Description
[1]コクサッキーウイルスB3野生型(CVB3-WT)のゲノムに、組織特異的microRNA(miRNA)の標的配列からなるポリヌクレオチドが少なくとも一つ挿入された変異ゲノムを含む、組織特異的に増殖が抑制された改変コクサッキーウイルス。
[2]挿入される位置が、CVB3-WTゲノムの3'UTR領域内である、1に記載の改変コクサッキーウイルス。

<miR標的配列挿入によるCVB3の安全性向上>
本発明によって提供される改変コクサッキーウイルスの一態様は、コクサッキーウイルスB3野生型(CVB3-WT)のゲノムに、組織特異的microRNA(miRNA)の標的配列からなるポリヌクレオチドが少なくとも一つ挿入された変異ゲノムを含む、改変コクサッキーウイルスである。このような改変コクサッキーウイルスは、組織特異的に増殖が抑制されうる。

<GM-CSFの挿入による攻撃性向上>
本発明によって提供される改変コクサッキーウイルスの別の一態様では、CVB3-WTのゲノムに、顆粒球単球コロニー刺激因子(GM-CSF)をコードする領域が発現可能に挿入されている。GM-CSFをコードする領域を発現可能に挿入するためには、mGM-CSFの遊離方法、遺伝子の挿入部位、プロテアーゼ認識配列の改変について、考慮することができる。mGM-CSFの遊離方法の例としては、3Cプロテアーゼ認識配列の利用、2Aプロテーゼ認識配列の利用、IRESの利用、2Apeptideの利用が挙げられ、遺伝子の挿入部位の例としては、CVB3-WTのゲノムの翻訳開始地点ATG直下(VP4の上流)、2A遺伝子の上流、3'UTRが挙げられ、プロテアーゼ認識配列の改変としては、CVB3以外のウイルス由来配列への改変が挙げられる。
本発明の改変ウイルスは、WT-CVB3を遺伝子操作することにより調製できる。WT-CVB3は、検体等から既知のウイルス単離方法によって単離できる。ウイルス単離方法の例は、遠心分離、培養細胞によるウイルスの増殖等である。いったん改変コクサッキーウイルスが調製されれば、ウイルスの生産のための生物学的な種々の方法を用いて、当該改変コクサッキーウイルスを増やすことができる。
<適用疾患>
本発明によって提供される医薬組成物の一態様は、上述の改変CVB3を有効成分として含む。医薬組成物が処置の対象とする癌種は、特に限定されず、固形癌および液性癌を含む。CVB3は、固形癌および液性癌の癌細胞に対する細胞傷害性を有する。CVB3による癌細胞に対する細胞傷害性は、ウイルスが癌細胞に感染し、癌細胞の細胞質で複製することによる癌細胞の溶解によるか、ウイルスの感染に起因する癌細胞内のカスパーゼの活性化によるアポトーシスによる。CVB3は、細胞表面のCARを認識して、当該細胞に感染することができる。なお、本発明に関し、疾患または状態に関連して「処置(する)」というときは、予防および治療を含む。
また、本実施の形態に係る医薬組成物は、種々の剤形とし、種々の投与経路を採ることができる。すなわち、本実施の形態に係る医薬組成物は、局所用の製剤とすることもでき、全身投与用の製剤とすることもできる。例えば、注射剤または点滴剤とし、癌種に応じて腫瘍内投与、静脈投与、胸腔内投与、腹腔内投与とすることができる。特に、食道癌、大腸癌等の消化器癌の多くは、内視鏡等で腫瘍組織を視認しながら、医薬組成物を腫瘍組織に直接注射できる。この場合、内視鏡等で注射した部位を確認できるため、出血しても対処しやすいという利点もある。他に、経口投与でもよいし、筋肉、皮下、直腸、膣 、鼻腔等を介して投与してもよい。
本実施の形態に係る医薬組成物は、抗癌剤と併用してもよい。当該医薬組成物と異なる作用機序を有する抗癌剤を併用することで、抗腫瘍効果の向上が期待できる。抗癌剤は、特に限定されないが、小細胞肺癌、非小細胞肺癌、肺扁平上皮癌、悪性中皮腫、大腸癌、結腸直腸癌、食道癌、下咽頭癌、ヒトBリンパ腫、子宮頸癌および膵癌等の治療に用いられるものが望ましい。具体的には、抗癌剤は、CDDP(シスプラチン)、ゲフィチニブ、オキサリプラチンなどである。

本研究の目的は腫瘍溶解性野生型CVB3(CVB-WT)で見られた膵臓・心筋における副作用軽減である。CVB3-WTは膵臓に特に激しい炎症を引き起こし、血清生化学検査においてAMYの著しい上昇、H&E染色組織像において外分泌腺の破壊が確認される。これらの問題を克服するため、我々はCVB3-WTのゲノム3' untranslated region(UTR)に組織特異的microRNA(miRNA)の標的配列を挿入し、予想される副作用の軽減を目指した。挿入したmiRNAの標的配列に組織特異的なmiRNAを有したRISC複合体が結合しウイルスタンパク質の翻訳等を阻害するため、組織特異的な増殖抑制が可能となる。
我々は2つのmiRNAに着目し、1つは膵臓特異的に発現していると言われているmiR-217、もう1つは筋組織および正常細胞特異的に発現していると言われているmiR-1を組み合わせ、計4つmiRNA標的配列が連続している二種類のmiRNA標的配列(miR-1×2&miR-217×2、miR-217×4)を作製した。二種類の標的配列をそれぞれCVB3ゲノムの7304-7305bpの間にoverlap extension PCR法を用いて挿入した。また、同時に概出の標的配列をCVB3ゲノムの7344-7345bpの間にoverlap extension PCR法にて挿入した(図1)。
以降、遺伝子改変CVB3-miRTは7304-7305 bpに挿入したものを指す。また、特に記載した場合を除き、CVB3-miR-1&217Tとは、図1に示したmiR-1とmiR-217Tとを2つずつ(配列番号1のポリヌクレオチド)挿入したものを指し、またCVB3-miR-217Tとは、図1に示したmiR-217Tを4つ(配列番号2の配列のポリヌクレオチド)挿入したものを指す。
次に、HeLa細胞を用いて遺伝子改変CVB3-miRTを作製した際、他の二種類(CVB3-WT、CVB3-miR-217T)と比較してCVB3-miR-1&217Tにおいてのみ増殖の遅延が認められた(図3)。
上記の結果を踏まえ、CVB3-miRTの適切な産生細胞を決定するため、HeLa細胞およびmiRNAの発現が低かったH1299細胞を用いてウイルス産生および力価の測定を行った。H1299細胞で産生したCVB3-miR-1&217TはHeLa細胞で産生したウイルスよりも5倍ものウイルス力価を呈した(図6)。また、HeLa細胞で産生したCVB3-miR-1&217Tを用いて、H1299細胞およびHeLa細胞でウイルス力価の測定を行ったところ、H1299細胞でのウイルス力価はHeLa細胞でのウイルス力価よりも10倍以上高い値を示した(図7)。これらのことから、CVB3-miRTのウイルス産生およびウイルス力価測定にはH1299細胞が適切であることが明らかとなった。
遺伝子改変CVB3のin vivo抗腫瘍効果および毒性を検討した。0日目にヌードマウスの右側腹部に腫瘍(非小細胞肺癌、NCI-H1299細胞)を接種し、2日目より計5回、CVB3ウイルスを隔日で腫瘍内に投与(5×106 TCID50 / 回)した。
以下のプロトコールにしたがって、本研究は行われた。

10.翌日、コロニーを拾い、LB培地にて培養する。

1.室温で以下の順に0.2 ml PCRチューブに混合する。(Total 20 μl)
3.3 h後、反応中のチューブにTURBO DNaseを1 μl加え、よく攪拌し、追加で37℃、15 min。
4.事前に1.5 mlチューブにUltraPure 114 μlと付属のAmmonium Acetate Stop Solutionを15 μlを混合した溶液を本数分×1.1倍作製しておく。
5.反応後、チューブに上記の混合液を129 μl加え、ピペッティング。
6.全量を0.7 mlチューブに移す。

1.分注したOpti-MEMにRNAを80 μgになるように液中に加え、もう別チューブに分注したOpti-MEMにLipofectamine 2000を120 μl液中に加える。
2.両者、室温、5 min静置。
3.両チューブを混合し、室温、20 min静置。
4.20 min後、混合液全量をNCI-H1299細胞に添加する。
5.4 h後、Growth mediumに置換する。
6.37℃、5% CO2、24 hインキュベート。
7.Transfectionから24 h後、培地を捨てないまま各NCI-H1299 cellsをセルスクレーパーで剥離する。
8.細胞ごと上清を50 mlチューブに回収する。
9.50 mlチューブをLiquid N2に浸けて凍結させる。
10.37℃ウォーターバスで解凍。
11.5〜6の凍結融解を2回繰り返す。 (計3回Freeze-Thaw)
12.4℃、3,000 rpm、15 min遠心。
13.上清を移すNCI-H1299 cellsの培地を新鮮な培地に置換する。(15 ml)
14.遠心後の上清全量をNCI-H1299 cellsに加える。
15.37℃、5% CO2、5〜6 hインキュベート
16.CPEが60〜70%以上確認された時点に、培養上清を除去。
17.Opti-MEM(登録商標)Iを2 ml加える。
18.セルスクレーパーで細胞を剥離。
19.細胞ごとOpti-MEM(登録商標)Iを全量、15 mlチューブに回収する。
20.この15 mlチューブをLiquid nitrogenに浸けて凍結させる。(約2 min)
21.37℃ウォーターバスで解凍。(約10 min)
22.5〜6の凍結融解を2回繰り返す (計3回Freeze-Thaw)
23.4℃、3,000 rpm、15 min遠心。
24.上清を-80℃で保存。

5.ウイルス10-2液を96 well Round plateの1列目に120 μlずつ添加。
6.1列目のウイルス液を、電動8連ピペッターを使用して20 μlずつ2列目に移す。
7.そこで一度ピペッティングを行い、チップを変える。
8.直前に希釈した列から20 μlずつ取り、隣の列に移す。
9.一度ピペッティングを行い、チップを変える。
10.8.〜9.を11列まで繰り返す。
11.NCI-H1299細胞を播種したFlat plateをインキュベーターから取り出す。
12.8連ピペッターを用いて、Round plateの薄い12列目から順に、Flat PlateのNCI-H1299細胞に50 μlずつ加える。
13.37℃、5% CO2、120 h(5日間)インキュベート。
14.50% CPEが見られたwellをカウントする。以下の式を用いてTiterを算出する

1.NCI-H2199 cellsを150 mm dish 16枚に播種。
2.4枚を剥がし50 mlチューブに回収。計4本の50mlチューブを遠心 (4℃、1,000 rpm、5 min)。
3.2本のペレットをPBS 20 mlに懸濁。これらを混ぜ合わせ計40 mlの懸濁液を作製。
4.4℃、1,000 rpm、5 min遠心。
5.NCI-H1299 cellsを5×107 cells/mlになるようにPBSで再懸濁。
6.BALB/c nu/nuの右側腹部に上記懸濁液を100 μl皮下注射する。
7.2日後、5×106 TCID50/50 μlになるようにOpti-MEMで希釈した各々のウイルスを腫瘍内投与する。
8.上記のウイルス投与は隔日で行い、計5回投与する。
9.隔日で腫瘍径(長径、短径)、体重を測定する。
8.更に2日後、マウスから血液および臓器を採取する。
9.血液は遠心分離後、上清を生化学検査に用いる。臓器はPFA固定後、脱水を行い、H&E染色を行う。
我々は野生型CVB3(CVB-WT)では腫瘍の完全治療が認められなかった事例に対して、更に腫瘍退縮効果を高める目的で、顆粒球単球コロニー刺激因子(GM-CSF)が挿入された遺伝子改変ウイルスの作製を試みた。
我々は以下の三点に注目して遺伝子改変の条件を検討した。1つ目はmGM-CSFの遊離方法(3Cpro、2Apro、IRES、2A peptide)。2つ目は遺伝子の挿入部位(CVB3-WTのゲノムの翻訳開始地点ATG直下、2A遺伝子前、もしくは3'UTR)。3つ目はプロテアーゼ認識配列の違い(他ウイルス由来配列との比較)。検討結果をまとめた表を示す。
まず、エンテロウイルス属の遺伝子改変で報告のある3Cproを利用しATG直下に挿入した。その結果、mGM-CSF遺伝子を挿入したCVB3は細胞変性効果(CPE)が見られなかった。しかしながら、対照として作製し確認できたEGFP挿入CVB3(CVB3-EGFP)は、既存の報告通りEGFPの発現が確認されCPEが見られた(図10)。このことから、mGM-CSFを挿入するには3Cproを利用しATG直下に挿入することは不適切である可能性が示唆された。なお、3rd Generationおよび4th Generationとして、IRESまたは2A peptideを利用し、3'UTR(STOPコドン上流または下流)に挿入することを試みたが、同様の結果であった。
上の結果を受け、蛋白質遊離方法として3Cpro、2Apro、IRES、2A peptideのどれがCPEを誘導しmGM-CSFの遊離ができるかを検討したところ、2Aproを利用し2A遺伝子前の直前にmGM-CSFを挿入したCVB3がCPEを誘導し、かつmGM-CSFの発現がELISA法によって確認できた。さらに、mGM-CSFと同時に挿入した2Apro認識配列の検討を行ったところ、CVB3由来2Apro認識配列を用いるより、Poliovirue由来2Apro認識配列を用いたものがCPEを誘導し(図12)、かつより高いウイルス力価のCVB3が得られることが判明した。しかしながら、2Aproを利用し2A遺伝子前の直前にmGM-CSFを挿入したCVB3においても、in vivo実験に適用可能なウイルス力価が得られなかった。
上の結果を受け、in vivo実験に使用可能な高いウイルス力価を有するCVB3-EGFPの挿入部位であるATG直下に再び着目した。このATG直下に、2Aproを利用したCVB3-mGM-CSFを作製した。in vivo実験に使用可能な高力価のCVB3-mGM-CSFを得ることができた。なお、Poliovirue由来3Cpro利用およびATG直下へのmGM-CSFの挿入(7th Generation)では、十分なウイルス力価が期待できなかった。
遺伝子改変CVB3-GM-CSF(挿入した配列は、上記の8th Generation CVB3-GM-CSF-LTTY。配列番号4のポリヌクレオチド。)のin vivo抗腫瘍効果を検討した。0日目にC57BL/6マウスの右側腹部に腫瘍(マウス肺癌、TC-1細胞)を接種し、4日目より計2回、CVB3-WTあるいはCVB3-GM-CSFを隔日で腫瘍内に投与(5×106 TCID50 / 回)した。その結果、遺伝子改変CVB3-GM-CSFにより、WTより高い腫瘍溶解性が奏された。
[小括]
本発明はmGM-CSFをエンテロウイルス属に挿入した初めて発明であり、他のウイルスでmGM-CSF挿入しているこれまでの報告とは全く違う方法でmGM-CSFを発現遊離している。エンテロウイルスの細胞変性効果を保持したまま外来性タンパク質を発現させるという考え方は病原性の研究が行われていたエンテロウイルスでは極めて異端であり、本研究の独自性および新規性は極めて高いものと考えられる。

1.Milli-Qを600 mlほどオートクレーブ滅菌。
2.冷めたら、Wash Buffer Concentrate(25×)を25 ml加えて全量625 ml(1×)に希釈する。
3.必要分の付属のmicroplate stripをプレートフレームにセットする。不要分はホイル内に戻す。
4.各wellにAssay Diluent RD1Wを50 μlずつ添加する。
5.スタンダードを薄い方から順に各well(Assay Diluent添加済)に50 μl添加。サンプルを同様に各well(Assay Diluent添加済)に50 μl添加し、フレームを緩やかに1 minタッピング。
6.カバーシールをしっかりとし、室温で2 hours静置。
7.各wellの液を吸引し、Wash Buffer(上記2.で作製済み)を400 μl添加する。
8.上記7の作業を計5回繰り返す。最後の時に清潔なキムタオルで水気を取る。
9.各wellにMouse GM-CSF Conjugateを100 μlずつ加える。
10.新しいカバーシールを張りつけ、室温で2 hours静置。
11.上記の洗浄作業を繰り返す。
12.各wellにSubstrate Solutionを100 μl添加する。
13.シールはせずに室温、暗所で30 min静置する。
14.各wellに100 μlのStop Solutionを添加する。
15.混ざるまで緩やかにプレートをタッピング。
16.30 min以内にプレートリーダーで450 nmにおける吸光度を測定する。
(Materials & reagents)
・150 mm dish
・TC-1 cells (1 ×106 cells/100 μl PBS/mouse) (03/20 Thawing T175 2 passage)
・RPMI 1640 w/10% FBS、1 mM sodium pyruvate (Growth medium)
・PBS
・18Gニードル Invitrogen Cat# 11668-019 Lot# 890730
・27Gニードル
・1 ml テルモシリンジ
・ウイルス
CVB3-WT (3.33×108 TCID50/ml : 約15 μl/mouse)
CVB3-mGM-CSF (1.12×108 TCID50/ml : 約45 μl/mouse)
・1 ml テルモシリンジ (29G)
・Opti-MEM(登録商標)I
1.TC-1 cellsを150 mm dish に播種。
2.4枚を剥がし50 mlチューブに回収。計4本の50mlチューブを遠心 (4℃、1,000 rpm、5 min)。
3.2本のペレットをPBS 20 mlに懸濁。これらを混ぜ合わせ計40 mlの懸濁液を作製。
4.4℃、1,000 rpm、5 min遠心。
5.TC-1 cellsを1×107 cells/mlになるようにPBSで再懸濁。
6.C57BL/6の右側腹部に上記懸濁液を100 μl皮下注射する。
7.4日後、5×106 TCID50/50 μlになるようにOpti-MEMで希釈した各々のウイルスを腫瘍内投与する。
8.上記のウイルス投与は隔日で行い、計2回投与する。
9.隔日で腫瘍径(長径、短径)、体重を測定する。

Claims (11)
- コクサッキーウイルスB3野生型(CVB3−WT)のゲノムに、組織特異的microRNA(miRNA)の標的配列からなるポリヌクレオチドが少なくとも一つ挿入された変異ゲノムを含む、組織特異的に増殖が抑制され、
前記ポリヌクレオチドが挿入される位置が、CVB3−WTゲノムの位置7304と位置7305との間であり、
前記組織特異的miRNAが、miR−1および/またはmiR−217であると共に、
挿入されるポリヌクレオチドが、下記(a)若しくは(b)、または(a)若しくは(b)において1〜数個のヌクレオチドを欠失、置換若しくは付加した配列からなるポリヌクレオチドであり、
CVB3−WTが感染可能な癌細胞を標的細胞とする改変コクサッキーウイルス。

- 組織特異的miRNAが、膵臓および/または心筋に発現しているものである、請求項1または2に記載の改変コクサッキーウイルス。
- 標的配列からなるポリヌクレオチドが複数個(例えば2〜6個)挿入された、請求項1〜3のいずれか1項に記載の改変コクサッキーウイルス。
- 変異ゲノムがさらに、顆粒球単球コロニー刺激因子(GM−CSF)をコードする領域とその下流に連結された2Aプロテアーゼ認識配列をコードする領域とを含むポリヌクレオチドが、翻訳開始地点のATGの直下に、機能可能に挿入されたものであり、
前記2Aプロテアーゼ認識配列が、ポリオウイルス由来のLTTYで表されるアミノ酸配列である、請求項1〜4のいずれか1項に記載の改変コクサッキーウイルス。 - CVB3−WTゲノムにおいて、GM−CSFをコードする領域とその下流に連結された2Aプロテアーゼ認識配列をコードする領域とを含むポリヌクレオチドが、翻訳開始地点のATGの直下に、機能可能に挿入された変異ゲノムを含み、
前記2Aプロテアーゼ認識配列が、ポリオウイルス由来のLTTYで表されるアミノ酸配列である、
改変コクサッキーウイルス。 - 請求項1〜6のいずれか1項に記載の改変コクサッキーウイルスを含む医薬組成物。
- 癌またはその前癌状態の処置(予防または治療)のための、請求項7に記載の医薬組成物。
- 癌またはその前癌状態の処置(予防または治療)において使用するための、請求項1〜6のいずれか1項に記載の改変コクサッキーウイルス。
- 請求項1〜6のいずれか1項に記載の改変コクサッキーウイルスの、力価測定または増殖のための、対応する組織特異的miRNAの発現量が高くない細胞(例えば、H1299)細胞の使用。
- 局所用または全身投与用の製剤である、請求項7または8に記載の医薬組成物。
