Description
非特許文献1〜2では、血清タンパク質であるトランスフェリン(TFと略すことがある)に近赤外波長領域の光を吸収する有機色素(以下、近赤外色素と略すことがある)を結合させた化合物が開示されている。なお、本明細書における近赤外色素とは、近赤外波長領域の光を吸収する有機色素である。本明細書において近赤外波長領域の光とは600nm乃至1300nmの波長の光を意味する。
ICGなどの近赤外色素は、血中に投与した場合、血中のタンパク質と吸着した上で体外へと排出されやすい。その結果、単独の近赤外色素を生体の血中に投与しても、血中滞留性が低く、腫瘍への集積性が低い。したがって、単独の近赤外色素を光音響イメージング用造影剤として用いた場合、腫瘍から発せられる光音響信号強度は小さい。
本明細書においてトランスフェリン1分子に対して共有結合している近赤外色素の平均の数を、色素標識数とよぶ。本実施形態に係る化合物が1分子の場合は、色素標識数nは整数値である。一方、本実施形態に係る化合物が複数集まって混合物を形成している場合、化合物における色素標識数とは、その混合物を構成する化合物の色素標識数の平均値で表わす。よって、本明細書における色素標識数は整数に限らないが、実際のトランスフェリンに標識されている数は整数値をとる。本実施形態に係るPAI用造影剤において、色素標識数は、1以上が好ましく、9以下が好ましい。さらには、2以上が好ましく、8以下が好ましい。特に、4以上が好ましく、7以下が好ましい。これは、色素標識率が上記範囲内にあるときに、腫瘍集積性ならびに腫瘍血中比が高いからである。
なお、色素標識数は近赤外色素の濃度およびトランスフェリンの濃度をそれぞれ求め、それらの比(近赤外色素の濃度/トランスフェリンの濃度)から算出することで求める。近赤外色素の濃度は、その色素の特異的な吸収波長における吸光度と吸収係数から算出した。特異的な吸収波長とは、例えば、近赤外色素としてICG−Sulfo−OSu(下記式(d5−1)で示される化合物)を用いた場合は790nmを採用することができるが、特異的な吸収波長は他の値を採用してもよい。トランスフェリンの濃度はタンパク質特有の吸収である280nmの吸光度と、その波長におけるトランスフェリンの吸収係数を用いて求めることができるし、BCA法等によって求めることもできる。なおトランスフェリンの280nmにおけるモル吸収係数は、非特許文献1に記載された、92300(M−1×cm−1)を用いた。
本実施形態に係るトランスフェリンは、血漿に存在し、分子量が約80kDaからなるタンパク質である。トランスフェリンは主に鉄イオンを輸送する役割を有する。本実施形態に係るトランスフェリンとしては、ヒト血清由来以外にも、ウシ血清由来、ラット血清由来、マウス血清由来など、人以外の動物種由来のものであってもよく、またこれらの断片であってもよい。本実施形態に係るトランスフェリンとしては、人体での安全性が高いと考えられる、ヒト血清由来トランスフェリン、その改変体、その断片のいずれかであることが好ましい。また、本実施形態に係るトランスフェリンはヒト血液からの抽出物であってもよいし、遺伝子組換え体でもよく、大腸菌、酵母、あるいは培養細胞等からの生産物でもあっても構わない。

本実施形態において近赤外色素としては、近赤外波長領域の光を吸収して音響波を発する有機色素であれば特に限定されない。本実施形態における近赤外色素としては、例えば、アジン系色素、アクリジン系色素、トリフェニルメタン系色素、キサンテン系色素、ポルフィリン系色素、シアニン系色素、フタロシアニン系色素、スチリル系色素、ピリリウム系色素、アゾ系色素、キノン系色素、テトラサイクリン系色素、フラボン系色素、ポリエン系色素、BODIPY(登録商標)系色素、インジゴイド系色素を挙げることが出来る。上記シアニン系色素としては、例えば、インドシアニングリーン(ICG)やその誘導体が挙げられる。
本実施形態において、トランスフェリンと近赤外色素は、アミノ基、チオール基、カルボキシル基、またはヒドロキシル基を介して公知のカップリング反応によって共有結合させることができる。特に、トランスフェリンのアミノ基を介して結合させることが好ましい。アミノ基はトランスフェリン中に複数存在し、特にリジン残基の1級アミノ基は、弱アルカリ性のpH領域において効率的かつ選択的に近赤外色素のN−ヒドロキシスクシンイミド基に求核反応する。前記反応によりトランスフェリンと結合した近赤外色素は、限外濾過法、サイズ排除カラムクロマトグラフィー法、透析等の公知のタンパク質精製法により洗浄、精製することができる。
本実施形態に係るPAI用造影剤において、トランスフェリンと近赤外色素の結合はトランスフェリン表面に存在する前記アミノ基、チオール基、カルボキシル基、またはヒドロキシル基と近赤外色素の誘導体とで直接結合されていても良いし、種々のリンカー(架橋材やクロスリンカーともいわれる)を介してトランスフェリンと近赤外色素が結合されていても良い。
本実施形態に係るPAI用造影剤は、上記化合物以外に分散媒を有していてもよい。なお、PAIは、光音響トモグラフィー(断層撮影法)を含む概念である。分散媒として例えば、生理食塩水、注射用蒸留水、リン酸緩衝生理食塩水、ブドウ糖水溶液が挙げられる。また本実施形態に係るPAI用造影剤は、必要に応じて薬理上許容できる添加物、例えば界面活性剤などを有していても良い。
本実施形態における化合物あるいはPAI用造影剤は標的部位に特異的に結合する捕捉分子を有していてもよい。捕捉分子とは、腫瘍などの標的部位に特異的に結合する物質、標的部位の周辺に存在する物質に特異的に結合する物質などであり、生体分子や医薬品等の化学物質などから任意に選択することができる。具体的には、タンパク質、抗体、抗体フラグメント、酵素、生物活性ペプチド、グリコペプチド、ポリペプチド、ペプチド、糖鎖、脂質、分子認識化合物などが挙げられる。これらの物質は単独で用いることもできるし、あるいは複数を組み合わせて用いることもできる。捕捉分子が化学結合された化合物を用いることで、標的部位の特異的な検出、標的物質の動態、局在、薬効、代謝等の追跡を行うことができる。本実施形態において、タンパク質とは、アミド結合によって天然または非天然アミノ酸が90個以上連結されたものを言う。本実施形態においてポリペプチドとは、アミド結合によって天然または非天然アミノ酸が30個以上90個未満で連結されたものを言う。本実施形態において、ペプチドとは、アミド結合によって天然または非天然アミノ酸が30個未満で連結されたものを言う。本実施形態において、タンパク質とポリペプチドとペプチドの分類は、アミノ酸の連結数によってなされるものであり、種々の修飾の有無を問わない。本実施形態において捕捉分子はタンパク質、または、ポリペプチド、または、ペプチドであることが好ましい。
本実施形態に係る光音響イメージング用造影剤は、凍結乾燥時に使用する添加剤を含んでいてもよい。添加剤の一例としてグルコース、ラクトース、マンニトール、ポリエチレングリコール、グリシン、塩化ナトリウム、リン酸水素ナトリウムが挙げられる。添加剤は1種類のみを用いても、複数種類を併用してもよい。
生体内に投与された本実施形態に係るPAI用造影剤を、光音響イメージング装置を用いて検出する方法について説明する。本実施形態に係るPAI用造影剤を検出する方法は以下の(a)、(b)の工程を有する。但し、本実施形態に係る光音響イメージング方法は、以下に示す工程以外の工程を含んでいても良い。
(a)本実施形態に係るPAI用造影剤が投与された検体に600nm乃至1300nmの波長領域の光を照射する工程
(b)前記検体内に存在する前記造影剤から発生する音響波を検出する工程
本発明の実施例においてトランスフェリンに対する色素標識数は化合物の吸光度測定により算出した。つまり、化合物の水溶液を調製し、色素ならびにトランスフェリンの吸光度を測定することで、色素ならびにトランスフェリン濃度を求め、色素濃度をトランスフェリン濃度で割ることにより、色素標識数が求められる。
具体的には、ICG−Sulfo−OSu(下記式(d5−1)で示される化合物)を用いて調製された化合物の場合は、5%ドデシル硫酸ナトリウム(Sulfuric acid dodecyl、以後SDSと略す)で希釈された化合物の水溶液中における790nmの吸光度を計測した。この吸光度はICG由来のものである。ICG濃度は予め作成しておいたSDS中での検量線から算出した790nmの吸収係数120000(M−1×cm−1)を用いて色素濃度を算出した。トランスフェリン濃度は、リン酸緩衝生理食塩水(PBSと略すことがある)中でトランスフェリンの280nmにおける吸光度を測定し、トランスフェリンのモル吸収係数92300(M−1×cm−1)を用いて算出した。色素標識数は、色素濃度をトランスフェリン濃度で割ることで求めた。なお、トランスフェリンの280nmの吸光度は、測定された280nmの吸光度から、ICGの790nmの吸光度に0.4をかけた値を引くことで、トランスフェリンの280nmの吸光度に対する、ICG−Sulfo−OSuの280nmの吸光度の寄与を補正した。
本実施例における化合物はトランスフェリン表面のアミノ基に対して近赤外色素を共有結合させたものである。以下に調製方法の一例を示す。
まず、10mgのトランスフェリン(トランスフェリン ヒト、T3309、SIGMA社製)を1mLの炭酸緩衝液(pH9.0)に溶解してトランスフェリン溶液とした。下記式(d5−1)で示される1mgの近赤外色素(ICG−Sulfo−OSu、I254、Dojindo Laboratories製)を0.1mLのジメチルスルホキシド(Dimethyl sulfoxide、以下DMSOと略すことがある)に溶解した。それぞれの溶液をプラスチックチューブ内で混合して室温で3時間、回転撹拌した。得られた反応溶液を0.22μmシリンジフィルターでろ過し、ろ液を回収した。回収した溶液に、セファクリルゲルS−400を加え、室温で1時間回転撹拌した。次いで、このゲル含有溶液を遠心分離し、プラスチックチューブ底にあるゲルを分取しないようにして、上清部分を回収した。最後に、回収した緑色溶液を0.22μmシリンジフィルターでろ過することで本発明にかかる近赤外色素とトランスフェリンが共有結合した化合物(以後iTFと略すことがある)を得た。
実施例1で得られたiTF(2.0)、iTF(4.8)、iTF(7.0)、iTF(8.3)、iTF(8.4)の水溶液、ならびに比較例として、下記式で表されるICGの水溶液の光音響信号を測定した。
実施例1で得られたiTF(2.0)、iTF(4.8)、iTF(7.0)、iTF(8.3)、iTF(8.4)、ならびに比較例として、Alexa Fluor(登録商標) 680近赤外色素が結合したトランスフェリン(Transferrin From Human Serum, Alexa Fluor(登録商標) 680 Conjugate、ライフテクノロジー社製、製品番号:T35357、色素標識数2.0)のがん細胞への取り込み能を測定した。以後、Alexa Fluor(登録商標) 680近赤外色素が結合したトランスフェリンをAlexaFluor680−TF(2.0)と略す。
実施例1で得られたiTF(2.0)、iTF(4.8)、iTF(7.0)、iTF(8.3)、iTF(8.4)、ならびに比較例として、ICGとAlexaFluor680−TF(2.0)について、担癌マウスを用いた蛍光イメージングにより、腫瘍のイメージングコントラストを評価した。蛍光イメージング実験においては、雌の非近交系BALB/c Slc−nu/nuマウス(購入時6週齢)(日本エスエルシー株式会社)を用いた。マウスに担癌させる前の1週間、標準的な食餌、寝床を用い、自由に食餌および飲料水を摂取できる環境下でマウスを順応させた。イメージング実験の約1週間前に1×106個のcolon26マウス腸癌細胞(理研)を、マウスの大腿部に皮下注射することで担癌モデルマウスを調製した。
実施例4で実施した腫瘍造影実験のマウスの腫瘍中色素量と血中色素量を定量することで、化合物の腫瘍集積率と血中残存率を評価した。

- 分散媒をさらに含む請求項1乃至6のいずれか一項に記載の光音響イメージング用造影剤。
- 腫瘍の造影に用いられる請求項1乃至7のいずれか一項に記載の光音響イメージング用造影剤。
