Description
本発明はまた、内在性OSR1の発現に連動して発現するレポーター遺伝子を染色体内に有するヒト多能性幹細胞ならびに本細胞を用いた分化誘導剤のスクリーニング方法にも関する。
る(特許文献1または特許文献2)。そこで、腎不全の治療方法として、これらの多能性幹細胞から分化誘導して得られた腎細胞を移植する治療法が検討されている。さらに、これらの多能性幹細胞由来の均一な腎細胞を用いて治療薬の開発を行うことも視野に入れられている。
[1]以下の工程(i)および(ii):
(i)Activin AおよびWntを含む培地中でヒト多能性幹細胞を培養する工程、および
(ii)BMPおよびWntもしくはWntの機能等価物を含む培地中で、工程(i)で得られた細胞を培養する工程
を含む、ヒト多能性幹細胞から中間中胚葉細胞の製造方法。
[2]前記中間中胚葉細胞がOSR1陽性細胞である、[1]に記載の方法。
[3]前記工程(i)での培養においてヒト多能性幹細胞を浮遊培養して細胞集合体または
細胞塊を形成させ、前記工程(ii)での培養において該細胞集合体または細胞塊を接着培養する、[1]または[2]に記載の方法。
[4]前記工程(i)での培養において、ヒト多能性幹細胞の細胞集合体または細胞塊を実質

本発明は、内在性OSR1の発現に連動して発現するレポーター遺伝子を染色体内に有するヒト多能性幹細胞、および、増殖因子を加えた培地を用いてヒト多能性幹細胞から中間中胚葉細胞を分化誘導する方法に関する。
本発明で使用可能な多能性幹細胞は、生体に存在するすべての細胞に分化可能である多能性を有し、かつ、増殖能をも併せもつ幹細胞である。それらの多能性幹細胞の具体例は、以下のものに限定されないが、胚性幹(ES)細胞、核移植により得られるクローン胚由来の胚性幹(ntES)細胞、精子幹細胞(「GS細胞」)、胚性生殖細胞(「EG細胞」)、人工多能性幹(iPS)細胞などが含まれる。好ましい多能性幹細胞は、ES細胞、ntES細胞、およびiPS細胞である。
ES細胞は、ヒトやマウスなどの哺乳動物の初期胚(例えば胚盤胞)の内部細胞塊から樹立された、多能性と自己複製による増殖能とを有する幹細胞である。

用し、37℃、5% CO2湿潤雰囲気下でヒトES細胞を維持することができる。また、ES細胞は、3〜4日おきに継代する必要があり、このとき、継代は、例えば1mM CaCl2及び20% KSRを含有するPBS(リン酸緩衝生理食塩水)中の0.25% トリプシン及び0.1mg/mlコラゲナーゼIVを用いて行うことができる。
精子幹細胞は、精巣由来の多能性幹細胞であり、精子形成のための起源となる細胞である。この細胞は、ES細胞と同様に、種々の系列の細胞に分化誘導可能であり、例えばマウス胚盤胞に移植するとキメラマウスを作出できるなどの性質をもつ(M. Kanatsu-Shinohara et al. (2003) Biol. Reprod., 69:612-616; K. Shinohara et al. (2004), Cell, 119:1001-1012)。精子幹細胞は、神経膠細胞系由来神経栄養因子(glial cell line-derived neurotrophic factor (GDNF))を含む培地で自己複製可能であるし、ES細胞と同様の培養
条件下で継代を繰り返すことによって精子幹細胞を得ることができる(竹林正則ら(2008),実験医学,26巻,5号(増刊),41〜46頁,羊土社(東京、日本))。
胚性生殖細胞は、胎生期の始原生殖細胞から樹立される、ES細胞と同様な多能性をもつ細胞であり、LIF、bFGF、幹細胞因子(stem cell factor)などの物質の存在下で始原生殖
細胞を培養することによって樹立しうる(Y. Matsui et al. (1992), Cell, 70:841-847;
J.L. Resnick et al. (1992), Nature, 359:550-551)。
人工多能性幹 (iPS) 細胞は、ある特定の1種もしくは複数種の核初期化物質を、DNAま
たはタンパク質の形態で体細胞に導入することによるか、または、ある特定の1種もしく
は複数種の薬剤の使用により該核初期化物質の内在性のmRNAおよびタンパク質の発現量を上昇させることによって作製することができる、ES細胞とほぼ同等の特性、例えば分化多能性と自己複製による増殖能を有する体細胞由来の人工の幹細胞である(K. Takahashi and S. Yamanaka (2006) Cell, 126: 663-676、K. Takahashi et al. (2007) Cell, 131: 861-872、J. Yu et al. (2007) Science, 318: 1917-1920、M. Nakagawa et al. (2008) Nat. Biotechnol., 26: 101-106、国際公開WO 2007/069666および国際公開WO 2010/068955)。核初期化物質は、ES細胞に特異的に発現している遺伝子もしくはES細胞の未分化維
持に重要な役割を果たす遺伝子、またはそれらの遺伝子産物であればよく、特に限定されないが、例えば、Oct3/4, Klf4, Klf1, Klf2, Klf5, Sox2, Sox1, Sox3, Sox15, Sox17, Sox18, c-Myc, L-Myc, N-Myc, TERT, SV40 Large T antigen, HPV16 E6, HPV16 E7, Bmil, Lin28, Lin28b, Nanog, Esrrb, EsrrgおよびGlis1が例示される。これらの初期化物質
は、iPS細胞樹立の際には、組み合わされて使用されてもよい。そのような組合せは、上
記初期化物質を、少なくとも1種、2種もしくは3種含む組み合わせ、好ましくは3種もしくは4種を含む組み合わせとすることができる。

ベクター、リポフェクション、リポソームの使用、マイクロインジェクションなどの手法によって体細胞内に導入することができる。ウイルスベクターとしては、レトロウイルスベクター、レンチウイルスベクター(これらのベクターは、Cell, 126, pp.663-676, 2006; Cell, 131, pp.861-872, 2007; Science, 318, pp.1917-1920, 2007に準ずる。)、アデノウイルスベクター(Science, 322, 945-949, 2008)、アデノ随伴ウイルスベクター、
センダイウイルスベクター(Proc Jpn Acad Ser B Phys Biol Sci. 85, 348-62, 2009)
などが例示される。また、人工染色体ベクターとしては、例えばヒト人工染色体(HAC)、
酵母人工染色体(YAC)、細菌人工染色体(BACおよびPAC)などが含まれる。プラスミドとし
ては、哺乳動物細胞用プラスミドを使用しうる(Science, 322:949-953, 2008)。ベクター
には、核初期化物質が発現可能なように、プロモーター、エンハンサー、リボゾーム結合配列、ターミネーター、ポリアデニル化サイトもしくはポリアデニル化シグナルなどの制御配列を含むことができる。使用されるプロモーターとしては、例えばEF1αプロモータ
ー、CAGプロモーター、SRαプロモーター、SV40プロモーター、LTRプロモーター、CMV(
サイトメガロウイルス)プロモーター、RSV(ラウス肉腫ウイルス)プロモーター、MoMuLV(モロニーマウス白血病ウイルス)LTR、HSV-TK(単純ヘルペスウイルスチミジンキナーゼ)プロモーターなどが用いられる。なかでも、EF1αプロモーター、CAGプロモーター、MoMuLV LTR、CMVプロモーター、SRαプロモーターなどが好ましい例として挙げられる。
さらに、必要に応じて、薬剤耐性遺伝子(例えばカナマイシン耐性遺伝子、アンピシリン
耐性遺伝子またはピューロマイシン耐性遺伝子)、チミジンキナーゼ遺伝子、およびジフ
テリアトキシン遺伝子もしくはその断片などの選択マーカー配列、並びに、緑色蛍光タンパク質(GFP)、βグルクロニダーゼ(GUS)またはFLAGなどのレポーター遺伝子配列などを含むことができる。また、上記ベクターには、体細胞への導入後、核初期化物質をコードする遺伝子もしくはプロモーターとそれに結合する核初期化物質をコードする遺伝子を共に切除するために、それらの前後にLoxP配列を有してもよい。別の好ましい実施態様においては、トランスポゾンを用いて染色体に導入遺伝子を組み込んだ後に、プラスミドベクターもしくはアデノウイルスベクターを用いて細胞に転移酵素を作用させ、導入遺伝子を完全に染色体から除去する方法が用いられ得る。好ましいトランスポゾンとしては、例えば、鱗翅目昆虫由来のトランスポゾンであるpiggyBac等が挙げられる(Kaji, K. et al., (2009), Nature, 458: 771-775、Woltjen et al., (2009), Nature, 458: 766-770 、WO 2010/012077)。さらに、ベクターには、染色体への組み込みがなくとも複製されて、エ
ピソーマルに存在するように、リンパ指向性ヘルペスウイルス(lymphotrophic herpes virus)、BKウイルスおよび牛乳頭腫ウイルス(Bovine papillomavirus)の起点とその複
製に関わる配列を含んでいてもよい。例えば、EBNA-1およびoriP、または、Large Tおよ
びSV40ori配列を含むことが挙げられる(WO 2009/115295、WO 2009/157201およびWO 2009/149233)。また、2種またはそれ以上の核初期化物質を同時に導入するために、ポリシストロニックに発現させることができる発現ベクターを用いてもよい。ポリシストロニックに発現させるためには、遺伝子をコードする配列と配列の間は、IRESまたは口蹄病ウイルス(FMDV)2Aコード領域により結合されていてもよい(Science, 322:949-953, 2008およびWO 2009/0920422009/152529)。

ることができる。
トロ-3'-オキシム、バルプロ酸、2-(3-(6-メチルピリジン-2-イル)-lH-ピラゾール-4-イ
ル)-1,5-ナフチリジン、1-(4-メチルフェニル)-2-(4,5,6,7-テトラヒドロ-2-イミノ-3(2H)-ベンゾチアゾリル)エタノンHBr(pifithrin-alpha)、プロスタグランジンJ2およびプロ
スタグランジンE2等が例示される(WO 2010/068955)。
ピューロマイシン、L-グルタミン、非必須アミノ酸類、β-メルカプトエタノールなどを
適宜含むことができる。)、(2) bFGFまたはSCFを含有するES細胞培養用培地、例えばマ
ウスES細胞培養用培地(例えばTX-WES培地、トロンボX社)または霊長類ES細胞培養用培地(例えば霊長類(ヒトまたはサル)ES細胞用培地(販売先:リプロセル、京都、日本)、mTeSR-1)、などが含まれる。
培養し、その後、細胞をフィーダー細胞 (例えば、マイトマイシンC処理STO細胞、SNL細
胞等) 上にまきなおし、体細胞と核初期化物質の接触から約10日後からbFGF含有霊長類ES細胞培養用培地で培養し、該接触から約30〜約45日またはそれ以上ののちにES細胞様コロニーを生じさせることができる。また、iPS細胞の誘導効率を高めるために、5〜10%と低い酸素濃度の条件下で培養してもよい。
培地(すなわち、選択培地)で細胞の培養を行うことによりマーカー遺伝子発現細胞を選択することができる。またマーカー遺伝子が蛍光タンパク質遺伝子の場合は蛍光顕微鏡で観察することによって、発光酵素遺伝子の場合は発光基質を加えることによって、または、発色酵素遺伝子の場合は発色基質を加えることによって、マーカー遺伝子発現細胞を検出することができる。
細胞(例、血管内皮細胞)、運搬能をもつ繊毛のある細胞(例、気道上皮細胞)、細胞外マトリックス分泌用細胞(例、線維芽細胞)、収縮性細胞(例、平滑筋細胞)、血液と免疫系の細胞(例、Tリンパ球)、感覚に関する細胞(例、桿細胞)、自律神経系ニューロ
ン(例、コリン作動性ニューロン)、感覚器と末梢ニューロンの支持細胞(例、随伴細胞
)、中枢神経系の神経細胞とグリア細胞(例、星状グリア細胞)、色素細胞(例、網膜色素上皮細胞)、およびそれらの前駆細胞 (組織前駆細胞) 等が挙げられる。細胞の分化の程度や細胞を採取する動物の齢などに特に制限はなく、未分化な前駆細胞 (体性幹細胞も含む) であっても、最終分化した成熟細胞であっても、同様に本発明における体細胞の起源として使用することができる。ここで未分化な前駆細胞としては、たとえば神経幹細胞、造血幹細胞、間葉系幹細胞、歯髄幹細胞等の組織幹細胞(体性幹細胞)が挙げられる。
nt ES細胞は、核移植技術によって作製されたクローン胚由来のES細胞であり、受精卵
由来のES細胞とほぼ同じ特性を有している(T. Wakayama et al. (2001), Science, 292:740-743; S. Wakayama et al. (2005), Biol. Reprod., 72:932-936; J. Byrne et al. (2007), Nature, 450:497-502)。具体的には、nt ES(nuclear transfer ES)細胞は、未受精卵の核を体細胞の核と置換することによって得られたクローン胚由来の胚盤胞の内部細胞塊から樹立される。nt ES細胞の作製のためには、核移植技術(J.B. Cibelli et al. (1998), Nat. Biotechnol., 16:642-646)とES細胞作製技術(上記)との組み合わせが利用
される(若山清香ら(2008),実験医学,26巻,5号(増刊), 47〜52頁)。核移植においては、哺乳動物の除核した未受精卵に、体細胞の核を注入し、数時間培養することで再プログラム化することができる。
融合幹細胞は、体細胞と卵子もしくはES細胞とを融合させることにより作製され、融合させたES細胞と同様な多能性を有し、さらに体細胞に特有の遺伝子も有する幹細胞である(Tada M et al. Curr Biol. 11:1553-8, 2001; Cowan CA et al. Science. 2005 Aug 26;309(5739):1369-73)。
本発明によれば、ES細胞、iPS細胞などの多能性幹細胞から中間中胚葉細胞への分化誘
導に際して、以下の工程(A)および(B)を含む方法を用いることができる。
本工程は、2種類の培養方法のいずれかを使用して実施されうる。すなわち、第1の方法は、培養の間に、ヒト多能性幹細胞の細胞集合体(すなわち、細胞凝集体)または細胞塊を形成することを含む方法であり、あるいは、第2の方法は、培養の間に、互いに実質的
に分離されるヒト多能性幹細胞を培養することを含む方法である。
幹細胞を、一旦、複数の小細胞塊に分離し、その後に再び凝集させることも可能である。このとき、小細胞塊に分離するために、分離溶液を用いるか、あるいは機械的に、微細になるまで細胞塊を分離する。好適な方法は、最初に分離溶液を使用し、その後に細胞塊を複数の小塊に機械的に分離することを含む。ここで使用される分離溶液の具体例には、プロテアーゼ活性およびコラゲナーゼ活性を有する分離溶液(例えば、AccutaseTMやAccumaxTM(その各々は、例えばフナコシ(日本)から入手しうる)などの、トリプシンとコラ
ゲナーゼを含有する溶液)、ならびに、コラゲナーゼ活性のみを有する分離溶液が挙げられる。この方法では、上記の分離溶液として、コラゲナーゼ活性のみを有する分離溶液を使用することが好ましい。
胞塊から、ヒト多能性幹細胞を実質的に分離(もしくは解離)し、その後にActivin AとWntもしくはWntの機能等価物とを含有する培地を用いて接着培養により該細胞を培養する
。このとき、ヒト多能性幹細胞を、多能性幹細胞培養用培地を用いて80%コンフルエントとなるまで培養してコロニーを形成したあとか、あるいは、ヒト多能性幹細胞を分離(もしくは解離)した直後に、Activin AとWntもしくはWntの機能等価物とを含有する培地を
用いて接着培養により該細胞を培養してもよい。ここで使用される分離法の具体例には、機械的方法、ならびに、プロテアーゼ活性およびコラゲナーゼ活性を有する分離溶液(例えば、AccutaseTMやAccumaxTMなどの、トリプシンとコラゲナーゼを含有する溶液)を使
用するか、または、コラゲナーゼ活性のみを有する分離溶液を使用する方法が包含される。そのような方法の好適例として、プロテアーゼ活性およびコラゲナーゼ活性を有する分離溶液を用いて細胞を解離することを含む方法、ならびに、プロテアーゼ活性およびコラゲナーゼ活性を有する分離溶液を用いて細胞を解離し、その後に機械的に微細になるまで細胞を分離することを含む方法が挙げられる。
浮遊培養では、細胞は、培養ディッシュに接着させずに培養される。浮遊培養は、細胞との接着性を向上させる目的で人工的に処理(例えば、細胞外マトリックス等によるコーティング処理)されていない培養ディッシュ、あるいは、人工的に接着を抑制する処理(例えば、ポリヒドロキシエチルメタクリル酸(poly-HEMA)によるコーティング処理)し
た培養ディッシュを使用して行うことができるが、これらの具体例に特に限定されない。第1の方法では、ヒト多能性幹細胞を浮遊培養することによって細胞集合体(もしくは細
胞塊)を形成する。
されていてもよいし、あるいは無血清でもよい。必要に応じて、例えば、アルブミン、トランスフェリン、ノックアウト血清代替物(Knockout Serum Replacement(KSR))(ES細胞培養時のFBSの血清代替物)、N2サプリメント(Invitrogen)、B27サプリメント(Invitrogen)、脂肪酸、インスリン、コラーゲン前駆体、微量元素、2-メルカプトエタノール、3’
-チオールグリセロールなどの1つ以上の血清代替物を含んでもよいし、脂質、アミノ酸、L-グルタミン、GlutaMAXTM(Invitrogen)、非必須アミノ酸、ビタミン、増殖因子、抗生物質、抗酸化剤、ピルビン酸、緩衝剤、無機塩類、それらと同類の物質の1つ以上の物質
も含有しうる。好ましい増殖因子の例としては、Wnt1、Wnt3、Wnt3a、Wnt4、Wnt7a、TGF-β、Activin A、Nodal、BMP2、BMP4、BMP6、BMP7およびGDFが挙げられる。少なくとも、
本工程ではWnt3aおよびActivin Aを増殖因子として用いることが望ましい。
物」という用語は、FzレセプターとLRP5/6膜タンパク質のリガンドとの複合体の形成、あるいは、βカテニンと、Axin、GSK3β、APC(adenomatous polyposis coli)などの分子と
の複合体の形成を抑制する物質を指す。そのような物質の例は、WntアゴニストまたはGSK3β阻害剤である。
GSK-3βインヒビターIX; 6-ブロモインジルビン-3'-オキシム)などのインジルビン(indirubin)誘導体、SB216763 (すなわち、3-(2,4-ジクロロフェニル)-4-(1-メチル-1H-インドール-3-イル)-1H-ピロール-2,5-ジオン)などのマレイイミド誘導体、GSK-3βインヒビタ
ーVII (すなわち、4-ジブロモ-アセトフェノン)などのα-ブロモメチルケトン化合物、CHIR99021(すなわち、6-[(2-{[4-(2,4-ジクロロフェニル)-5-(4-メチルイミダゾール-2-イル)ピリミジン-2-イル]アミノ}エチル)アミノ]ピリジン-3-カルボニトリル)(WO1999/65897; CAS番号 252917-06-9)、L803-mts (別称: GSK-3βペプチドインヒビター; Myr-N-GKEAPPAPPQSpP-NH2)などの細胞膜透過性リン酸化ペプチド、およびそれらの誘導体が挙げら
れる。これらの化合物は、Calbiochem, Biomol, Stemgenなどによって上市されており、
容易に使用可能なものである。しかし、上記の化合物の例には特に限定されない。さらにまた、Wntアゴニストの例は、2-アミノ-4-(3,4-(メチレンジオキシ)ベンジルアミノ)-6-(3-メトキシフェニル)ピリミジンである。
もしくはGSK3βインヒビター(例えばCHIR99021)、およびActivin Aを含有するDMEM/Ham's F12混合培地が例示される。

No. 14-8993-62など))、 Nodal (例:NM_018055)、 BMP2 (例:NM_001200)、BMP4 (例: NM_001202, NM_130850, NM_130851)、BMP6 (例:NM_001718)、BMP7 (例:NM_001719 (SEQ ID NOs: 1および2)、ならびに GDF (例:NM_001492, NM_016204, NM_020634, NM_000557, NM_001001557, NM_182828, NM_005259, NM_005260, NM_004962, NM_005811, NM_004864)などが挙げられる。本工程では、少なくともWnt3aおよびBMP7を増殖因子として用いることが好ましい。

中間中胚葉細胞は、OSR1、PAX2、WT1、EYA1およびSIX2などの中間中胚葉細胞のマーカ
ーを用いて同定されうる。
OSR1の発現に連動して発現するレポーター遺伝子を染色体内に有するヒト多能性幹細胞は、ターゲッティングベクターを用いて相同組換え方法により製造することができる。その典型例は、次の通りである。
基に、相同組換え用ターゲティングベクターを構築することができる。該ターゲティングベクターは、染色体上のOSR1遺伝子そのものの塩基配列、もしくは染色体上のOSR1遺伝子の一部を欠いた塩基配列を含む核酸を有するように、あるいは、OSR1遺伝子とは異なる異種遺伝子の5’側及び3’側が染色体上のOSR1遺伝子の上流及び下流の塩基配列によって隣接された塩基配列を含む核酸を有するように、ベクター上に塩基配列を設計することによって構築されうる。
蛍光タンパク質をコードするDNA等のような、一般に用いられるものであればいずれを用
いてもよい。
しいBACクローンは、RP11-458J18、RP11-203M1およびRP11-802J2などが挙げられる。BAC
クローンを用いる場合、例えば、Red/ET相同組換え法を用いて、所望の場所へマーカー遺伝子等を導入することができる。

本発明はさらに、工程(A)および工程(B)を含む上記の方法によってヒト多能性幹細胞から中間中胚葉を誘導し、その後、工程(B)を継続して、後腎細胞を生成することを含んで
なる、後腎細胞の作製方法を提供する。
上記の条件等と同じである。間葉系細胞である後腎細胞を得ることができるまで培養を継続することができる。
OSR1の発現に連動して発現するレポーター遺伝子を染色体内に有するヒト多能性幹細胞は、中間中胚葉細胞への分化誘導剤(例えば医薬化合物、溶媒、低分子量物質、ペプチド、またはポリヌクレオチド)のスクリーニングに用いることもできる。例えば、候補分化誘導剤を単独でまたは他の薬剤と組み合わせて、上述のOSR1の発現に連動して発現するレポーター遺伝子を染色体内に有するヒト多能性幹細胞に接触させ、その際のレポーター遺伝子の発現量の変化により、評価を行うことができる。本発明では、レポーター遺伝子の発現を上昇させる薬剤を中間中胚葉の分化誘導剤として同定することができる。好ましくは、ディッシュへ接着させた当該レポーター遺伝子を染色体内に有するヒト多能性幹細胞の培地に候補薬剤を添加して、当該レポーター遺伝子の発現を上昇可能にする物質を選択する。
ヒトiPS細胞(201B7)は、京都大学(京都、日本)の山中博士より受領し、従来の方法で培養した(Takahashi K, et al. Cell. 131:861-72)。続いて、BACクローン(RP11-458J18)(BACPAC RESOURCES)のOSR1の開始コドンの下流へpRed/ET(Gene Bridges GmbH)を用いてGFP-PGK-Neoカセットを挿入し、OSR1-GFP BAC トランスジーンを作製した(図1a)
。作製した改変BACクローンを導入した約130個のiPS細胞株の各染色体をテンプレートと
して用いて、開始コドンを挟むように設計されたプライマー(正方向プライマー:5'-GGATTGAGAAGCCACTGCAACT-3'(配列番号5)および逆方向プライマー:5'-CCGTTCACTGCCTGAAGGA-3'(配列番号6)により定量PCRを行った。4つのiPS細胞株において対照としての201B7
と比較して増幅産物量が1/2であることが確認された。この4つの株のうち、図1bに3D36および3D45の結果を示す。以上より、内在性のOSR1の発現と連動してGFPを発現するOSR1-GFPレポーターiPS細胞株の樹立に成功した。
上記のように作製したOSR1-GFPレポーターiPS細胞(3D36)を、MEFをフィーダー細胞として用いてコンフルエントになるまで培養した。DMEMで希釈したコラゲナーゼ溶液(1mg/ml)を加えて解離させた。コラゲナーゼ溶液を除去後、Gluta MAXTM(Invitrogen)、ペニ
シリン、ストレプトマイシンおよび2%のFBS(Hyclone社製)を含有したDMEM/F12を加えて
ピペッティングした。続いて、培地を除去したのち、100 ng/ml Wnt3a、100 ng/ml ActivinA、Gluta MAXTM(Invitrogen)、ペニシリン、ストレプトマイシンおよび2%のFBS(Hyclone社製)を含有したDMEM/F12中にて浮遊培養により胚様体(EB)を形成させた。浮遊培養2
日後、EBを回収し、100 ng/ml Wnt3a、100 ng/ml BMP7、0.055mM 2-メルカプトエタノー
ル、0.1mM 非必須アミノ酸、GlutaMAXTM、ペニシリン、ストレプトマイシンおよび10% KSR(Invitrogen)を含むDMEM/F12を用いてゼラチンコートディッシュにて接着培養を16日間
行った。この時、3日に1度培地交換を行った。
分化誘導後の細胞をフローサイトメトリーによりGFP陽性細胞数を測定して得られた結果を図2aに示す。これより、上記の分化誘導によりGFP陽性細胞(つまりOSR1陽性細胞)への誘導が成功したことが確認された。
以上より、本方法を用いることでヒトiPS細胞よりOSR1陽性細胞を作製することできた
。
実施例1で得られたOSR1-GFPレポーターiPS細胞(3D36)をSNL細胞(McMahon, A.P. およびBradley, A. (1990) Cell 62;1073-1085)をフィーダー細胞として有する10cmディ
ッシュ上でコンフルエントになるまで培養し、CTK溶液(0.25%トリプシン、0.1%コラゲナーゼIV、20% KSRおよび1 mM CaCl2含有PBS)を加えて解離させた。続いて、MatrigelTM(BD)でコーティングした24ウエルデッィシュの各ウエルに、回収した全細胞に対して1/24量になるよう細胞を添加し付着させた。細胞をさらに、bFGFを含有した霊長類ES細胞用培地(リプロセル、日本)にて、コンフルエントになるまで培養し、100 ng/ml Wnt3a、100 ng/ml ActivinA、GlutaMAXTM(Invitrogen)、ペニシリン、ストレプトマイシンおよび2%FBS(Hyclone社製)を含有したDMEM/F12に交換し2日間培養した。さらに、培地を100 ng/ml Wnt3a、100 ng/ml BMP7、0.055mM 2-メルカプトエタノール、0.1mM 非必須アミノ酸、GlutaMAXTM、ペニシリン、ストレプトマイシンおよび10% KSR(Invitrogen)を含むDMEM/F12へ交換し、細胞を16日間培養した。培養の間、3日に1度培地交換を行った。培養後、GFPの発現を指標として細胞をFACSにより解析したところ、GFP陽性細胞の存在が確認された
。
実施例1で取得したOSR1-GFPレポーターiPS(3D45)を、MEFをフィーダー細胞として有す
る10cmディッシュにて、コンフルエントなるまで培養した。解離のために、CTK溶液を添
加し、さらに、AccutaseTMを添加したのち、ピペッティングにより細胞を解離した。GlutaMAXTM、ペニシリン、ストレプトマイシンおよび2%FBSを含有したDMEM/F12を加えて酵素
反応を止め、それに続いて、ピペッティングを行った。遠心分離により培地を除いた後、10μM Y27632、100 ng/ml Activin A、3μM CHIR99021、GlutaMAXTM、ペニシリン、スト
レプトマイシンおよび2%FBSを含有するDMEM/F12を有する、コラーゲンIでコーティングした24ウエルのプレートにて、細胞を2日間培養した。その後、100 ng/ml BMP7、3μM CHIR99021、0.055mM 2-メルカプトエタノール、0.1mM 非必須アミノ酸、GlutaMAXTM、ペニシ
リン、ストレプトマイシンおよび10% KSRを含有するDMEM/F12に培地を交換し、細胞を8日間培養した。培養の間、3日に1度培地交換を行った。培養後、GFPの発現を指標として用いて、細胞をFACSにより解析し、抗GFP抗体で免疫染色した後、蛍光顕微鏡によって細胞
を評価し、これにより、GFP陽性細胞が確認された(図3aおよび図3b)。さらに、OSR1に
ついてin situハイブリダイゼーションにより細胞を評価した結果、分化誘導後の多くの
細胞においてOSR1の発現が確認された(図3c)。

Claims (13)
- 以下の工程(i)および(ii):
(i)Activin AおよびWntもしくはGSK3β阻害剤を含む培地中でヒト多能性幹細胞を培養する工程、および
(ii)BMPおよびWntもしくはGSK3β阻害剤を含む培地中で、工程(i)で得られた細胞を培養する工程
を含む、ヒト多能性幹細胞から中間中胚葉細胞の製造方法であって、前記中間中胚葉細胞がOSR1陽性細胞である、上記方法。 - 前記工程(i)での培養においてヒト多能性幹細胞を浮遊培養して細胞集合体または細胞塊を形成させ、前記工程(ii)での培養において該細胞集合体または細胞塊を接着培養する、請求項1に記載の方法。
- 前記工程(i)での培養において、ヒト多能性幹細胞の細胞集合体または細胞塊を実質的に個々の細胞に分離することをさらに含む、請求項1に記載の方法。
- 前記工程(i)が、分離した細胞を、MatigelTMでコーティングした培養ディッシュまたはコラーゲンでコーティングした培養ディッシュに接着し、次いで、該細胞を培養することをさらに含む、請求項3に記載の方法。
- 前記工程(i)において、培地が、牛胎児血清(FBS)をさらに含む、請求項1〜4のいずれか1項に記載の方法。
- 前記工程(ii)において、培地が、ノックアウト血清代替物(KSR)をさらに含む、請求項1〜5のいずれか1項に記載の方法。
- 前記WntがWnt3aである、請求項1〜6のいずれか1項に記載の方法。
- 前記GSK3β阻害剤がCHIR99021である、請求項1〜6のいずれか1項に記載の方法。
- 前記BMPがBMP7である、請求項1〜8のいずれか1項に記載の方法。
- 前記工程(i)での培養が2〜5日間の培養期間行われる、請求項1〜9のいずれか1項に記載の方法。
- 前記ヒト多能性幹細胞が、ヒトiPS細胞またはヒトES細胞である、請求項1〜10のいずれか1項に記載の方法。
- 前記ヒト多能性幹細胞が、内在性OSR1の発現に連動して発現する外来レポーター遺伝子を染色体内に有する、請求項1〜11のいずれか1項に記載の方法。
- 前記レポーター遺伝子が、蛍光タンパク質、発光タンパク質、GUSまたはLacZをコードするDNAである、請求項12に記載の方法。
