Description
本出願は、2012年12月12日に出願された米国特許仮出願第61/736,067号の優先権を主張する、2013年3月14日に出願された米国特許出願第13/803,878号の一部継続出願であり、これらの各々はあらゆる目的のためにその全体が本明細書中に組み込まれている。
本発明は、国立保健研究施設(NIH)及び国立癌研究所(NCI)により授与された、グラント番号2R01CA115483−06、並びにVA Career Development Award−2の下で米国政府の支援により行われた。米国政府は、本発明に一定の権利を有する。
様々な種類の癌の治療のためのいくつかの有効な化学療法薬は、非常に水に溶けにくく、望ましくない副作用を誘発するような製剤化を必要としている。近年、アブラキサン(登録商標)(パクリタキセル−内包型アルブミンナノ粒子)、ドキシル(登録商標)(ドキソルビシン−内包型リポソーム)などのナノ療法製剤が、これらの薬物の臨床毒性プロファイルは改善するが、それらの抗腫瘍作用は、当初の薬物製剤をわずかに上回るのみであることが示されている。これは一部、ナノ療法製剤の比較的大きいサイズ(一般に>100nm)に起因し、このことは薬物が腫瘍塊に透過することができる程度を制限する。場合によっては、この大きいサイズはまた、ナノ治療薬が肝臓及び細網内皮系(RES)に捕獲されることを引き起こす。従ってインビボにおける抗癌剤の効果的送達のために、より小さい(20〜80nm)ステルス性及び生体適合性のナノ担体を開発することが必要とされている。

I. 定義
本明細書において使用される用語「デンドリマー」及び「樹状分岐高分子」とは、焦点、複数の分岐したモノマー単位、及び複数の末端基を含む、分岐したポリマーをいう。モノマーは、一緒に連結され、焦点から伸び、且つ末端基で終着する腕(又は「デンドロン」)を形成する。デンドリマーの焦点は、本発明の化合物の他のセグメントへ結合されることができ、並びにこれらの末端基は、追加の化学部分により更に官能基化されてよい。
本発明は、親水性ポリ(エチレングリコール)(PEG)セグメント及び疎水性セグメント、並びに少なくとも1個のポルフィリンを有する、両親媒性テロデンドリマーコンジュゲートを提供する。PEGセグメントは、1個又は複数のPEG鎖を含む、分岐した又は線状アーキテクチャーを有することができる。テロデンドリマーの疎水性セグメントは、疎水面及び親水面を有するコール酸により提供され得る。ポルフィリン、コール酸及びPEGは、様々な酸反復単位を含むことができるオリゴマー及び/又はポリマーにより結合される。典型的には、オリゴマー及びポリマーは、ジアミノカルボン酸、リジンを含む。テロデンドリマーは、溶液中で凝集し、疎水性内側及び親水性外側を持つミセルを形成することができる。これらのミセルは、水への溶解度が低い薬物又は他の物質を送達するためのナノ担体として使用することができる。

本発明のテロデンドリマーは、それらの焦点で共に連結されている、2つの分岐したセグメントを含む。一般にテロデンドリマーは、先に記載されたか又は既報(WO 2010/039496)の任意のテロデンドリマー及びオリゴマー焦点に結合された2個以上のPEG鎖を含む分岐したPEGセグメントを含む。
本発明のテロデンドリマーは、凝集し、疎水性コア及び親水性外側を持つナノ担体を形成する。一部の実施態様において、本発明は、内側及び外側を有するナノ担体、複数の本発明のデンドリマーコンジュゲートを含むナノ担体を提供し、ここで各化合物は、水性溶媒中で自己組織化しナノ担体を形成し、その結果疎水性ポケットが、ナノ担体の内側に形成され、且つここで各化合物のPEGは、ナノ担体の外側上に自己組織化する。
本発明のナノ担体は、ナノ担体の内側に疎水性薬物を封鎖することによるか、又は薬物のナノ担体のコンジュゲートへの共有的結合によるなど、薬物の投与を必要とする任意の疾患を治療するために使用することができる。ナノ担体はまた、ナノ担体の内側に造影剤を封鎖することによるか、又は造影剤のナノ担体のコンジュゲートへ結合することにより、造影のために使用することができる。
本発明のナノ担体は、当業者に公知の様々な異なる様式で製剤化することができる。医薬として許容し得る担体は、投与される特定の組成物により、更には組成物を投与するために使用される特定の方法により、一部決定される。従って、本発明の医薬組成物の多種多様な好適な製剤が存在する(例えば「レミントン薬科学(Remington 's Pharmaceutical Sciences)」、第20版、2003年、前掲参照)。有効な製剤は、経口及び経鼻製剤、非経口的投与のための製剤、及び徐放性を持つよう製剤化された組成物を含む。
本発明のナノ担体は、必要に応じ頻繁に投与することができ、これは毎時、毎日、毎週又は毎月を含む。本発明の医薬方法において利用される化合物は、初回量約0.0001mg/kg〜約1000mg/kgを毎日で投与される。1日投与量の範囲約0.01mg/kg〜約500mg/kg、又は約0.1mg/kg〜約200mg/kg、又は約1mg/kg〜約100mg/kg、又は約10mg/kg〜約50mg/kgを使用することができる。しかしこれらの用量は、患者の必要性、治療される病態の重症度、及び利用される化合物に応じ、変動することができる。例えば用量は、特定の患者において診断された疾患の型及び病期を考慮し、経験的に決定することができる。本発明の状況において、患者へ投与される投与量は、ある期間にわたり患者において有益な治療反応を作用するのに十分でなければならない。投与量のサイズはまた、特定の患者における特定の化合物の投与に付随する何らかの有害副作用の存在、性質、及び程度により決定されるであろう。特定の状況に関する適量の決定は、熟練開業医の技術の範囲内である。一般に治療は、本化合物の最適量よりも少ない比較的小用量で開始される。その後この用量は、状況下で最適効果に達するまで、少しずつ増加される。便宜上、総1日用量を、望ましいならば、分割し、その日のうちに部分毎に投与することができる。投与量は、治療担当医の決定により、毎日、又は隔日で与えることができる。投与量はまた、皮下カプセル剤、サシェ剤若しくはデポ剤の使用により、又は貼付剤若しくはポンプを介してなどで、長期間にわたり(数週、数ヶ月又は数年)、規則的又は連続して与えることができる。
一部の実施態様において、本発明は、本発明のナノ担体の有効量を、造影される対象へ投与することを含む、造影方法を提供し、ここで該ナノ担体は、造影剤を含む。他の実施態様において、治療方法及び造影方法は、薬物及び造影剤の両方を有するナノ担体を同時に使用し達成される。
材料
モノメチル−末端ポリ(エチレングリコール)モノアミン(MeO−PEG−NH2、Mw:5000Da)及びα−アミノ−ω−Boc−アミノポリ(エチレングリコール)(Boc−NH−PEG−NH2、Mw:5000Da)は、Rapp Polymere社(独国)から購入した。ピロフェオホルビド−aは、Santa Cruz Biotechnology社(サンタクルズ)から入手した。塩酸ドキソルビシン(DOX・HC1)(Novaplus)は、UC Davis Cancer Center Pharmacyから入手した。(Fmoc)lys(Boc)−OH、(Fmoc)Lys(Dde)−OH、(Fmoc)Lys(Fmoc)−OH、(Fmoc)Cys(Trt)−OH及び(Fmoc)Ebes−OHは、AnaSpec社(サンノゼ、CA)から入手した。3,3’−ジヘキサデシルオキサカルボシアニン過塩素酸塩(DiO)及び4,6−ジアミジノ−2−フェニルインドール(DAPI、青色)は、Invitrogen社から購入した。コール酸、MTT[3−(4,5−ジメチルジアゾール−2−イル)−2,5−ジフェニルテトラゾリウムブロミド]、エルマン試薬[DTNB、5,59−ジチオビス(2−ニトロ安息香酸)]及び他の全ての化学物質は、Sigma−Aldrich社(セントルイス)から購入した。
テロデンドリマーは、既報のように(Bioconjug Chem 21, 1216−1224 (2010);Biomaterials 30, 6006−6016 (2009))、ビルディングブロックとしてMeo−PEG−NH2、Boc−NH−PEG−NH2、リジン、コール酸及びピロフェオホルビド−aを使用する、液相縮合反応により合成した。
金属性ナノポルフィリンの調製。キレートされた金属を伴うテロデンドリマーを生成するために、既報の方法6,49に従い、過剰な遊離金属イオン(例えば10倍過剰)を、テロデンドリマーと一緒に、メタノール/クロロホルム中で、1〜5時間、窒素下で、インキュベーションした。遊離金属は、分子量カットオフ値3,500のカラム濾過により除去した。その後この金属ポルフィリン−脂質を、アリコートとし、乾燥させ、アルゴン下、−20℃で貯蔵した。
金属性NPsを、音波処理により、PBS中に金属−テロデンドリマーを溶解することにより、作製した。チオール遊離ポルフィリン−テロデンドリマー20mgを、リン酸緩衝食塩水(PBS)1mL中に溶解し、引き続き10分間音波処理し、非架橋のナノポルフィリンを形成した。標的化ミセルを作製するために、PLZ4−コンジュゲートされたテロデンドリマー(PLZ4−PEG5k−CA8)を、PEG5k−Por4−CA4と混合した。自己組織化後、より親水性の標的化PLZ4リガンドが、ミセルの表面上に提示された。
NPsの合成及び特徴決定。NPsは、線状ポリエチレングリコール(PEG)並びにピロフェオホルビド−a(Por、ポルフィリン類似体)の樹状オリゴマー及びコール酸(CA)で構成された、新規クラスのハイブリッド両親媒性ポリマー(テロデンドリマーと称す)の自己組織化により形成した(図12〜図15)。代表的テロデンドリマーであるPEG5k−Por4−CA4を、図12aに示す。透過型電子顕微鏡(TEM)は、このNPsが、サイズ20nmの球形であることを示した(図12c)。このNPsは、2つの吸収ピークを有し、一つは405nmで、及び一つは近赤外(NIR)範囲680nmであった(図12d)。PEG5k−Por4−CA4は、銅(Cu(II))、パラジウム(Pd(II))、ガドリニウム(Gd(III))及びガリウム(Ga(III))などの様々な金属イオンをキレートする固有の能力を有する。PBS中のこれらの金属−テロデンドリマーの自己組織化により作製される、金属イオン−内包型NPsは、吸光度ピーク(650〜690nm)に独自のシフトを示すことがわかった(図12d)。金属イオンが、NPsのポルフィリン成分に内包された場合、これらの重原子は、電子を大きい角度で散乱させ、このことはより大きい位相シフトにつながり、TEMのコントラストを増大する。従って金属イオン層は、TEM下で、染色なしでも、観察することができる(図19)。リンタングステン酸による染色及びTEMによる観察後、金属層が、NPsの疎水性コアと親水性コロナの境界面に認められた(図12e、f)。
薬物内包されたミセルの調製
ドキソルビシンを、先に説明した溶媒蒸発法により、ミセルへ内包させた。DOXの高分子ミセルへの封入前に、DOX・HClを、クロロホルム(CHCl3)/メタノール(MeOH)(1:1、v/v)中のトリエチルアミン3モル当量と共に一晩攪拌し、DOX・HClからHClを除去した。テロデンドリマー20mgを、異なる量の中和したDOXと一緒に、最初にCHCl3/MeOHに溶解し、混合し、回転蒸発器上で蒸発させ、均質な乾燥ポリマーフィルムを得た。このフィルムを、リン酸緩衝溶液(PBS)1mL中に再構成し、引き続き30分間音波処理し、試料フィルムを、ミセル溶液に分散させた。それらのインビボでの運命を追跡するために、疎水性NIRF色素DiDを、同様の方法を用い、一部のミセルに封入した。最後に、これらのミセル製剤を、0.22μmフィルターにより濾過し、試料を滅菌した。DOX量を決定するために、DOX−内包型ミセルを、DMSO(ミセル溶液/DMSO:1:9、v/v)に希釈し、ミセルナノ粒子を解離し、蛍光を、NanoDrop 2000分光計(Thermo Scientific社)により測定し、ここで様々な濃度の一連のDOX/DMSO標準溶液を使用し、較正曲線を得た。DOX内包型ジスルフィド架橋されたミセルは、同じ方法により調製し、その後、過酸化水素によりチオールを酸化し、ミセル内ジスルフィド結合を形成した。
本発明者らは更に、PEG5k−Por4−CA4によって形成されたNPsは、様々な水に溶けにくい化学療法薬物及び分子標的化された薬物を効率的に運搬することができることを明らかにした。例えば、化学療法薬(ドキソルビシン、パクリタキセル及びビンクリスチン)並びに分子標的化された薬物、例えばプロテアソーム阻害薬(ボルテゾミブ)、チロシンキナーゼ阻害薬(ソラフェニブ)及び熱ショックタンパク質90(Hsp90)阻害薬(17−アリルアミノ−17−デメトキシゲルダナマイシン、17AAG)などは、NPsへ容易に取り込まれ、水への溶解度の有意な増加を生じる。薬物−内包型NPsの最終粒子サイズは、全ておよそ12〜20nmであり、これは空のNPsのサイズに類似していた(図25)。
方法
インビトロにおける細胞毒性及び機序。SKOV−3卵巣癌細胞株及びPC3前立腺癌細胞株を使用し、NPsの光増感機能を評価した。細胞を、NPsにより24又は72時間処理し、引き続き処理後24時間の時点で光照射した。細胞生存度を、WST−8キットを使用し決定した。熱及びROS生成を、照射時点で調べた。
本発明者らは、NPsの独自のアーキテクチャー−依存性ROS生成(図12i)は、光増感剤それ自身の光毒性を軽減するために使用することができることを認めた。光照射がないと、無傷のNPsは、SKOV−3卵巣癌細胞に対し1.0mg/mLまで観察可能な細胞毒性を示さないのに対し、遊離Porは、投与量−依存様式で有意な細胞毒性を示した(図29)。光曝露後のNPsのインビトロ抗腫瘍作用は、光−線量及びNP−投与量に依存し(図30a、b)、且つ同じ線量の照射時に、細胞毒性は、5−アミノレブリン酸(図30−ALA、PDTに関するFDA承認薬)よりもはるかに強力であった(図30b)。細胞生存度の喪失は、細胞内ROS生成に関連し(図30c)、ミトコンドリア膜電位の喪失、細胞損傷、及びカスパーゼ3/7活性化により証明される細胞アポトーシスを生じた(図30d−f)。更に本発明者らは、NP−DOX媒介性の化学療法と光線療法の併用は、NPs媒介した光線療法単独、遊離DOX、又はNM−DOX(ポルフィリンを伴わないDOX内包型標準ミセル)11よりも、同じ光曝露後に有意により効果的であることを発見した(図30g)。本発明者らは更に、NP−媒介型光線療法は、Hsp90阻害薬17AAGなどの分子標的化された薬物と、相乗的に組み合わせることができることを明らかにした。強力な相乗作用が、アンドロゲン依存性PC3前立腺癌細胞(図30h、下側)及びアンドロゲン依存性LNCAP前立腺癌細胞(図31、左下側)の両方で、17AAG内包型NPs(NP−AAG)により認められた。不死化された正常前立腺細胞RWPE1は、この療法に対し比較的抵抗性であった(図31、右側)。NP−AAGは、アネキシンV及びPI染色により測定した場合(図30i)、PC3細胞におけるアポトーシスの誘導のより大きい作用を有する。興味深いことに、HIF1α、Akt、サバイビン及びMMP2は、PDT上でNPsにより増大し、この誘導並びにSrcは、NP−AAGにより枯渇され(図30j)、これはNP−AAGの強力な相乗作用の機序を暗示している。
SKOV−3ヒト卵巣癌細胞株及び5637ヒト膀胱癌細胞株を、ATCCから購入し、推奨培地において維持した。イヌの膀胱癌細胞株K9TCC−Pu−Inは、2009年7月に、Dr. Deborah Knapp(Purdue大学)により最初に開発され、博士から直接提供された。これらの細胞株は、Dr. Knapp研究室において、2009年に、形態学、免疫組織化学、遺伝子発現及び腫瘍原性アッセイを使用し、試験され、及び正当性が認証された。これらの細胞は、細胞株の特徴決定を行ったDr. Knappから直接得られ、且つ蘇生(resuscitation)後6ヶ月未満使用者の研究室において継代したので、再認証は不要であった。
癌細胞を、8ウェル組織培養チャンバースライド(BD Biosciences社、ベッドフォード、MA、米国)において播種し、引き続き10%FBSを含有する細胞培養培地において24時間インキュベーションした。培地を交換し、ナノ−ポルフィリンミセルを、各ウェルへ添加した。予め決定した時点で、細胞をPBSにより3回洗浄し、4%パラホルムアルデヒドで固定し、細胞核をDAPIにより染色した。スライドにカバースリップを載せ、且つDelta Vision画像処理システム(AppliedPrecision社、CA)により、製造業者のプロトコールに従い、観察した。
K9TCC−Pu−In及び5637膀胱癌細胞株並びにSKOV−3卵巣癌細胞株を使用し、ナノ−ポルフィリンの光増感機能を評価した。本発明者らは、癌細胞を、2.2μMナノ−ポルフィリンにより2時間処理した。完全に洗浄した後、細胞を赤色光(650nm)に曝した。細胞生存度を、WST−8増殖キット(Caymen社)を用い、照射後24時間決定した。従来の光線力学診断/治療薬である5−アミノレブリン酸(5−ALA)を、対照として使用した。別の実験において、DOX内包型ナノポルフィリンを使用し、これらの癌細胞を処置し、引き続き光曝露した。暗毒性及びテロデンドリマー関連毒性を評価するために、これらの細胞はまた、テロデンドリマー及び空の架橋されたミセルにより、異なる希釈で処理し、且つ光曝露せずに合計72時間インキュベーションした。
6〜8週齢の雌の無胸腺ヌードマウス(Nu/Nu系統)を、Harlan社(リバーモア、CA)から購入した。全ての動物を、AAALAC指針に従い、無病原体−条件下で維持し、実験前に少なくとも4日間馴化させた。全ての動物実験は、施設の指針を順守し、並びにカリフォルニア大学デービス校の動物の使用と飼養に関する管理諮問委員会(Animal Use and Care Administrative Advisory Committee)により承認されたプロトコール第07−13119号及び第09−15584号に従い、実施した。卵巣癌の皮下異種移植片モデルを、PBS及びマトリゲル(1:1v/v)の混合液100μL中のSKOV−3卵巣細胞7×106個を、雌ヌードマウスの右脇腹に皮下注射することにより、樹立した。
直径およそ8〜10mmの皮下腫瘍を持つ、K9TCC−Pu−In及び5637膀胱癌並びにSKOV−3卵巣癌の異種移植片マウスモデルに、インビボNIRF光学造影を施した。ナノ−ポルフィリンミセルの注射後異なる時点で、Kodakマルチモーダル造影システムIS2000MMを、励起帯域通過フィルター625nm及び発光700nmで用い、マウスを走査した。マウスは、各造影前に、ペントバルビタール(60mg/kg)の腹腔内注射により麻酔をかけた。インビボ造影後、動物を注射の24、48及び72時間後にCO2過剰投与量により安楽死させた。腫瘍及び主要臓器を摘出し、Kodak画像処理ステーションにより画像処理した。
方法
動物モデル。6〜8週齢の雌の無胸腺ヌードマウス(Nu/Nu系統)を、Harlan社(リバーモア、CA)から購入した。これらのマウスは、4〜40月齢で、乳癌を自然発症することがわかっている。全ての動物を、AAALAC指針に従い、無病原体−条件下で飼育し、実験前に少なくとも4日間馴化させた。全ての動物実験は、施設の指針を順守し、並びにカリフォルニア大学デービス校の動物の使用と飼養に関する管理諮問委員会により承認されたプロトコール第07−13119号及び第09−15584号に従い、実施した。卵巣癌の皮下異種移植片モデルを、PBS及びマトリゲル(1:1v/v)の混合液100μL中のSKOV−3卵巣細胞又はA549肺癌細胞2×106個を、雌ヌードマウスの右脇腹に皮下注射することにより、樹立した。乳癌のトランスジェニックマウス(FVB/n Tg(MMTV−PyVmT))は、Jackson Laboratory社に発注した。
NPs及びジスルフィド架橋されたNPsの粒子サイズは、およそ20〜30nmであり(図12c、図16b)、これは、腫瘍を標的化し透過するのに最適な範囲である11,14,20。それらの可逆的に架橋する性質及び独特なアーキテクチャー−依存性蛍光特性を基に、架橋されたNPsは、血液中のバックグラウンド抑制及び腫瘍部位での優先的蓄積及びシグナル増幅により、改善された癌検出に関するNIRFI感度を増大するための、活性化可能な光学ナノプローブとして使用するのに特に適している。架橋されたNPsは、血液循環中で最小バックグラウンド蛍光シグナルを伴い、サイレント(又は「オフ」状態)で存在することができる(図38a、下側パネル)。血液中の架橋されたNPsの消光状態は更に、SDS及びGSHの添加による、蛍光の回復により明らかにされた(図39)。予想されたように、SKOV−3卵巣癌マウスモデルにおいて、全身内の全体的蛍光シグナルは、静脈内注射後、架橋されたNPsについて、非架橋のNPsよりも、劇的に低下した(図38a)。サイズにより媒介された増強された透過性亢進及び残存(EPR)効果による腫瘍部位での蓄積時に、これらの架橋されたナノプローブは、腫瘍部位での又は癌細胞内の、内在性還元剤GSHによるジスルフィド結合の切断により、「オン」に変化し、その後ミセル解離及び蛍光シグナル増幅が続く。架橋したNPsは、注射後24時間で、非架橋のNPsよりも、有意に高い腫瘍蓄積を示した(図38a)。注射後24時間でのエクスビボ造影は、非架橋NPs及び架橋されたNPsの両方が、正常臓器と比べ勝った腫瘍における蛍光シグナルを有することを示した(図38b)。腫瘍部位での架橋されたNPsの平均蛍光は、同じ群のマウスの筋肉における値よりも15倍高く、且つ腫瘍部位の非架橋のNPsの値よりも3倍高かった(図38c)。架橋されたNPsと非架橋のNPsの間の示差的蛍光シグナルは、循環内の架橋されたNPsの構築体の安定性、従ってより高い腫瘍への取り込みにより説明することができる。
方法
インビボ治療試験。乳癌のトランスジェニックマウス(FVB/n Tg(MMTV−PyVmT)及びSKOV−3卵巣癌異種移植片を保有するヌードマウスを、インビボ治療試験のために使用した。トランスジェニックマウスにおいて腫瘍容積が4〜5mmに到達した時点で、架橋されたNPsを、2.5mg/kg DOXを伴う又は伴わずに、尾静脈を介して毎週1回、3投与量を注射した。24時間後、全身麻酔下で、腫瘍を、ダイオードレーザーシステム(Applied Optronics社、ニューポート、CT)により690nmで照射した。光線量は、全腫瘍をおおう0.8cm2ビームスポットを生じる光ファイバーを通じて、1.25W/cm2で2分間であった。腫瘍温度を、赤外カメラ(Flir)により記録した。照射後の腫瘍内のROS生成を、NPs及びPBSで処理した腫瘍に由来する組織溶解液100μLと混合した2’,7’−ジクロロフルオレシンジアセタート(DCF)を用いて測定した。場合によっては、マウスの血液を採取し、照射後の血液中のROS及び熱発生を、予め決められた時点で測定した。引き続き溶血した。腫瘍容積を、週2回測定し、一旦腫瘍サイズが1000mm3に到達したならば、マウスを屠殺した。腫瘍を、照射後24時間の組織病理学的評価のために摘出した。SKOV−3卵巣癌異種移植片モデルは、100万個の細胞を、下方脇腹部位に皮下に注射することにより、樹立した。腫瘍が150〜200mm3に到達した後、マウスに、PBS、2.5mg/kg DOX、並びに2.5mg/kg DOX内包型又は非内包型架橋されたNPsを、3日毎に、3投与量与えた。レーザー光0.2W/cm2を、投薬の1及び3回目後に2分間あてた。腫瘍サイズ及び体重を、週2回測定し、同時に毒性の潜在的徴候について毎日マウスをモニタリングした。最終投薬の2日後、CBC及び血清化学試験を行った。また各群から1匹のマウスを屠殺し、組織病理学的評価のために、主要臓器を摘出した。
PDT及びPTTなどの光−ベースの技術は、癌治療の有望な機会をもたらす27-36。光線療法の臨床使用は、以下の点により妨げられている:1)光増感剤は、腫瘍と正常組織の間の選択性が悪く、その結果限定された有効性及び正常組織への光毒性をもたらす37-41;2)光線療法が媒介した酸化的ストレスは、腫瘍微小環境において、サバイビン、Akt、低酸素症誘導因子1α(HIF−1α)、マトリクスメタロプロテイナーゼ−2(MMP−2)及び血管内皮増殖因子(VEGF)などの、生存促進性及び血管新生性シグナル伝達分子の発現を誘導し、このことは治療効果に負の影響を及ぼし且つ腫瘍再発に繋がり得る27,42,43。ミセル及びリポソームなどのナノ担体は、光線療法に関して、腫瘍への光増感剤の標的化された送達のために使用されている6,44。しかしインビボにおける結果は、これらのナノ担体の腫瘍標的化特異性は、恐らくインビボにおける不安定性及び正常組織における非特異的標的化のために、予想されたほど良好ではないことを示している44。より最近になって、有機ポルフィソーム(porphysome)ナノ小胞の新規クラス6が、PTTのための光を熱に効率的に変換する、多機能性生体光工学薬剤として報告された。残念ながら、ポルフィソームは、それらのかなり大きいサイズ(〜100nm)のために、一部高い肝臓及び脾臓蓄積を有し、結果的に細網内系(RES)による非特異的クリアランスを有する傾向がある11,14,20。対照的に、架橋されたNPsは、極めて低い肝臓及び脾臓取り込み、並びに高い腫瘍標的化特異性を示した(図38b、c)。架橋されたNPsの使用は、光増感剤の光−毒性を最小化し、且つ光線療法の有効性を大きく増強することができることが予想される。

からなる群から選択される、請求項6記載の化合物。 - 内側及び外側を有するナノ担体であって、該ナノ担体が、複数の第一のコンジュゲートを含み、ここで各コンジュゲートは、請求項1記載の化合物であり、
ここで各コンジュゲートは、水性溶媒中で自己組織化し、ナノ担体を形成し、その結果疎水性ポケットが、各両親媒性化合物の疎水面の互いへの配向により、ナノ担体の内側に形成され、ここで各コンジュゲートのPEGは、ナノ担体の外側上に自己組織化している、ナノ担体。 - ナノ担体が、
(a)疎水性薬物又は造影剤を更に含み、その結果疎水性薬物又は造影剤が、ナノ担体の疎水性ポケット内に封鎖され、前記疎水性薬物が、ボルテゾミブ、パクリタキセル、SN38、カンプトテシン、エトポシド及びドキソルビシン、ドセタキセル、ダウノルビシン、VP16、プレドニソン、デキサメタゾン、ビンクリスチン、ビンブラスチン、テムシロリムス、カルムシン、ソラフィニブ、ラパチニブ、及びボルテゾミオブからなる群から選択される、
(b)ポルフィリンにキレートされた金属陽イオンを含み;
(c)64Cu、67Cu、177Lu、67Ga、111In、及び90Ytからなる群から
選択される放射性−金属陽イオンを含み、ここで前記放射性金属が、ポルフィリンへキレートされている、
(d)コンジュゲートが、架橋基を介して架橋され、
(e)各コンジュゲートが、
少なくとも2個のコール酸;
少なくとも2個のピロフェオホルビド−a基;及び
少なくとも2個の架橋基を含み、
ここで、ナノ担体のコンジュゲートが、架橋基を介して架橋される、
(f)ポリエチレングリコール(PEG)ポリマー;
PEGポリマーに連結された結合リガンド;
親水面及び疎水面の両方を有する、少なくとも2個の両親媒性化合物;
PEG及び両親媒性化合物へ、共有結合された、樹状分岐高分子、を含む、少なくとも1個の結合コンジュゲートを含み、
ここで、各結合コンジュゲートは、水性溶媒中で第一のコンジュゲートと自己組織化し、ナノ担体を形成し、その結果疎水性ポケットが、各両親媒性化合物の疎水面の互いへの配向により、ナノ担体の内側に形成され、ここで各コンジュゲートのPEGが、ナノ担体の外側上に自己組織化している、請求項9記載のナノ担体。 - 治療有効量の請求項9又は10記載のナノ担体を含む、音波力学療法により疾患を治療するための医薬組成物であって、該医薬組成物はそれを必要とする対象へ投与され、且つ該対象は音波に曝露される、医薬組成物。
- 対象における腫瘍検出を補助する方法であって、
投与された有効量の請求項9又は10記載のナノ担体に第一波長の放射線を曝露した対象由来の試料を準備し、
ここで該放射線は、ナノ担体上に存在するポルフィリンを励起し、その結果ポルフィリンが、第二波長の放射線を放出すること;並びに
励起されたポルフィリンにより放出された放射線を検出すること
を含む、腫瘍検出方法。
