Description
本発明に係るサソリ毒耐熱合成ペプチドの製造方法は以下のとおりである。
1.サソリ毒耐熱ペプチドのアミノ酸配列の測定
(1)BmKサソリ毒の凍結乾燥粉末を溶かして遠心し、上清液を取って100℃の水浴で加熱した後、遠心し、上清液即ちサソリ毒耐熱成分抽出液を取り、順次に分画分子量が50kDaと30kDaの濾過膜を用いる遠心式限外濾過管で、サソリ毒耐熱成分抽出液を遠心限外濾過し、上槽液を収集し、サソリ毒耐熱ペプチド抽出液を得る。Superdex Peptide 10/300GL分子ふるいカラム(Optimum Separation range(peptides) M、100−7000Da)で上記サソリ毒耐熱ペプチド抽出液を粗分離した後(図1をご参照)、得られた試料をHPLCにより更に細分離する(図2を参照)。クロマトグラフィー条件は、クロマトグラフィーカラムはZorbax SB−C18 4.6*250 5μm(AgilentUSA)であり、移動相はA液とB液を含み、その中、A液は0.1%トリフルオロ酢酸を含むアセトニトリル/水=2:98であり、B液は0.08%トリフルオロ酢酸を含むアセトニトリル/水=98:2であり、順次に0−40%のB液を含む移動相3−6カラム体積、40−100%のB液を含む移動相0.5−1カラム体積、100%B液1−3カラム体積(CV)を流し、流速は0.8mL/minであり、紫外線検出器の検出波長は280nm/258nm/214nmである。
(1)該当ポリペプチドの合成とは、上記サソリ毒耐熱ペプチドのアミノ配列に従い、アミド結合の指向形成方法で目標分子を得るものである。固相合成とは、原料薬の一つのアミノ酸のカルボキシル基を共有結合の形式で固相担体(Fmoc 樹脂)と連結して、このアミノ酸のアミノ基を合成起点として、隣接するアミノ酸(アミノ基保護)のカルボキシル基とアシル化反応させ、ペプチド結合を形成するものである。その後、この二つのアミノ酸を含んだ樹脂ペプチドのアミノ基を脱保護させた後、次のアミノ酸のカルボキシル基と反応させる。目標ペプチドが得られるまで、この過程を繰り返す。
(3)質量スペクトルを用い、合成ポリペプチドの構造解析を行い、検索及び解析を経て、サソリ毒耐熱合成ペプチド(SVHRSP)のアミノ酸配列を確定し、該当SVHRSPのアミノ酸配列は、SEQ ID No.1に示す。
サソリ毒耐熱ペプチドのアミノ酸配列の測定:
(1)三次蒸留水で河南宜昌由来のBmKサソリ毒の凍結乾燥粉末を溶解した後、遠心し、上清液、即ちBmKサソリ毒抽出液を得る。カバー付きの耐熱プラスチックチューブに分注し、恒温水浴槽において、100℃で4h加熱した後取り出し、自然に室温まで冷却させて、遠心分離及び高速遠心分離を行って、上清液、即ちサソリ耐熱成分抽出液を得る。その抽出液を順次に分画分子量が50kDaと30kDaの濾過膜を用いる遠心限外濾過管で遠心限外濾過し、上槽液、即ちサソリ毒耐熱ポリペプチド抽出液を得る。これを遠心し、上清液、即ちサソリ毒耐熱成分抽出液を得る。
質量スペクトルより採集した原始wiffグラフファイルに対して、Protein Pilot Software v.4.5(AB SCIEX、USA)ソフトウェアでデータ加工処理と検索分析を行う。データベースはuniprotベース中のサソリであり、検索方式は徹底分析であり、仮陽性率を1%FDRに制御する。得られたサソリ毒耐熱合成ペプチド(SVHRSP)のアミノ酸配列は、配列表SEQ ID No.1に示す。検索結果は表1のとおりである。
合成サソリ毒耐熱ポリペプチドの高速液体クロマトグラフィー(HPLC)による純化:
ポリペプチドの合成過程において目標ペプチドの構造に類似する不純ペプチドが生成される可能性があり、例えばアミノ酸のラセミ化によるジアステレオ異性体、一部のアミノ酸が連結されないことによる欠乏ペプチド、ペプチド連鎖の断裂による断裂ペプチドなどが生成される。従ってRP−HPLCで純化を行う。

Morrisウォーターメイズ実験法でSVHRSPが(AD)マウスの空間学習記憶能力に与える影響の測定:
モリスウォーターメイズ実験において行動学の変化を検出する。定位・航行実験を5d持続する。毎日4回訓練し、マウスがプラットホーム(プラットホームは第1象限に置く)を探し出す時間、即ち、脱出潜伏期を記録し、且つマウスをプラットホームで20s停留させる。マウスが60s内にプラットホームを探し出せない場合、脱出潜伏期を60sと記録する。空間捜索実験は、定位・航行実験の第5dにおいての4回訓練後に行う。プラットホームを取り外した後、第3象限の入水点でマウスをプールに入れて、試験を開始する。マウスの目標象限(第1象限)内の水泳距離と水泳時間との百分比を算出し、総時間は60sにする。マウスが隠しプラットホームを通過する回数を測定する。
モリスウォーターメイズ実験からSVHRSPがADマウスの空間学習記憶能力に対して促進作用を有することがわかった。モリスウォーターメイズの隠しプラットホーム獲得実験のテスト日(第5日)において、ADモデル薬物投与組マウスの脱出潜伏期(第5d、12±2回)が明らかにADモデル組(第5日、22±2回)の脱出潜伏期**p<0.01より短く、その水泳距離はモデル組の第5dの500cmから薬物投与組の300cmに低下し、プラットホームを通過する回数はモデル組の5回から薬物投与組の8回に向上した。
線虫走化行動学の実験方法:
同期化した遺伝子組み換え線虫CL2355及びそのコントロール株CL2122に対してそれぞれ異なる濃度のSVHRSP(2、20、40μg/mL)及び等量の三次蒸留水を投与する。虫卵である時から投与し、毎日線虫を新しい薬物含有のNGM培養皿に移転する。すべての線虫を16℃で36時間培養した後、インキュベーターの温度を23℃に向上し、継続して36時間培養する。上記線虫を収集し、M9緩衝液で3回洗浄する。走化実験を行う時に100mmの大きい培養皿を用いる。固体培地は1.9%の寒天、1mMのCaCl2、1mMのMgSO4、25mMのリン酸塩緩衝液を含み、pHは6.0であり、マイクロ波で均一に混合した後、皿に入れて、冷却させる。培養皿の底部に、中心位置を経由するようにマジックペンで二本の互いに垂直する直線を描くことにより、皿全体を均一に四つの象限に分ける。
各組の約60本の線虫を走化培養皿の中心位置に置いて、1μLのベンズアルデヒド(濃度が0.1%であり、無水エタノールで希釈する)と1μLの0.25Mアジ化ナトリウムを1、3象限の中心位置に滴下し、1μLの無水エタノールと1μLの0.25Mアジ化ナトリウムを2、4象限に滴下し、その中、該点から中心点までの距離は2センチメートル以上である必要がある。走化培養皿を23℃のインキュベーターに移動させ、1時間が経過した後、走化指数を計数する。ベンズアルデヒドは高濃度気味を有する化学物質であり、線虫はベンズアルデヒドの気味に吸引されその区域へ移動するはずであり、そこでアジ化ナトリウムに接触すると麻痺して原地に止まる。走化指数の算出方法は、1、3象限(吸引物が所在する象限)に計数する線虫の数から2、4象限での線虫の数を差し引き、得られた数を線虫の総数で割るものである。
サソリ毒耐熱合成ペプチド(SVHRSP)は投与量依頼性でCL2355のCIを向上させることができ、40μg/mLのSVHRSPのニューロンに対する保護作用が最も顕著であり、ほとんど完全にAβの発現による走化性行動損傷を逆転させた。コントロール組の39.45±6.2に比べて、薬物投与組は7±5.66にまで減少され、p<0.01であった。これらの結果は、SVHRSPは、ニューロンを保護し、Aβによる毒性作用に対抗し、ニューロンのAβの発現による走化性行動異常を改善させる作用があることを表明する。結果は、表2に示す。
SVHRSPが6−OHDAによるSH−SY5Y酸化ストレスに与える影響:
蛍光プローブDCFH−DAによりROSを検出し、SVHRSPが6−OHDAによるSH−SY5Y酸化ストレスに与える影響を観察する。実験は、対照組、ROSup陽性対照組、6−OHDAモデル組及び異なる濃度(2μg/mL、5μg/mL、10μg/mL、20μg/mL)のSVHRSP薬物組に分けて行う。指数増殖期にあるSH−SY5Y細胞を取り、培地を除き、滅菌PBSで一回洗浄した後、0.25%のパンクレアチンを加えて消化し、ピペッチングして細胞を分散させる。細胞計数板で細胞をカウントして、1.5×104個/mLになるように調製した後、96ウエルプレートに巻き込み、細胞がプレートの底に沈んだ後、5%CO2、37℃のインキュベーターに入れ、24h培養する。濃度が20μg/mLのSVHRSP薬物組は、薬物を加えて1h作用させた後、6−OHDAを加え、5%CO2、37℃のインキュベーターに入れて24h継続して培養する。1:500の希釈比率でROSupを培地で希釈して、各ウエルに100μLずつ加え、27分間培養する。1:2000の希釈比率でDCFH−DAを無血清培地で希釈し、最終濃度が5μmol/Lになるようにする。培地を捨て、各ウエルに50μLずつ上記希釈したDCFH−DA溶液を加えて、軽く均一に揺れてから5%CO2、37℃のインキュベーターに入れ、20min培養する。上清液を捨て、PBSで3回洗浄し、蛍光マイクロプレートリーダーにおいて、488nmの励起波長、525nmの発射波長で蛍光測定を行う。OD値を測定し、比較する。
サソリ毒耐熱合成ペプチド(SVHRSP)の難治性てんかんピロカルピンモデルラットに対する予防・治療作用:
1)体重180gの健康なオスSDラットを用い、リチウム−ピロカルピンを300mg/kgの用量でipより投与して、側頭葉てんかんの発作を誘発する。てんかんが急性に発作した後の15日目に、てんかんラットをモデル組とモデル薬物投与組にランダムにグルーピングする。てんかんモデル組の動物は生理塩水(NS)を腹腔内に注射し、15日間連続する。てんかんモデル薬物投与組のラットは更に3つのモデル薬物投与組にグルーピングし、それぞれSVHRP抽出液を50μg/kg/dの用量でipより投与した組、SVHRSPを50μg/kg/dの用量でipより投与した組、陽性対照薬物−バルプロ酸ナトリウム投与組である。即ち、ピロカルピンてんかんモデル+NS組、リチウム−ピロカルピンてんかんモデル+SVHRP組、リチウム−ピロカルピンてんかんモデル+SVHRSP組、リチウム−ピロカルピンてんかんモデル+バルプロ酸ナトリウム組である。各組において、てんかんレベルを観察して記録し、Racine’sレベルを評価標準とし、表3に示す。
(1)SVHRSPは、リチウム−ピロカルピンてんかんラットの慢性モデルてんかん発作に対して著しい制御効果を有し、発作回数を著しく低減させる。
(2)SVHRSPがリチウム−ピロカルピンてんかんラット慢性モデルの発作レベル及び発作回数を低下する能力は、いずれも陽性対照組であるバルプロ酸ナトリウムより強い。
サソリ毒耐熱合成ペプチド(SVHRSP)の初代培養ニューロンのナトリウム電流に対する抑制作用:
(1)初代培養による海馬ニューロンの培養:
生まれて24h後のSDラットを75%アルコールで消毒した後、氷上で断頭し、解剖顕微鏡下で解剖し快速に両側の海馬を完全に剥離し、解剖液中に置いて体積が1mm3の小塊になるように切って、終濃度が0.125%のパンクレアチン消化液に入れ、37℃、10%CO2のインキュベーターに入れて30min消化させる。消化された組織を取り出して、播種液を入れて、消化を終止し、ピペッチングで懸濁させ、濾過を行うことによって、密度が1×105/mLの細胞懸濁液を作り、ポリリシン(0.1mg/mL)で事前にコーティングして置いた培養プレートに播種し、37℃、10%CO2のインキュベーターにおいて培養する。第2dに播種液を全部培地に切替えて、培養する。培養第4dに培地にシタラビン(3μg/mL)を加え、非神経細胞の過度増殖を抑制する。その後3dごとに50%の新鮮な培地を切替える。体外培養により、海馬ニューロンは第9−12d培養することで、成熟ニューロンになり、本実験においては、培養して第10d目に得られたニューロンを用いる。
初代培養した海馬ニューロンを10d培養した後、全細胞パッチクランプ記録を行う。記録用ガラス管微小電極の外径は1.5mm、内径は0.6−0.8mm、長さは8cmであり、PP−830型微小電極引張機で両段階垂直引張を行って製作する。引張後の電極直径が約1μmであり、電極内液を充填した後、電極抵抗が3−5MΩである。参照電極はAg−AgCl電極である。全細胞パッチクランプ記録において、電極と細胞膜との間に安定な高抵抗封止を形成した後、快速のコンデンサー補償(C−fast)を行い、やや負圧を加えて破膜させ、全細胞構造を形成した後、減速コンデンサー補償(C−slow)を行う。減速コンデンサー補償率は80%−85%である。リーク電流法でリーク電流(Leak Substraction)を差し引く。設定された刺激電圧で電流のアクティブ情況を観察する。全細胞パッチクランプ記録に用いられる機器の設置条件は以下の通りである。EPC−10二重チャンネルパッチクランプ増幅器実験パラメータ、データの採集と刺激の与えは、いずれもPulseソフトウェアによって制御され、Besselフィルター1の周波数は10KHzであり、Besselフィルター2の周波数は2.9KHzである。実験過程中に、所用する薬物はいずれもBPS−8潅流装置により投与され、排出管毎個の直径は0.2mmであり、管口は記録に用いる細胞より約100μm離れる。初代培養したニューロンシステムに対して、パッチクランプを行いナトリウムチャンネル電流を記録する。同じ細胞に対してサソリ毒耐熱合成ペプチド(SVHRSP)を加える前後の電流を比較する。
全細胞パッチクランプ記録において、同じ細胞に対してサソリ毒耐熱合成ペプチド(SVHRSP)を加える前後の電流を比較する。比較結果により、2μm/mLのSVHRSPは初代培養した海馬神経細胞体のナトリウムチャンネル電流を顕著に抑制し、その抑制率は90%以上に達し(n=6)(p<0.0001)、細胞外液で洗浄した後電流が回復できることが分かった。
SH−SY5Y細胞は人の神経芽腫細胞株由来の細胞株である。NMDAモデル:NMDA20mM+Gly10μM。
実験過程:実験前、実験目的に応じてグルーピングする。SH−SY5Yを96ウェルのプレートに0.8×104/穴になるように巻き込み、24h培養した後、SVHRSPとpeptideを加え24hプレインキュベーションする。その後、グルーピングされた各組において、NMDA20mM+Gly10μMを加えて24h培養した後、MTTアッセイにより各組細胞の生存率を測定する。測定結果は図11に示す。測定結果により、SVHRSPは、NMDAによるSH−SY5Y細胞損傷に対して保護作用を有することが判明された。その中、SVHRP濃度は2μg/mL、20μg/mLであり、SVHRSP濃度は2μg/mL、20μg/mLであり、NMDA20mM+Gly10μMであり、p<0.05(n=3)である。
II型星状膠細胞は逆分化によって増殖及び分化能力を有する多能性神経幹細胞を形成する。
実験過程:生まれて24h後のSDラットを用い、大脳皮層を無菌分離し、細胞懸濁液を得、10%ウシ胎仔血清を含有するDMEM培地において、混合膠細胞の初代培養を行う。7−9d培養した後、まずL−ロイシンメチル塩酸塩(L−leucine methyl ester hydrochloride)で1h処理し、新鮮な無菌の細胞培地を切替えて、37℃の恒温振とう台で180r/minの速度で18時間揺らした後、上清液を200メッシュの濾過網で濾過し、得られた上清液をポリリシンで予備処理した6ウェルプレートに播種して、相対的に純化されたOPCを得る。細胞をインキュベーターに中で自然沈降させ、2h後に細胞が壁に接着したか否かを観察する。壁に接着した後、上清液を捨ててII型星状膠細胞誘導液(20%DMEM)を切替え、4−5d継続して培養して、相対的に純化されたII型星状膠細胞を得る。
5d後、II型星状膠細胞を消化させ超低接着6ウェルプレートに播種し、培地を幹細胞培養培地に切替えて、12d継続して培養して、幹細胞コロニーを形成させる。この超低接着6ウェルプレート中のコロニーを取って、ポリリシンを処理した細胞クライミングスライドを敷設した24ウェルプレートに巻き込んで24h培養した後、細胞クライミングスライドを取出し、4%のパラホルムアルデヒドで固定し、免疫蛍光染色を行う。Nestin(green)でコロニーが幹細胞であるか否かを確認する。Nestinは、神経幹細胞の特異性マーカーであり、サソリ毒耐熱合成ペプチド(SVHRSP)はII型星状膠細胞が逆分化して神経幹細胞を形成する過程のマーカーであるNestinに影響する。
サソリ毒耐熱合成ペプチド(SVHRSP)の急性毒性実験:
1)主な観察指標:
i)最大耐性量(Maximal tolerancedose,MTD):即ち最大非致死投与量、受験動物が死に至らない最大の用量を言う。
ii)最小致死投与量(Minimal lethal dose,MLD):個別の受験動物が死亡し始める用量を言う。
iii)最大無毒性量(最高無毒):即ち毒性反応が現れない最大の用量(No observed adverse effect level,NOAEL)であり、即ち一定の時間内に、動物に損害作用を与えない最大の用量を言う。
iv)最小毒性量:動物が毒性反応を現す最低用量を言い、毒性反応が現れたばかりの用量、即ち動物が毒性反応を現す最低用量を言う。
2)注:
i)受験動物:昆明種マウス、雌と雄がそれぞれ半分ずつである(健康な成年であり、体重が19−20gである)。
ii)受験試薬:SVHRSP:
a.出所:河南省宜昌XINXINサソリ養殖工場(2012−05−28)からBmKサソリ毒抽出液を購入する。
b.Lot No:P160418−CQ455216。
c.含有量(又は仕様):10mg/1つ。
d.保存条件及び配合方法:−20℃で長期保存でき、4℃では7−10dで保存でき、使用時に超純水で溶かす。
iii)投与経路:それぞれipで投与する(マウスip経路投与において、常用する最大容積は0.1−1.0mL/20gBWである)。
投与経路に基づいて当該動物種の最適な投与容量(投与量が異なっても、投与容量は同じにする)を選択することであり、今回実験において、ip投与経路は、いずれも0.2mL/20gBWにする。
iv)採用方法の選択
近似致死量法によって、可能な配列表を作成する。
従来サソリ毒耐熱ポリペプチドの昆明種マウスに対する急性毒性試験結果に基づき,サソリ毒耐熱合成ペプチド(SVHRSP)の可能な最大耐性量と最小致死量を算出し、複数の投与量を含んだ投与量配列表を設計する。
可能な最大耐性量(Maximal tolerancedose,MTD):76.8、51.2、34.1mg/kg。
可能な最小致死量(Minimal lethal dose,MLD):115.2、76.8、51.2mg/kg。
算出した投与量配列表から可能な致死量範囲(115.2、76.8、51.2、34.1mg/kg)を探し出し、この範囲内にある各投与量を一匹の動物に投与し、最小致死量と最大耐性量を測定した後、両者の間にいる投与量を一匹の動物に投与する。当該投与量で動物が死亡に至らない場合、当該投与量と最小致死量との間の範囲を近似致死量範囲とする。当該投与量で動物が死亡する場合、当該投与量と最大耐性量との間の範囲を近似致死量範囲とする。
(付記1)
完全な遺伝子配列がSEQ ID No.1である、ことを特徴とするサソリ毒耐熱合成ペプチド。
BmKサソリ毒の凍結乾燥粉末を溶かして遠心し、上清液を取り100℃の水浴で加熱した後、遠心し、上清液を回収してサソリ毒耐熱成分抽出液を得、該サソリ毒耐熱成分抽出液を順次に分画分子量が50kDaと30kDaである遠心式限外濾過管において遠心限外濾過を行って上槽液を回収し、サソリ毒耐熱ポリペプチド抽出液を得るステップと、 Superdex Peptide 10/300GL分子ふるいカラム(Optimum Separation range(peptides)M,100−7000Da)により前記サソリ毒耐熱ポリペプチド抽出液を粗分離した後、得られた試料をHPLCによりさらに分離し、その中、クロマトグラフィー条件は、クロマトグラフィーカラムはZorbax SB−C18 4.6*250 5μm(Agilent USA)、移動相はA液とB液を含み、その中、A液は0.1%トリフルオロ酢酸を含むアセトニトリル/水=2:98であり、B液は0.08%トリフルオロ酢酸を含むアセトニトリル/水=98:2であり、順次に0−40%のB液を含む移動相3−6カラム体積、40−100%のB液を含む移動相0.5−1カラム体積、100%B液1−3カラム体積(CV)を流し、流速は0.8mL/minであり、紫外線検出器の検出波長は280nm/258nm/214nmであるステップと、
冷凍乾燥したポリペプチド試料をNano−RPLC BufferAに再び溶解させ、オンラインNano−RPLC液体クロマトグラフィーはEksigentnanoLC−U1traTM 2Dシステム(AB SCIEX)で行い、溶解させた前記試料は2μL/minの流速でC18予備カラム(100μm×3cm、C18、3μm、150Å)に掛け、流速を保持しながら10min洗浄して脱塩し、質量スペクトルはTriple TOF 5600システム(AB SCIEX)とナノリットル噴霧IIIイオン源(AB SCIEX、USA)を連用するシステムを用い、質量スペクトルより採集した原始wiffグラフファイルに対して、Protein Pilot Software v.4.5(AB SCIEX、USA)ソフトウェアでデータ加工処理を行って、アミノ酸配列がSEQ ID No.1であるサソリ毒耐熱合成ペプチドを得るステップと、
固相化学合成法で前記サソリ毒耐熱合成ペプチドを製造するステップと、
前記ポリペプチド合成過程において、目標ペプチドの構造に類似するポリペプチドが生成され、それに対して、高速液体クロマトグラフィー(HPLC)により純化を行い、その中、クロマトグラフィー条件は、クロマトグラフィーカラムはInertsil ODS−SP 4.6mm*250mmであり、移動相は0.1%トリフルオロ酢酸を含むアセトニトリル−0.1%トリフルオロ酢酸を含む水であり、紫外線検出器の検出波長は214nmであるステップと、
質量スペクトルを用い、前記合成ポリペプチドの構造解析を行い、検索及び解析を経て、サソリ毒耐熱合成ペプチドのアミノ酸配列を確定し、そのアミノ酸配列がSEQ ID No.1であるステップを含む、ことを特徴とするサソリ毒耐熱合成ペプチドの製造方法。
付記1に記載のサソリ毒耐熱合成ペプチドのてんかん、アルツハイマー病やパーキンソン病の治療薬としての用途。

- アミノ酸配列がSEQ ID No.1である、ことを特徴とするサソリ毒耐熱合成ペプチド。
- 請求項1に記載のサソリ毒耐熱合成ペプチドのてんかん、アルツハイマー病やパーキンソン病の治療薬の製造のための使用。
