Lactobacillus crustorumはラクトバチルス属に属する乳酸菌の一種で、小麦サワードウから分離され、新規な種として分類することが提唱されたものである(International Journal of Systematic and Evolutionary Microbiology (2007), 57, 146101467)。サワードウとは、乳酸菌および酵母を含む複数の微生物を用いて、小麦粉やライ麦粉と水を混ぜた生地を発酵させたパン種をいう。
形態的性質
(1)桿菌、2−15μmの長さ、0.5−1μmの幅
(2)運動性:なし
(3)胞子形成:なし
培養的性質
(1)液体または固体MRS培地中、嫌気的条件又は好気的条件においてよく生育する
(2)15℃での生育:5、6、および7%食塩の存在下で生育する
(3)45℃での生育:5、6、および7%食塩の存在下で生育する
生理学的性質
(1)グラム染色:陽性
(2)カタラーゼ試験:
(3)発酵形式:ホモ乳酸発酵
(4)アンモニウム産生(アルギニンから):なし
(5)ガス産生(グルコースまたはグルコン酸塩):なし
(6)酸の産生の有無(LMG 23699株、LMG 23701株、LMG 23702株、R-30103株について):
ガラクトース、グルコース、フルクトース、マンノース、N−アセチルグルコサミン、エスクリンおよびサリシン:あり
グリセロール、エリスリトール、D−アラビノース、L−アラビノース、リボース、D−キシロース、L−キシロース、アドニトール、β−メチル−D−キシロシド、ソルボース、ダルシトール、イノシトール、マンニトール、ソルビトール、α−メチル−D−マンノシド、α−メチル−D−グルコシド、メリビオース、ショ糖、イヌリン、メレジトース、ラフィノース、スターチ、グリコーゲン、キシリトール、D−ツラノース、D−リキソース、D−フコース、L−フコース、D−アラビトール、L−アラビトール、グルコネート、2−ケトグルコネート、および5−ケトグルコネート:なし。
本発明の腸管バリア機能亢進剤は、Lactobacillus crustorumの菌体または菌体処理物を含み、腸管バリア機能亢進作用を有する。
本発明は、上記腸管バリア機能亢進剤を含む医薬組成物も提供する。
本発明は、上記腸管バリア機能亢進剤を含む飲食品も提供する。
本発明の時計遺伝子の腸管バリア機能亢進剤は、有効成分として、Lactobacillus crustorumの菌体または菌体処理物を含むことにより、腸管バリア機能を亢進し、腸管バリア機能の異常に基づく状態もしくは疾患を予防または治療することができる。そして、医薬組成物または飲食品として提供することができる。
Lactobacillus crustorumとして、Lactobacillus crustorum SAM2695株、Lactobacillus crustorum SAM2697株、Lactobacillus crustorum SAM2698株、およびタイプカルチャーであるLactobacillus crustorum JCM 15951Tを用いた。
菌の調製
前培養では、各菌株の保存バイアルから100 μlを10 mlのMRS培地に接種し、24時間、30 ℃で培養した。次に、本培養では1リットルのMRS培地に前培養液10 mlを接種し、24時間、30 ℃で培養した。24時間の培養後、培養液を8000 rpmで10分間遠心分離し、上清を除いた。滅菌水を加えて洗浄し、同条件で遠心分離した。上清を除き、更に1回洗浄し、最後に滅菌水で縣濁した。
ミリセル24ウェルセルカルチャープレート(MERCK MILLIPORE)に0.7×105 細胞/wellの条件でアピカルウェルにCaco2を播種し、37 ℃、5% CO2の条件下で2週間培養した。培地はDulbecco's Modified Eagle Medium(DMEM)(Sigma-Aldrich, D6429)、Fetal Bovine Serum(FBS)(Equiteck Bio, Inc)および抗生物質としてAntibiotic‐Antimycotic Mixed Stock Solution(100x)(ナカライテスク株式会社)を用いた。培地は、500 mlのDMEMに50 mlのFBSおよび5 mlの抗生物質を添加して調製した。培地は、アピカルウェルに400 μl、バソラテラルウェルに800 μl添加した。培地は2〜3日おきに交換した。
2週間培養したCaco2細胞に、1×109 細胞/mlに調整した菌縣濁液を40 μl(コントロールは培地)添加し、37 ℃、5 % CO2の条件下で2時間共培養した。膜抵抗値の測定には、ミリセルERS-2電圧抵抗計(MERCK MILLIPORE)を用い、Lactobacillus crustorumの添加前および添加24時間後の膜抵抗値を測定した。実験は各n=6で実施した。
結果を図1に示す。
Caco2とLactobacillus crustorum SAM 2698株を共培養し、腸管バリア関連遺伝子の発現を調べた。
菌の調製
Lactobacillus crustorumとして、Lactobacillus crustorum SAM 2698株を用いた。
12ウェルセルカルチャープレート(BD Falcon)に0.63×105 細胞/ 2 ml / ウェルの条件でCaco2細胞を播種し、37 ℃、5 % CO2の条件下で2週間培養した。培地はDulbecco's Modified Eagle Medium(DMEM)(Sigma-Aldrich, D6429)、Fetal Bovine Serum(FBS)( Equiteck Bio, Inc)および抗生物質としてAntibiotic‐Antimycotic Mixed Stock Solution(100x)(ナカライテスク株式会社)を用いた。培地は、500mlのDMEMに50mlのFBSおよび5mlの抗生物質を添加して調製した。培地は2〜3日おきに交換した。
実験方法
2週間培養したCaco2細胞に、DMEMで縣濁したLactobacillus crustorumを1×109細胞/2ml/well添加し、5時間共培養した。実験は全てn数を3で実施した。5時間後、上清を除き、PBS(-)で洗浄後した。回収した細胞から、RNeasy Mini Kit(QIAGEN)を用いてRNAを調製した。
タカラバイオ株式会社にAgilent Expression Array解析を依頼し、Human SurePrint G3 (8×60k) ver2.0を実施した。
タイトジャンクション関連遺伝子クラウディンに着目して解析した結果を図2に示す。コントロールと比較すると、Lactobacillus crustorum SAM 2698と共培養したCaco2細胞ではクラウディン1 (CLDN 1)の発現量が増加した(約1.1倍)。また、CLDN 3およびCLDN4の発現量も有意に増加した(各約1.6倍)。
Lactobacillus crustorum SAM 2698 生菌をマウスに投与し、小腸上皮組織におけるCLDN1の発現変化を調べた。
Lactobacillus crustorumとして、Lactobacillus crustorum SAM 2698株を用いた。
実験方法
生菌は120億細胞/mlの縣濁液250 μl (30億cells/mouse) を単回経口投与した。投与5時間後に小腸上皮組織を採取した。
小腸上皮組織に150-212 μmのグラスビーズ(SIGMA社製)を添加し、マルチビーズショッカー(YASUI KIKAI社製)を用いて3000rpm, 3分間処理した。その後、RNeasy Mini Kit(QIAGEN)を用い、プロトコルに従ってRNAを調製した。
マウスのCLDN1遺伝子のためのプライマー配列は、公開されている遺伝子配列情報をもとに設計した。配列は以下の通りである。マウスCLDN1フォワードプライマー : CCTATGCTGGGGACAACATC/マウスCLDN1リバースプライマー : TGCACCTCATCATCTTCCAG。発現解析はGAPDH遺伝子発現をコントロールとした。GAPDH遺伝子のためのプライマー配列は、ShineGene Molecular Biotech, Incが公開しているHousekeeping Gene Primers for RT-PCRを参考にして設計した。配列は以下の通りである。Mouse GAPDH F49 : GGTGAAGGTCGGTGTGAACG/R281 : CTCGCTCCTGGAAGATGGTG。
発現解析はリアルタイムPCR (Step One Plus, Applied Biosystems社製)を用いて実施した。
タイトジャンクション関連遺伝子CLDN1に着目して解析した結果を図3に示す。滅菌水投与群と比較してLactobacillus crustorum SAM 2698を投与したマウスの小腸上皮において、クラウディン1 (CLDN 1)の発現が有意に増加した。
Lactobacillus crustorumとして、Lactobacillus crustorum SAM2695株、Lactobacillus crustorum SAM2698株、およびタイプカルチャーであるLactobacillus crustorum JCM 15951Tを用いた。
実験方法
試験期間は25日間で、生菌は120億細胞/mlの縣濁液250 μl (30億細胞/日/匹) を経口投与した。
正常食および60%高脂肪食はリサーチダイエット社の精製飼料10kcal%の脂肪(ラード)および精製飼料60kcal%の脂肪(ラード)を用いた。
試験25日目に糖負荷試験を実施した。20%グルコース溶液を1g/kg(5ml/kg)の条件で経口投与し、投与前、投与後15、30、60及び90分後に尾静脈より採血した。血糖はグルテストneoスーパー(株式会社三和化学研究所)で測定した。
結果を図4および図5に示す。
Claims (3)
- Lactobacillus crustorumの菌体または菌体処理物を含む、腸管バリア機能亢進作用を有する飲食品であって、Lactobacillus crustorumが受託番号NITE BP-01721として寄託されたLactobacillus crustorum SAM2695株、受託番号NITE BP-01722として寄託されたLactobacillus crustorum SAM2697株、受託番号NITE BP-01723として寄託されたLactobacillus crustorum SAM2698株、または2以上のこれらの組み合わせである、前記飲食品。
- 前記Lactobacillus crustorumの生菌体を含む、請求項1記載の飲食品。
- 糖尿病、高血圧、高脂血症、および肥満からなる群より選択される1以上の状態もしくは疾患の予防または治療用である、請求項1または2記載の飲食品。
