Description
本出願は、2011年2月24日に提出された欧州特許出願第EP11305197.3号の優先権を主張し、これはその全体が参照により本明細書に組み入れられる。
IGF(インスリン様成長因子)系は、細胞がその生理学的環境と情報交換するために使用する、十分に特徴付けられたポリペプチドのセットからなる。この系は、2つの細胞表面レセプター(IGF1RおよびIGF2R)、2つのペプチド性リガンド(IGF−IおよびIGF−II)、6つの高親和性IGF結合タンパク質ファミリー(IGFBP−1〜6)、並びに関連したIGFBP分解酵素(まとめてプロテアーゼと呼ばれる)を含む。IGFシグナル伝達経路は、哺乳動物の身長の増加に主要な役割を果たしているだけでなく、細胞の増殖および生存にも関与している。成長ホルモン(GH)は多くの組織においてIGF−I産生の主要なレギュレーターであるが、IGFは、ほぼ遍在的に産生され、そして血清中においてIGFBPとほぼ結合して高い濃度で循環している。IGFが、そのチロシンキナーゼ細胞表面レセプターであるIGF1Rとの会合を通してその効果を発揮するためには、それらはまず、IGFBPと形成された複合体から解離しなければならず、IGF−IおよびIGF−IIに対するIGFBPの親和性は、時にIGF1Rよりも高い。従って、レセプター−リガンド相互作用は、「遊離」IGFレベルに高度に依存し、前記レベルは、血清および他の生物学的液体中に存在するIGFBPによって堅固に調節されている。それ故、IGFとIGFBPの相互作用は、制御されない細胞増殖または低血糖などの望ましくないIGF作用を防ぐことができる。逆に、IGF/IGFBP複合体の破壊は、IGFレセプターを通してIGFがその分裂促進作用および代謝作用を発揮するためのほぼ確実な必要条件である。
本発明は、種々の治療的および/または診断的適用において有用であるIGFBP−3の誘導体に関する。
生物学的試料を、IGFBP−3ポリペプチド誘導体に対して融合したSeAPのアミノ酸配列を含むIGFBP−3ポリペプチド誘導体と接触させ、これにより、IGFBP−3誘導体と、生物学的試料中に存在するあらゆるpro−IGF−IIとの間の複合体の形成を可能とさせる工程、ここで、pro−IGF−IIは、IGF−IIの部分的にプロセシングされた形態である;および

前記したように、IGFは、ほぼ全体が、IGF利用能および作用の重要な決定因子である6個の特異的で高親和性の結合タンパク質(IGFBP−1〜6)と会合して、ヒトの身体全体に存在している。血漿中のIGF−Iの90%超が、同定された6個のIGFBPの中で最も豊富であるIGFBP−3と結合している。IGFBP−3は、主要な循環担体タンパク質としての作用に加えて、多くの組織において産生され、ここでIGF作用を調節するその能力を介しておよびまた直接的な内因性作用に因り、細胞機能に対して複数の効果を及ぼす。
本発明のIGFBP−3誘導体は、タンパク質分解的開裂に対する抵抗性によって特徴付けられる。前記したように、野生型IGFBP−3の中間ドメインはゆるく構築され、そしてタンパク質分解的開裂のターゲットである。いくつかのプロテアーゼがIGFBP−3を開裂することが示されており、これにはプラスミン、トロンビン、カリクレイン、前立腺特異的抗原、マトリックスメタロプロテアーゼおよびカテプシンが挙げられる。IGFBP−3のタンパク質分解は、IGFに対するその親和性を低下させ、従って、完全に結合したIGFに対する遊離IGFの全体的な比は増加する。
前記したIGFBP−3誘導体は、少なくとも2つの主要な特性を共有する:(1)それらはタンパク質分解的開裂に対して抵抗性である、そして(2)それらは、野生型IGFBP−3の対応する結合親和性と同一、実質的に類似またはさらにはより高い、IGF−I、IGF−II、ヘパリンおよびALSに対する結合親和性を示す。本発明者らは、これらのIGFBP−3誘導体を「ミニ−IGFBP−3」と呼ぶ。
従って、1つの局面において、本発明は、(1)タンパク質分解的開裂に対して抵抗性であり、(2)野生型IGFBP−3の対応する結合親和性と同一、実質的に類似またはさらにはより高い、IGF−I、IGF−IIおよびヘパリンに対する結合親和性を示すが、(3)ALSには結合しない(または実質的に結合しない)、IGFBP−3誘導体を提供する。これらのIGFBP−3誘導体は、本明細書において、「ミニ−IGFBP−3ALS−」誘導体と呼ばれる。

本発明はまた、タンパク質分解的開裂に対して抵抗性であるIGFBP−3/IGF−I複合体を提供する。
本発明はまた、(1)タンパク質分解的開裂に対して抵抗性であり、(2)野生型IGFBP−3の対応する結合親和性と同一、実質的に類似またはさらにはより高い、IGF−I、IGF−II、ヘパリンおよびALSに対する結合親和性を示し、そして(3)BirA酵素によるビオチニル化を受け入れられる、IGFBP−3誘導体を提供する。これらのIGFBP−3誘導体は、「ミニ−IGFBP−3−avitag」誘導体と呼ばれる。

本発明によるIGFBP−3誘導体は、本発明者らによって使用されたような部位特異的突然変異誘発および組換え技術を含む、当技術分野において公知の多種多様な適切な方法のいずれかを使用して調製され得る(実施例の章を参照)。本発明は、本発明者らによって開発された発現ベクター並びにこのような発現ベクターを含む宿主細胞を包含する。
本発明のIGFBP−3ポリペプチド誘導体は、種々の治療的および/または診断的適用において使用され得る。それらは、ヒト起源のタンパク質から誘導されたという利点を有し、これはいくつかの治療適応症のためにFDAによってすでに認可され、そしてこれは大きな二次的作用を全く伴わないことが証明された。
A.適応症
本発明のIGFBP−3ポリペプチド誘導体は、IGF(IGF−Iおよび/またはIGF−II)を捕捉し、そしてこれによりそのバイオアベイラビリティ−を低下させる薬剤として有用であり得る。従って、それらは、ヒト悪性腫瘍の化学療法または放射線療法におけるアジュバントとして、既存の抗IGF1−Rモノクローナル抗体およびチロシンキナーゼ阻害剤の代替物を構成し得る(Klinakis et al., Proc. Natl. Acad. Sci. USA, 2009, 106: 2359-2364; Pollack et al., Nat. Rev. Cancer, 2004, 4: 505-518; Ryan and Goss; Oncologist, 2008, 13: 16-24; Sachdev and Yee, Mol. Cancer Ther., 2007, 6: 1-12; Samani et al., Endocr. Rev.2006)。本発明のいくつかのIGFBP−3ポリペプチド誘導体、例えばミニ−IGFBP−3−Fcは、血流中に存在するIGF−IおよびIGF−IIの両方を捕捉することができ、一方、ミニ−IGFBP−3 ALS−などのその他は、生物学的に利用可能な組織中IGFを減少させることができる。この後者の特徴はさらに、IGFBP−3誘導体にビオチン残基を付加することによって(ミニ−IGFBP−3−ALS−avitag−ビオチン)腫瘍組織それ自体へとターゲティングさせることができる。両方の戦略において、本発明のIGFBP−3ポリペプチド誘導体の投与により、IGFシグナル伝達経路のダウンレギュレーションが起こる。このアプローチは、インタクトなIGFBP−3の循環が強く抑制されている転移性腫瘍患者の場合には特に関連性がありそして見込みがあると思われる(Fowlkes et al., Endocr., 2004, 145: 620-626; Miyamoto et al., Cancer Res., 2004, 64: 665-371; Nakamura et al., Biochem. Biophys. Res. Commun., 2005, 333: 1011-1016)。プロテアーゼに抵抗性のIGFBP−3ポリペプチド誘導体の投与は、腫瘍にとって有害な持続的なIGFの欠乏をもたらし、これは腫瘍量を減少させるための他の化学療法剤の効果を増強するはずである。
IGFBP−3ポリペプチド誘導体(場合により1つ以上の適切な薬学的に許容される担体または賦形剤を用いて製剤化した後)を、所望の投与量で、それを必要とする被験体に任意の適切な経路によって投与することができる。種々の送達システムが知られており、そしてこれを使用して、本発明のIGFBP−3誘導体を投与することができ、これには錠剤、カプセル剤、注射溶液、封入(リポソーム、マイクロ粒子、マイクロカプセルなどへの)が含まれる。投与法としては、経皮、皮内、筋肉内、腹腔内、病巣内、静脈内、皮下、鼻腔内、肺内、硬膜外、眼内および経口経路が挙げられるがそれらに限定されない。本発明のIGFBP−3誘導体または組成物は、任意の簡便なまたは他の適切な経路によって、例えば点滴またはボーラス注入によって、上皮または皮膚粘膜裏打ち(mucocutaneous lining)(例えば経口、粘膜、直腸および腸管粘膜など)を通した吸収によって投与され得る。投与は全身的であっても局所的であってもよい。非経口投与は、カテーテル法によるなどの、患者の特定の組織に向けられ得る。当業者には理解されるであろうように、本発明のIGFBP−3誘導体が追加の治療剤と共に投与される態様において、IGFBP−3誘導体および治療剤は、同じ経路(例えば経口)によって、または異なる経路(例えば経口および静脈内)によって投与され得る。
本発明のIGFBP−3誘導体(またはその組成物)の投与は、送達される量が意図する目的のために効果的であるような投与量である。投与経路、投与される製剤および投与量は、所望の治療効果、処置する疾患の重度、あらゆる感染の存在、患者の年齢、性別、体重および全般的な健康状態並びに使用されるIGFBP−3誘導体の効力、バイオアベイラビリティおよびin vivoにおける半減期、併用療法の使用(または不使用)および他の臨床因子に依存する。これらの因子は、療法の過程で担当の医師によって容易に決定され得る。代替的にまたは追加的に、投与しようとする投与量は、動物モデルを使用した研究から決定され得る。これらまたは他の方法に基づいて最大効力を達成するように投与量を調整することは、当技術分野において周知であり、そしてこれは訓練された医師の能力の範囲内である。研究は本発明のIGFBP−3誘導体を使用して実施されるので、適切な投与量レベルおよび処置期間に関するさらなる情報が現れるだろう。
本発明のIGFBP−3ポリペプチド誘導体のいくつかは、診断適用において有用であり得る。特に、本発明は、生物学的試料中のpro−IGF−II濃度を決定するための方法を提供し、前記方法は、(1)生物学的試料を、IGFBP−3ポリペプチド誘導体に対して融合した、SeAPのアミノ酸配列を含む、IGFBP−3ポリペプチド誘導体と接触させて、IGFBP−3誘導体と、生物学的試料中に存在するあらゆるpro−IGF−IIとの間の複合体の形成を可能とする工程、ここで、pro−IGF−IIはIGF−IIの部分的にプロセシングされた形態である;および(2)複合体中のSeAPのアルカリ活性を測定することによって生物学的試料中のpro−IGF−IIの濃度を決定する工程を含む。
前記したように、本発明のIGFBP−3ポリペプチド誘導体は、それ自体または薬学的組成物として投与され得る。従って、本発明は、有効量の少なくとも1つのIGFBP−3ポリペプチド誘導体および少なくとも1つの薬学的に許容される担体または賦形剤を含む、薬学的組成物を提供する。いくつかの態様において、前記組成物はさらに、1つ以上の追加の生物学的に活性な薬剤を含む。
注射調製物、例えば無菌注射水性または油性懸濁液を、公知の技術に従って、適切な分散剤または湿潤剤、および懸濁化剤を使用して製剤化し得る。無菌注射調製物はまた、無毒性の非経口的に許容される希釈剤または溶媒中の無菌注射溶液、懸濁液またはエマルション、例えば2,3−ブタンジオール溶液などであり得る。使用され得る許容されるビヒクルおよび溶媒の中には、水、リンガー液、U.S.P.および等張塩化ナトリウム溶液がある。さらに、無菌の不揮発性油が溶液または懸濁媒体として慣用的に使用される。この目的のために、合成モノまたはジグリセリドを含む、任意の無刺激性の不揮発性油を使用することができる。オレイン酸などの脂肪酸もまた、注射製剤の調製に使用し得る。無菌液体担体は、非経口投与のための無菌液体形組成物に有用である。
特定の態様において、本発明のIGFBP−3ポリペプチド誘導体は、本発明の薬学的組成物における唯一の活性成分である。他の態様において、薬学的組成物はさらに、1つ以上の生物学的に活性な薬剤を含む。適切な生物学的に活性な薬剤の例としては、抗癌剤、抗炎症剤、免疫調節剤、鎮痛剤、抗微生物剤、抗細菌剤、抗生物質、抗酸化剤、消毒剤およびその組合せが挙げられるがそれらに限定されない。
別の局面において、本発明は、本発明のIGFBP−3ポリペプチド誘導体の投与を可能とする、本発明の薬学的組成物の1つ以上の成分を含む1つ以上の容器(例えばバイアル、アンプル、試験管、フラスコまたは瓶)を含む医薬パックまたはキットを提供する。本発明はまた、生物学的試料中のpro−IGF−II濃度の決定を可能とする1つ以上の成分を含む1つ以上の容器を含むキットを提供する。このようなキットは、一般的に、IGFBP−3ポリペプチド誘導体に対して融合したSeAPのアミノ酸配列を含む本発明のIGFBP−3ポリペプチド誘導体、およびSeAPのアルカリ活性を測定するための少なくとも1つの試薬を含む。

導入ベクターの構築
以下の導入ベクターを本研究に使用した:pDB−3s−H6、pMJ−SeAPおよびpYM−Fc−IgG1。pDB−3s−H6は、以前に記載されているように(Netchine et al., J. Clin. Endocrinol. Metab., 2009, 94: 3913-3921)本発明者らの研究室で構築された分泌タンパク質発現のためのバキュロウイルス導入ベクターである。同様に、アルカリホスファターゼ(SeAP)またはヒトIgG1のFcフラグメントのいずれかを有する分泌融合タンパク質を産生するために使用されたpMJ−SeAPおよびpYM−Fc−IgG1ベクターもまた、本発明者らの研究室で構築され、そしてこれは以前に記載されている(Bara et al., Int. J. Cancer, 1998, 75: 767-773; Mahiou et al., Biochem. J., 1998, 330: 1051-1058、これはその全体が参照により本明細書に組み入れられる)。
(終止コドンに下線を引いている)をそれぞれセンスおよびアンチセンスプライマーとして;および、IGFBP−3cDNAを有するプラスミドを鋳型として、使用した。上のプライマーに加えられた制限酵素部位SfiIおよびXhoIは小文字で提示されている。815bpのPCR産物を、SfiIおよびXhoI制限エンドヌクレアーゼで消化し、そして同酵素で前以て消化しておいたpDB−3s−H6導入ベクターにライゲーションした。ライゲーション産物を使用して、エレクトロコンピテントなE.coli細胞を形質転換した。pDB−3s−H6−IGFBP−3と呼ばれる組換えプラスミドを、ヌクレオチドシークエンスの後に選択し、そしてバキュロウイルスDNAと一緒にSf9細胞に同時トランスフェクションして、H6−IGFBP−3組換えタンパク質をコードする対応する組換えウイルスを産生した。
部位特異的突然変異誘発を、実質的には以前に記載されているように(Kunkel, Proc. Natl. Acad. Sci. USA, 1985, 82: 488-492)変異原性オリゴヌクレオチドおよび一本鎖DNA鋳型を使用して、「MutaGene(登録商標)Phagemid in vitro mutagenesis kit version 2」(BioRad)を使用して実施した。簡潔に言うと、導入ベクターpMJ−H6−IGFBP−3およびpDB−3s−H6−IGFBP−3(各々、M13複製起点を含む)を使用して、CJ236(ung−dut−)突然変異株を形質転換した。ファージミドクローンを選択して、ヘルパーファージM13KO7を用いての感染後にウイルス粒子を産生させた。抽出後、対応する一本鎖DNAは、Kunkelによって記載されたような5’−リン酸化変異原性オリゴヌクレオチド、T7DNAポリメラーゼおよびT4DNAリガーゼを含む重合反応における鋳型として作用した。
にアニーリングさせることによって、ヒトIGFBP−3の中間ドメインを短くした。得られたムテインでは、野生型IGFBP−3中間ドメインに存在する85アミノ酸(残基95から残基179)は、15アミノ酸のスペーサー(その配列は
である)によって置換されている。得られた突然変異したプラスミド(pDB−ミニ−IGFBP−3)は、ネイティブな中間ドメインの大半を欠失した、「ミニ−IGFBP−3」と呼ばれる、このIGFBP−3ムテインをコードする組換えバキュロウイルスを生成するように作用した。
とを使用して、配列AGGSGに置換した。
と併せて使用して、ミニ−IGFBP−3のC末端に、BirA酵素のための基質(Schatz, Biotechnology, 1993, 11: 1138-1143)であるGLNDIFEAQKIEWHEペプチド(Beckett et al., Protein Sci., 1999, 8: 921-929)を付加した。得られたタンパク質、すなわち−ミニ−IGFBP−3−avitagを酵素的にビオチニル化することができた。
ミニ−IGFBP−3リポータータンパク質を得るために、ミニ−IGFBP−3コード配列を、SfiIおよびXhoI制限エンドヌクレアーゼを用いてのその二重消化後に得られたベクターpDB−3s−H6−ミニ−IGFBP−3からのミニ−IGFBP−3の切り出し、およびその後の同酵素を用いて前以て消化しておいたpMJ−SeAPベクターへの得られたフラグメントのライゲーションによって、分泌型アルカリホスファターゼ(SeAP)の3’末端に融合させた。得られた組換えプラスミドを使用して、「SeAP−ミニ−IGFBP−3」と呼ばれるリポーター融合タンパク質をコードする組換えバキュロウイルスを生成した。
前記した組換えウイルスのいずれかを用いて感染させた昆虫細胞の培養培地を収集し、そして組換えタンパク質を、実質的に以前に記載されたように(Mahiou et al., 1998、上を参照)IMACによって精製した。組換えタンパク質は、イオン交換クロマトグラフィーによるさらなる精製工程を必要としたので、IGFBP−3−Fcイムノアドヘシンを、プロテインA−セファロースアフィニティクロマトグラフィーによってさらに精製した(Mahiou et al., 1998、上を参照)。
図2は、野生型IGFBP−3(H6−IGFBP−3)および突然変異した組換えIGFBP−3(ミニ−IGFBP−3)の図解を示し、これは、後者が、部位特異的突然変異誘発によって達成された示された15アミノ酸の可動性リンカーによるその中間ドメインの大半の置換により、野生型H6−IGFBP−3から誘導されるという事実を示す。また、図2には、H6−IGFBP−3およびミニ−IGFBP−3のアミノ酸配列も提示されている。
図5は、部位特異的突然変異誘発によって得られたpRH−3s−H6−avitag導入ベクターの制限酵素地図の図解を示す。このベクターにおいて、SfiIおよびPvuII制限酵素部位によってフランキングされるあらゆるオープンリーディングフレーム配列をPCRによって同酵素によって消化されたベクターに導入することができる。構築物の翻訳から生じた分泌ポリペプチドは、そのC末端にAvitagペプチドを有し、そしてBirA酵素を使用したターゲティングされた酵素的ビオチニル化を受け入れられる。ミニ−IGFBP−3のコード配列をこのベクターに導入することにより、順にpRH−3s−H6−ミニ−IGFBP−3ベクター、AcMNPV−ミニ−IGFBP−3組換えウイルスおよびミニ−IGFBP−3−avitag組換えタンパク質が得られた。
図8(A)は、SeAP−ミニ−IGFBP−3リポーター融合タンパク質を産生するために構築および使用されたpMJ−SeAP−ミニ−IGFBP−3導入ベクターの制限酵素地図を示す。
図9(A)は、ミニ−IGFBP−3−FcIgG1融合タンパク質(図9(B)に図解により提示する)を産生するために使用された、構築されたpYM−ミニ−IGFBP−3導入ベクターの制限酵素地図を示す。
INA−6骨髄腫細胞株を、10%FCSの存在下で増殖させた。細胞を、ミニ−IGFBP−3(100nm)またはppp(1μM)と共に3、6または24時間インキュベーションした。インキュベーション後、細胞を、溶解緩衝液を使用して溶解し、そして細胞抽出物(タンパク質含量について標準化された)をSDS−PAGEゲルにローディングし、ニトロセルロース膜に転写し、これをさらにAKTタンパク質のホスホ−S273残基に対して作られたモノクローナル抗体と共にインキュベーションした。膜をまた、等価なローディングを確認するために、ストリッピング後に抗アクチン抗体を用いて再プローブした。得られた結果を図11Aに提示する。
ミニ−IGFBP−3のin vivo活性の将来の評価のための根拠を築くために、ミニ−IGFBP−3およびその誘導体をマウスに注入した。ヒト特異的IGFBP−3ELISA試験を使用して測定した場合、いくつかの動態を観察することができた。
エストロゲンに対するその依存性を回避する細胞の出現を妨げるミニ−IGFBP−3の能力は、現在処理中である。Luminal A型の腫瘍に典型的なMCF−7細胞はエストロゲンレセプター陽性であり、そしてその増殖をエストロゲンに依存する。これらの実験において、これらの細胞を、ミニ−IGFBP−3の存在下または非存在下においてエストロゲンを枯渇させた培地中に蒔く。エストロゲン非依存性増殖を、クリスタルバイオレットを用いての染色によって対照および試験プレートにおいて評価する。いくつかの対照細胞はそうした条件下で増殖するが、ミニ−IGFBP−3で処理されたプレートにおいては全く細胞は増殖しないと予期される。

- 生物学的試料中のpro−IGF−II濃度を決定するための方法であって、前記方法は、
生物学的試料を、請求項8記載のIGFBP−3ポリペプチド誘導体と接触させて、IGFBP−3ポリペプチド誘導体と、生物学的試料中に存在するあらゆるpro−IGF−IIとの間の複合体の形成を可能とさせる工程、ここで、pro−IGF−IIはIGF−IIの部分的にプロセシングされた形態である;および
複合体におけるSeAPのアルカリ活性を測定することによって、生物学的試料中のpro−IGF−IIの濃度を決定する工程
を含む、前記方法。 - 請求項8記載のIGFBP−3ポリペプチド誘導体と、アルカリ活性を測定するための少なくとも1つの試薬とを含むキット。
- N末端ドメイン、中間ドメインおよびC末端ドメインを含むIGFBP−3ポリペプチド誘導体であって、
N末端ドメインは、野生型ヒトIGFBP−3の、または、その生物学的に活性なフラグメントの、配列番号1に示されたN末端ドメインのアミノ酸配列からなり、ここで前記野生型ヒトIGFBP−3の生物学的に活性なフラグメントのN末端ドメインは、IGF−I及びIGF−IIに結合する野生型ヒトIGFBP−3のN末端ドメインの能力を保持し、;
中間ドメインは、野生型ヒトIGFBP−3の中間ドメインの配列番号2に示されたアミノ酸配列の85個又は85個超の連続したアミノ酸残基がタンパク質分解的開裂に対して抵抗性のあるリンカーで置換されたアミノ酸配列からなり、ここで前記タンパク質分解的開裂に対して抵抗性のあるリンカーは、配列番号4に示された配列を有し、;そして
C末端ドメインは、野生型ヒトIGFBP−3の、または、その生物学的に活性なフラグメントの、配列番号3に示されたC末端ドメインのアミノ酸配列からなり、ここで前記野生型ヒトIGFBP−3の生物学的に活性なフラグメントのC末端ドメインは、IGF−I、IGF−IIおよびヘパリンに結合する野生型ヒトIGFBP−3のC末端ドメインの能力を保持し、
配列番号3におけるアミノ酸残基43〜47が、ALSに結合しないAGGSG(配列番号5)またはその任意の変異体で置換されている、
前記IGFBP−3ポリペプチド誘導体。 - IGFBP−3ポリペプチド誘導体に対して融合した、免疫グロブリンIgG1Fcフラグメントをさらに含む、請求項14記載のIGFBP−3ポリペプチド誘導体。
- IGF−Iのアミノ酸配列をさらに含み、IGF−IがIGFBP−3ポリペプチド誘導体と複合体を形成している、請求項14記載のIGFBP−3ポリペプチド誘導体。
- IGFBP−3ポリペプチド誘導体のC末端ドメインの末端に共有結合したBirA酵素基質をさらに含む、請求項14記載のIGFBP−3ポリペプチド誘導体。
- BirA酵素基質が配列番号6に示された配列を有する、請求項17記載のIGFBP−3ポリペプチド誘導体。
- BirA酵素基質に共有結合したビオチンをさらに含む、請求項17または請求項18記載のIGFBP−3ポリペプチド誘導体。
- IGFBP−3ポリペプチド誘導体に対して融合した、SeAP(分泌型アルカリホスファターゼ)のアミノ酸配列をさらに含む、請求項14項記載のIGFBP−3ポリペプチド誘導体。
- 癌および増殖網膜症から選択された疾患の処置のための医薬品を製造するための請求項14、15、17および18記載のIGFBP−3ポリペプチド誘導体の使用。
- 成長ホルモン抵抗性、IGF−I欠乏症、重度の熱傷、HIV消耗、嚢胞性線維症、セリアック病、神経性食欲不振症、筋肉消耗疾病、筋緊張性ジストロフィー、筋萎縮性側索硬化症、骨粗鬆症、重度のインスリン抵抗性、I型糖尿病、II型糖尿病、脳虚血、心虚血、および移植からなる群より選択された疾患の処置のための医薬品を製造するための請求項16記載のIGFBP−3ポリペプチド誘導体の使用。
- 治療有効量の請求項14〜18のいずれか1項記載の少なくとも1つのIGFBP−3ポリペプチド誘導体と、薬学的に許容される担体または賦形剤とを含む、薬学的組成物。
- 生物学的試料中のpro−IGF−II濃度を決定するための方法であって、前記方法は、
生物学的試料を、請求項20記載のIGFBP−3ポリペプチド誘導体と接触させて、IGFBP−3ポリペプチド誘導体と、生物学的試料中に存在するあらゆるpro−IGF−IIとの間の複合体の形成を可能とさせる工程、ここで、pro−IGF−IIはIGF−IIの部分的にプロセシングされた形態である;および
複合体におけるSeAPのアルカリ活性を測定することによって、生物学的試料中のpro−IGF−IIの濃度を決定する工程
を含む、前記方法。 - 請求項20記載のIGFBP−3ポリペプチド誘導体と、アルカリ活性を測定するための少なくとも1つの試薬とを含むキット。
