Claims (4)
- 閉鎖した細胞増殖システムに細胞を直接再播種する方法であって、
毛細管内空間及び毛細管外空間を有する中空糸型バイオリアクターの細胞成長空間において、完全培地で細胞を成長させるステップであって、前記細胞成長空間は、前記毛細管内空間又は前記毛細管外空間のうち一方である、ステップと、
成長した前記細胞の細胞収集バッグへの収集を開始する前に、前記細胞収集バッグに収集培地を添加するステップであって、前記収集培地は代謝産物及び一定量のタンパク質を含み、前記一定量のタンパク質はFBSを含む、ステップと、
前記細胞成長空間から前記完全培地を除去するステップと、
前記細胞成長空間にトリプシンを添加することにより、前記細胞成長空間に対する前記細胞の付着を緩和するステップと、
前記細胞成長空間から前記細胞及び前記トリプシンを除去するステップと、
前記除去した細胞及びトリプシンを、前記収集培地が添加された前記細胞収集バッグに添加するステップであって、前記収集培地が細胞環境中のイオンの不足を補い、前記一定量のタンパク質が前記トリプシンを不活性化するステップと、
前記中空糸型バイオリアクターの前記細胞成長空間に前記除去した細胞を直接再播種するステップと、
を有する直接再播種する方法。 - 請求項1記載の方法において、前記細胞及び前記トリプシンを除去するステップはさらに、前記毛細管内空間又は前記毛細管外空間のうち前記細胞成長空間ではない方から前記中空糸型バイオリアクターの中空糸を通って前記細胞成長空間に流体を押し流すステップを含むことを特徴とする方法。
- 請求項1記載の方法において、
前記細胞収集バッグは、前記中空糸型バイオリアクターの前記細胞成長空間に繋がる流体循環路に接続されたチューブラインに接続されており、
前記除去した細胞及びトリプシンを前記細胞収集バッグに添加するステップでは、前記チューブラインを介して前記除去した細胞及びトリプシンを前記細胞収集バッグへと運ぶ、
ことを特徴とする方法。 - 請求項1記載の方法において、前記細胞収集バッグに前記収集培地を添加するステップはさらに、前記収集培地を容器から前記細胞収集バッグにポンピングするステップを含むことを特徴とする方法。
