Description
本発明は、リポタンパク質媒介性取込み遮断が解決され得る脂質集合体またはリポソームに関する。より詳しくは、本発明は、カルボキシル頭基(carboxylic head group)またはリン酸頭基(phosphate head group)を極性領域内に有する負電荷脂質と、イミノ部分もしくはグアニド部分またはその誘導体を極性領域内に有する正電荷脂質の両方に含むリポソームの改善に関する。
リポソームは、活性成分の担体として広く使用されている。中性または負電荷のリポソームは、多くの場合、小分子薬物の送達に使用されるが、正電荷の(カチオン性)または最近紹介された類型である両性のリポソームは、主に、プラスミドまたはオリゴヌクレオチドなどの核酸の送達に使用される。運搬体(cargo)核酸の送達に使用されているカチオン性リポソームの重要な例としては、限定されないが、非特許文献1;非特許文献2;非特許文献3;de Fougerolles,Nat.Rev.Drug Discov.(2007)6:443−453;Kimら,Mol.Ther.(2006)14:343−350;Morrissey,Nat.Biotech.(2005)23:1002−1007;Peer,Science(2008)319:627−630およびSantel,Gene Ther.(2006)13:1222−1234が挙げられる。核酸の送達のための両性リポソームの適用は、Andreakosら,Arthritis Rheum.(2009)60:994−1005に示されている。
したがって、本発明の目的は、種々の血清の存在下で細胞をトランスフェクトすることが可能な脂質集合体またはリポソームを提供することであった。
本発明は、脂質集合体、リポソームおよびその細胞トランスフェクションのための使用を提供する。ここで、前記脂質集合体は、アニオン性両親媒性物質とカチオン性両親媒性物質を含み、該カチオン性両親媒性物質の少なくとも一部分は、pH7.5で実質的に荷電したイミノ脂質であり、該アニオン性両親媒性物質はカルボキシル脂質またはリン酸脂質(phosphate lipid)であり、さらに、該カチオン性両親媒性物質と該アニオン性両親媒性物質との間の電荷比率は1.5以上である。
本発明は、例えば以下の項目を提供する。
(項目1)
アニオン性両親媒性物質とカチオン性両親媒性物質とを含む脂質集合体であって、前記カチオン性両親媒性物質の少なくとも一部分が、生理学的条件下で実質的に荷電したイミノ脂質であり、前記アニオン性両親媒性物質はカルボキシル脂質またはリン酸脂質であり、さらに、前記カチオン性両親媒性物質と前記アニオン性両親媒性物質との間の電荷比率が1.5以下である、脂質集合体。
(項目2)
アニオン性両親媒性物質とカチオン性両親媒性物質を含む、項目1に記載の脂質集合体であって、前記カチオン性両親媒性物質の少なくとも一部分が、生理学的条件下で実質的に荷電したイミノ脂質であり、さらに、前記アニオン性両親媒性物質の少なくとも一部分がカルボキシル脂質であり、前記カチオン性両親媒性物質と前記アニオン性両親媒性物質との間の比率が1.5であるか、またはそれより小さい、脂質集合体。
(項目3)
脂質の組合せを含む、項目1または2記載の脂質集合体であって、前記組合せの前記カチオン性脂質がグアニド部分を含み、前記組合せの前記アニオン性脂質がカルボキシル基を含み、さらに、前記グアニド部分と前記カルボキシル基との間の比率が1.5であるか、またはそれより小さいことを特徴とする、脂質集合体。
(項目4)
アニオン性両親媒性物質とカチオン性両親媒性物質を含む、項目1に記載の脂質集合体であって、前記カチオン性両親媒性物質の少なくとも一部分が、生理学的条件下で実質的に荷電したイミノ脂質であり、さらに、前記アニオン性両親媒性物質の少なくとも一部分がリン酸脂質であり、前記カチオン性両親媒性物質と前記アニオン性両親媒性物質との間の電荷比率が1.5であるか、またはそれより小さい、脂質集合体。
(項目5)
さらに、前記カチオン性両親媒性物質の前記荷電したイミノ基が、7.5より大きいpKを有し、イミン、アミジン、ピリジン、2−アミノピリジン、複素環式窒素塩基、グアニド部分、イソ尿素またはチオイソ尿素から選択されることを特徴とする、項目1〜4のいずれかに記載の脂質集合体。
(項目6)
カチオン性両親媒性物質がI1〜I113の構造、構造A1〜A21または構造L1〜L17を含む群から選択され、前記群のメンバーが、さらに、7.5より大きいpKに従って選択される、項目1〜5のいずれかに記載の脂質集合体。
(項目7)
さらに、前記カチオン性脂質がPONA、CHOLGUA、GUADACA、MPDACAまたはSAINT−18の群から選択されることを特徴とする、前記項目のいずれかに記載の脂質集合体。
(項目8)
さらに、前記アニオン性脂質がCHEMS、DMGS、DOGS、DOPAまたはPOPAの群から選択されることを特徴とする、前記項目のいずれかに記載の脂質集合体。

脂質の化学
「荷電性の」により、両親媒性物質が4〜pH8の範囲のpKを有することを意図する。したがって、荷電性の両親媒性物質は弱酸または弱塩基であり得る。電荷両親媒性物質に関連する「安定な」は、pKがこの範囲外であり、pH4〜pH8の範囲で実質的に安定な電荷をもたらす強酸または強塩基を意味する。

このような脂質の一部は、以前に文献に示されている(例えば、グアニド脂質は、WO91/16024、WO97/43363、WO98/05678、WO01/55098、WO2008/137758(アミノ酸脂質)、EP 0685234(ジアシルグリセロール系)、US5965434(これもジアシルグリセロール系)またはピリジニウム化合物はUS6726894に)。さらに、WO29086558に示されたように、または構造(15)に図示したように、択一的な脂質主鎖(例えば、ジオキソランリンカーセグメントを含むが、それぞれの頭基の官能性は維持されているもの)を使用することも可能である。
本発明では、アニオン性両親媒性物質とカチオン性両親媒性物質を含み;該カチオン性両親媒性物質の少なくとも一部分が、生理学的条件下で実質的に荷電したイミノ脂質であり、さらに、該アニオン性両親媒性物質の少なくとも一部分がカルボキシル脂質またはリン酸脂質である脂質混合物を開示する。
本明細書に記載する成分は、集合して、当業者に知られた種々の構造になるものであり得る。該成分は、1つまたはいくつかの個々の二重層を含むリポソーム、他の超分子脂質集合体、または水相を提供するかなり大きな内部容積を有する小胞であり得る。また、これは、エマルジョン滴、またはリポプレックス集合体の形態の構造であってもよく、後者は、多くの実施形態において、脂質と核酸の静電複合体を含むものである。好ましい実施形態では、このような構造はリポソームまたは小胞である。多くの実施形態において、リポソームまたは小胞は、かなり大きな水性内部を有する。本発明の多くの態様では、活性医薬成分が脂質集合体に錯化、封入され、封鎖あるいは会合される。
本発明のリポソームまたは脂質集合体は、少なくとも1種類の活性薬剤を封鎖または封入し得るものである。前記活性薬剤は薬物を含むものであり得る。一部の実施形態では、前記活性薬剤は1種類の以上の核酸を含むものであり得る。好ましい実施形態では、活性成分は核酸からなる。
コレステロール系脂質
本発明の教示を例示するため、グアニド部分(荷電したイミノ基,CHOL−GUA)、イミダゾール部分(非荷電したイミノ基,CHIM)またはジメチルアミノもしくはトリメチルアンモニウム部分(非イミノだが電荷基,DC−CHOLもしくはTC−CHOL)を含むコレステロールのカチオン性誘導体を、システマテックに種々のアニオン性脂質と組み合わせた。
頭基の化学に対する血清抵抗性トランスフェクションの依存性をさらに調べるため、共通のジアルキル−カルボン酸(DACA)アンカーをその疎水性ドメインとして使用し、以下の脂質を合成した。
US6726894に記載のピリジニウム脂質 SAINT−18(構造31)を使用すると、同様の所見が得られた。
本発明の教示をさらに例証するため、カチオン性グアニド脂質PONA(パルミトイル−オレオイル−ノル−アルギニン,構造21)を、CHEMSまたはDMGSなどの種々の脂質アニオンと組み合わせた。カチオン性脂質とアニオン性脂質の比率をシステマテックな様式で種々に変え、得られた二元混合物に、任意選択で、さらに20mol%のコレステロールを加えた。個々の脂質混合物でリポソームを形質転換し、活性siRNAおよび対照siRNAの封入に使用した。HeLa細胞にて試験すると、実施例5に示すように、細胞生存性の効率的かつ特異的な阻害が、ほとんどの試験製剤で観察された。活性は、ヒトまたはマウス血清の存在によって全く影響されなかったか、または影響はほんのわずかであった。
リポソームのトランスフェクションと競合するリポタンパク質は、その密度に応じてさまざまな構造を含むものである。これは、カイロミクロン、VLDL、LDL、IDLまたはHDL粒子として知られている。内因性経路では、カイロミクロンは小腸の上皮内層で合成され、合成には、ApoB遺伝子産物の短鎖バリアントであるApoB−48が使用される。さらに、リポタンパク質をHDL粒子で交換されると、ApoC−IIおよびApoEがカイロミクロン粒子に転移され、該粒子からの脂質の放出に必要とされる酵素リポタンパク質リパーゼの活性化という最初の媒介がもたらされる。加水分解されたカイロミクロンは、いわゆるレムナントを形成し、これは、主に、そのApoE部分の認識によって肝臓に取り込まれる。VLDL粒子の合成、成熟、使用および再利用もまさに同じ経路に従うが、肝臓から始まり、ApoB−100タンパク質がその構造形成単位として使用される。この場合も、ApoEによってVLDL−レムナント(いわゆる、IDL粒子)の最終の取込みおよび再利用が媒介される(http://en.wikipedia.org/wiki/Lipoproteinも参照のこと)。
本発明の教示は、以下の実施例を検討すると、よりよく理解され得よう。しかしながら、本発明の教示は、この実施例によってなんら限定されるべきでない。
リポソームを、WO07/107304に開示された方法を用いて調製した。より詳しくは、脂質をイソプロパノールに溶解させ、NaAc 20mM、スクロース300mM(pH4.0(HAcでpH調整))のsiRNA溶液をアルコール性脂質ミックスに添加し、アルコールの最終濃度を30%にすることにより、リポソームを生成させた。形成されたリポソーム懸濁液を、2倍容量のNa2HPO4 136mM、NaCl 100mM(pH9)でpH7.5にシフトさせ、脂質の最終濃度を3mMに、イソプロパノールの最終濃度を10%にした。
対照siRNA(SCR):同じ供給源の21量体 。
HeLa細胞をDSMZ(German Collection of Micro Organism and Cell Cultures)から取得し、DMEM(Gibco−Invitrogen)中に維持し、10%FCSを補給した。細胞を2.5×104細胞/mlの密度でプレーティングし、100μlの培地中、5%CO2下、37℃で培養した。16時間後、siRNA含有リポソームを希釈し、10μlを細胞に添加し、最終濃度を0.4〜100nM Plk1またはスクランブル型siRNAにした;また、未処理細胞および細胞なしのウェルにも10μlの希釈バッファーを添加した。細胞培養皿を5%CO2下、37℃で72時間インキュベートした。
DODAP:DMGS:コレステロール(24:36:40mol%)のリポソームに、上記の活性siRNAおよび対照siRNAを負荷し、25μlのリポソームを、異なる種に由来する75μlの血清(SIGMA−Aldrich)とともに30分間インキュベートした。その後、リポソームを細胞に添加し、インキュベーションを72時間継続し、細胞生存性を上記のようにして測定した。
実施例3と同様のリポソームを、特定の補体因子またはリポタンパク質を除いたヒト血清(SIGMA−Aldrich)とともに上記のようにしてインキュベートし、HeLa細胞に対するRNAiの効果を媒介する能力について解析した。
一連のリポソームを、PONA:アニオン性脂質:コレステロール(x:y:20mol%)から構築し、実施例1の場合と同じ活性siRNAおよび対照siRNAを負荷した。この一連のものにおいて、カチオン性成分PONAとアニオン性脂質CHEMSまたはDMGSの比を、0.33〜2の間で表に示したとおりにシステマテックに変えた。1より大きいカチオン性:アニオン性脂質比を有するリポソームに、さらに、2mol%DMPE−PEG2000(日本油脂)を加えた(粒子の凝集を回避するため)。この改良を表中に「+」で示す。C/A<1を有する粒子での対照反応では、PEG脂質の存在下または非存在下で、トランスフェクション特性の変化は示されなかった。
カチオン性脂質成分とアニオン性脂質成分の比率をシステマテックに変えた一連のリポソームを作製し、実施例5の場合と同じsiRNAを負荷した。カチオン性脂質成分はPONA、PONアミンおよびPONアンモニウムとし、アニオン性脂質はCHEMSとし、コレステロール含有量は20mol%に固定した。1より大きいカチオン性:アニオン性脂質比を有するリポソームに、さらに、2mol%DMPE−PEG2000(日本油脂)を加えた(粒子の凝集を回避するため)。この改良を表中に「+」で示す。
カチオン性脂質成分とアニオン性脂質成分の比率をシステマテックに変えた一連のリポソームを作製し、実施例5の場合と同じsiRNAを負荷した。カチオン性脂質成分はPONAとし、アニオン性脂質は、CHEMS、DMGSまたはDOGSとし、コレステロール含有量は0〜40mol%の間で変えた。1より大きいカチオン性:アニオン性脂質比を有するリポソーム、また、一部の他のリポソームに、さらに、2mol%DMPE−PEG2000(日本油脂)を加えた(粒子の凝集を回避するため)。この改良を表中に「+」で示す。
SAINT−18をカチオン性脂質として使用した。そのメチル化ピリジニウム構造は電荷イミノ部分を提供する。CHEM、DMGSおよびDOGSを、カルボキシル官能基を提供するアニオン性脂質として、個々に使用した。カチオン性脂質成分とアニオン性脂質成分の比率をシステマテックに変えた一連のリポソームを作製し、実施例5の場合と同じsiRNAを負荷した。脂質混合物に、さらに、20または40mol%のコレステロールを加えた。1より大きいカチオン性:アニオン性脂質比を有するリポソームに、さらに、2mol%DMPE−PEG2000(日本油脂)を加えた(粒子の凝集を回避するため)。この改良を表中に「+」で示す。
9.1 PONA:CHEMS:CHOLで形成されたリポソームのゼータ電位の解析
x mol%のPONA、y mol%のCHEMSおよび20mol%のコレステロール(総脂質濃度20mM,溶媒:イソプロパノール)を含む100μlの脂質ミックスを、10mMの酢酸と10mMのリン酸を含む900μlのバッファー(pH4)中に注入した。PONAおよびCHEMSのモルパーセンテージであるXおよびYは、表17のC/A比が得られるように調整した。
脂質粒子を、GUADACAとDOPA(脂質頭基がイミノ/リン酸の組合せ)の二元混合物からも調製した。この粒子は、9.1に記載のものと同様にして調製し、異なるC/A比を有する混合物について、表18のゼータ電位が記録された。
比較のため、同じ測定を、PONAをDOTAPで置き換えた脂質混合物で行なった。結果を表19に示す。PONA:CHEMSとは対照的に、DOTAP:CHEMSの両性粒子はC/A<1でのみ認められる。
25gのコレステロールクロロホルミエートと50当量(eq)のエチレンジアミンをジクロロメタンに溶解させ、20℃で6時間反応させた。アミノエチルカルバモイル−コレステレロール(cholestererol)を、クロマトグラフィーと晶出を用いて単離した。収量は28.7gであった(純度90%)。
42.4gのオレイルアルコール、2.5eqのジイソプロイルアゾジカルボキシレート、2.5eqのトリフェニルホスフィンおよび5eqのLiIを、テトラヒドロフラン(THF)中で、20℃にて24時間反応させた。ヨウ化オレイルをクロマトグラフィーによって単離し、純度は90%、収量は13.4gであった。
第1工程で、3.5gのDACAと1.5eqの1,1’−カルボニルジイミダゾールをジクロロメタンに溶解させ、20℃で16時間インキュベートした後、30eqのエチレンジアミンを添加した。反応混合物を20℃で4時間インキュベートした後、アミノエチル−DACAを精製した(クロマトグラフィーなど)。純度は90%であった(収量 3.2g)。
BADACAはDACAから、以下の手順に従って合成した。4.15gのDACA、1.2eqのp−アミノベンズアミジン、1.2eqのN.N’−ジシクロヘキシルカルボジイミドおよび3eqの4−ジメチルアミノピリジンを乾燥ジメチルホルムアミド中で混合し、70℃で16時間インキュベートした。BADACAを反応液から、クロマトグラフィーを用いて単離した。純度:95%,収量:1.62g。
カチオン性脂質成分とアニオン性脂質成分の比率をシステマテックに変えた一連のリポソームを作製し、実施例5の場合と同じsiRNAを負荷した。カチオン性脂質成分はCHOLGUA、CHIM、DC−CHOL、TC−CHOL、GUADACA、MPDACA、BADACAおよびPDACAとした。アニオン性脂質は、CHEMS、DMGSまたはDOGSとし、コレステロール含有量は、20mol%または40mol%のいずれかとし、すべての脂質混合物をデータ表に特定する。1より大きいカチオン性:アニオン性脂質比を有するリポソーム(C/A>=1)に、さらに、1.5mol%DMPE−PEG2000(日本油脂)を加えた。
0.75または1のいずれかのC/A比を有する一連のリポソームを作製し、実施例5の場合と同じsiRNAを負荷した。カチオン性脂質成分は、CHOLGUA、CHIM、DC−CHOL、GUADACA、MPDACA、BADACA、PONA、DOTAPまたはDODAPとした。アニオン性脂質はDOPAとし、コレステロール含有量は40mol%とし、すべての脂質混合物を表20に特定する。リポソームに、さらに、1.5mol%DMPE−PEG2000(日本油脂)を加えた。
一連のリポソームを、カチオン性脂質および中性脂質としてのコレステロールから作製した。これらの調製物にアニオン性脂質は使用しなかった。
カチオン性脂質成分は、CHOLGUA、CHIM、DC−CHOL、ADACA、GUADACA、MPDACA、BADACA、PONA、DOTAPおよびDODAPとし、リポソームは、実施例5に記載の手順により作製した。
Claims (15)
- アニオン性両親媒性物質とカチオン性両親媒性物質とを含む脂質集合体であって、前記カチオン性両親媒性物質の少なくとも一部分が、生理学的条件下で実質的に荷電したグアニド脂質であり、ここに、前記グアニド脂質は、下式:
で示されるCHOLGUAまたは、下式:
で示されるGUADACAであり、前記アニオン性両親媒性物質はカルボキシル脂質またはリン酸脂質であり、さらに、前記カチオン性両親媒性物質と前記アニオン性両親媒性物質との間の電荷比率が<1である、脂質集合体。 - 前記アニオン性両親媒性物質の少なくとも一部分がカルボキシル脂質である、請求項1に記載の脂質集合体。
- 前記アニオン性両親媒性物質がカルボキシル脂質であり、前記グアニド脂質のグアニド基と前記カルボキシル脂質のカルボキシル基との間の電荷比率が<1であることを特徴とする、請求項1または2記載の脂質集合体。
- 前記アニオン性両親媒性物質の少なくとも一部分がリン酸脂質である、請求項1に記載の脂質集合体。
- さらに、前記カチオン性両親媒性物質の前記荷電したグアニド基が、7.5より大きいpKを有することを特徴とする、請求項1〜4のいずれかに記載の脂質集合体。
- さらに、前記アニオン性両親媒性物質がCHEMS、DMGS、DOGS、DOPAおよびPOPAの群から選択されることを特徴とする、請求項3に記載の脂質集合体。
- さらに、0.5と<1との間である前記カチオン性両親媒性物質と前記アニオン性両親媒性物質との電荷比率を有することを特徴とする、請求項3に記載の脂質集合体。
- 請求項1〜7のいずれかに記載の脂質集合体であることを特徴とする、リポソーム。
- さらに、(a)コレステロール、ホスファチジルコリン、ホスファチジルエタノールアミン、スフィンゴミエリンおよびそれらの混合物から選択される中性脂質もしくは両性イオン脂質、または(b)PEG脂質を含む、請求項8に記載のリポソーム。
- さらに、前記中性脂質がコレステロールであり、前記脂質混合物中のコレステロールのモル分率が10mol%〜50mol%であるか、または、前記PEG脂質が最外膜リーフレット(outermost membrane leaflet)内に存在していることを特徴とする、請求項9に記載のリポソーム。
- さらにオリゴヌクレオチドを含む、請求項8〜10のいずれかに記載のリポソーム。
- 前記オリゴヌクレオチドが、デコイオリゴヌクレオチド、アンチセンスオリゴヌクレオチド、siRNA、転写に影響を及ぼす因子、リボザイム、DNAザイムまたはアプタマーである、請求項11に記載のリポソーム。
- 前記オリゴヌクレオチドが、そのホスホジエステル形態またはホスホチオエート形態においてDNA、RNA、LNA、PNA、2’OMe RNA、2’MOE RNA、2’F RNAなどの修飾ヌクレオシドを含む、請求項12に記載のリポソーム。
- 細胞のインビトロまたはエキソビボでのトランスフェクションのための請求項8〜13のいずれかに記載のリポソームの使用。
- 肺細胞のトランスフェクションに使用するための請求項8〜13のいずれかに記載のリポソームを含むエアロゾル。
