Description
合成されたペプチドは、Genscript(New Jersey、USA)から得た。ペプチドは、Flexpeptide technology方法で合成され、高速液体クロマトグラフィー及び質量分析計を用いて確認した。合成されたペプチドのアミノ酸配列は、以下の表1の通りである。
SGGAG(配列番号8)、
GAGGAGGAG(配列番号9)
スコポラミン(Scopolamine)は、Sigma-Aldrich(St. Louis、MO、USA)から購入した。4週齢の雄ICRマウスをKorean BioLink Co. (Chungbuk、Korea)から購入した。 1週間の適応期間の後、試験に使用し、すべての試薬は、腹腔内投与した。記憶障害は、検査30分前のスコポラミンの注入によって誘導され、合成されたペプチドは、スコポラミンの注入30分前に注入した。
受動回避反応検査は、同一の明るい部屋及び暗い部屋で行った。各部屋の床は、2-mmのステンレス棒が1-cm間隔で離れている形になっている。明るい部屋(20×20×20cm)は、100-Wの電球を備えている。これらの部屋は、ギロチンドア(guillotine door)で連結されている。
マウスを一方の枝の長さが30cm、幅5cm、高さ13cmからなるY迷路の片端に置いて各枝に入る順番を記録した。変更(alternation)は、3つの他の枝に順次入った場合、成功したものと判断した。変更行動力(spontaneous alternation)は、次の数式で定義した。
Claims (14)
- GGAG、QGAG、SGAGAG、QAGAG、SGGAG、及びGAGGAGGAGからなる群から選択されるアミノ酸配列からなる、ペプチド。
- 前記ペプチドは、人工的に合成されたことを特徴とする、請求項1に記載のペプチド。
- GGAG、AGAG、QGAG、SGAGAG、QAGAG、SGGAG、及びGAGGAGGAGからなる群から選択されるアミノ酸配列からなるペプチドを有効成分として含んでなる、記憶、認知、又は学習障害の予防用又は治療用薬学的組成物。
- 前記記憶、認知、又は学習障害は、老化、アルツハイマー病、精神分裂症、パーキンソン病、ハンチントン病、ピック病、クロイツフェルト-ヤコブ病、うつ病、頭部外傷、脳卒中、CNS低酸素症、脳虚血症、脳炎、健忘症、外傷性脳損傷、低血糖症、ウェルニッケ・コルサコフ症候群、薬物中毒、雷電症、癲癇、海馬硬化症、頭痛、脳老衰症、痴呆、前頭側頭葉変性、腫瘍、正常圧水頭症、HIV、脳血管性疾患、脳神経疾患、心血管系疾患、記憶喪失、被曝、代謝性疾患、甲状腺機能低下症、軽度認知障害、認知欠乏、又は注意力欠陥による記憶、認知、又は学習障害を特徴とする、請求項3に記載の薬学的組成物。
- GGAG、AGAG、QGAG、SGAGAG、QAGAG、SGGAG、及びGAGGAGGAGからなる群から選択されるアミノ酸配列からなるペプチドを有効成分として含んでなる、頭脳又は認知機能の増進用食品組成物。
- 前記頭脳又は認知機能は、学習能力、記憶能力、又は集中力であることを特徴とする、請求項5に記載の食品組成物。
- 請求項1又は請求項2に記載されたペプチドをコードする、ポリヌクレオチド。
- 請求項7に記載されたポリヌクレオチドを含んでなる、組換えベクター。
- 請求項8に記載された組み換えベクターで形質転換されてなる、宿主細胞。
- 請求項9に記載された宿主細胞を培養するステップを含んでなる、ペプチドの製造方法。
- 記憶、認知、又は学習障害の予防用又は治療用薬剤の製造のための、GGAG、AGAG、QGAG、SGAGAG、QAGAG、SGGAG、及びGAGGAGGAGからなる群から選択されるアミノ酸配列からなる、ペプチドの使用。
- 前記記憶、認知、又は学習障害は、老化、アルツハイマー病、精神分裂症、パーキンソン病、ハンチントン病、ピック病、クロイツフェルト−ヤコブ病、うつ病、頭部外傷、脳卒中、CNS低酸素症、脳虚血症、脳炎、健忘症、外傷性脳損傷、低血糖症、ウェルニッケ・コルサコフ症候群、薬物中毒、雷電症、癲癇、海馬硬化症、頭痛、脳老衰症、痴呆、前頭側頭葉変性、腫瘍、正常圧水頭症、HIV、脳血管性疾患、脳神経疾患、心血管系疾患、記憶喪失、被曝、代謝性疾患、甲状腺機能低下症、軽度認知障害、認知欠乏又は注意力欠陥による記憶、認知、又は学習障害を特徴とする、請求項11に記載のペプチドの使用。
- 頭脳又は認知機能の増進用食品の製造のための、GGAG、AGAG、QGAG、SGAGAG、QAGAG、SGGAG、及びGAGGAGGAGからなる群から選択されるアミノ酸配列からなる、ペプチドの使用。
- 前記頭脳又は認知機能は、学習能力、記憶能力又は集中力であることを特徴とする、請求項13に記載のペプチドの使用。
