料100gに対して95%エタノールで60℃、9時間抽出した。その抽出液(ホスファチジルコリン含量は40%以上)に水をエタノール濃度70%となるように添加し、5℃達温後に1時間保持した後、ろ紙(ADVANTEC社、NO.5C)で濾過し不溶物を取り除いた。さらに、ろ過液をロータリーエバポレーターにて濃縮乾固した。その乾固物5.5gをエタノールで溶解した後、シリカゲルカラム(容量100ml、FL60BD、富士シリシア化学社)にアプライし、95%エタノールを通液した。ホスファチジルコリンが含まれる画分を取り分けて、再度濃縮し、組成物3gを得た。組成物のリン脂質含量は95%、ホスファチジルコリン含量88.3%となった。リン脂質組成物の脂肪酸組成として、αリノレン酸46%、リノール酸26%、オレイン酸11%、パルミトレイン酸7.7%となった。
高脂肪食(脂質20%)に試料油脂を2%添加した飼料をマウス(C57BL/6Jマウス、3週齢オスを2週間予備飼育)に実施例2で取得した本願組成物とアマニ油(製品名「アマニ油、和光純薬社製」)、クリルオイル(製品名「コーヨークリルオイル」甲陽ケミカル社製)を投与し、肝機能及び脂質代謝改善の動物試験を行った。7週経過後にマウスを解剖し、血漿脂質に対する効果を評価した。7週間の経過中の体重の推移も測定した。体重の推移は、図3に示す。また、40日後の体重比較を図4に示す。本願組成物は、40日後の体重においては、他の組成物よりも体重の減少傾向が見られた。血液の生化学分析結果、GOT(AST)値、GPT(ALT)値については、本願組成物(5)、アマニ油(6)、クリルオイル(7)は、コントロール(4)と比較し同等(図5)の低下がみられた。(図5、図6) さらに、本願組成物は、肝臓中のトリグリセリド濃度がクリルオイルと近い低値を示した(図7)。

- トルラ酵母由来のリン脂質型αリノレン酸含有組成物であって、
一般式(1)の化合物を含み、前記リン脂質型αリノレン酸組成物中における一般式(1)の化合物含量が30質量%以上である組成物。
R2は、水素または任意の脂肪酸から選択され、
R1又はR2うち、少なくとも1つは、脂肪酸であり、
Xは、コリン残基(PC)、エタノールアミン残基(PE)、イノシトール残基(PI)、又はセリン残基(PS)から1つが選択される) - 一般式(1)の前記化合物のうちコリン残基を有するホスファチジルコリン(PC)が、50質量%(PC/全リン脂質)以上である請求項1に記載の組成物。
