Description
・(i)パッケージング、逆転写、及び転写に必要なレンチウイルス、特にHIV−1のシス作用配列を含み、更に機能性のレンチウイルス、特にHIV−1由来のDNA flapエレメント、及び(ii)制御発現配列の制御下で、本明細書に開示されるマラリア寄生虫の抗原のポリペプチドをコードするポリヌクレオチドを含み、任意で統合のための配列を含む、ベクタープラスミド;
・RNAウイルス由来の偽型化エンベロープをコードする発現プラスミド、ここで前記発現プラスミドは、偽型化のためのエンベロープタンパク質(単数)又はタンパク質(複数)をコードするポリヌクレオチドを含み、前記エンベロープ偽型化タンパク質は、有利にVSV由来であり、特にVSV−G若しくはそれらの変異体由来である;及び
・レンチウイルス、特にHIV−1の、統合能力のあるベクター粒子の産生に好適なgag−polパッケージング配列、又は統合欠損ベクター粒子の産生に好適な改変されたgag−polパッケージング配列のいずれかを含む、キャプシド形成プラスミド、
を用いた、哺乳動物細胞の共トランスフェクションから回収される生成物である。
・(i)パッケージング、逆転写、及び転写に必要なレンチウイルス、特にHIV−1のシス作用配列を含み、機能性のレンチウイルス、特にHIV−1のDNA flapエレメントを更に含み、任意で統合に必要なシス作用配列を含むベクタープラスミド、ここで前記ベクタープラスミドは更に、(ii)制御発現配列、特にプロモーターの制御下で、プラスモディウム属寄生虫のcs遺伝子の切断された、哺乳動物、特にヒトコドン最適化配列のポリヌクレオチドを含む;
・VSV−Gエンベロープタンパク質(単数)又はエンベロープタンパク質(複数)をコードするポリヌクレオチドを含むVSV−Gエンベロープ発現プラスミド、ここで前記ポリヌクレオチドは、制御発現配列、特に誘導可能プロモーターを含む制御発現配列の制御下にある;及び
・キャプシド形成プラスミドがレンチウイルス、特にHIV−1の統合能力のあるベクター粒子の産生に好適なgag−polコーディング配列、又は統合欠損ベクター粒子の産生に好適な改変されたgag−polコーディング配列のいずれかを含み、前記gag−pol配列がDNA flapエレメントとして同一のレンチウイルスサブファミリー由来であり、前記レンチウイルスgag−pol若しくは改変されたgag−pol配列が制御発現配列の制御下にある、キャプシド形成プラスミド;
を用いた安定的な細胞株から回収された生成物である。
1.逆転写に要求されるLTR配列(長い末端反復)、該配列は、転写のために要求され、ウイルスDNA統合のための配列を任意で含む。3’LTRは、二つの主な理由:第一に、DNAがゲノムに統合されてすぐの、宿主遺伝子のトランス活性化を避けるため、及び第二に、逆転写(retrotranscription)後、ウイルスのシス配列の自己不活性化を可能にするために、遺伝子輸送のために必要な機能を撹乱させることなく、少なくとも(自己不活性化する)SINベクターを提供するために、プロモーターについて、U3領域において欠失している。任意で、ゲノムの転写を促す5’−LTRからのtat−依存U3配列は、非内因性プロモーター配列により置き換えられる。従って、標的細胞において、内在プロモーターからの配列のみが転写される(導入遺伝子)。
2.ウイルスRNAキャプシド形成のために必要なΨ領域。
3.Revタンパク質の結合後に、核から細胞質ゾルまでのウイルスのRNAメッセンジャーの輸送を可能にする、RRE配列(REV応答エレメント)。
4.核内への輸送を容易にするDNA flapエレメント(cPPT/CTS、通常Polに含まれる)
5.任意で、転写後エレメント、例えばmRNAの安定性を最適化するためにまた付加されるWPREシス作用配列(ウッドチャック肝炎B型ウイルス応答後エレメント)、マトリックス又はスキャホールド付着領域(SAR及びMAR配列)、例えば免疫グロブリン−カッパ遺伝子の配列(Park F.et al Mol Ther 2001;4:164−173)。
1.マトリックス(MA、見かけの分子量 p17)、キャプシド(CA、p24)及びヌクレオキャプシド(NC、p6)の結合のためのGAGタンパク質。
2.酵素:インテグラーゼ、プロテアーゼ、及び逆転写酵素をコードするPOL。
3.TAT及びREV制御タンパク質、ここでTATは、LTR媒介転写の開始に必要であり;5’LTRのU3領域がtat−独立転写に促されるプロモーターと置換される場合、TAT発現は除かれてもよい。REVは改変されてもよく、例えばドメインの認識がベクターゲノム中のRRE配列を置き換える得る、組み換えタンパク質に適宜用いられてもよく、又はそのRBD(RNA結合ドメイン)を通してはRRE配列と結合し得るフラグメントとして用いられてもよい。
(i)第一の決定された異種の、ウイルスエンベロープ偽型化タンパク質(単数)又はウイルスエンベロープ偽型化タンパク質(複数)で偽型化された本発明のレンチウイルスベクター粒子;
(ii)(i)におけるレンチウイルスベクター粒子とは別に提供される、前記第一の異種のウイルスエンベロープ偽型化タンパク質(単数又は複数)とは異なる、第二の決定された異種のウイルスエンベロープ偽型化タンパク質(単数)又はウイルスエンベロープ偽型化タンパク質(複数)で偽型化された本発明のレンチウイルスベクター粒子;
ここで、前記第一及び第二のウイルスエンベロープ偽型化タンパク質(単数又は複数)は、互いに血清中和せず、哺乳動物細胞、特にヒト細胞のインビボ形質導入に好適である;
を含む。
・ヒト宿主において、特にプラスモディウム・ファルシパルム(Plasmodium falciparum)、プラスモディウム・マラリアエ(Plasmodium malariae)、プラスモディウム・ビバックス(Plasmodium vivax)、プラスモディウム・ノウレシ(Plasmodium knowlesi)又はプラスモディウム・オバール(Plasmodium ovale)によるマラリア寄生虫感染に対する、無菌保護を誘発すること;
・マラリア寄生虫の細胞外形態を阻害すること;
・マラリア寄生虫による肝細胞感染を予防すること、又は感染の肝臓ステージ増幅の阻害;
・マラリア寄生虫抗原(単数又は複数)に対する特異的T細胞免疫応答、特にCD8+T細胞応答及び/又は特異的CD4+T細胞応答を誘発すること;
・寄生虫抗原(単数又は複数)に対するB細胞応答を誘発すること;
・マラリア寄生虫による感染の影響を減少又は緩和するために寄生虫血症を制御すること;
・寄生虫による感染又は寄生虫により誘導される病理に対する保護細胞性免疫を誘発すること;
・メモリーT細胞免疫応答を誘発すること;
・マラリア寄生虫による感染の間、CD4+及びCD8+T細胞応答の早期の及び高い回復を誘発すること;
・プラスモディウム属感染に関して肝臓内のメモリーリンパ球を刺激することによる早期の強力なCT(CD8+T)応答を誘発すること;
・マラリア寄生虫が免疫応答から回避することを防ぐこと、従って、マラリア寄生虫による感染の長期の制御を可能にすること;
の少なくとも1つを得るために好適である用量、及び投与計画において、哺乳動物宿主、特にヒト宿主に、マラリア寄生虫感染又は寄生虫に誘導される病理に対する予防的免疫付与のために、特に用いられる。
動物及び寄生虫
Balb/c Ola Hsd(6週齢、雌性)は、Harlan Laboratories(Gannat,France)から購入された。全ての動物実験は、Animal Care at the Pasteur Instituteのガイドラインに従って実行された。感染実験は、プラスモディウム・ヨエリ(Plasmodium yoelii)(17XNL株)野生型、又は生きている蚊中でオーシスト及びスポロゾイトの検出を可能にするために、緑色蛍光タンパク質を発現する、遺伝的改変体を用いて行なわれた。プラスモディウム・ヨエリ(Plasmodium yoelii)は、ステフェンスハマダラカ(Anopheles stephensi)及びBalb/cマウスにおいて、代替のサイクルの推移(cyclic passages)により、維持された。蚊は、標準的な手順を用いて、Pasteur InstituteのCenter for Production and Infection of Anopheles (CEPIA)において飼育された。
Py CSタンパク質(アミノ酸1−367;GenBank登録番号M58295)の全長についてコードする遺伝子の哺乳動物コドン最適化形態は、Geneartにより合成された。GPIアンカーリングモチーフの欠失が、CSタンパク質の免疫原性を向上させることが示されているので、我々は、最後の11個のアミノ酸をコードする配列の欠失したcs遺伝子のコドン最適化形態を構築した。この配列は、以下のオリゴヌクレオチド(Sigma−Proligo):(フォワード)5’GGTACCGGATCCGCCACCATGAAGAAATGCACC−3’(下線はBamHI部位);(リバース)5’−AGCTCGAGTCATCACAGGCTGTTGGACACGATGTTGAAGATGC−3’(下線はXhoI部位)を用いて、コドン最適化cs遺伝子のフラグメントのPCR増幅により得られた。得られた増幅産物は、pCR2.1−TOPOプラスミド(Invitrogen)及び配列(pCR2.1−TOPO CSと称されるプラスミド)にクローンされた。pTRIP CSベクタープラスミドは、BamHI/XhoI消化されたpTRIP CMV−GFP−WPREからのGFP配列を、pCR 2.1−TOPO CSの BamHI/xhoI消化後得られる切断されたコドン最適化CS配列と置き換えることにより生成された。pTRIP Hep17について、kozak配列を含み、5’にBamH1部位、及び3’にXhoI部位を有する、Py Hep17遺伝子(GenBank登録番号 U43539)の哺乳動物コドン最適化配列(Geneart)は、BamH1/XhoI消化されたpTRIP CMV−WPREにクローンされた。MSP1構築物について、合成の哺乳動物コドン最適化配列(Geneart)は:Py MSP142(GenBank登録番号 JO4668)のコドン最適化配列に融合されたカルレチクリン(MLLSVPLLLGLLGLAVA)の分泌シグナルについてコードする配列を含むために設計された。BamH1/XhoIで消化される全体配列は、BamH1/XhoIで消化されるpTRIP CMV−WPREにクローンされた。
ベクター粒子は、前述のように15、ベクタープラスミドpTRIP CS、VSV−Gエンベロープ発現プラスミド(pHCMV−G)、及び統合が欠損しているベクター(インテグラーゼの触媒ドメインにおけるD64V置換が、統合ステップのDNA切断及び連結反応を妨げる)の生産のための、pD64Vキャプシド形成GAG POLプラスミドを用いて、293T細胞の一時的リン酸カルシウム共トランスフェクションにより、生産された。濃縮されたベクター粒子のp24抗原含有量の定量は、市販のHIV−1 p24酵素結合免疫吸着測定法(ELISA)キット(Perkin Elmer Life Sciences)を用いて行なわれた。TRIP.I及びTRIP.NI粒子のベクター力価は、アフィジコリン(SIGMA)で処理されたHeLa細胞を形質導入すること、及び前述のように15定量PCRを行なうことにより、測定された。統合性及び非統合性レンチウイルスベクターの力価は、増殖を停止した細胞において測定されたp24含量、及び定量PCRに従って、同様であった。
6週齢のBALB/cマウスは、0.1mlのダルベッコのリン酸緩衝生理食塩水に希釈された、VSV−G Indianaエンベロープで偽型化された100ngのTRIP.NI CSベクター粒子で腹腔内に(i.p.)免疫付与された。8週間後、マウスは、VSV−G New Jerseyエンベロープで偽型化された1500ngのTRIP.NI CSベクター粒子で腹腔内にブーストされた。免疫付与された、及び対照のマウスのチャレンジは、最後の免疫付与から4週間又はそれ以上後の、静脈内に80,000のスポロゾイトの注射からなった。チャレンジの結果は、後に詳述する定量的なリアルタイムRT−PCR法を用いることにより、マウスの肝臓における寄生虫負担を測定することにより、決定された。我々はまた、マウスの対照及び免疫付与された群において、3日目からチャレンジ後14日まで、毎日得られるギムザ染色された薄い血液塗布標本の顕微鏡検査により、500スポロゾイトの静脈内接種後、マウスが寄生虫血症を発症したかどうか測定した。簡潔に言うと、チャレンジされたマウスからの血液の小液滴は、顕微鏡スライド上に置かれた。液滴は、第二のスライドを用いて塗りつけられ、空気乾燥され、30秒間100%メタノール中で固定された。固定されたスライドは、水(Volvic)で希釈された10%ギムザ(Reactfs RAL)の新鮮な溶液中で30分間染色され、水でリンスされ、空気中で乾燥された。スライドは、x100油浸対物レンズで観察された。
チャレンジされたマウスの肝臓における寄生虫負荷の定量は、幾つかの修正ととともに、前述のように16行なわれた。チャレンジの40時間後、肝臓は回収され、RNAはRNeasyミニキット(Qiagen)を用いて抽出された。2μgのRNAは、寄生虫特異的18S rRNAの定量に用いられた。リアルタイムRT−PCRの反応は、EXPRESS One−Step SYBR(登録商標)GreenER(商標)キット(invitrogen)、及びP.ヨエリ(P.yoelii)の18S rRNAの増幅に特異的なプライマーを用いて行なわれた。プライマー(Sigma−Proligo)の配列は:5’−GGGGATTGGTTTTGACGTTTTTGCG−3’(フォワードプライマー)、及び5’−AAGCATTAAATAAAGCGAATACATCCTTAT−3’(リバースプライマー)である。実験は、LightCycler(商標)装置(Roche diagnostics)を用いて行なわれた。寄生虫RNAコピーの量は、寄生虫の18S cDNA PCR増幅フラグメントを含むプラスミド構築物(pCR 2.1−TOPOプラスミド−Invitrogen)の段階希釈により調製される内部標準曲線上への閾蛍光値の外挿法により評価された。
ニトロセルロースマイクロプレート(MAHA S4510,Millipore)は、捕捉抗体(Mouse IFNg Elispot pair,BD Pharmingen)でコートされ、10%FCSが添加されたRPMI 1640 Glutamax、抗体、Hepes、非必須アミノ酸、b−メルカプトエタノール、及びピルビン酸ナトリウムからなる完全培地でブロックされた。ベクター免疫付与マウスからの脾細胞は、0.125x106細胞/ウェルで3重にプレートに添加された。CS特異的CD8+T細胞応答の定量のために、脾細胞は、2μg/mlのペプチド(PolyPeptide Laboratories France)、SYVPSAEQI(Py CS280-288)又はIYNRNIVNRL(Py CS58-67)とインキュベートされた。CS特異的CD4+T細胞応答を評価するために、脾細胞は、2μg/mlのペプチドSYVPSAEQILEFVKQI(Py CS280-295)とインキュベートされた。20時間後、スポットは、ビオチンコンジュゲート化抗体(Mouse IFNg Elispot pair,BD Pharmingen)、続いてストレプトアビジン−AP(Roche)、及びBCIP/NB基質溶液(Promega)を用いて明らかにされた。スポットは、Bioreader 2000(Biosys,Karben,Germany)を用いて計測され、結果は、100万個の脾細胞あたりのIFNγスポット形成細胞(sfc)として表わされた。同一のプロトコールは、Hep17特異的T細胞応答の定量に適用された。Elispot及びインビボ細胞傷害性アッセイにおいて刺激のために用いられるペプチドは、表1に要約される。
標的細胞調製のために、ナイーブマウスからの脾細胞は、様々な濃度(高濃度、5μM;低濃度、1μM)のCFSE(カルボキシフルオレセイン二酢酸スクシンイミジルエステル、Vybrant CFDA−SE 細胞追跡キット,Molecular Probes)で標識された。高濃度のCFSEで標識された脾細胞は、5μg/mlにおいてペプチドの組み合わせでパルスされた。低用量のCFSEで染色された対照集団は、ペプチドを含まない培地中でインキュベートされた。各マウスは、眼窩後静脈を通して、各画分から等量の細胞を含む混合物の107CFSE−標識細胞を接種された。15〜18時間後、脾臓からの単一細胞の懸濁液は、フローサイトメトリー(Becton Dickinson,CellQuest software)により分析された。ペプチド−パルスされた細胞の消失は、免疫付与されたマウスに対するナイーブマウスにおける、パルスされた集団(High CFSE蛍光強度)と未パルスの集団(低濃度 CFSE蛍光強度)の比を比較することにより決定された。特異的な死亡のパーセンテージは、以下の計算:

に従って確立された。
P.ヨエリ(P.yoelii)YM MSP119(aa1649−1757)は、pTRIP MSP142からのフォワードプライマー5’−CGTGGATCCATGGACGGCATGGATCTGCTG−3’及びリバースプライマー5’−GATGAATTCGGAGCTGCTGCTGCAGAACACG−3’を用いてPCRにより増幅され、グルタチオンS−トランスフェラーゼ(GST)−融合タンパク質発現ベクターpGEX−2T(Amersham Biosciences,Bucks,UK)にクローンされた。大腸菌(Escherichia coli)BL21 star(Invitrogen)は、pGEX−2T MSP119で形質転換され、増殖及び誘導は、製造者の指示(pGEXベクター、GST遺伝子融合システム、Amersham)に従って行なわれた。BL21においてタンパク質の発現の誘導後、細胞は回収され、BugBuster試薬(Novagen)を用いて溶解された。組み換えタンパク質は、製造者の指示に従って、GST結合タンパク質精製キット(Novagen)を用いて、GST結合レジンクロマトグラフィーにより精製された。
血清は、酵素結合免疫吸着測定法(ELISA)によるMSP119特異的抗体の評価のために、最後の免疫付与の3週間後に回収された。組み換えGST−MSP119融合タンパク質又はGST対照は、PBS中2μg/mlにおいて、4℃で96ウェルNunc−Immuno Maxisorpプレート(FischerScientific,Wohlen,Germany)に一晩吸着させた。PBS中0.05%Tween 20で3回の洗浄後、ウェルは、室温で1時間、10%のウシ胎児血清(FBS)を含む100μlのPBSの添加により、ブロックされた。プレートは、PBS中0.05%Tween 20で3回洗浄され、100μlの血清の10倍段階希釈液が、ウェルに添加された。室温で2時間のインキュベーション後、ウェルは洗浄され、PBS10%FBSに1/4000に希釈された100μlのペルオキシダーゼヤギ抗−マウス免疫グロブリン(H+L)(Jackson Immuno Research)は、各ウェルに添加された。室温で1時間のインキュベーション後、ウェルは洗浄され、100μlのテトラメチルベンジジン基質試薬(BD Pharmingen)は、各ウェルに添加された。プレートは、室温で30分間インキュベートされ、100μlの1N H2SO4は、反応を停止するために添加された。プレートは、450nmにおいて、光学密度について測定された。終点(endpoint)力価は、2倍の非免疫付与マウスからの血清の光学密度を誘発する、最後の希釈の逆数として計算された。

Claims (13)
- (i)RNAウイルス由来の第一の決定された異種のウイルスエンベロープタンパク質(単数)又はウイルスエンベロープタンパク質(複数)で偽型化されるレンチウイルスベクター粒子であって、そのゲノム中に、哺乳動物宿主を感染し得るプラスモディウム属(Plasmodium)寄生虫の前赤血球ステージのサーカムスポロゾイトタンパク質(CSP)のエピトープ(単数又は複数)を有する少なくとも1つのポリペプチド(単数又は複数)をコードする少なくとも1つの組み換えポリヌクレオチドを含み、ここで前記エピトープ(単数又は複数)は、T−エピトープ(単数又は複数)及びB−エピトープ(単数又は複数)を含む、非統合的で、複製能力の無い、前記レンチウイルスベクター粒子;
(ii)(i)におけるレンチウイルスベクター粒子と別に提供される、前記第一の異種のウイルスエンベロープ(単数又は複数)と異なる、RNAウイルス由来の第二の決定された異種のウイルスエンベロープタンパク質(単数)又はウイルスエンベロープタンパク質(複数)で偽型化されるレンチウイルスベクター粒子であって、そのゲノム中に、哺乳動物宿主を感染し得るプラスモディウム属(Plasmodium)寄生虫の前赤血球ステージのサーカムスポロゾイトタンパク質(CSP)のエピトープ(単数又は複数)を有する少なくとも1つのポリペプチド(単数又は複数)をコードする少なくとも1つの組み換えポリヌクレオチドを含み、ここで前記エピトープ(単数又は複数)は、T−エピトープ(単数又は複数)及びB−エピトープ(単数又は複数)を含む、非統合的で、複製能力の無い、前記レンチウイルスベクター粒子;
ここで、前記第一及び第二のウイルスエンベロープタンパク質(単数又は複数)は、互いに血清中和せず、哺乳動物細胞のインビボ形質導入に好適である、
を少なくとも含む、哺乳動物宿主に別々に投与するための化合物の組み合わせであって、
前記(i)及び(ii)におけるレンチウイルスベクター粒子は、
・パッケージング、逆転写、及び転写に必要なHIV−1のシス作用配列を含み、さらに、機能性のHIV−1のDNA flapエレメントを含み、さらに、制御発現配列の制御下にある、GPI−アンカーリングモチーフを欠失したプラスモディウム属寄生虫の切断されたcs遺伝子のヒトコドン最適化配列のポリヌクレオチドを含む、ベクタープラスミドであって、ここで前記シス作用配列は、切断され、U3領域を欠損している、又はU3領域において部分的に欠失している、HIV−1ゲノムからの3’LTR配列を少なくとも含む、前記ベクタープラスミド;
・Indiana株のVSV−Gタンパク質(単数又は複数)、New Jersey株のVSV−Gタンパク質(単数又は複数)、Cocal株のVSV−Gタンパク質(単数又は複数)、Isfahan株のVSV−Gタンパク質(単数又は複数)、Chandipura株のVSV−Gタンパク質(単数又は複数)、Pyri株のVSV−Gタンパク質(単数又は複数)およびSVCV株のVSV−Gタンパク質(単数又は複数)から選択される、VSV−Gエンベロープタンパク質(単数又は複数)をコードするポリヌクレオチドを含むVSV−Gエンベロープ発現プラスミドであって、前記ポリヌクレオチドは、制御発現配列の制御下にある、VSV−Gエンベロープ発現プラスミド;及び
・統合能力が欠損したベクター粒子の産生に好適な改変されたHIV−1のgag−polコーディング配列を含むキャプシド形成プラスミドであって、ここで前記改変されたgag−pol配列が制御発現配列の制御下にある、キャプシド形成プラスミド;
を用いた、哺乳動物細胞の共トランスフェクションから回収された生成物であり、
・前記第一及び第二のウイルスエンベロープタンパク質がそれぞれIndiana株のVSV−G及びNew Jersey株のVSV−G、若しくはその逆である、又は
・前記第一及び第二のエンベロープタンパク質の1つ又は双方が、天然のIndiana株のVSV−G若しくは/及びNew Jersey株のVSV−Gの改変型である、又は
・前記第一及び第二のエンベロープタンパク質の少なくとも一つが、キメラのVSV−Gタンパク質であって、ここで以下のドメイン:輸送決定因子YTDIE、細胞質尾部、膜貫通ドメイン、又は細胞質ドメインの少なくとも一つが、Indiana株由来である、又は
・第一のウイルスエンベロープタンパク質が、Indiana株のVSV−G若しくはNew Jersey株のVSV−Gのいずれかであり、第二のウイルスエンベロープタンパク質が、Cocal株のVSV−Gタンパク質(単数又は複数)、Isfahan株のVSV−Gタンパク質(単数又は複数)、Chandipura株のVSV−Gタンパク質(単数又は複数)、Pyri株のVSV−Gタンパク質(単数又は複数)、及びSVCV株のVSV−Gタンパク質(単数又は複数)の群から選択される、前記組み合わせ。 - 前記第一及び第二のウイルスエンベロープタンパク質がそれぞれIndiana株のVSV−G及びNew Jersey株のVSV−G、若しくはその逆である、請求項1に記載の化合物の組み合わせ。
- 前記レンチウイルス粒子が、(i)CSP抗原のポリペプチド、又はGPIアンカーリングモチーフを欠損したCSP抗原のポリペプチド、及び(ii)スポロゾイト表面タンパク質2(TRAP/SSP2)、肝臓ステージ抗原(LSA)、Pfエクスポーティッドタンパク質1(Pf Exp1)、Pf抗原2の群において選択されるマラリア寄生虫の抗原の少なくとも一つの更なるポリペプチドを含む、異なるポリペプチドをコードし、ここで、前記抗原の異なるポリペプチドが、同一のレンチウイルス粒子又は異なるレンチウイルス粒子から発現される、請求項1又は2に記載の、別々に提供される化合物の組み合わせ。
- 前記レンチウイルス粒子が、メロゾイト表面タンパク質1(MSP−1)、メロゾイト表面タンパク質2(MSP−2)、頂端膜抗原1(AMA−1)、セリンリピート抗原(SERA)、GLURP抗原、Pf155/RESA(リング−感染赤血球表面抗原)、又は桿小体関連タンパク質1(RAP−1)若しくは桿小体関連タンパク質2(RAP−2)の群において選択されるポリペプチドをコードする、請求項1〜3のいずれか一項に記載の、別々に提供される、化合物の組み合わせ。
- 哺乳動物宿主において、マラリア寄生虫感染又は寄生虫に誘導される病理に対する予防的免疫付与のための、別々に提供される請求項1〜4のいずれか一項に記載の化合物の組み合わせ。
- 哺乳動物宿主において、マラリア寄生虫感染又は寄生虫に誘導される病理に対する予防的免疫付与に使用のための好適な投与ビヒクルで製剤化され、ここで前記使用は、宿主の細胞性免疫応答をプライムするために有効量のレンチウイルス粒子を投与すること、及び宿主の細胞性免疫応答をブーストするために有効量のレンチウイルス粒子を後で投与することを含む免疫付与パターンを含み、ここでプライム又はブートするステップのそれぞれにおいて投与されるレンチウイルス粒子は、互いに血清中和をしない異なるエンベロープタンパク質(単数又は複数)で偽型化され、ここで前記プライム又はブーストするステップは、少なくとも6週間空けられる、別々に提供される請求項1〜4のいずれか一項に記載の化合物の組み合わせ。
- 前記免疫付与パターンが、ブーストについての前記投与ステップを繰り返すことをさらに含む、別々に提供される請求項6に記載の化合物の組み合わせ。
- 別々に提供される用量の前記レンチウイルス粒子を含む投与計画において、哺乳動物宿主においてマラリア寄生虫感染又は寄生虫に誘導される病理に対する予防的免疫に用いられ、ここで、統合能力のないベクター粒子が用いられる場合、細胞性免疫応答をプライム及びブーストすることを目的とする用量は、108〜1010のレンチウイルス粒子を含む、別々に提供される請求項1〜4のいずれか一項に記載の化合物の組み合わせ。
- 宿主において、少なくとも1つの以下の効果:
(XII)ヒト宿主において、マラリア寄生虫感染に対する無菌保護を誘発すること;
(XIII)マラリア寄生虫の細胞外形態を阻害すること;
(XIV)マラリア寄生虫による肝細胞感染を予防すること、又は感染の肝臓ステージ増幅を阻害すること;
(XV)マラリア寄生虫抗原(単数又は複数)に対する特異的T細胞免疫応答を誘発すること;
(XVI)寄生虫抗原(単数又は複数)に対するB細胞応答を誘発すること;
(XVII)マラリア寄生虫による感染の影響を減少又は緩和するために寄生虫血症を制御すること;
(XVIII)寄生虫による感染又は寄生虫により誘導される病理に対する保護細胞性免疫を誘発すること;
(XIX)メモリーT細胞免疫応答を誘発すること;
(XX)マラリア寄生虫による感染の間、CD4+及びCD8+T細胞応答の早期の及び高い回復を誘発すること;
(XXI)プラスモディウム属感染に関して肝臓内のメモリーリンパ球を刺激することによる早期の強力なCT(CD8+T)応答を誘発すること;
(XXII)マラリア寄生虫が免疫応答を回避することを防ぐこと、従って、マラリア寄生虫による感染の長期の制御を可能にすること;
を得るために好適である、投与用量及び投与計画において、哺乳動物宿主における、マラリア寄生虫感染又は寄生虫に誘導される病理に対する予防的免疫付与に用いられる、別々に提供される請求項1〜4のいずれか一項に記載の化合物の組み合わせ。 - 前記マラリア寄生虫感染が、プラスモディウム・ファルシパルム(Plasmodium falciparum)、プラスモディウム・マラリアエ(Plasmodium malariae)、プラスモディウム・ビバックス(Plasmodium vivax)、プラスモディウム・ノウレシ(Plasmodium knowlesi)又はプラスモディウム・オバール(Plasmodium ovale)による感染である、別々に提供される請求項9に記載の化合物の組み合わせ。
- 前記特異的T細胞免疫応答が、CD8+T細胞応答及び/又は特異的CD4+T細胞応答である、別々に提供される請求項9に記載の化合物の組み合わせ。
- アジュバント化合物、及び/又は免疫付与化合物、及び適切な送達ビヒクルと関連して、哺乳動物宿主において、マラリア寄生虫感染又は寄生虫に誘導される病理に対する予防的免疫付与に用いられる、別々に提供される請求項1〜4のいずれか一項に記載の化合物の組み合わせ。
- ヒト宿主において、マラリア寄生虫感染又は寄生虫に誘導される病理に対する予防的免疫付与のために用いられる、別々に提供される請求項1〜4のいずれか一項に記載の化合物の組み合わせ。
