Abstract
Description
(1)本発明の接着性分子
本発明の接着性分子は、NRVDNYKEVYNKIYNKRN(配列番号1)、又は配列番号1に対して保存的アミノ酸置換、欠失、挿入及び/又は修飾を含む配列を有する。本発明において、「保存的アミノ酸置換、欠失、挿入及び/又は修飾」とは、ペプチドの特性を失わない、つまり接着性分子の接着性を失わない置換、欠失、挿入及び/又は修飾を意味する。本発明において、接着性とは、物体表面に付着し、水洗しても物体表面から脱離しない性質を表し、実施例2に記載の試験で青色部分が確認された場合に、接着性を有すると判断する。この接着性を利用し、接着性のある物質に同時に二つの物体を接着させることによって、二つの物体を接着させ、結合させることができる。

あるアミノ酸配列に対する1から数個のアミノ酸残基の置換、欠失、挿入及び/又は修飾を含むアミノ酸配列を有するペプチドが、元のペプチドの性質を失わないことはすでに知られている(例えば、Mark, D. F. et al., Proc. Natl. Acad. Sci. USA (1984) 81, 5662-5666、Dalbadie-McFarland, G. et al., Proc. Natl. Acad. Sci. USA (1982) 79, 6409-6413を参照)。
(2)本発明の接着性分子の製造方法
本発明の接着性分子の製造方法は特に限定されないが、化学合成法や、細胞に産生させる方法が例として挙げられる。
(3)本発明の接着性分子の用途
本発明の接着性分子は、物体同士を接着する接着剤の有効成分として用いることができる。接着する物体は特に限定されないが、ガラス、プラスチックや樹脂等の高分子化合物、金属、木材、生体試料が例として挙げられる。接着する物体は、異なる物体を接着しても、同じ物体を接着してもよい。
(4)本発明の接着性分子を用いた接着方法及びコーティング方法
本発明の接着分子を含む接着剤を用いて、物体同士を接着することができる。例えば、本発明の接着剤を、物体表面に任意の方法、例えば滴下や塗布等によって付着させ、その上に別の物体を配置し、乾燥させると、それら物体同士を接着することができる。接着に用いる接着剤の量は目的に応じて任意に設定してよい。
配列番号1のアミノ酸配列に対応する塩基配列を決定し、その塩基配列の両末端に制限酵素BamHIの認識配列を付加した人工ポリヌクレオチド(配列番号2)を作製した。
<実施例2> 本発明の接着性分子のガラスへの接着性試験
精製した39s1Cを純水と混合して得たペプチド液、及びBSAを純水に溶解したペプチド液の、2種類のペプチド液を調製した。どちらのペプチド液も、ペプチド濃度は1mg/mlとした。各ペプチド液1μlをスライドグラス上に滴下し、次いで、凍結乾燥状態のチロシナーゼをリン酸カリウムバッファー(50mM、pH6.5)に溶解して調整したチロシナーゼ溶液(1mg/ml)1μlを滴下した各ペプチド液に加えてピペッティングによって軽く混合した。次いで、スライドグラスを22〜25℃の飽和水蒸気下に静置し、0分又は30分インキュベートした。インキュベート後、室温にて30分の減圧乾燥を行った。次いで、スライドグラスを蒸留水に浸したまま、45回/分の振とうで30分間洗浄を行い、再度、室温にて30分減圧乾燥を行った。乾燥後、スライドグラスを0.25%CBB(R‐250)溶液に一晩浸し、次いで蒸留水でスライドグラスを数回洗浄し、CBBによる染色で青色になった部分があるかを目視で確認した。ペプチドがガラス表面に付着していれば、CBB染色による青色部分が確認される。
<実施例3> 本発明の接着性分子の金属への接着性試験
精製した39s1Cを純水と混合して得たペプチド液を調製した。ペプチド濃度は1mg/mlとした。ペプチド液1μlをアルミニウム(Al)板、銅(Cu)板及び鉄(Fe)板表面に滴下した。次いでチロシナーゼ溶液1μl(1mg/ml、凍結乾燥状態のチロシナーゼをリン酸カリウムバッファー(50mM、pH6.5)に溶解し調製したもの)を加えてピペッティングによって軽く混合した。次いで、スライドグラスを22〜25℃の飽和水蒸気下に静置し、30分インキュベートした。インキュベート後、室温にて30分の減圧乾燥を行った。次いで、スライドグラスを蒸留水に浸したまま、45回/分の振とうで30分間洗浄を行い、再度、室温にて30分減圧乾燥を行った。乾燥後、スライドグラスを0.25%CBB(R-250)溶液に一晩浸し、次いで蒸留水でスライドグラスを数回洗浄し、CBBによる染色で青色になった部分があるかを目視で確認した。ペプチドが金属表面に付着していれば、CBB染色による青色部分が確認される。図4に示すように、どの金属板に対しても、39s1Cは付着していた。

- 前記接着性分子が少なくとも1つのジヒドロキシフェニルアラニンを含む、請求項1に記載の接着性分子。
- 請求項1又は2に記載の接着性分子を含む接着剤。
- 請求項1又は2に記載の接着性分子をコードする塩基配列を有するポリヌクレオチド。
- 請求項4に記載のポリヌクレオチドを含むベクター。
- 第1の物体を第2の物体に接着させる接着方法であって、請求項3に記載の接着剤を、第1の物体表面に付着させることを含む、接着方法。
- 請求項3に記載の接着剤を、物体表面に付着させることを含む、物体表面のコーティング方法。
- 接着性分子の接着性を増強する方法であって、チロシナーゼと、請求項1の接着性分子と、を反応させることを含む、方法。
- 配列番号1のアミノ酸配列、又は配列番号1の配列に対して保存的アミノ酸置換、欠失、挿入及び/又は修飾を含むアミノ酸配列を有する分子の接着性を調べる方法。
