Description
本発明は、第1の局面において、非形質転換組織または非形質転換細胞の増殖の増強および/または治癒および/またはアポトーシスの阻害に用いるための配列番号1のアミノ酸配列を含むタンパク質またはその断片もしくは配列番号1と少なくとも80%の配列同一性を有する変異体を提供する。
本発明を詳細に説明する前に、本発明は本明細書に記載の特定の方法論、プロトコール、および試薬に限定されず変化しうると理解すべきである。本明細書で用いている用語は特定の態様を説明するだけのためであり、本発明の範囲はそれに限定されるものではなく、特許請求の範囲によってのみ限定されると理解すべきである。特記しない限り、本明細書で用いているすべての技術および科学用語は、当業者が一般に理解しているのと同じ意味を有する。
好ましくは、本明細書で用いている用語は、「A multilingual glossary of biotechnological terms:(IUPAC Recommendations)」、H.G.W. Leuenberger、B. Nagel、and H. Koelbl編、Helvetica Chimica Acta、CH-4010 Basel、Switzerland、(1995)に記載のごとく定義される。
以下に本発明の要素について説明する。該要素は具体的態様と共に記載するが、さらなる態様を生み出すために幾つでも如何様にも組み合わせることができると理解すべきである。種々の記載した実施例および好ましい態様は明示的に記載した態様のみに限定されると解釈すべきではない。本明細書は、明示的に記載した態様とあらゆる数の開示したおよび/または好ましい要素を組み合わせる態様を支持し含むと理解すべきである。さらに、文脈がそうでないと示さない限り、本願中の記載したすべての要素のあらゆる置換と組み合わせが本願の明細書に開示されていると考えるべきである。
別の好ましい態様において、該非形質転換細胞または該非形質転換組織は、消化器系、内分泌系、排泄系、免疫系、外皮系、筋肉系、神経系、生殖系、呼吸器系、または骨格系を含む群から選ばれる個体の身体の特定の系に属するかまたは由来する。本発明の別の好ましい態様において、該細胞は、個体の記載した系の2、3、4、5、6、7、8、9、10、またはすべてに属する。
別の好ましい態様において、該調節は細胞タイプまたは組織タイプ特異的および生理的条件特異的である。特に好ましい態様において、該発現は心臓特異的発現である。特に別の態様において、該発現は心臓特異的および創傷特異的である。
その貫膜領域によりエフェクター細胞の膜と結合する。典型的には、本明細書で用いている用語「抗体」は、貫膜領域を欠き、血流および体腔中に放出されることができる分泌免疫グロブリンを表す。抗体は、それが持つ重鎖に基づいて種々のアイソタイプに分類される。ギリシャ文字、α、δ、ε、γ、およびμで示される5種類のヒトIg重鎖がある。存在する重鎖の種類は抗体のクラスを定義し、これらの鎖は、それぞれ異なる役割を果たし、異なるタイプの抗原に対する適切な免疫反応をもたらすIgA、IgD、IgE、IgG、およびIgM抗体にみられる。別個の重鎖は大きさと組成が異なり、αとγは約450アミノ酸を含むが、μとεは約550アミノ酸を有する(Janeway et al. (2001) Immunobiology、Garland Science)。抗体は、4つのポリペプチド鎖、すなわちジスルフィド結合で互いに結合した2つの重(H)鎖と2つの軽(L)鎖を含む。各重鎖は、重鎖可変領域(本明細書ではHCVRまたはVHと略す)と重鎖定常領域からなる。重鎖定常領域は、3つのドメイン、CH1、CH2、およびCH3からなる。各軽鎖は、軽鎖可変領域(本明細書ではLCVRまたはVLと略す)および軽鎖定常領域からなる。軽鎖定常領域は1つのドメインCLからなる。VHおよびVL領域は、さらに、フレームワーク領域(FR)と呼ばれるより保存された領域が組み込まれた相補性決定領域(CDR)と呼ばれる超可変領域に細分することができる。各VHおよびVLは、以下の順番でアミノ末端からカルボキシ末端に並んだ3つのCDRと4つのFRからなる:FR1、CDR1、FR2、CDR2、FR3、CDR3、FR4。重鎖と軽鎖のCDRは当該分野で知られたごとく決定することができる。例えば、以下の一連の規則を用いて各抗体軽鎖配列と重鎖配列内のCDRをみいだすことができる:

10〜12週齢の雄C57BL/6マウスをイソフルラン(1〜2%)で麻酔および換気し、一時的左前下行枝冠動脈結紮を1時間行い、次いで28日間再灌流した。マウスに、再灌流時に組換えファクター1またはファクター2(各10μg)を単回s.c.注射した(PBSをコントロールマウスに注射した)。次いで、Alzetミニポンプを用いて組換えファクター1またはファクター2を7日間連続s.c.注入した(10μg/日)。コントロールマウスにPBSを注入した。マウスを28日間毎日検査して梗塞後の生存を評価した。N=29 PBS処置マウス;N=25 ファクター1処置マウス;N=15 ファクター2処置マウス。
用いたパラメーターはデフォルトパラメーターである:11エクステンション:1、組成調整=非冗長タンパク質配列(nr)のデータベースと共に条件組成スコアマトリックス調整。
同定した配列から、種々の脊椎動物、主として哺乳類からの例と両生類、鳥類および魚類から1配列を選んだ。各配列を表1に記載する。
選んだアミノ酸配列をhttp://www.ebi.ac.uk/Tools/clustalw2/index.htmlのCLUSTALW2アルゴリズムを用いてアラインメントした。用いた各パラメーターはデフォルトパラメーターである:アラインメントタイプ=スロー、タンパク質重量マトリックス=Gonnet、ギャップオープン=10、ギャップエクステンション=0,20、ギャップ間隔=5、末端ギャップなし=なし、アウトプットオプション:フォーマット=Aln w/numbers、順序=アラインド。すべての配列について得られた複数のアラインメントを図6に、哺乳類配列に関するものを図7に示す。
用いたパラメーターはデフォルトパラメーターである:11エクステンション:1、組成調整=非冗長タンパク質配列(nr)のデータベースと共に条件組成スコアマトリックス調整。同定した配列から、種々の脊椎動物種、主として哺乳類からの例と、両生類および魚類から一配列を選んだ。各配列を表2に示す。
選んだアミノ酸配列をhttp://www.ebi.ac.uk/Tools/clustalw2/index.htmlのCLUSTALW2アルゴリズムを用いてアラインメントした。用いた各パラメーターはデフォルトパラメーターである:アラインメントタイプ=スロー、タンパク質重量マトリックス=Gonnet、ギャップオープン=10、ギャップエクステンション=0,20、ギャップ間隔=5、末端ギャップなし=なし、アウトプットオプション:フォーマット=Aln w/numbers、順序=アラインド。
すべての配列について得られた複数のアラインメントを図8に、哺乳類配列に関するものを図9に示す。
選んだアミノ酸配列を、http://www.ebi.ac.uk/Tools/clustalw2/index.htmlのCLUSTALW2アルゴリズムを用いてアラインメントした。用いた各パラメーターはデフォルトパラメーターである:アラインメントタイプ=スロー、タンパク質重量マトリックス=Gonnet、ギャップオープン=10、ギャップエクステンション=0,20、ギャップ間隔=5、末端ギャップなし=なし、アウトプットオプション:フォーマット=Aln w/numbers、順序=アラインド。すべての配列について得られた複数のアラインメントを図10に、哺乳類配列に関するものを図11に示す。
Claims (12)
- 虚血、再灌流傷害、機械的負荷、心筋梗塞または心不全を処置するための医薬の製造のための、配列番号1のアミノ酸配列を含むタンパク質または配列番号1のアミノ酸配列を含むタンパク質の断片もしくは変異体の使用であって、配列番号1のアミノ酸配列を含むタンパク質の断片もしくは変異体が、配列番号1と少なくとも90%の配列同一性を有し、抗アポトーシス潜在能力を示し、アポトーシスによる細胞死から細胞または組織を保護し、抗アポトーシス潜在能力が、配列番号1で示されるアミノ酸配列を有するタンパク質のそれの少なくとも50%である、使用。
- タンパク質が配列番号2のアミノ酸配列を含む、請求項1に記載のタンパク質、断片または変異体の使用。
- タンパク質が配列番号1のアミノ酸配列からなる、請求項1に記載のタンパク質、断片または変異体の使用。
- 心臓の虚血または再灌流傷害を処置するための医薬の製造のための、請求項1に記載のタンパク質、断片または変異体の使用。
- 心筋梗塞または急性心筋梗塞を処置するための医薬の製造のための、請求項1に記載のタンパク質、断片または変異体の使用。
- 虚血、再灌流傷害、機械的負荷、心筋梗塞または心不全を処置するための医薬の製造のための、請求項1〜3のいずれかに記載のタンパク質、断片または変異体をコードする核酸の使用。
- 虚血、再灌流傷害、機械的負荷、心筋梗塞または心不全を処置するための医薬の製造のための、請求項6に記載の核酸を含むベクターの使用。
- ベクターが以下からなる群から選ばれる請求項7に記載のベクターの使用:プラスミドベクター;コスミドベクター;ファージベクター、ラムダファージ、糸状ファージベクター;ウイルスベクター、アデノウイルスベクター、アデノ関連ウイルス(AAV)ベクター、アルファウイルスベクター、ヘルペスウイルスベクター、麻疹ウイルスベクター、ポックスウイルスベクター、水疱性口内炎ウイルスベクター、レトロウイルスベクターおよびレンチウイルスベクター;ウイルス様粒子;および細菌胞子。
- 請求項1〜3のいずれかに記載のタンパク質、断片または変異体、請求項6に記載の核酸または請求項7に記載のベクター、および適切な医薬的賦形剤を含む、虚血、再灌流傷害、機械的負荷、心筋梗塞または心不全を処置するための医薬組成物。
- 経口、静脈内、皮下、粘膜内、動脈内、筋肉内、または冠動脈内経路で投与される、請求項9に記載の医薬組成物。
- 再灌流療法の前、後またはそれと同時に投与される、請求項9または10に記載の医薬組成物。
- 該投与が1またはそれ以上のボーラス注射および/または注入によるものである、請求項11に記載の医薬組成物。
