Claims (17)

前記検体結合領域の較正された親和性,結合が検出された単一分子検体要素の数,及び前記閉じられた検出領域の体積に基づいて,前記閉じられた検出領域中の前記単一分子検体要素の数を決定するステップとを含む,方法。 - 請求項1に記載の方法であって,前記単一分子検体要素の数を決定するステップは,複数の区画された検体結合領域を用いる多重化によって,複数の前記単一分子検体要素を定量化するステップを含む,方法。
- 請求項1又は請求項2に記載の方法であって,前記単一分子検体要素の存在を示す検出信号に応じて前記単一分子検体要素の数のカウントを増加又は減少させて,前記単一分子検体要素のリカウントを防止するステップをさらに含む,方法。
- 請求項1から請求項3のいずれか1項に記載の方法であって,前記少なくとも一つの区画された検体結合領域は,マイクロアレイを含む,方法。
- 請求項1から請求項4のいずれか1項に記載の方法であって,前記単一分子検体要素は,タンパク質,糖タンパク質,タンパク質の後翻訳修飾体,DNA及びmRNAの少なくとも一つを含む生体分子を含む,方法。
- 請求項1から請求項5のいずれか1項に記載の方法であって,前記少なくとも一つの区画された検体結合領域は,親和性パッチを含み,前記単一分子検体要素の存在は前記親和性パッチで検出される,方法。
- 請求項1から請求項6のいずれか1項に記載の方法であって,前記検体は,細胞のタンパク質を含み,前記細胞は,前記閉じられた検出領域で捕捉され,前記細胞のタンパク質を遊離するために光学的又は化学的に溶解される,方法。
- 請求項2から請求項7のいずれか1項に記載の方法であって,前記検出された単一分子検体要素をブリーチングすることによって,又は画像比較によって,前記リカウントが防止され,前記画像比較のステップは,連続画像減算ステップを含む,方法。
- 請求項1から請求項8のいずれか1項に記載の方法であって,前記検体要素の蛍光体が検出される,方法。
- 請求項1から請求項9のいずれか1項に記載の方法であって,前記検体は,静的ハイブリダイゼーション又は連続フローレジームのいずれかの結合領域に存在するか,又は前記結合領域で液相の物質に結合している,方法。
