Description
本開示は、ミトコンドリアに薬剤を輸送するように構成されたナノ粒子、ならびに診断的および治療的使用を含む、その使用方法に関する。

本開示は、とりわけ、ミトコンドリアに薬剤を輸送するように構成されたナノ粒子について記載する。細胞質または細胞の他の細胞小器官とは対照的に、ミトコンドリアに薬剤を輸送することにより、ミトコンドリア中の薬剤の相対濃度が増大して、他の部位における効果が低下する一方、ミトコンドリアに対する薬剤の効果が増強する可能性がある。したがって、副作用または毒性が低下する一方、効果が増強する可能性がある。ミトコンドリアに送達される薬剤は、治療薬、診断薬等であってもよい。
概略図面では、尺度は必ずしも同じではない。図中で使用される同様の番号は、同様の構成要素、工程等を指す。しかしながら、所与の図で構成要素を指すための番号の使用は、同じ番号で標識した別の図の構成要素を限定することを意図するものではないことは、理解されよう。加えて、構成要素を指す異なる番号の使用は、異なる番号付けがなされた構成要素が同じかまたは同様でありえないことを示すことを意図するものではない。
ナノ粒子のコアは、任意の適切な1つまたは複数の成分から形成することができる。好ましくは、コアは、疎水性ポリマーまたはポリマーの疎水性部分などの疎水性成分から形成される。コアは、加えてまたは代替的に、疎水性コアおよび親水性外表面を有する粒子に水性環境中で自己集合しうる、疎水性部分および親水性部分を有するブロックコポリマーを含むことができる。実施形態では、コアは1つまたは複数の生分解性ポリマーまたは生分解性部分を有するポリマーを含む。
本明細書に記載するナノ粒子は、場合によっては、親水性コアを包囲する親水性層を含むことができる。親水性層は、ナノ粒子が免疫系による認識を回避するのを助けることができ、かつナノ粒子の血液循環半減期を増大させることができる。
本明細書に記載するナノ粒子は、ミトコンドリアにナノ粒子を標的化する1つまたは複数の部分を含む。本明細書で使用する場合、ミトコンドリアへのナノ粒子の「標的化」は、ナノ粒子が実質的に同様の非標的化ナノ粒子よりも高濃度で、他の細胞小器官または細胞質に比較して、ミトコンドリアに蓄積することを意味する。実質的に同様の非標的化ナノ粒子は、標的化ナノ粒子と実質的に同じ相対濃度(例えば、約5%以内)で同じ成分を含むが、標的化部分を欠いている。
本明細書に記載するナノ粒子は、任意の1つまたは複数の治療薬を含むことができる。治療薬は、ナノ粒子のコア中に埋め込まれることも、コア内に含有されることもある。好ましくは、治療薬は所望の速度でコアから放出される。コアがポリマー(PLGAなど)または公知の放出速度を有するポリマーの組合せから形成される場合、放出速度は容易に制御することができる。
本明細書に記載するナノ粒子は、イメージング、視覚化、または診断を目的とする1つまたは複数の造影剤を含むことができる。実施形態では、治療用ナノ粒子がミトコンドリアに適切に輸送されていることを確認するためにイメージングを実施する。任意の適切な造影剤を使用することができる。実施形態では、造影剤は、in vivo磁気共鳴イメージング(MRI)に適しており、例えば、酸化鉄(IO)ナノ結晶またはガドリニウム錯体がある。実施形態では、造影剤は、ex vivo/in vivo光学イメージングに適しており、例えば、蛍光色素の量子ドット(QD)(蛍光)、cdot、pdot等がある。実施形態では、ナノ粒子は、MRI用造影剤および蛍光光学イメージング用薬剤の両方を含む。
本明細書に記載するナノ粒子は、任意の適切なプロセスにより合成または構築することができる。好ましくは、ナノ粒子はプロセスの変動を最小化するために単一工程で構築される。単一工程プロセスは、ナノ沈殿および自己集合を含むことができる。
一般に、本明細書に記載するナノ粒子は、ミトコンドリアに、特に特定のミトコンドリアに標的化することができる。ナノ粒子は、ミトコンドリアの機能障害に関連した疾患を視覚化、イメージング、モニタリング、診断、または治療するために使用することができる。
A.PLGA−b−PEG−TPPの合成
PLGA−b−PEG−TPPの合成。HO−PEG−OH(0.75g;0.23mmol)、PLGA−COOH(0.50g;0.1mmol)、および4−ジメチルアミノピリジン(0.01g;0.08mmol)を、7mLの乾燥CH2Cl2に溶解した。N,N’−ジシクロヘキシルカルボジイミド(DCC)(0.02g;0.1mmol)のCH2Cl2溶液2mLを、0℃で撹拌しながら反応混合物に滴下して加えた。この混合物を室温に加温し、一晩撹拌した。不溶性のジシクロヘキシル尿素を濾過し、混合物を50mLの50:50ジエチルエーテルおよびメタノールから沈殿させた。得られた固体を4℃で15分間、1,400×gで遠心分離した。最終的な精製として、PLGA−b−PEG−OHのメタノール溶液を繰り返し沈殿させ、冷ジエチルエーテルで洗浄し、白色固体として30%(0.2g)の収率で単離した。1H−NMR(CHCl3−d):δ5.3[m,(OCHCH3C(O)],4.9[m,(OCH2C(O))],3.6[s,(OCH2)],1.9[m,(CH3CH)]。13C−NMR(CHCl3−d):δ169.6,166.5,66.0,61.1,60.9,16.89,15.46。ゲル透過クロマトグラフィー:Mn=6,900g/mol、MW=9,200g/mol、Mz=12,300g/mol、PDI=1.33。PLGA−b−PEG−OH(0.29g;0.03mmol)。(5−カルボキシペンチル)トリフェニルホスホニウムカチオン(31)(0.11g;0.29mmol)および4−ジメチルアミノピリジン(0.007g;0.06mmol)を3mLの乾燥CH2Cl2に溶解した。DCC(0.06g;0.29mmol)をCH2Cl2(1mL)に溶解し、0℃で撹拌しながら反応混合物に滴下して加えた。この混合物を室温で一晩攪拌し、その後、形成したいずれのジシクロヘキシル尿素も濾去した。次いで、得られた混合物に50mLの冷ジエチルエーテルを加え、ポリマーを沈殿させた。固体を4℃で15分間、1,400×gで遠心分離した。溶媒を除去し、固体を凍結乾燥した。ポリマーを99%(0.3g)の収率で白色固体として単離した。1H−NMR(CHCl3−d):δ7.9−7.6[m,15H(Ar)],5.3[m,(OCHCH3C(O)],4.9[m,(OCH2C(O))],3.6[s,(OCH2)],1.9[m,(CH3CH)]。13C−NMR(CHCl3−d):δ166.5,135.3,133.9,130.7,66.0,61.0,16.89,15.46。IR:νcm−1 3,029(C−H sp2),2,944(C−H sp3),2,200−2,000(C−H Ar),1,741(C=O)。31P−NMR(CHCl3−d):δ24.37。
PLGA−COOH(0.4g;80μmol)、1−エチル−3−(3−ジメチルアミノプロピル)カルボジイミド(EDC)(12.4mg;80μmol)、およびNHS(9mg;80μmol)を、3mLのジメチルホルムアミド(DMF)に溶解し、8μM QD−PEG−NH2の250μL溶液を加えた。この混合物を室温で24時間撹拌し、次いで100kDaカットオフのAmiconフィルター(Millipore)で濾過した。得られた固体をH2Oに溶解し、一晩凍結乾燥し、次いでDMF中に再懸濁した。動的光散乱(DLS)を使用して、PLGA−b−PEG−QDを特性評価し、10.1±0.1nmの流体力学的径および0.3のPDIを得た。
ナノ粒子は、Marrache and Dhar(2012),Proc.Natl.Acad.Sci.USA 109:16288−16293;Kolishetti et al.(2010)Proc.Natl.Acad.Sci.USA 107:17939−17944;およびDhar et al.(2011)Proc.Natl.Acad.Sci.USA 108:1850−1855に全般的に記載されているナノ沈殿法により合成した。手短に言えば、アセトニトリル中のPLGA−b−PEG−OHまたはPLGA−b−PEG−TPPを、最終ポリマー濃度5mg/mLに、一定に攪拌しながらナノ純水に滴下して加えた。次いで、ナノ粒子を2時間撹拌した。有機溶媒を3回の洗浄により除去し、100kDaカットオフのAmiconフィルター(Millipore)を通して濾過した。ナノ粒子をナノ純水に再懸濁し、次の使用まで4℃に保存した。ナノ粒子のサイズ、PDI、およびゼータ電位を決定するためにDLS測定を行った。サイズが調整可能なPLGA−b−PEG−TPPナノ粒子を、ナノ沈殿法に従い、所定の比のPLGA−COOH(0、10%、20%、35%、50%、70%、および90%)とPLGA−b−PEG−TPPとをブレンドすることにより合成した。PLGA−b−PEG−TPPナノ粒子の表面電荷は、前述のナノ沈殿法に従い、所定の比のPLGA−b−PEG−TPP(0、15%、35%、50%、65%、80%、90%、および100%)とPLGA−b−PEG−OHとをブレンドすることにより変動させた。QD−ブレンドナノ粒子は、このナノ沈殿法に従い、最終ポリマー濃度5mg/mLで、DMF:アセトニトリル(1:9比)中のPLGA−b−PEG−OHまたはPLGA−b−PEG−TPPとPLGA−b−PEG−QD(10mmolのDMF溶液)とを使用して合成した。サイズ、PDI、およびゼータ電位を決定するためにDLS測定を行った。ナノ粒子はすべてTEMを使用して特性評価した。治療薬負荷ナノ粒子の合成の場合は、PLGA−b−PEG−OHまたはPLGA−b−PEG−TPP(DMF中50mg/mL)を、所定量の治療薬(DMF中10mg/mL)と混合し、最終ポリマー濃度5mg/mLにアセトニトリルで希釈した。この混合物を、ナノ沈殿法に従い、一定に攪拌しながら、ナノ純水に滴下して加えた。サイズ、PDI、およびゼータ電位を決定するためにDLS測定を行った。ポリマーのコアを溶解し、ナノ粒子中の治療薬の量を定量するためにHPLCを使用することにより、薬物負荷および封入効率を決定した。
種々のサイズおよびゼータ電位のQD−ブレンドナノ粒子(10μM)を、HeLa細胞(1.5×107細胞)に12時間内部移行させた。内部移行させた後、哺乳動物細胞用のミトコンドリア単離キットを使用してミトコンドリアおよびサイトゾルを単離した。次いで、サイトゾル画分およびミトコンドリア画分について、ICP−MSによりQD中のCd濃度を分析した。単離したミトコンドリア画分およびサイトゾル画分についてビシンコニン酸(BCA)アッセイを行い、単離したタンパク質1マイクログラム当たりのCd量を算出した。
脂肪生成を誘導するために、3T3−L1前脂肪細胞を1ウェル当たり5,000細胞の密度で96ウェルプレート上に播種し、コンフルエンスになるまで増殖させた。2,4−DNPを負荷した標的化および非標的化ナノ粒子ならびに遊離2,4−DNPの抗脂肪生成特性を、脂肪生成アッセイの1日目にナノ粒子を内部移行させることにより評価した。また、1日目に、0.5mmol 3−イソブチル−1−メチルキサンチン(IBMX)、0.5μMデキサメタゾン、および20nMインスリンを添加した、10%FBS含有分化DMEM培地中で細胞を分化誘導した。3日目に、誘導培地を、DMEM、10%FBSおよび20nMインスリンを含有するインスリン培地と交換した。処理の6日後に、製造業者の説明書に従い、AdipoRed(Lonza)アッセイを使用して、細胞についてトリグリセリド蓄積を分析した。
スプラーグドーリーラットにPLGA−b−PEG−TPP−QDナノ粒子を静脈内投与した。血漿中のCdレベルの経時変動、臓器分布、および排泄特性を決定した。対照ラットには生理食塩水を注射した。
データはすべて、平均±SDとして表した。標的化、非標的化、および遊離薬物の間の差は、一元配置分散分析により評価した。P<0.05を、統計的有意性を示すとみなした。

ミトコンドリア取り込みに対するPLGAベースのナノ粒子のサイズ、形状、および表面電荷の重要性の理解が進むと、ミトコンドリアの機能障害に関連した疾患のための標的化送達ビヒクルを開発する機会が生み出される可能性がある。本発明者らは、得られるナノ粒子のサイズおよび表面電荷が変動するように、ミトコンドリア標的化の機能を有するポリマーであるPLGA−b−PEG−TPPとPLGA−COOHまたはPLGA−b−PEG−OHとをブレンドして、ミトコンドリア取り込みに対するこれらの特性の効果を試験し、最適な製剤を決定した。ナノ粒子の特性が変動する可能性は、合成に必要とされる処理工程の数と共に増大する。本発明者らは、ナノ粒子合成の前にTPP標的化部分をポリマー中に組み込むことがそのような変動を最小化するであろうと予想した。ミトコンドリア内部に治療薬を高効率に送達するために、本発明者らは、TPPのコンジュゲーションを可能にする末端OH基(PLGA−b−PEG−OH)を有する生分解性ポリマーを合成して、PLGA−b−PEG−TPPを得た(図1)。標的化リガンドのコンジュゲーションは、ゼータ電位、サイズ、細胞取り込み、および細胞内輸送を含む、ナノ粒子の基本特性に影響を及ぼした。本発明者らは、上記に記載したナノ沈殿法を使用して、ブレンドナノ粒子を合成し(図2)、4回以上の独立したナノ沈殿実験で、再現可能な直径および表面電荷を有する標的化および非標的化ナノ粒子を得た。
本発明者らは、ミトコンドリア取り込みに対するサイズおよび表面電荷の効果に取り組むために、代表的かつ包括的な試験に着手した。本発明者らは、サイズおよび表面電荷は異なるが、他の特性はすべて同一かまたは実質的に同一であるナノ粒子を作製することが重要であると考えた。本発明者らは、種々の量のPLGA−COOHとPLGA−b−PEG−TPPとをブレンドして、種々のサイズおよび一定の表面電荷を有するナノ粒子のライブラリーを構築した。ブレンド技術を使用して、本発明者らは、一定の表面電荷により証明されるように(図3)、TPP部分の数を一定に維持しながら、ナノ粒子サイズを約80〜約410nmに調整することができた(図3)。ミトコンドリア取り込みに対する表面電荷の効果を調べるために、本発明者らは、所定量のPLGA−b−PEG−OHとPLGA−b−PEG−TPPとをブレンドした。主要なスキャフォールドおよびナノ粒子サイズを変化させることなく、表面電荷を変動させることに成功した(図4)。これらのナノ粒子ライブラリーは、同様の形状の均質集団を示し(TEM画像;図5A〜F)、ミトコンドリア取り込みに対するナノ粒子のサイズおよび電荷の効果を調べることを可能にした。
ナノ粒子系の細胞取り込みプロファイルは、系の効率およびバイオアベイラビリティを反映する。細胞取り込みの定量的評価と共に、細胞内局在は、目下のナノ粒子プラットフォームの有効性の評価にとって重要である。本発明者らは、ヒト子宮頸癌HeLa細胞中の標的化および非標的化ナノ粒子の分布を検討するために、強固な蛍光性レポーター量子ドット(QD)を使用した。本発明者らは、QD−コンジュゲートのアミン末端PEGであるNH2−PEG−QDを使用して、細胞内区画中のナノ粒子を追跡した。PLGA−COOHをNH2−PEG−QDにコンジュゲートして、トリブロックコポリマーであるPLGA−b−PEG−QDを得た(図1)。本発明者らは、PLGA−b−PEG−QDとPLGA−b−PEG−TPP、ならびにPLGA−b−PEG−QDとPLGA−b−PEG−OHとをブレンドすることより、それぞれ標的化ならびに非標的化ナノ粒子の内部移行をモニターした。処理した細胞の共焦点顕微鏡分析により、非標的化ナノ粒子よりも、標的化ナノ粒子の細胞ミトコンドリアへの取り込みが顕著に多いことが示された(図7A〜B)。蛍光強度を比較すると、非標的化ナノ粒子に比較して、正に荷電した標的化ナノ粒子の全体的な取り込みが顕著に多いことが示された。ImageJ「共局在ファインダー」プラグインを使用する定量分析により、細胞ミトコンドリア中での標的化ナノ粒子とMitoTracker Green(Invitrogen)との有意な共局在(ピアソンの相関係数、ρ=0.53)が明らかになった。
ナノ粒子の取り込みおよび細胞内輸送は、いくつかの競合する経路に沿って起こる。ミトコンドリアを標的化するナノ粒子の使用は、多くの場合、ナノ粒子がエンドソーム経路に取り込まれるという事実、およびエンドソームがミトコンドリア輸送に対して障害として働くという事実により限定される。本発明者らは、標的化ナノ粒子の効率的なエンドソーム回避に必要な融合特性を検討するために、初期エンドソームマーカーのEAA−1を使用して、時間依存的な取り込み試験を行った。標的化ナノ粒子については、エンドソームとの共局在が最初の1時間に観察され、2時間までに減少した。時間経過と共に、標的化ナノ粒子は完全なエンドソーム回避を示し、細胞のミトコンドリアに局在した(図7A〜B);しかしながら、非標的化ナノ粒子では、初期エンドソームとの顕著な共局在が4時間後でも観察された。標的化ナノ粒子の高度に効率的なエンドソーム回避は、これらのナノ粒子を「プロトンスポンジ」として作用させうる高い緩衝能に帰することができる。正に荷電したPEGの緩衝作用によるプロトンの吸収は、エンドソーム小胞の酸性化を防止し、それによって、プロトンおよび対イオンのATPアーゼ媒介流入が増大するはずであり、次には、浸透圧膨潤、エンドソーム膜の破裂、およびナノ粒子のサイトゾルへの最終的な漏出がもたらされ、ミトコンドリア取り込みにナノ粒子が利用可能になる。
種々の正に荷電したナノ粒子は、血清タンパク質との相互作用を示し、負に荷電したタンパク質の表面吸着に起因する凝集が誘発される。血清タンパク質は、DMEM中の10%(容量/容量)FBSまたはH2O中の10%(容量/容量)FBSとの7日間インキュベーションにおいて、本発明者らの標的化ナノ粒子のサイズおよび多分散性指数(PDI)に対して効果がなかった。この発見は、標的化ナノ粒子のゼータ電位が、DMEM中の10%FBSまたはH2O中の10%FBSとの7日間の接触の後に、最小限変化したのみであり、粒子の目に見える凝集もないという観察によってさらに支持される。これらの発見は、血清中の標的化ナノ粒子の優れた安定性が、それらをin vivo用途に適したものにすることを示唆する。
ミトコンドリア膜を横断するために、ナノ粒子は、一般移入細孔を通って外膜を横切り輸送される。本発明者らは、管状クリステ、小胞状クリステ、および板状クリステの複雑な構造、ならびにミトコンドリア内膜へのそれらの弱い接続により、ナノ粒子の易動度が制約され、それらの拡散が極めて複雑でサイズ依存的なプロセスになる可能性があることを予測した。内膜電位(ΔΨm)は、内部が負であり、正に荷電した化学種に対して電気泳動効果を及ぼすことにより、移入において主要な機能を果たす。高い正電荷を有するナノ粒子は、低い正電荷を有するナノ粒子よりも、低いΔΨmで移入されると期待される;したがって、HeLaモデル細胞株を使用して、本発明者らは、ミトコンドリア内膜の横断における、ナノ粒子のサイズおよび電荷の効果を比較評価した(図9A〜D)。本発明者らは、サイズは異なるがゼータ電位は同様の標的化PLGA−b−PEG−TPP/PLGA−b−PEG−QDブレンドナノ粒子を用いてHeLa細胞を処理し、次いで誘導結合プラズマ質量分析(ICP−MS)を使用して定量的検討を行い、細胞により内部移行されたQDに由来するカドミウム(Cd)量を評価した。直径80〜330nmのナノ粒子のミトコンドリア取り込みを評価すると、直径80〜100nmの粒子が最大の取り込みを示す傾向があることがわかった。ナノ粒子サイズに対するサイトゾル画分およびミトコンドリア画分中のナノ粒子数を示すヒストグラムにより、ナノ粒子の細胞取り込みは、粒子サイズに極度に依存することがわかる(図9A〜B)。本発明者らは、表面電荷は異なるが流体力学的径は同様のナノ粒子ライブラリーを使用して、細胞およびミトコンドリアの取り込みに対するナノ粒子表面電荷の効果を検討し、負に荷電したナノ粒子のミトコンドリア取り込みを検出せず(図9C)、また全体的な細胞取り込み(図9D)もほとんど検出しなかった。細胞取り込みは、表面電荷が1.3mVに達すると増大し、表面電荷が約22mVまで一定のままであったが、ミトコンドリア取り込みは増大した。本発明者らは、表面電荷が約34mVに増大し、飽和に達するときに、細胞取り込みのもう一つのジャンプを見出した。ミトコンドリア取り込みは、より正に荷電したナノ粒子によって顕著に増大した。ナノ粒子の直径および表面電荷の効果に関するこの系統的検討は、ミトコンドリア輸送のために最適化されたナノ粒子プラットフォームの設計に有用になりうる。
この系の多用途性の概念実証として、本発明者らは、神経変性、肥満症、および癌の管理のためのミトコンドリア作用治療薬の送達を検討した。本発明者らは、ナノ沈殿法を使用して、高い負荷および封入効率で治療薬を負荷した標的化および非標的化ナノ粒子を合成した。
アミロイド様斑により、ADの神経病理学が定義される。ベータアミロイド(Aβ)ペプチドの凝集がアミロイド様病変を形成するため、Aβ形成の防止によるアミロイド蓄積の低減は、ADに対する治療薬蓄積を改善するための魅力的なアプローチである。クルクミンは、Aβおよびそれに関連したミトコンドリア酸化ストレスを阻害することが知られている;しかしながら、その低いバイオアベイラビリティおよび光分解が主要な問題となっている。そのような問題を念頭において、本発明者らは、光安定性を提供し、かつミトコンドリア取り込みを増強するために、標的化クルクミン負荷ナノ粒子を製剤化した。MTT[3−(4,5−ジメチルチアゾール−2−イル)−2,5−ジフェニルテトラゾリウムブロマイド]アッセイを使用して、20μM Aβで処理されたヒト神経芽細胞腫IMR−32細胞のin vitro生存を評価すると、非標的化クルクミンナノ粒子または遊離クルクミンと比較して、標的化クルクミンナノ粒子によるAβに対する神経保護が増強していることが実証され(図10)、これにより、細胞ミトコンドリアへのクルクミンの標的化された送達が説明される。
ミトコンドリアが、癌治療の具体的な標的として浮上した。本発明者らの系が癌に適用可能であることを実証するために、ミトコンドリア作用LNDおよびα−TOSを選択した。癌細胞に対するLNDおよびα−TOSの選択性および効率は、細胞ミトコンドリアを標的化するそれらの能力に依存する。LNDおよびα−TOSの送達における、本発明者らの標的化ナノ粒子の有効性を評価するために、HeLa細胞のMTTアッセイを行なった。標的化LNDナノ粒子のIC50値は、非標的化ナノ粒子の約1/5、遊離形態のLNDの1/108であり(図11A)、細胞毒性効果がより高いことが示された。より高い細胞毒性効果はまた、標的化α−TOSナノ粒子に関して、非標的化α−TOSナノ粒子または遊離α−TOSと比較して観察された。(IC50はそれぞれ、75±2nM、230±4nM、および381±5nM)(図11B)。標的細胞小器官への優先的な局在化により、標的化ナノ粒子中に封入されたLNDおよびα−TOSの両方の細胞毒性が説明される。空のPLGA−b−PEG−TPPナノ粒子が高濃度においても細胞毒性効果がないという本発明者らの発見により、いかなる細胞毒性に対しても送達系または高いゼータ電位の寄与は除外される(図11C)。
肥満症は、II型糖尿病、心臓血管疾患、および特定の癌タイプを含む、種々の代謝障害と関連するために世界的な健康問題になっている。現在入手可能な抗肥満症薬に付随する、限定的な有効性および望ましくない副作用のために、エネルギー代謝を調節するために細胞内部位に薬物を直接送達することができる伝達ビヒクルの開発に注目が集まっている。ヒト肥満症の治療として、ミトコンドリア脱共役剤2,4−DNPが著しい成功を収めたことにより、エネルギー消費に対する脱共役の有益な効果が、熱量摂取量の代償的な増大により圧倒されないことが実証される。しかしながら、2,4−DNPの狭い治療濃度域により、その使用が中止された。最近の研究では、TPPに共有結合的に連結された2,4−DNPが、脱共役に効果がないことが見出されている。共有結合で連結した脱共役剤の狭い治療濃度域および失敗に関する事態が、細胞ミトコンドリアにこの脱共役剤を指向させることについてのミトコンドリア標的化ナノ粒子の評価に結びついた。標的化ナノ粒子中への2,4−DNPの封入が、細胞毒性を及ぼすには不十分な低濃度で、3T3−L1前脂肪細胞の分化誘導を抑制することができるか否かを調べるために、本発明者らは、1μM、4μM、25μM、および100μMの2,4−DNPナノ粒子(図12A〜Dおよび13)に、7日間の分化期間の間、3T3−L1細胞を継続的に曝露し、細胞生存率および細胞内脂質蓄積を評価した。非標的化2,4−DNPナノ粒子および遊離2,4−DNPを対照として使用した。分化期間の間、7日間の1μM、4μM、または100μMの標的化2,4−DNPの存在では、細胞生存率に影響はなかったが、非標的化2,4−DNPナノ粒子または遊離2,4−DNPと比較して、脂質蓄積の顕著な低下がもたらされた(図12A〜Dおよび13)。遊離2,4−DNPは、100μM濃度で細胞毒性を示した。これらの結果から、標的化ナノ粒子中の2,4−DNPは、低濃度(1または4μM)で、3T3−L1細胞の脂肪細胞分化を抑制することができ、かつこれらの細胞に対して細胞毒性効果を及ぼさないことがわかる。本発明者らは、標的化ポリマーナノ粒子系を使用して、細胞ミトコンドリアに2,4−DNPを送達し、肥満症の管理に適用可能な遊離形態と比較して低濃度で、脂質蓄積を低下させることができることを実証した。
静脈内投与後の、雄性スプラーグドーリーラットにおける標的化PLGA−b−PEG−TPP−QDナノ粒子(NP)の生体内分布および薬物動態および排泄を評価した。血漿中のCdレベルの経時変化、臓器分布、および排泄特性を、図14に示す。これらの標的化NPの血漿中でのNP滞留が延長していることが観察された。試験した臓器の中で、最大のCdは肝臓で観察された。これらの高度に正に荷電したNPは、迅速な肝胆汁排泄を示す。これらのNPは、おそらく高い正電荷のために、肝臓内に通常隔離されたままである負に荷電したPLGA−b−PEG−COOH−NPと比較して、肝臓から胃腸管の中に迅速に排泄される。
この概念実証研究において、本発明者らは、適切に操作されたミトコンドリア標的化生分解性PLGAベースナノ粒子送達系を、ナノ粒子の表面電荷およびサイズを微調整することにより高効率で細胞ミトコンドリアに入るように作製することができることを実証した。すべてのタイプのナノ粒子がミトコンドリアに入る能力を有しているとは限らない。その理由は、表面電荷およびサイズのプログラミングに制限があるため、ナノ粒子が複雑な二重膜を横断することができないためである。以前の報告では、ミトコンドリアへの効率的な移入に対する、ナノ粒子のサイズおよび電荷の関係を検討していない。PLGAベースのナノ粒子の細胞内輸送に対する表面電荷および直径の効果に取り組む、この合理的な試験により、ミトコンドリア送達に適用するための生分解性ナノ担体の設計への一般化されたアプローチが提供される。これらの標的化ナノ粒子は、AD、肥満症、および癌を含む、種々のミトコンドリア機能障害に関連した障害に使用することができる。この研究では、いくつかの例外的に有望な研究方向が強調されるが、イメージング用に統合することができる、PLGAベースのナノ粒子の多様な適用のためのプラットフォームが提供される。
Claims (32)

ここで、PLGA-b-PEG-TPP分子のPLGA-COOHポリマー分子に対する重量比は、1:1以上であり、
