Description
Claims (17)
- 細胞成長チャンバー内を循環する流体から成分を濃縮する方法であって、該方法は、
第1のポンプで前記細胞成長チャンバー内に流体を循環させる工程と、
前記細胞成長チャンバー内で細胞を増殖させる工程と、
第1の流体を用いて、前記細胞成長チャンバーから、増殖した細胞を取り出す工程と、
前記増殖した細胞を含む前記第1の流体を、前記細胞成長チャンバーと流体的に関連付けられた接線流フィルタに、前記細胞成長チャンバーのポートから直接流入させ、前記接線流フィルタに沿って前記第1のポンプで循環させることで、濃縮された流体成分と濃縮された細胞成分とを生成する工程と、
前記濃縮された流体成分の少なくとも一部を、容器に集める工程と、
を有し、
前記増殖させる工程は、
前記細胞成長チャンバー内の毛細管内側空間において、第1の流体培地を前記第1のポンプにより循環させる工程と、
前記細胞成長チャンバー内の毛細管外側空間に、第2の流体培地を第2のポンプにより循環させる工程と、
を有し、
前記毛細管外側空間は、酸素供給器と流体連通していることを特徴とする方法。 - 請求項1記載の方法において、前記濃縮された細胞成分を、前記接線流フィルタに沿って前記第1のポンプで再循環させることで、前記濃縮された細胞成分中に前記細胞を濃縮する工程をさらに有することを特徴とする方法。
- 請求項1又は2記載の方法において、前記第1の流体を前記接線流フィルタに沿って循環させる前に、前記第1の流体に第2の流体を加える工程をさらに有することを特徴とする方法。
- 請求項2記載の方法において、前記濃縮された細胞成分を前記接線流フィルタに沿って再循環させる前に、前記濃縮された細胞成分に第2の流体を加える工程をさらに有することを特徴とする方法。
- 請求項1記載の方法において、前記細胞成長チャンバーと前記接線流フィルタは、閉鎖系の一部であることを特徴とする方法。
- 請求項2記載の方法において、前記再循環させる工程は、前記濃縮された細胞成分中における細胞濃度が所定濃度に達するまで、行われることを特徴とする方法。
- 請求項1記載の方法において、前記濃縮された細胞成分を、前記細胞成長チャンバーに戻す工程をさらに有することを特徴とする方法。
- 請求項1記載の方法において、
前記増殖工程は、前記酸素供給器で、前記第2の流体培地に酸素を供給する工程を有することを特徴とする方法。 - 請求項1〜8のいずれか1項に記載の方法において、前記細胞成長チャンバーは、中空糸膜装置を備えることを特徴とする方法。
- 請求項1〜9のいずれか1項に記載の方法において、前記接線流フィルタは、中空糸膜装置を備えることを特徴とする方法。
- 請求項1又は8記載の方法において、前記酸素供給器は、中空糸膜装置を備えることを特徴とする方法。
- 複数の細胞を増殖させるシステムであって、該システムは、
第1流体流路と第2流体流路とを有する細胞成長チャンバーと、
前記細胞成長チャンバーの前記第1流体流路と流体的に関連付けられた接線流フィルタと、
前記細胞成長チャンバーの前記第1流体流路において流体を循環させ、且つ前記細胞成長チャンバーで増殖した細胞を含む流体を前記接線流フィルタに沿って循環させる第1のポンプと、
を備え、
前記第1流体流路は、少なくとも両端部を有し、
前記第1流体流路の第1端部は、中空糸膜の第1ポートと流体的に関連付けられ、前記第1流体流路の第2端部は、前記中空糸膜の第2ポートと流体的に関連付けられ、前記第1流体流路は、前記中空糸膜の毛細管内側部分を有し、
前記第2流体流路は、少なくとも両端部を有し、
前記第2流体流路の第1端部は、前記中空糸膜の第3ポートと流体的に関連付けられ、前記第2流体流路の第2端部は、前記中空糸膜の第4ポートと流体的に関連付けられ、前記第2流体流路は、前記中空糸膜の毛細管外側部分を有し、
前記流体が周囲環境に曝されることなく前記中空糸膜から前記接線流フィルタに直接流れることを特徴とするシステム。 - 請求項12記載のシステムにおいて、前記第2流体流路と流体的に関連付けられた酸素供給器をさらに備えることを特徴とするシステム。
- 請求項12又は13記載のシステムにおいて、前記細胞成長チャンバーの前記第2流体流路に流体を循環させる第2のポンプをさらに備えることを特徴とするシステム。
- 請求項12〜14のいずれか1項に記載のシステムにおいて、前記細胞成長チャンバーは、中空糸膜装置を備えることを特徴とするシステム。
- 請求項12〜15のいずれか1項に記載のシステムにおいて、前記接線流フィルタは、中空糸膜装置を備えることを特徴とするシステム。
- 請求項13に記載のシステムにおいて、前記酸素供給器は、中空糸膜装置を備えることを特徴とするシステム。
