5%のパラジウム/炭素(15g;7.0mmol)をN2でパージしたフラスコに加え、そして、EtOAc(250mL)を加える。溶液としての3−(メトキシメチル)−6−[1−(1H−ピラゾール−5−イル)エテニル]−3H−1,3−ベンゾチアゾール−2−オン(29g、101mmol)を、EtOAc(250mL)中に加える。吸引下で脱気して、そして、バルーンを介して水素をチャージする。一晩撹拌し、加圧下で過剰な水素を排気し、そして、N2をフラッシュする。ケイ藻土を通して濾過し、そして、濃縮して3−(メトキシメチル)−6−[1−(1H−ピラゾール−5−イル)エチル]−3H−1,3−ベンゾチアゾール−2−オンを濃黄色オイル(27g、93mmol;収率92%)として得る。さらに精製することなく、物質を次の工程に持ち込んでもよい。LCMS(低)、rt=1.73分、M+1=290。

3−(メトキシメチル)−6−{1−[1−(5−{2−(テトラヒドロ−2H−ピラン−2−イルオキシ)エトキシ}ピリジン−2−イル)−1H−ピラゾール−3−イル]エチル}−3H−1,3−ベンゾチアゾール−2−オン(140mg、274.18μmol)を、トリフルオロ酢酸(0.05M;73mmol;5.5mL)中に溶解し、そして、(〜72℃)N2下2時間還流加熱する(オレンジ色が、約10分後70℃にて観察される)。反応物を室温に冷却し、そして、減圧下で濃縮する。残渣をTHF(0.05M;67mmol、5.5mL)に溶解し、28%の水酸化アンモニウム(0.2M;9.9mmol;1.4mL)を加え、そして、室温で〜1時間撹拌する。EtOAc(100ml)で希釈し、ブラインで洗浄し、硫酸ナトリウムで乾燥し、濾過し、そして、濃縮する。へキサン中の60〜80%EtOAcの濃度勾配を使って溶出しながら、25gのシリカゲルカラム上での液体クロマトグラフィーにより精製して、6−(1−{1−[5−(2−ヒドロキシ−エトキシ)−ピリジン−2−イル]−1H−ピラゾール−3−イル}−エチル)−3H−1,3−ベンゾチアゾール−2−オンを白色フォーム(80mg;209μmol;収率76%)として得る。LCMS(低)、rt 1.8分、M+1=383。
6−(1−{1−[5−(2−ヒドロキシ−エトキシ)−ピリジン−2−イル]−1H−ピラゾール−3−イル}−エチル)−3H−1,3−ベンゾチアゾール−2−オン(330mg)を、MeOH(3.0mL)およびDCM(2.5mL)中に溶解する。そして、一回のラン当たり300μLのアリコートを注入し、225nmで検出し、70mL/分で40%MeOH/液体CO2を使って溶出しながら、Chiralcel(登録商標)OJ−H(2.1×15cm、5μmカラム、Chiral Technologies Europe)を使用するキラルクロマトグラフィーにより複数の光学異性体へと分離する。分離した光学異性体を含む画分を合わせて、そして吸引下で濃縮して、異性体1(150mg)および異性体2(169mg)を非晶質白色固形物として得る。225nmで検出して、5mL/分で40%MeOH/液体CO2を使って溶出しながら、Chiracel(登録商標)OJ−H(4.6×150mm、5μmのカラム、Chiral Technologies Europe)を使用する分析的キラルクロマトグラフィーは、異性体1をrt=2.03で、異性体2をrt=2.66minで特徴づけた。LCMS(低):異性体1、rt 1.8分、M+1=383;異性体2、rt 1.8分、M+1=383。
70℃で20分間撹拌して、6−((S)−1−{1−[5−(2−ヒドロキシ−エトキシ)−ピリジン−2−イル]−1H−ピラゾール−3−イル}−エチル)−3H−1,3−ベンゾチアゾール−2−オン(異性体1)(56.9mg)を、1:1のEtOH:ヘプタン(3.0mL)に溶解する。熱い間に溶液を濾過し、二つの等量ポーションへと分割する;その一つを、外気温のバイアルに移し、蓋をして、そして、5℃で一晩撹拌する。固形物を濾過により回収し、そして、窒素下で乾燥して、結晶型I(23.9mg、収率84%)を得る。
結晶性固形物のXRPDパターンを、CuKa源(λ=1.54060Å)とVantec検出器を備えたBruker D4 Endeavor粉末X線回折装置上で35kVおよび50mAで稼働しながら得る。サンプルを、2Θでの4〜40°で、2Θでのステップサイズ0.0087°及びスキャン速度0.5秒/ステップを用い、並びに0.6mmの発散、5.28mmの固定アンチスキャッターおよび9.5mmの検出器スリットを用いてスキャンする。乾燥粉末を石英サンプルホルダに詰めて、滑面をガラススライドを用いて得る。結晶学技術分野において周知なのは、任意の所定の結晶形に関して、回折ピークの相対強度が、複数因子(例、結晶形態および晶癖)から生じる優先配向に起因して異なる可能性があることである。優先配向の効果が存在する場所では、ピーク強度が変化するが、多形体の特徴的ピーク位置は変化しない。例えば、米国薬局方33−国民医薬品集28章<941>粉末X線回折法(XRPD)による結晶性固形物の特徴解析、公定、2010年10月1日〜2011年2月1日を参照されたい。さらに、任意の所定の結晶形に関して、ピーク角度位置が若干変化する場合があるということも結晶学技術分野では周知である。例えば、サンプルを解析する温度若しくは湿度、サンプル変位、または内部標準の存在若しくは非存在の変動に起因して、ピーク位置はシフトする場合がある。本件では、2Θでの±0.2のピーク位置変動は、記載される結晶形の明白な同定を妨げることなく、これらの潜在的変動を考慮にいれることにする。結晶形の確認は、区別可能な(°(2Θ)単位での)複数ピーク、典型的にはより顕著な複数ピークの任意のユニークな組み合わせに基づいて実行する。外気温および相対湿度で得られる結晶形回折パターンを、8.85および26.77°(2Θ)でのNIST675標準ピークに基づいて調整する。
AMPA受容体アンタゴニスト化合物のTARPサブタイプ選択性を、海馬に多く含まれるTARP(TARPγ8)または小脳に多く含まれるTARP(TARPγ2)のいずれかを有するCHO−S細胞に共発現するAMPA GluA1フロップアイソフォーム受容体サブユニットにおける化合物活性を比較することにより実証する。AMPA受容体アンタゴニスト化合物のTARP依存性を、共発現TARPが存在しないCHO−S細胞に発現するAMPA GluA1フリップアイソフォーム受容体サブユニットにおける活性と、上記活性とを比較することにより実証する。
スプラーグドーリーラットの雄(Harlan Sprague Dawleyより、インディアナポリス、IN)で試験時の体重が90〜110グラムのものを、標準的照明サイクル(午前6時に点灯、午後6時に消灯)の大型コロニー部屋の中で、自由に飼料と水にアクセスさせて、1ケージに5匹飼育する。動物は、試験前少なくとも3日間コロニー部屋で維持される。動物を試験開始1時間前に静かな部屋に移動する。動物の使用は一度きりである。

2 動物がスクリーンから落ちてしまった場合。
スクリーン試験を完了した後、動物に生理食塩水中の35mg/kgのPTZを1ml/kgの容量で皮下投薬する。その動物を、その後観察ケージに入れて、PTZ後30分間観察する。クローヌスは、ラットが起き上がれなくなっている際の前脚と後脚の間代性発作として定義される。強直性発作は、強直に後脚が伸びて起き上がれなくなったものとして定義される。観察期間中に死んだ場合も記録される。観察期間中の任意の時間での特異的発作型の存在に従って、動物を点数化する。所定の発作型を有する動物の数としてデータを報告する(例えば、4/5間代性発作とは、動物5匹中4匹が観察期間中の任意の時間で任意の期間少なくとも一回の間代性発作を示したことを意味する)。
マニュアルホルマリン誘導性疼痛モデルは、化合物を鎮痛剤特性に基づいてスクリーニングすることに関して周知である(Mogil J.S.ら、「Heritability of nociception I: Responses of 11 inbred mouse strains on 12 measures of nociception.(痛覚の遺伝率I:11種類の純系マウスの痛覚の12種類の測定に関する応答)」、Pain、80、(1999)、67−82)。アッセイを、Plexiglas(登録商標)箱(約10cm×10cm×10cmのサイズのもの)の中で実施する。ケージの背面に設置されるミラーは、ホルマリンを注入した脚の観察が妨げられないことを可能にする。非絶食雄マウス(Harlan(HSD)CD1−Icr)を、実験前少なくとも60分間それぞれ個人用小室に入れる。全ての試験は、08:00〜16:00時の間に行い、試験室の温度を21〜23℃に維持する。複数用量の試験化合物(3、10、および30mg/kg)、ビヒクル(10%アカシア中の5%DMSO、0.05%消泡剤)、およびポジティブコントロール(1%HEC中のトラマドール80mg/kg、0.25%Tween80、0.05%消泡剤)を、ホルマリン注入する前の異なる時間に末梢(経口)投与する。ホルマリン(0.9%生理食塩水中の5%溶液20μL)を、左後脚の足底に27ゲージ針を使って皮下注射する。ホルマリン注射直後に、観察を開始する。5分間の間隔中における舐める行為のそれぞれが持続する秒数を記録することにより、ホルマリン誘導性疼痛を定量する。疼痛スコアを、ホルマリン注射後60分間測定する。二相の疼痛行動が、以前に記載されるように観察される(Wheeler−Aceto, H.、Porreca, F.およびCowan, A.、「The rat paw formalin test: comparison of noxious agents(ラット脚ホルマリン試験:有害物質の比較)」、Pain、40(1990)、229−238)。初期相は、ホルマリン注射直後に始まり、約5分間持続する。その後、後期相が10〜15分で開始し、最大応答がホルマリン注射後約25〜40分で典型的に観察される。60分間の観察期間の後、動物をCO2とその後頚椎脱臼により屠殺する。ホルマリン試験に関して報告される様々なスコアパラメータの中で、注射された脚を舐めたり噛んだりするのに使われた合計時間が最も適切であると考えられる。(Abbottら、「The formalin test: scoring properties of the first and second phases of the pain response in rats(ホルマリン試験:ラットの疼痛応答の第一および第二相のスコア特性)」、Pain、60(1995)、91−102;Coderreら、「The formalin test: a validation of the weighted−scores method of the behavioral pain rating(ホルマリン試験:行動的疼痛評価の重み付けスコア方法の検証)」,Pain、54(1993)、43−50)。初期相スコアは、0時〜5分の間に舐めるのに要した時間(秒)の合計である。後期相スコアは、観察期間の15分〜55分の間に舐めるのに要した合計秒数を加えて得られる。データを、平均値と平均値の標準誤差(±SEM)として提示する。データを、一元配置分散分析(ANOVA)と両側比較ダネット「t」検定により解析される適切な対比とにより評価する。もしP値が0.05未満の場合、差異は有意であると考えられる。(Abbott(上記);Coderre(上記);およびWheeler−Aceto(上記))。
