− 網膜の脈絡膜部分における血管新生部位への上皮細胞のホーミングおよび付着、
− SDF−1は、成体の骨髄における幹細胞および前駆細胞、例えば造血前駆(通常CD34+)細胞の維持に必要とされる、
− SDF−1は、プレB細胞の増殖を支持し、骨髄B細胞前駆細胞の成長を増大させ、プレB細胞およびプロB細胞の特異的遊走を誘導するが、成熟B細胞の有意な化学誘引物質として作用していない、
− SDF−1は、最も効果的なT細胞化学誘引物質の1種である、
− SDF−1およびその受容体CXCR4は、胚発生に不可欠である。

さらに、「A型」、「B型」および「C型」のSDF−1結合モチーフを共有していないさらなる3つのSDF−1結合核酸を特定し、本明細書で「D型」と称する。それらを、プルダウン結合アッセイを使用してアプタマーとして分析した(図9)。
<小規模合成>
2’TBDMS RNAホスホラミダイト化学(DamhaおよびOgilvie、1993)を使用してABI394合成装置(Applied Biosystems、Foster City、CA、USA)を用いた固相合成によって、アプタマーおよびスピーゲルマーを生成した。D−立体配置およびL−立体配置のrA(N−Bz)−ホスホラミダイト、rC(Ac)−ホスホラミダイト、rG(N−ibu)−ホスホラミダイト、およびrU−ホスホラミダイトは、ChemGenes、Wilmington、MAから購入した。アプタマーおよびスピーゲルマーはゲル電気泳動によって精製した。
2’TBDMS RNAホスホラミダイト化学(DamhaおよびOgilvie、1993)を使用して、AktaPilot100合成装置(Amersham Biosciences、General Electric Healthcare、Freiburg)を用いた固相合成によって、スピーゲルマーを生成した。L−rA(N−Bz)−ホスホラミダイト、L−rC(Ac)−ホスホラミダイト、L−rG(N−ibu)−ホスホラミダイト、およびL−rU−ホスホラミダイトは、ChemGenes(Wilmington、MA、USA)から購入した。5’−アミノ−修飾剤は、American International Chemicals Inc.(Framingham、MA、USA)から購入した。スピーゲルマーの合成を、L−リボG、L−リボC、L−リボAまたはL−リボU修飾CPG孔径1000Å(Link Technology、Glasgow、UK)でそれぞれ開始した。カップリング(1サイクル当たり15分)について、アセトニトリル中の0.3Mのベンジルチオテトラゾール(American International Chemicals Inc.、Framingham、MA、USA)、およびアセトニトリル中のそれぞれの0.2Mのホスホラミダイト溶液3.5当量を使用した。酸素−キャッピングサイクルを使用した。オリゴヌクレオチド合成のためのさらなる標準的な溶媒および試薬は、Biosolve(Valkenswaard、NL)から購入した。スピーゲルマーをDMT−ON合成し、脱保護後、Source15RPC媒体(Amersham)を使用して分取RP−HPLC(Wincott F.ら、1995)によって精製した。5’DMT−基を、80%酢酸(室温で90分)で除去した。続いて、水性2MのNaOAc溶液を加え、5K再生セルロース膜(Millipore、Bedford、MA)を使用した接線流濾過によってスピーゲルマーを脱塩した。
in vivoでのスピーゲルマーの血漿残留時間を延長するために、スピーゲルマーを、40kDaのポリエチレングリコール(PEG)部分の5’末端に共有結合した。
<直接プルダウン結合アッセイ>
ビオチン化ヒトD−SDF−1に対するアプタマーの親和性を、37℃にてプルダウン結合アッセイ形式において測定した。アプタマーを、[γ−32P]−標識ATP(Hartmann Analytic、Braunschweig、Germany)を使用して、T4ポリヌクレオチドキナーゼ(Invitrogen、Karlsruhe、Germany)によって5’−リン酸標識した。標識アプタマーの比放射能は、200,000〜800,000cpm/pmolであった。アプタマーを、変性および復元後、低濃度で平衡に達するために、10、20、30または40pMの濃度、37℃で選択緩衝液(20mMのTris−HCl pH7.4、137mMのNaCl、5mMのKCl、1mMのMgCl2、1mMのCaCl2、0.1%[w/vol]のTween−20)中で、様々な量のビオチン化ヒトD−SDF−1と一緒に4〜12時間インキュベートした。結合相手の使用されたプラスチック容器または固定化マトリクスの表面との吸着を避けるために、選択緩衝液に10μg/mlのヒト血清アルブミン(Sigma−Aldrich、Steinheim、Germany)および10μg/mlの酵母RNA(Ambion、Austin、USA)を添加した。ビオチン化ヒトD−SDF−1の濃度範囲は8pM〜100nMに設定し、全反応容量は1mlであった。ペプチドおよびペプチド−アプタマーの複合体を、選択緩衝液で予め平衡化された1.5μlのストレプトアビジンUltralink Plus粒子(Pierce Biotechnology、Rockford、USA)上に固定し、全容量6μl中に再懸濁した。粒子を、それぞれの温度で30分間、サーモミキサー中で懸濁し続けた。上清の分離および適切な洗浄後、固定された放射能をシンチレーションカウンターで定量した。結合の割合をビオチン化ヒトD−SDF−1の濃度に対してプロッティングし、1:1の化学量論比を仮定して、ソフトウェアアルゴリズム(GRAFIT、Erithacus Software、Surrey U.K.)を使用することによって解離定数を得た。
異なるD−SDF−1結合アプタマーを比較するために、競合的順位付けアッセイを実施した。この目的のために、利用可能な大部分のアフィンアプタマーを放射性標識(上記参照)し、参照とした。変性および復元の後、これをビオチン化ヒトD−SDF−1と一緒に37℃で1mlの選択的緩衝液において、競合なしでニュートラアビジンアガロースまたはストレプトアビジンUltralink Plus(共にPierce製)における固定化および洗浄後にペプチドに対する約5〜10%の結合をもたらす条件でインキュベートした。過剰の変性および復元された標識されていないD−RNAアプタマー変異体を、結合反応を並行させるために標識された参照アプタマーと一緒に、種々の濃度(例えば2、10、および50nM)に添加した。試験されるアプタマーは、標的結合に関して参照アプタマーと競合したため、それらの結合特性に依存して結合シグナルを減少させた。したがって、このアッセイにおいて最も活性であると判明したアプタマーは、さらなるアプタマー変異体の競合分析のための新しい参照として機能することができた。
Biacore 2000装置(Biacore AB、Uppsala、Sweden)を使用して、ヒトSDF−1αに対するスピーゲルマーの結合を分析した。ヒトSDF−1αのカップリングをアミン基により達成した場合、ヒトSDF−1αを1〜2時間水に対して透析して(Millipore VSWP混合セルロースエステル、孔径、0.025μM)、干渉アミンを除去した。CM4センサチップ(Biacore AB、Uppsala、Sweden)を、1:1希釈の0.4MのNHSおよび0.1MのEDC35μlを5μl/分の流量で注入することによりタンパク質カップリングの前に活性化した。次いで装置の反応が1000〜2000RU(相対単位)の範囲になるまで、ヒトMCP−1またはヒトSDF−1αを2μl/分の流量で0.1〜1.5μg/mlの濃度で注入した。未反応のNHSエステルを、5μl/分の流量で35μlのエタノールアミン塩酸塩溶液(pH8.5)を注入することにより非活性化した。センサチップを、結合緩衝液を用いて2回プライミングして、ベースラインが安定に見えるまで10μl/分で1〜2時間平衡化した。全てのタンパク質について、選択緩衝液(Tris−HCl、20nM、NaCl、137mM、KCl、5mM、CaCl2、1mM、MgCl2、1mM、Tween20、0.1%「w/v」、pH7.4)中の1000、500、250、125、62.5、31.25、および0nM濃度の一連のスピーゲルマー注入によって、動態パラメータおよび解離定数を評価した。全ての実験において、10μl/分の流量で180秒の結合時間および360秒の解離時間を定量するKinjectコマンドを使用して37℃で分析を実施した。データ分析および解離定数(KD)の算出は、Langmuir1:1化学量論的適合アルゴリズムを使用して、BIAevaluation3.0ソフトウェア(BIACORE AB、Uppsala、Sweden)により行った。
ヒトT細胞白血病細胞株Jurkat、ヒト白血病単球リンパ腫細胞株U937、ヒトpre−B細胞白血病細胞株BV−173およびヒトpre−B ALL細胞株Nalm−6はCXCR4を発現する。Jurkat細胞はCXCR7を発現しないが、白血病株BV−173およびU−937はCXCR7発現について検査で陽性が出た。使用した全ての細胞はDSMZ(Braunschweig)から得た。全ての細胞株を、10%のウシ胎仔血清、100単位/mlのペニリシンおよび100μg/mlのストレプトマイシン(Invitrogen、Karlsruhe、Germany)を含有するGlutamax(Invitrogen、Karlsruhe、Germany)を含むRPMI 1640培地中で37℃および5%CO2にて培養した。実験の1日前に、細胞を、0.3×106/ml(Jurkat、U937、BV−173)または0,75×106/ml(Nalm−6)のそれぞれの密度で新たなT175フラスコ中で播種した。
ヒトSDF−1は、約0.3nMにて最大半数刺激を有し、用量依存的にJurkat細胞の移動を刺激することを見出した。
CXCR4以外に、SDF−1もまた、ケモカイン受容体CXCR7に結合する。CXCR7に対するSDF−1結合スピーゲルマーNOX−A12の阻害可能性を、β−ガラクトシダーゼの断片に融合したCXCR7およびβ−アレスチンの両方を安定に発現するCHO細胞を用いた相補性アッセイにおいて試験した(PathHunterTM−β−アレスチンアッセイ、DiscoveRX、CA、USA)。SDF−1結合β−アレスチンがCXCR7と複合体化すると、それにより化学発光基質を用いて測定したβ−ガラクトシダーゼの相補性および活性を導いた。
PathHunter eXpress CHO−K1ヒトCXCR7 β−アレスチン細胞をOCC2培地中で48時間プレーティングし、10nMのSDF−1および種々の濃度のSDF−1結合スピーゲルマーNOX−A12を用いて90分間刺激した。刺激後、PatHUnter検出キットおよび製造業社の推薦によるプロトコル(DiscoveRX、CA、USA)を使用してシグナルを検出した。
10nMのヒトSDF−1を用いたβ−ガラクトシダーゼおよびそれによるCXCR7活性化の刺激は、5.4nMのIC50でSDF−1結合スピーゲルマーNOX−A12により効果的に遮断した(図13)。
ヒトSDF−1結合スピーゲルマー193−G2−012−5’−PEGはまた、標準的な血管形成器官培養アッセイにおいても機能するかどうかを試験するために、大動脈輪発芽アッセイを実施した。外植片からの血管状伸長の長さおよび量を評価するこのアッセイは、血管形成のために最も広範に使用されている器官培養モデルである(Auerbachら、2003)。SDF−1がこの種のアッセイにおいて発芽を誘導することは既に示されている(Salcedoら、1999)。
オスのラットからの大動脈はBagheri Life sciences(Berlin、Germany)から得た。大動脈を新たに調製し、50単位/mlのペニシリン、50μg/mlのストレプトマイシン(共にInvitrogen、Karlsruhe、Germany)および2.5μg/mlのファンギゾン(fungizone)(Cambrex、USA)を含有するMCDB131−培地(Invitrogen、Karlsruhe、Germany)中に氷上で移した。
SDF−1は発芽を誘導し、この作用はヒトSDF−1結合スピーゲルマー193−G2−012−5’−PEGを用いて遮断できることが実証され得る。非機能的PEG化対照スピーゲルマーによってSDF−1誘導性発芽の遮断は観察されなかった(図14および15)。
白血病細胞が、それらのCXCR4受容体と、骨髄(略語、BM)ニッチなどの特定の組織微環境内の間質細胞によって分泌されたSDF−1との間の相互作用によって従来の化学療法から保護できるという重要な証拠が存在している。したがって、SDF−1結合スピーゲルマーNOX−A12を使用することによってCXCR4−SDF−1軸を標的にすることは、SDF−1分泌間質細胞の保護効果を破壊するため、および後の化学療法に対して白血球細胞を感受性化するための魅力的なアプローチである。
マウス間質細胞株MS−5(ACC441)はDSMZから購入し、多発性骨髄腫細胞株RPMI8226(CCL−155)はATCCから購入した。10%のFBS(Biochrom)およびペニシリン−ストレプトマイシンを添加したRPMI培地1640GlutaMAX(Invitrogen)中に多発性骨髄腫細胞株RPMI8226を維持し、10%のFBSおよびペニシリン−ストレプトマイシンを含むMEMアルファGlutaMAX(Invitrogen)中でMS−5細胞を培養した。化学増感共培養実験のために、MEMアルファGlutaMAX培地(+10%のFBS)を含む8×104/mL/ウェルの濃度の24ウェルプレート(内側に8個のウェル)上で前日に間質MS−5細胞を播種し、5%のCO2中で37℃にてインキュベートした。コンフルエントな間質細胞層を洗浄し、0.5mLのRPMI培地1640(+1%FBS)をウェルに加えた。続いてSDF−1結合スピーゲルマーNOX−A12またはrevNOX−A12を100nMの最終濃度までウェルに加え、4時間インキュベートした。RPMI培地1640(+1%のFBS)中の3.5×105のRPMI8226細胞を間質細胞層に加えた。4時間後、示した場合、1μMのF−ara−A(Sigma Aldrich)を細胞に加えた。40時間のインキュベーション後、15mLのチューブに細胞を回収し、最初に上清を収集し、次いでMS−5細胞を含む付着した細胞をトリプシン処理した。回収した細胞をPBS(+1%のBSA)で2回洗浄し、2mLのPBS(+1%のBSA)中で再懸濁した。150μLの細胞懸濁液をu字形96ウェルプレートに移し、次いで室温にて15分間、50μLのViaCount試薬(Millipore)と共にインキュベートした。Guava EasyCyte 6HT/2L(Millipore)を使用したフローサイトメトリーによって細胞生存率および細胞数を決定した。
間質MS−5細胞と共培養したRPMI−8226細胞の細胞生存率は、SDF−1結合スピーゲルマーNOX A12によりわずかだけ影響を受けた。1μMのF−ara−AはRPMI−8226細胞の生存率に対して有意な効果を示さなかった。しかしながら、NOX−A12およびF−ara−Aを合わせた場合、細胞生存率の相乗的減少が観察された(図16)。したがって、SDF−1結合スピーゲルマーNOX−A12は、BM間質細胞株MS5と共培養した場合、化学療法剤F−ara−Aの処理に対してMM細胞株RPMI−8226を感受性化することが示された。間質MS−5細胞の生存率は、F−ara−AにもNOX−A12にも影響を受けなかった(データは示さず)。これらの結果により、BM間質細胞により分泌されたSDF−1の保護効果を破壊するために破壊する際のNOX A12の可能性の原理の証明が実証される。
この実験の目的は、SDF−1結合スピーゲルマーNOX−A12が、骨髄(略語、BM)間質細胞との共培養において白血病細胞の増殖に対して影響を与えるかどうかを示すことであった。SDF−1を分泌するマウス間質MS−5細胞を、SDF−1結合スピーゲルマーNOX−A12または非機能的スピーゲルマーrevNOX−A12とインキュベートした。白血病T細胞株Jurkatをコンフルエントな間質細胞層に加え、37℃および5%のCO2にて40時間インキュベートした。Guava EasyCyteおよびViaCount試薬を使用したフローサイトメトリーによって細胞数を定量した。
マウスの間質細胞株MS−5(ACC 441)はDSMZから購入し、10%のFBSおよびペニシリン−ストレプトマイシンを含むMEMアルファGlutaMAX(Invitrogen)中で培養した。増殖共培養実験のために、MEMアルファGlutaMAX培地(+10%のFBS)を含む8×104/mL/ウェルの濃度の24ウェルプレート(内側の8個のウェル)上で、前日に間質MS−5細胞を播種し、5%CO2中で37℃にてインキュベートした。コンフルエントな間質細胞層を洗浄し、0.5mLのRPMI培地1640(+1%のFBS)をウェルに加えた。続いてSDF−1結合スピーゲルマーNOX−A12またはスピーゲルマーrevNOX A12を100nMの最終濃度までウェルに加え、4時間インキュベートした。RPMI培地1640(+1%のFBS)中の2×105のJurkat細胞(ほぼ対数増殖期、一度洗浄した)をコンフルエントな間質細胞層に加え、5%CO2を用いて37℃にて48時間インキュベートした。次いで細胞を15mLのチューブに回収し、MS−5細胞を含む付着した細胞をトリプシン処理した。回収した細胞をPBS(+1%のBSA)を用いて2回洗浄した。150μLのこの細胞懸濁液をu字形96ウェルプレートに移し、次いで50μLのViaCount試薬(Millipore)と共に室温にて15分間インキュベートした。Guava EasyCyte6HT/2Lを使用したフローサイトメトリーによって細胞生存率および細胞数を決定した。
培養の40時間後、1nMのSDF−1結合スピーゲルマーNOX−A12は、Jurkat細胞数に対する効果を示さなかったが、間質MS−5細胞を、10または100nMのSDF−1結合スピーゲルマーNOX−A12とプレインキュベートした場合、細胞数は20%まで減少した(図17)。したがって、間質細胞によって分泌されたSDF−1はJurkat細胞の増殖を刺激しているように見える。増殖のSDF−1依存性誘導はSDF−1結合スピーゲルマーNOX−A12により遮断され得、より少数の白血病細胞の検出を導く。
白血病細胞と細胞外マトリクス(略語、ECM)タンパク質との相互作用は、白血病発病において重要な役割を果たす。したがって、我々は、ECMタンパク質フィブロネクチン上での白血病細胞の接着に対するSDF−1結合スピーゲルマーNOX−A12の効果を試験した。SDF−1によるJurkat白血病T細胞株の刺激は、フィブロネクチンに対する接着の用量依存性調節を導いた。
T細胞白血病Jurkat(ACC 282)はDSMZから購入し、10%のFBS(Biochrom)およびペニシリン−ストレプトマイシンを添加したRPMI培地1640GlutaMAX(Invitrogen)中に維持した。接着実験のために、PBS中の10μg/mLのヒトフィブロネクチン(R&D systems)を含む96ウェル培養プレートを37℃で2時間インキュベートした。100μLのPBSを用いてプレートを2回洗浄し、続いてPBS−BSA(0.1%)を用いて37℃で2時間遮断した。次いでRPMI培地を用いてウェルを洗浄した。RPMI培地(+0.1%のBSA)を用いて対数増殖期からのJurkat細胞を洗浄し、種々の濃度のヒトSDF−1(R&D systems)およびNOX−A12と共に37℃で15分間インキュベートした。NOX−A12およびSDF−1を30分間プレインキュベートした。1×105個の刺激したJurkat細胞を、フィブロネクチンでコーティングした96ウェルプレートに播種し、30分間インキュベートした。次いでRPMI培地を用いてプレートを5回洗浄した。Cell Titer Glo試薬(Promega)を使用することによって付着した細胞を定量した。そのために、50μLのRPMI培地、続いて50μLのCell Titer Glo試薬を各ウェルに加えた。プレートを2分間混合し、続いて室温にて10分間インキュベートした。相対発光シグナルによって細胞数を定量した。
SDF−1の培地濃度(1〜10nM)を低くすると、Jurkat細胞のフィブロネクチンに対する接着は減少したが、より高い濃度(30〜300nM)ではJurkat細胞の接着特性は増加した(図18A)。SDF−1結合スピーゲルマーNOX−A12はこの効果を反転するが、対照スピーゲルマーrevNOX−A12はそうではないことが示された(図18B)。したがって、SDF−1結合スピーゲルマーNOX−A12は、それらの保護的ECM環境で白血病細胞相互作用の破壊に対して影響を与え得る。さらに、この例は、骨髄ニッチ由来の造血細胞のSDF−1結合スピーゲルマーNOX−A12依存性分離および動員を説明できる。
SDF−1/CXCR4軸は、多発性骨髄腫(略語、MM)細胞の骨髄(略語、BM)へのホーミングおよび輸送において主要な役割を果たす。したがって、SDF−1阻害による周囲のBM環境からのMM細胞の脱接着は治療剤に対するMM感受性を向上させる。Azabらは、同様に化学療法剤に対するMM細胞の感受性を向上させる、局在化MM細胞に影響を与えるBM環境in vivoでMM細胞の相互作用を破壊するCXCR4阻害剤AMD3100の効果を試験するためのプロトコルを公開している。彼らは、CXCR4特異的アンタゴニストによるSDF−1受容体CXCR4の遮断が、治療に対するMM細胞の感受性を向上させ、結果としてボルテゾミブにより誘発される腫瘍減少を向上させるために、BM環境in vivoでMM細胞の相互作用の破壊を導くことを報告している(Azabら、2009)。このプロトコル(Azabら、2009)に基づいて、SDF−1結合スピーゲルマーNOX−A12を、BM環境in vivoでMM細胞の相互作用を破壊するその効果について試験し、それにより治療に対するMM細胞の感受性を向上させる。

- 前記血液癌が、白血病および骨髄腫を含む群から選択され、前記固形腫瘍が、神経膠芽腫、結腸直腸癌、乳癌、リンパ腫、前立腺癌、膵癌、腎癌、卵巣癌、および肺癌を含む群から選択される、請求項2に記載の医薬組成物。
- 前記補助治療が、対象を感受性化し、前記感受性化された対象が、前記疾患または障害の治療および/または予防のための治療法に対する応答性が高くなる、請求項1〜3のいずれか一項に記載の医薬組成物。
- 前記疾患または障害の治療および/または予防のための治療法が、さらなる薬学的活性剤の投与および/または対象の放射線照射および/または手術および/または細胞療法を含む、請求項4に記載の医薬組成物。
- 前記さらなる薬学的活性剤が、抗体、アルキル化剤、代謝拮抗薬、植物性アルカロイド、植物性テルペノイド、トポイソメラーゼ阻害剤、ロイコボリン、メトトレキサート、タモキシフェン、ソラフェニブ、レナリドマイド、ボルテゾミブ、デキサメタゾン、フルオロウラシル、およびプレドニゾンを含む群から選択される、請求項5に記載の医薬組成物。

- 前記B型のSDF−1結合L−核酸分子が、配列番号22および配列番号28のいずれか1つに記載のヌクレオチド配列から成る、請求項8に記載の医薬組成物。
- 前記C型のSDF−1結合L−核酸分子が、配列番号95から配列番号107、配列番号112から配列番号137、配列番号223、および配列番号224のいずれか1つに記載のヌクレオチド配列から成る、請求項1〜6のいずれか一項に記載の医薬組成物。
- 前記C型のSDF−1結合L−核酸分子が、配列番号120、配列番号128、配列番号129、配列番号134、配列番号135、配列番号223、および配列番号224のいずれか1つに記載のヌクレオチド配列から成る、請求項11に記載の医薬組成物。
- 前記A型のSDF−1結合L−核酸分子が、配列番号60から配列番号73、配列番号78から配列番号82、配列番号84から配列番号87、配列番号89から配列番号94、および配列番号145のいずれか1つに記載のヌクレオチド配列から成る、請求項1〜6のいずれか一項に記載の医薬組成物。
- 前記A型のSDF−1結合L−核酸分子が、配列番号60、配列番号63、配列番号66、配列番号78、配列番号84、および配列番号146のいずれか1つに記載のヌクレオチド配列から成る、請求項13に記載の医薬組成物。
- 前記A型のSDF−1結合L−核酸分子が、配列番号84および配列番号146のいずれか1つに記載のヌクレオチド配列から成る、請求項13に記載の医薬組成物。
- 前記L−核酸分子が、修飾を含む、請求項1〜15のいずれか一項に記載の医薬組成物
- 前記修飾が、好ましくは高分子量の部分であり、かつ/または前記修飾が、動物またはヒトの身体における滞留時間に関する前記L−核酸分子の特徴を修飾することを可能にする、請求項16に記載の医薬組成物。
- さらなる構成要素を含む請求項1〜17のいずれか一項に記載の医薬組成物であって、前記さらなる構成要素が、薬学的に許容される賦形剤、薬学的に許容される担体、およびさらなる薬学的活性剤を含む群から選択される、医薬組成物。
- 前記さらなる薬学的活性剤が、抗体、アルキル化剤、代謝拮抗薬、植物性アルカロイド、植物性テルペノイド、トポイソメラーゼ阻害剤、ロイコボリン、メトトレキサート、タモキシフェン、ソラフェニブ、レナリドマイド、ボルテゾミブ、デキサメタゾン、フルオロウラシル、およびプレドニゾンを含む群から選択される薬学的活性剤である、請求項18に記載の医薬組成物。
- 1つまたはいくつかの投与単位の少なくとも1つの第1の薬学的活性剤を含む薬剤であって、前記第1の薬学的活性剤が、請求項1〜19のいずれか一項に定義されているSDF−1と結合可能なL−核酸分子であり、前記薬剤が、疾患または障害の治療および/または予防のため、あるいは疾患もしくは障害を患っている対象または疾患もしくは障害を発症するリスクがある対象の補助治療のためのものである、薬剤であって、
前記疾患または障害がSDF−1受容体CXCR7を発現する癌である薬剤。 - 前記補助治療が、対象を感受性化し、前記感受性化された対象が、前記疾患または障害の治療および/または予防のための治療法に対する応答性が高くなる、請求項20に記載の薬剤。
- 前記疾患または障害の治療および/または予防のための治療法が、さらなる薬学的活性剤の投与および/または対象の放射線照射および/または手術および/または細胞療法を含む、請求項21に記載の薬剤。
- 前記薬剤が、さらなる薬学的活性剤を含む、請求項20から22のいずれか一項に記載の薬剤。
- 1つまたはいくつかの投与単位の前記さらなる薬学的活性剤を含み、前記さらなる薬学的活性剤が、抗体、アルキル化剤、代謝拮抗薬、植物性アルカロイド、植物性テルペノイド、トポイソメラーゼ阻害剤、ロイコボリン、メトトレキサート、タモキシフェン、ソラフェニブ、レナリドマイド、ボルテゾミブ、デキサメタゾン、フルオロウラシル、およびプレドニゾンを含む群から選択される、請求項23に記載の薬剤。
- 前記癌が、血液癌および固形腫瘍の群から選択される癌である、請求項20〜24のいずれか一項に記載の薬剤。
- 前記血液癌が、白血病、および骨髄腫を含む群から選択され、前記固形腫瘍が、神経膠芽腫、結腸直腸癌、乳癌、リンパ腫、前立腺癌、膵癌、腎癌、卵巣癌、および肺癌を含む群から選択される、請求項25に記載の薬剤。
- 疾患または障害の治療および/または予防のための、あるいは疾患もしくは障害を患っている対象または疾患もしくは障害を発症するリスクがある対象の補助治療のための薬剤の製造のための請求項1〜19のいずれか一項に定義されているL−核酸分子の使用であって、前記疾患または障害がSDF−1受容体CXCR7を発現する癌である使用。
- 前記補助治療が、対象を感受性化し、前記感受性化された対象が、前記疾患または障害の治療および/または予防のための治療法に対する応答性が高くなる、請求項27に記載の使用。
- 前記疾患または障害の治療および/または予防のための治療法が、さらなる薬学的活性剤の投与および/または対象の放射線照射および/または手術および/または細胞療法を含む、請求項28に記載の使用。
- 前記薬剤が、さらなる薬学的活性剤と組み合わせて用いられ、前記さらなる薬学的活性剤が、抗体、アルキル化剤、代謝拮抗薬、植物性アルカロイド、植物性テルペノイド、トポイソメラーゼ阻害剤、ロイコボリン、メトトレキサート、タモキシフェン、ソラフェニブ、レナリドマイド、ボルテゾミブ、デキサメタゾン、フルオロウラシル、およびプレドニゾンを含む群から選択される薬学的活性剤である、請求項27〜29の何れか一項に記載の使用。
- 前記癌が、血液癌および固形腫瘍の群から選択される癌である、請求項27〜30の何れか一項に記載の使用。
