Description
1) 特許US5,384,330に開示された以下のような構造を有する化合物。
本発明のまたもう一つの目的は、有効成分として前述化合物またはその薬学的に許容される塩と、薬学的に許容される補助剤とを含む医薬品組成物を提供することである。
本発明のさらにもう一つの目的は、神経系疾患などを治療する医薬品の製造における、上述化合物およびその薬学的に許容される塩またはこれらのいずれかを含む医薬品組成物の用途を提供することである。
Lは、存在しないか、あるいは任意にC1-C4アルキル基で置換されてもよい、
R3は、H、ハロゲン原子、トリフルオロメチル基、シアノ基、ニトロ基、C1-C4アルキルカルボニル基、C1-C4アルコキシカルボニル基から選ばれる。
Xは、O、SあるいはNHである。
Yは、存在しないか、あるいはOもしくはNR7である。ここで、R7は、H、C1-C6アルキル基、C2-C6アルケニル基、C2-C6アルキニル基、C3-C8シクロアルキル基から選ばれる。

トルエンスルホン酸のような弱酸の触媒下で、カルボニル化合物aとアニリンbとを縮合させてイミンcを得た後、後者を水素化ホウ素ナトリウムのような還元剤の作用によってイミン還元させて第二級アミンdを得る。第二級アミンdにハロゲン化炭化水素との置換反応、α,β-不飽和カルボニル化合物の付加反応またはアシル化反応などの手段で置換基R1を導入して中間体fを得る。当業者によく知られる公知の条件で中間体dまたはfにおけるアミノ保護基Pを脱離させて中間体fまたはgを得た後、アミンfまたはgをアシル化剤である
トルエンスルホン酸のような弱酸の触媒下で、カルボニル化合物aとニトロ基で置換されたフェニレンジアミン系化合物hとを縮合させてイミンiを得た後、後者を水素化ホウ素ナトリウムのような還元剤の作用によってイミン還元させて中間体であるアミンjを得る。当業者によく知られる方法、例えば、Pd-C触媒水素化やSnCl2(塩化第一スズ)還元でニトロ基を還元させてジアミノ中間体kを得て、2つのアミノ保護基Pを導入して中間体lを得て、方法1と類似の方法で置換基R1を導入して中間体mを得た後、保護基Pを除去して2つのアミノ基を露出させ、中間体nを得た後、アシル化剤である

一般式Iで表される化合物における置換基R1がアルケニル基あるいはアルキニル基である場合、さらにオレフィンメタセシス反応(olefin metathesis)で他の一般式Iで表される本発明に係る化合物を得る。前述通常のオレフィンメタセシス反応は、金属ルテニウムカルベン錯体(Grubbs触媒)の触媒下でオレフィン同士間、オレフィンとアルキンとの再結合反応を行うが、反応の一般式は以下の通りである。
本発明に係る一般式Iで表される化合物は、その薬学的に許容される塩、例えば、塩酸塩などに転換してもよいが、通常の方法は、相応の酸の溶液を上述化合物の溶液に入れ、塩形成が完了した後、減圧で溶媒を除去することで、本発明に係る化合物に相応の塩を得る。
本発明のまた一つは、KCNQカリウムチャネル作動薬としての上述化合物およびその薬学的に許容される塩またはこれらのいずれかを含む医薬品組成物の用途、特に神経系疾患などを治療する医薬品の製造における用途を提供する。
前述神経系疾患は、癲癇、痙攣、神経因性疼痛、急性虚血発作および神経変性疾患を含む。
本発明に記載の新規な化合物は、従来の医薬品であるレチガビン(Retigabine)と比べて、性質がより安定し、酸化変性しにくい。
同時に、本発明によって提供される化合物は、Retigabineのカリウムチャネル活性化の活性を維持するだけでなく、体内における抗癲癇効果が顕著で、より高い脳組織における分布濃度を有するため、より優れた治療効果がある。
以下、具体的な実施例で本発明をさらに説明するが、本発明はこれらの実施例に制限されない。
下述製造実施例において、NMRはVarian製のMercury-Vx 300Mで測定し、較正はδH 7.26 ppm(CDCl3)、2.50 ppm(DMSO-d6)、3.15 ppm (CD3OD)である。試薬は、主に上海化学試薬公司によって提供された。TLC薄層クロマトグラフィーは、山東煙台会友シリカゲル開発有限公司製、型式HSGF 254である。化合物精製に使用された順相カラムクロマトグラフィーのシリカゲルは、山東青島海洋加工工場分工場製、型式zcx-11、200-300メッシュである。

製造実施例8と類似の方法を使用し、それぞれ化合物K3の塩酸塩K3・HCl、K17の塩酸塩K17・HCl、K18の塩酸塩K18・HClおよび化合物K20の二塩酸塩K20・2HClを得た。
1. 細胞の培養とトランスフェクション
チャイニーズハムスター卵巣(CHO)細胞(中国科学院細胞庫)の培養液の配合:50/50 DMEM/F-12(Cellgro, Mamassas, VA)に10%ウシ胎児血清(FBS) (Gibco, オーストラリア)、2mM L-グルタミン酸(Invitrogen)を入れた。KCNQチャネルの発現およびその突然変異:トランスフェクションの24時間前に、トリプシン(Sigma, 中国)で消化した後、直径60mmの培養皿に展開した。トランスフェクションは、Lipofectamine2000TM試薬(Invitrogen)を使用し、そのプロトコールに従って操作した。トランスフェクションの24時間後、細胞を消化して再度poly-L-lysine(Sigma)に浸漬したスライドガラスに展開した。GFP(緑色蛍光タンパク質)と一緒にコトランスフェクションし、蛍光顕微鏡でトランスフェクション細胞を確認することができた。
室温で、Axopatch-200B増幅器(Molecular Devices, Sunnyvale, CA)で全細胞電圧クランプ記録を行った。ホウケイ酸ガラス毛細管(World Precision Instruments, Sarasota, FL)を伸ばして電極とし、電極内に細胞内液を充填した後の抵抗が3-5MΩである。細胞内液の配合:145mM KCl、1mM MgCl2、5mM EGTA、10mM HEPES、5mM MgATP(KOHでpH=7.3に調整した)。記録の間、BPSかん流システム(ALA Scientific Instruments, Westburg, NY)で細胞外液をかん流し続けた。細胞外液:140mM NaCl、5mM KCl、2mM CaCl2、1.5mM MgCl2、10mM HEPES、10mMグルコース(NaOHでpH=7.4に調整した)。電気信号を1kHzにおいてフィルターにかけた後、DigiData 1322AでpClamp 9.2ソフトウェア(Molecular Devices, Sunnyvale, CA)によってデジタル信号に転換した。抵抗を直列に接続して信号損失の60-80%補い、現在の研究に多段電圧スキームを使用した。スキームにおけるクランプ電圧は-100mVとした。細胞を一連の2000msの刺激電圧(-90mVから+50mVまで、間隔10mV)で電流を誘導した。
ここで、V1/2は半数活性化電圧で、ΔV1/2は半数活性化電圧の左へのシフト値で、負号(-)は電流活性化曲線の左へのシフトを、正号(+)は右へのシフトを表す。I/I0は活性化の倍率で、ここで、I0は-10mVのテスト電圧の刺激で生じた電流のピーク値の最大値で、Iは投与(化合物濃度が10μM)後同様に-10mVのテスト電圧の刺激で生じた電流のピーク値の最大値で、I/I0>1は作動活性、I/I0<1は阻害活性を表す。Nは測定した細胞の数である。NTは未測定を表す。
体内効果実施例1: 化合物K9とK1のペンチレンテトラゾール(PTZ)による痙攣に対する治療作用
1. 実験の目的:
本発明の化合物K9、K1とRetigabine(実験室でUS538,433,0に開示された方法を参照して自制したもの)の抗痙攣作用を比較することである。
2. サンプルの処理:
Retigabine: 淡灰色粉末で、水に溶けやすい。無色で、透明の液体である。
化合物K9: 黄色顆粒で、水に溶けにくい。0.2%CMC溶液を入れた後、30min超音波処理したところ、均一の懸濁液となった。
化合物K1: 淡黄色顆粒で、水に溶けにくい。0.2%CMC溶液を入れた後、30min超音波処理したところ、均一の懸濁液となった。
C57BL/6Jマウス(中国科学院実験動物センターから購入):16-18グラム。
4. 実験の手順
被験医薬品(30mg/kg)を胃かん流で投与した。1時間後、ペンチレンテトラゾール(PTZ、80mg/kg)を皮下注射した。すぐ動物の60min以内の反応を観察した。
1. 実験の目的:
K21と陽性化合物のRetigabineの抗痙攣作用を比較することである。
2. サンプルの処理:
Retigabine: 淡灰色粉末で、水に溶けやすい。無色で、透明の液体である。
K21: 茶青色層状顆粒で、水に溶けにくい。50ulのDMSOで溶解し、さらに0.5%HEC溶液を入れたところ、均一の懸濁液となった。
雄KMマウス(中国科学院実験動物センターから購入): (22±2)グラム。
4. 実験の手順:
(1) PTZによるテスト:各種の被験医薬品を腹腔注射した(10 mg/kg、5mg/kgm2.5mg/kg、0.1ml/10g)。30min後、ペンチレンテトラゾール(PTZ、100mg/kg)を皮下注射した。すぐ動物の1h以内の反応を観察して記録した。
化合物K21は、MESによるテストにおいて完全に大きい発作の発生を抑制し、PTZによるテストにおいて動物の痙攣の発生率および死亡率を顕著に低下させ、PTZによる急性痙攣のマウスのクローヌスの潜伏期間を延ばし、且つ一定の投与量に依頼する効果の関係が存在する。化合物K21は、抗痙攣効果が同じ投与量で陽性化合物Retigabineよりもやや強かった。両者は、いずれも一定の鎮静作用を示した。
1. 実験の目的:
K21と陽性化合物のRetigabineの経口投与による抗痙攣作用を比較することである。
2. サンプルの処理:
Retigabine: 淡灰色粉末で、水に溶けやすい。無色で、透明の液体である。
K21: 茶青色層状顆粒で、水に溶けにくい。50ulのDMSOで溶解し、さらに0.2%CMC溶液を入れ、20min超音波処理したところ、均一の懸濁液となった。
3. 実験動物:
雄KMマウス(中国科学院実験動物センターから購入):PTZ実験において、動物体重(22±2)グラム、MES実験において、動物体重18グラム。
(1) PTZによるテスト
動物を12h断食させ、胃かん流で被験化合物CF341とretigabine(30mg/kg、0.2ml/10g)を投与し、1h後、ペンチレンテトラゾール(PTZ、100mg/kg)を皮下注射し、すぐ動物の1h以内の反応を観察して記録した。
(2)MESによるテスト
YLS-9A型生理薬理電子刺激機を使用し、セット8とし、刺激電圧160V、波数90個(即ち、刺激時間5.4秒)とした。耳にクリップを挟んで電気刺激を1回与えた後、後ろ足の強直性伸長を痙攣を指標とし、実験の前日にスクリーニングした。
動物を12h禁食させ、胃かん流で各種被験医薬品(30mg/kg、0.2ml/10g)を投与し、1h後前日に設定したパラメーターでMESテストを行い、すぐ動物の反応を観察して記録した。
化合物K21とretigabineは、いずれもMESによるテストにおいて顕著に大きい発作の発生を抑制し、且つ効果が同等であった。PTZによるテストにおいて、retigabineは、動物の痙攣の発生率および死亡率を顕著に低下させたが、化合物K21は、顕著に動物の強直発作を抑制し、死亡率が対照群よりもすこし低下したが、顕著な差がなかった。
薬物動態学的研究実施例1: K9とRetigabineのマウスの脳組織における分布の試験研究
1. 投与スキーム
雄、体重18-20gの健康KMマウス108匹を、試験前に12h断食させ、自由に水を飲ませた。投与の2h後、同時に食事させた。具体的な手順は下述表に示す。
マウスの胃かん流と静脈投与後、以上の時刻で、各時刻に3匹ずつ腹大動脈から血を出して殺処分した。各動物から0.5mlの全血を収集し、ヘパリン化試験管に置き、3000rpmで10min遠心して血漿を分離し、-20℃の冷蔵庫に冷凍保存した。動物を殺処分した後、全脳を摘出して解剖し、冷やした生理食塩水で残留の血液を流し、乾燥した後、ラベルを貼り、-20℃の冷蔵庫に冷凍保存した。
液体クロマトグラフ-質量分析で血漿および脳組織におけるK9とRetigabineの濃度を測定した。
2.1. K9
マウスに胃かん流で20mg/kgの化合物K9を投与した後、血漿および脳組織におけるピーク濃度Cmaxはそれぞれ2197 ng/mlおよび4421 ng/gであった。濃度-時間曲線下の面積AUC0-tはそれぞれ1865 ng・h/mlおよび3565 ng・h/gであった。脳組織における被ばく量は、血漿の約1.9倍に相当した。消失半減期は約1.2hであった。
マウスに静脈注射で20mg/kgの化合物K9を投与した後、血漿および脳組織における濃度-時間曲線下の面積AUC0-tはそれぞれ7185 ng・h/mlおよび10694 ng・h/gで、血漿クリアランスは2.78 L/h/kgであった。化合物K9の脳組織における被ばく量は、血漿の約1.5倍に相当した。
投与量で修正した後、血漿AUC0-tで計算すると、マウスに胃かん流で20mg/kgの化合物K9を投与した後の生物学的利用能の絶対値が26%であった。
マウスに胃かん流で20mg/kgのRetigabineを投与した後、血漿および脳組織におけるピーク濃度Cmaxはそれぞれ2788 ng/mlおよび849 ng/gであった。濃度-時間曲線下の面積AUC0-tはそれぞれ21088 ng・h/mlおよび3460 ng・h/gであった。脳組織における被ばく量は、血漿の約16%に相当した。消失半減期は約7hであった。
マウスに静脈注射で20mg/kgのRetigabineを投与した後、血漿および脳組織における濃度-時間曲線下の面積AUC0-tはそれぞれ59987 ng・h/mlおよび8661ng・h/gで、血漿クリアランスは0.322 L/h/kgであった。Retigabineの脳組織における被ばく量は、血漿の約14%に相当した。
投与量で修正した後、血漿AUC0-tで計算すると、マウスに胃かん流で20mg/kgのRetigabineを投与した後の生物学的利用能の絶対値が35.2%であった。
1. 投与スキーム
雄、体重18-20gのKMマウス54匹を、試験前に12h断食させ、自由に水を飲ませた。投与の2h後、同時に食事させた。具体的な手順は下述表に示す。
マウスの胃かん流投与後、以上の時刻で、各時刻に3匹ずつ腹大動脈から血を出して殺処分した。各動物から0.5mlの全血を収集し、ヘパリン化試験管に置き、11000rpmで10min遠心して血漿を分離し、-20℃の冷蔵庫に冷凍保存した。動物を殺処分した後、全脳を摘出して解剖し、冷やした生理食塩水で残留の血液を流し、乾燥した後、ラベルを貼り、-20℃の冷蔵庫に冷凍保存した。
2.1. K20・2HCl
マウスに胃かん流で20mg/kgの化合物K20・2HClを投与した後、血漿および脳組織における元の形態の薬物K20・2HClと代謝物retigabineの濃度がピークに達した時間Tmaxはいずれも0.25hで、 代謝物retigabineの血漿および脳組織における被ばく量は、それぞれ元の形態の薬物K20・2HClの2.9%および1.8%であった。元の形態の薬物K20・2HClの脳組織における濃度は、血漿中濃度の2.3倍で、代謝物retigabineは1.4倍であった。
マウスに胃かん流で5mg/kgの化合物K21・2HClを投与した後、血漿および脳組織における元の形態の薬物K21・2HClと代謝物retigabineの濃度がピークに達した時間Tmaxはいずれも0.25hで、代謝物retigabineの血漿および脳組織における被ばく量は、それぞれ元の形態の薬物K21・2HClの5.4%および4.6%であった。元の形態の薬物K21・2HClの脳組織における濃度は、血漿中濃度の2.4倍で、代謝物retigabineは2.0倍であった。
1. 雄KMマウスモデルにおいて、腹腔注射による投与の場合、化合物K21・2HClの抗痙攣効果が陽性化合物Retigabineよりも優れた。
2. 同様に、雄KMマウスモデルにおいて、10mg/kgの投与量で胃かん流で投与した場合、化合物K21・2HClとRetigabineは、MESテストにおいて効果が同等で、顕著に大きい発作の発生を抑制することができた。PTZテストにおいて、Retigabineは、顕著に動物の痙攣の発生率と死亡率を低下させ、化合物K21・2HClは顕著に動物の強直発作を抑制することができた。
1. マウスに胃かん流で20mg/kgの化合物K9を投与した後、化合物の脳組織における被ばく量は、血漿の約1.9倍に相当した。マウスに静脈注射で20mg/kgの化合物K9を投与した後、化合物の脳組織における被ばく量は、血漿の1.5倍に相当した。同じ条件で、Retigabineの胃かん流および静脈注射による投与の場合、そのマウスの脳における被ばく量は、それぞれ血漿の16%および14%であった。
2. マウスに胃かん流で20mg/kgの化合物K20を投与した後、化合物の脳組織における被ばく量は、血漿濃度の2.3倍であった。
3. マウスに胃かん流で5mg/kgの化合物K21・2HClを投与した後、化合物の脳組織における被ばく量は、血漿濃度の2.4倍であった。
Claims (6)
- 以下の一般式IIIで表される構造を有する化合物またはその薬学的に許容される塩。
R1は、C 2-C8アルケニル基、任意にヒドロキシ基、アミノ基、ハロゲン原子、フェニル基、もしくはハロゲン化フェニル基で置換されたC2-C8アルケニル基、C 2-C8アルキニル基、任意にヒドロキシ基、アミノ基、ハロゲン原子、フェニル基、もしくはハロゲン化フェニル基で置換されたC2-C8アルキニル基から選ばれる。
R2は、F、Clあるいはメトキシ基から選ばれる。
R3は、H、ハロゲン原子あるいはトリフルオロメチル基から選ばれる。
Yは、Oである。
R4は、C1-C6アルキル基、C2-C6アルケニル基、C2-C6アルキニル基、あるいはC6-C10アリール基から選ばれる。
R5は、H、ハロゲン原子、アミノ基、
R6は、H、あるいはC1-C6アルキル基である。) - 前述化合物は、以下の化合物K4、K5、K7〜K25、K28〜K37およびK39から選ばれることを特徴とする請求項1に記載の化合物またはその薬学的に許容される塩。
- 前述化合物の薬学的に許容される塩は、前述化合物が酸と形成した塩で、前述酸は、マレイン酸、コハク酸、クエン酸、酒石酸、フマル酸、ギ酸、酢酸、プロパン酸、マロン酸、シュウ酸、安息香酸、フタル酸、メタンスルホン酸、ベンゼンスルホン酸、トルエンスルホン酸、ナフタレンスルホン酸、1,5-ナフタレンジスルホン酸、カンファー酸、カンファースルホン酸、サリチル酸、アセチルサリチル酸、アスパラギン酸、グルタミン酸、乳酸、グルコン酸、アスコルビン酸、没食子酸、アミグダリン酸、リンゴ酸、ソルビン酸、トリフルオロ酢酸、タウリン、ホモタウリン、2-ヒドロキシエタンスルホン酸、ケイ皮酸、塩酸、臭化水素酸、ヨウ化水素酸、硫酸、硝酸、リン酸または過塩素酸から選ばれることを特徴とする請求項1に記載の化合物またはその薬学的に許容される塩。
- 有効成分として請求項1に記載の化合物またはその薬学的に許容される塩と、薬学的に許容される補助剤とを含むことを特徴とする医薬品組成物。
- 神経系疾患を治療する医薬品の製造における、請求項1に記載の化合物またはその薬学的に許容される塩の使用。
- 前述神経系疾患は、癲癇、痙攣、神経因性疼痛、急性虚血発作および神経変性疾患を含むことを特徴とする請求項5に記載の使用。
