Tobacco Genome Initiativeにおいて見出されるタバコEST配列を用いてタバコトレオニン合成酵素遺伝子が同定される。タバコトレオニン合成酵素に近接する他のタバコESTコンティグ(NCBI_45312-v2pep-fl)をNCBI EST配列からアセンブルする。このコード配列は、タバコトレオニン合成酵素と正確に同じ長さ(1569bp)を有する。DNAマイクロアレイエクソンAffymetrixチップ(NtPMIa1g193542e1からの配列プローブ)を用いるデータは、タバコトレオニン合成酵素が、N.タバカムの根、トリコーム、緑葉および老化葉において等しく発現されていることを示す。鉄管乾燥(K326、K399)タバコとバーレー(TN90、Bu64)タバコの間で、遺伝子発現レベルの大きな変化は見られなかった。さらに、寒冷ストレスおよびカドミウムストレスは、タバコトレオニン合成酵素の発現レベルに影響を及ぼさない。このことは、このトレオニン合成酵素がタバコ葉において構成的に発現されていることを強く示唆する。
文献の記載に従い、古典的なリーフディスク法を用いて、アグロバクテリウム・ツメファキエンス(Agrobacterium tumefasciens)によって、カリフラワーモザイクウイルス35Sプロモーターの転写調節下、アラビドプシス・タリアナCGS(MT01、TAIR Accession AT3901120)がバーレータバコTN90に導入される。カナマイシン抗生物質選択遺伝子もまた挿入される。
RNAiアプローチを用い、タバコにおけるNtTSをサイレンシングするために、配列番号1のDNA断片を用いてプライマーを作成する。用いるプライマーは5'-ctgaaatcgacagcgatgata-3'(配列番号9)および5'-caaccaatagctaacggagctt-3'(配列番号10)である。対応するRNAi配列は、逆転写酵素-ポリメラーゼ連鎖反応(RT-PCR)によってcDNAから増幅し、次いで、製造業者(Invitrogen社)の指示に厳密に従って、エントリーベクターによってGatewayベクターpB7GWIWG2(II)に挿入される。このベクターは、植物のすべての組織における導入遺伝子の構成発現のためのプロモーター(カリフラワーモザイクウイルスCaMV 35Sプロモーター)および寒天平板上でのカナマイシン抗生物質選択(100mg/ml)のためのカナマイシン遺伝子を含む。次いで、古典的リーフディスク法を用いて、アグロバクテリウム・ツメファキエンスによって、バーレータバコTN90のゲノムにこの構築物を挿入する。カルスから、個々の系統が再生され、カナマイシンに対して選択される。RNAiサイレンシングに関して、(i)ゲノムDNAに対して35Sプロモーター内の1つのプライマー(5’-gagcatcgtggaaaaagaagac-3')およびサイレンシングに用いる断片内の1つのプライマー(5’-aagctccgttagctattggttg-3')を用いるRT-PCRによってT0系統をモニターし、種子を作らせ、次いで(ii)サイレンシングに用いる挿入物に隣接する特異的プライマー(5'-ttgattcacgtgtcggtaagac-3')を用いるRT-PCRによってモニターして種子を作らせる。T1種子を採取し、カナマイシン含有寒天上で再栽培し、T0小植物と全く同様にモニターする。ゲノムgDNAに対するPCRは、NtTS DNA断片がゲノムに挿入され、タバコトレオニン合成酵素を効果的にサイレンシングすることを示す。
カナマイシン抵抗性植物は、畑(ケンタッキー州、米国)における栽培の前に、フローティングトレイにおいて栽培される。サイレンシングされたトレオニン合成酵素系統の20本の植物(NtTS-RNAi系統;実施例2)、シスタチオニンγ合成酵素系統(実施例3)、ベクター対照(VC、pB7GWIWG2(II))およびTN90 USバックグラウンドタバコを、20本の植物を4連で栽培する。畑移植3ヶ月後(摘心36日後)、各連の20本の中から10本の同一の植物において幹中央部における1つの葉の試料を採取する。この葉(”緑葉”)を直ちにドライアイス中に保存し、凍結乾燥する。最良の2つの系統からの10本の植物を選択し、次いで、バーレーの農業規範に従って乾燥する。乾燥後、幹中央部の3枚の葉の試料を採取する。サイレンシングされたトレオニン合成酵素系統におけるサイレンシング効果をモニターし、シスタチオニンγ合成酵素系統と比較するために、3つの系統からの”緑葉”をすりつぶし、遊離アミノ酸解析に供する。
前述の系統を用い、LC-MSを用いて、トレオニン合成酵素のサイレンシングによって得られた緑葉において産生されるメチオニンと、乾燥タバコにおいて見出される化学化合物との相関を乾燥葉においてモニターする。メタノールで抽出を行う。用いる装置は、MSに連結されたWaters Acquity UPLCシステムである。カラムはWatersXBridge Shield RP18であり、移動層Aとして水:アセトニトリル(95:5v/v)を用い、移動層Bとしてメタノールを用いる。このLC-MSプロフィールは、緑葉におけるトレオニン合成酵素のサイレンシングが、乾燥葉における化学的シグナルを増強させることを示す。実際、対照と比較して、トレオニン合成酵素がサイレンシングされた系統において、4つのピークが改変されたプロフィールを示した(図1参照)。3つのピークは明確に存在量が増加し、1つは減少する。緑葉に蓄積するメチオニンは、乾燥葉において、分解硫黄産物またはメチオニン誘導体のいずれかに変換されることをこのことは示している。
開始コドンから終止コドンまでの1587塩基のコンセンサス配列に関して、配列番号1、2、3および20をアラインメントした。配列番号1、2および3間では相同であり、配列番号20とは異なる断片領域は、配列番号1、2および3のヌクレオチド1〜46と配列番号20のヌクレオチド1〜52;配列番号1のヌクレオチド99〜141;配列番号2および配列番号3のヌクレオチド102〜144と配列番号20のヌクレオチド102〜153;ならびに配列番号1、2および3のヌクレオチド1325〜1362と配列番号20のヌクレオチド1334〜1371である。これらの配列は、配列番号1、2、3に特異的なプライマーを設計するために用いられる。配列番号20に特異的な1組のプライマーもまた設計される。
Claims (16)
- タバコ植物の少なくとも一部分において遊離メチオニン濃度を増加させる方法であって、
(a)タバコ植物においてトレオニン合成酵素の発現または活性を低下させる段階であって、タバコ植物が、
(i)トレオニン合成酵素をコードし、かつ配列番号1、配列番号2もしくは配列番号3に少なくとも90%の配列同一性を有するか、または配列番号4もしくは配列番号5に少なくとも90%の配列同一性を有する配列を含むか、それからなるか、または本質的にそれからなるポリヌクレオチド;
(ii)(i)に記載の前記ポリヌクレオチドのいずれか1つによってコードされるポリペプチド;または
(iii)配列番号6、配列番号7もしくは配列番号8に少なくとも95%の配列同一性を有し、かつトレオニン合成酵素活性を有するポリペプチド;
を含む前記段階;
(b)段階(a)で得られた変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物の少なくとも一部分において遊離メチオニン濃度を測定する段階;および
(c)変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物を同定する段階であって、前記植物中の遊離メチオニン濃度が、トレオニン合成酵素の発現または活性が低下していない対照植物と比較して増加しており、畑移植3ヶ月後または摘心36日後に、前記変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物の視覚的外観が対照植物と実質的に同じである前記段階
を含む前記方法。 - 請求項1に記載の方法によって得られたか、または得ることができる、変異タバコ植物、天然に存在しないタバコ植物、またはトランスジェニックタバコ植物または前記タバコ植物から誘導されたか、誘導されることができる、タバコ植物材料。
- 変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物であって、
(i)トレオニン合成酵素をコードし、かつ配列番号1、配列番号2もしくは配列番号3に少なくとも90%の配列同一性を有するか、または配列番号4もしくは配列番号5に少なくとも90%の配列同一性を有する配列を含むか、それからなるか、または本質的にそれからなるポリヌクレオチド;
(ii)(i)に記載の前記ポリヌクレオチドのいずれか1つによってコードされるポリペプチド;
(iii)配列番号6、配列番号7もしくは配列番号8に少なくとも95%の配列同一性を有し、かつトレオニン合成酵素活性を有するポリペプチド;または
(iv)(i)に記載の前記ポリヌクレオチドを含む構築物、ベクターまたは発現ベクター
を含み、
トレオニン合成酵素の発現または活性が低下していない対照タバコ植物と比較して、トレオニン合成酵素の発現またはそれによってコードされるタンパク質の活性が低下し、植物の一部分が少なくともメチオニン含量の5%の増加を有し、エアゾール中のメチオナール濃度が、対照植物からのエアゾールと比較して少なくとも5%増加している変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物。 - 畑移植3ヶ月後または摘心36日後に、前記植物の視覚的外観が対照植物と実質的に同じである、請求項3に記載の変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物。
- 変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物の幹の高さが、畑移植3ヶ月後または摘心36日後に対照植物の幹の高さと実質的に同じであり、かつ/または変異タバコ植物、天然に存在しない植物またはトランスジェニック植物のクロロフィル含量が、畑移植3ヶ月後または摘心36日後に、対照植物のクロロフィル含量と実質的に同じである、請求項3又は4に記載の変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物。
- 植物の一部分において遊離トレオニン濃度が対照植物と比較して増加している、請求項3、4又は5に記載の変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物。
- (i)葉において遊離メチオニン濃度が少なくとも約0.03mg/gであり、(ii)葉において遊離トレオニン濃度が少なくとも約0.5mg/gであり、および(iii)加熱によるエアゾール中のメチオナール濃度が少なくとも約2000μg/gである、請求項6に記載の変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物。
- 請求項5〜7のいずれか1つに記載のタバコ植物からの細胞または組織を含む種子。
- 請求項5〜7のいずれか1つに記載のタバコ植物からの細胞または組織を含むバイオマスまたは葉を含む植物材料。
- 請求項5〜7のいずれか1つに記載のタバコ植物の一部分または請求項9に記載の植物材料を含むタバコ製品。
- メチオナールを製造する方法であって、
(a)請求項5〜7のいずれか1つに記載の変異タバコ植物、天然に存在しないタバコ植物もしくはトランスジェニックタバコ植物の一部分、請求項9に記載の植物材料または請求項10に記載のタバコ製品を提供する段階;および
(b)それらに熱を加える段階
を含む前記方法。 - 試料中のトレオニン合成酵素をコードし、かつ配列番号1、配列番号2、配列番号3、配列番号4、配列番号5または配列番号20に少なくとも95%の配列同一性を有するポリヌクレオチドを検出する方法であって、
(a)前記ポリヌクレオチドを含むか、またはそれを含むことが疑われる試料を提供する段階;
(b)前記ポリヌクレオチドの少なくとも一部を特異的に検出するための1以上のプライマーまたは1以上のプローブと前記試料を接触させる段階;および
(c)増幅産物の存在を検出する段階であって、ここで増幅産物の存在が、その試料中の前記ポリヌクレオチドの存在を示す前記段階
を含む前記方法。 - メチオニン濃度が、対照植物と比較して増加している、変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物において使用する為の、トレオニン合成酵素をコードし、かつ配列番号1、配列番号2もしくは配列番号3に少なくとも90%の配列同一性を有するか、または配列番号4もしくは配列番号5に少なくとも90%の配列同一性を有する配列を含むか、それからなるか、または本質的にそれからなる単離されたポリヌクレオチド。
- メチオニン濃度が、対照植物と比較して増加している、変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物において使用する為の、請求項13に記載のポリヌクレオチドのいずれか1つによってコードされる単離されたポリペプチド。
- メチオニン濃度が、対照植物と比較して増加している、変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物において使用する為の、配列番号6、配列番号7または配列番号8に少なくとも95%の配列同一性を有し、かつトレオニン合成酵素活性を有する、単離されたポリペプチド。
- メチオニン濃度が、対照植物と比較して増加している、変異タバコ植物、天然に存在しないタバコ植物またはトランスジェニックタバコ植物において使用する為の、請求項13に記載のポリヌクレオチドを含む構築物、ベクターまたは発現ベクター。
