Description
繊維は、例えば中間長さの糸もしくはモノフィラメントなどの連続長さの形態にあってよい、または結晶成長法などによって形成された、個別ピースへ連続長さの繊維を切断して、例えば不織布もしくは織布などのより短い個別繊維に形成されてもよい。
吸着性相互作用に基づく生体分子の分離および生物学的分離を可能にするために、このような繊維表面を官能化するための化学処理法が提供されている。化学処理法は、イオン親和性、または疎水性相互作用もしくは相互作用の組み合わせに基づいて、このような繊維に様々な表面化学的官能性を付与することができる。繊維製造および単純な表面化学処理工程を組み合わせた経済性は、生物薬剤学ならびにワクチン製造における精製作業のための経済的および容易に拡張可能な技術を生み出す。
図1から6は、非多孔質側壁34によって規定された内部区画32を有する、ならびに内面36、外面38、上部外周62で終了する第1端60、底面周囲72で終了する第2端70、液体サンプルを受け入れるための入口52、処理液体を放出するための出口54および本体の内部区画内に含有された高表面積ポリマー吸着素子40であって、ポリマー吸着素子が第1端42および第2端44を有する高表面積ポリマー吸着素子40を有する柔軟性折り畳み式本体であって、第1端および第2端は本体の内面に取り付けられている柔軟性折り畳み式本体30を含む使い捨ての柔軟性サンプル調製デバイス20を描出している。
材料は、単一層材料から構成できる、または二重壁容器を形成するために合わせてシールもしくは分離されているどちらかの2つ以上の層を含むことができる。層が合わせてシールされる場合、材料は積層もしくは押出材料を含むことができる。積層材料は、引き続いて接着剤によって一緒に固定される2枚以上の別個に形成された層を含んでいる。
一部の実施形態では、本発明は、サンプルが官能化成形繊維の吸着性カーテンに接触するにつれて、液体サンプルおよび/または供給物から関心対象の生体分子を単離するためのシステムを提供する。液体サンプルには、細胞、幹細胞、モノクローナル抗体(mAbs)、タンパク質、抗体、ペプチド、オリゴペプチド、核酸、オリゴヌクレオチド、RNA、DNA、オリゴ糖類および多糖類を含む関心対象の1つ以上の生体分子を含有する未浄化の液体供給物およびサンプルが含まれる。
ある種の実施形態では、本発明は、デバイス、デバイスを使用する方法およびデバイスを製造する方法を提供するが、このとき官能化成形繊維が関心対象の種を吸着すると、残りの固体を含む流体はデバイスから排出される。
空のバッグデバイス20内に溶離液バッファーを(完全に湿潤させるために十分に)添加する。
デバイス20を使用する固体クリアランス
本発明のある種の実施形態を使用して、液体サンプルからの細胞/細胞破片などの固体のクリアランスを証明するための実験を実施した。
同一バッファー量を使用して、タンパク質分離は、0.17mg/mLのmAb04および1mg/mLのBSAを用いてスパイクした20mM MES+0.1M NaCl(pH6)バッファーを対象に実施した。

Claims (11)
- 使い捨ての柔軟性液体サンプル調製デバイス20であって:
側壁34によって規定された内部区画32、内面36、外面38、入口52、出口54および内部区画内に含有された、カーテン様構造のポリマー成形繊維床を含むポリマー吸着素子40を有する折り畳み式本体30であって、ポリマー吸着素子は第1端42および第2端44を有し、第1端および第2端は側壁の内面に取り付けられている、折り畳み式本体30
を含む、使い捨ての柔軟性液体サンプル調製デバイス20。 - 本体は、二次元枕形バッグである、請求項1に記載のデバイス。
- ポリマー成形繊維床は、連続ポリマー成形モノフィラメント繊維を含む、請求項1に記載のデバイス。
- 成形繊維は、カチオン交換官能性またはアニオン交換官能性を提供する表面ペンダント官能基をさらに含む、請求項1に記載のデバイス。
- 成形繊維は、繊維の中心を上から下へ走る長手軸を含み、中央領域から外側へ半径方向に伸びる複数の突出部を有する中央領域を備える翼形断面を有する、請求項1に記載のデバイス。
- 成形繊維は、ポリオレフィン、熱可塑性ポリマー、ポリプロピレン、ポリエステル、ポリエチレン、ポリアミド、コポリエステル、液晶ポリマーおよび熱可塑性エラストマー、熱可塑性ポリウレタンエラストマーおよびエラストマーオレフィンを含む、請求項1に記載のデバイス。
- 成形繊維は、実質的に互いに平行なループに組み立てられる、請求項3に記載のデバイス。
- 使い捨ての柔軟性枕形液体サンプル調製デバイス(20)を製造する方法であって:
a.各シートが外周を有する2枚のポリマーシートを提供する工程、
b.少なくとも1つの吸着性ポリマー接着素子(46または48)に取り付けられたポリマー吸着素子(40)を提供する工程であって、該接着素子が第1端および第2端を有し、前記ポリマー吸着素子がカーテン様構造のポリマー成形繊維床を含む工程:
c.ポリマー吸着素子および接着素子を2枚のポリマーシートの間に、2枚のシートの外周が互いに重複し、接着素子の第1端および第2端が対向する重複する外周の間に挟まれるように配置する工程;
d.2枚のポリマーシートの対向する外周にしっかりと取り付けられたポリマー吸着素子を収容するための内部区画を有する枕形デバイスを形成する重複している外周にある2枚のポリマーシートならびに接着素子の第1端および第2端を一緒に接着する工程
を含む、使い捨ての柔軟性枕形液体サンプル調製デバイス(20)を製造する方法。 - 出口および入口が枕形デバイスにしっかりと取り付けられる、請求項8に記載の方法。
- 使い捨ての柔軟性枕形液体サンプル調製デバイスを製造する方法であって:
a.外周を有する単一ポリマーシートを提供する工程、
b.少なくとも1つの吸着性ポリマー接着素子に取り付けられたポリマー吸着素子を提供する工程であって、該接着素子が第1端および第2端を有し、前記ポリマー吸着素子がカーテン様構造のポリマー成形繊維床を含む工程、
c.ポリマー吸着素子および接着素子をポリマーシート上に配置する工程、
d.シートの外周が重複し、接着素子の第1端および第2端が対向する重複する外周の間に挟まれるように、ポリマーシートを折り畳む工程、
e.重複する外周ならびに接着素子の第1端および第2端でポリマーシートを結合する工程、
f.ポリマーシートの対向する外周にしっかりと取り付けられた吸着素子を収容するための内部区画を有する枕形デバイスを形成する工程
を含む、使い捨ての柔軟性枕形液体サンプル調製デバイスを製造する方法。 - 出口および入口が枕形デバイスにしっかりと取り付けられる、請求項10に記載の方法。
