Description
(a)本発明は、損傷した組織治療用移植組成物及び損傷した組織部位への治療用細胞のインビボ移動を誘導する方法を提供する。
(b)本発明によると、本発明の化学走性因子(例えば、IL−8またはMIP−3α)と反応させた生分解性支持体を損傷した位置(例えば、骨組織、関節軟骨または皮膚)に移植して、組織再生のための細胞のホーミングを誘導/促進することにより、損傷した組織を治療することができる。
(c)したがって、本発明の組成物は、それ自体として、従来技術と比較し、より簡便で効率的に損傷した骨組織、関節軟骨または皮膚組織の治療に適用できるだけではなく、他家細胞を利用した細胞治療において、希少性の高い治療用細胞資源を効率的に活用できるようにすることにより、有用な治療補助剤として利用できる。
細胞培養
ヒトの骨髄由来の間葉系幹細胞は、自家腸骨移植術が必要な患者の同意下、IRB審議を経て、後腸骨稜(posterior iliac crest)から骨髄を吸引して遠心分離し、骨髄内細胞を沈殿させてペレットを製造し、上済み液を除去した後、DMEM−LG(Dulbecco’s modified eagle’s medium−low glucose; GIBCO−BRL, Grand Island, NY, USA)培養液に10%(v/v)牛胎児血清(FBS, fetal bovine serum; GIBCO BRL, Grand Island, NY, USA), 100 units/mlのペニシリンG(GIBCO)及び0.1mg/mlストレプトマイシン(GIBCO)が添加された基本培養液で洗浄した後、T−75cm2細胞培養フラスコ(Falcon, Germany)に分株させて、4日間培養した。以後、フラスコ底に付着されていない細胞は除去し、底に付着された間葉系幹細胞は、3日間隔で培養液を入れ替えて、フラスコに約90%程度満たされると、継代培養を行った。分離された骨髄由来の間葉系幹細胞は、2または3継代培養まで細胞を増殖させて、全ての実験は、2または3継代の細胞を使用した。
体外軟骨損傷条件は、前炎症性サイトカインであるIL−1β(R&D Systems Inc., Minneapolis, MN, USA)またはTNF−α(R&D Systems Inc.)で処理することにより組成した。間葉系幹細胞を12時間牛胎児血清の含まれていないDMEM−LG培養液で培養した後、10ng/mlの濃度でIL−1βまたはTNF−αを培養液に添加して、4、24、48時間細胞を培養した後、実験に使用した。
正常間葉系幹細胞で発現するケモカイン受容体を選別するためにRNeasy kit(Qiagen, Valencia, CA, USA)を利用して間葉系幹細胞の総RNAを抽出して、逆転写PCR法を通じてケモカイン受容体の遺伝子発現を分析した。分析したケモカイン受容体は、CCR1−10、 CXCR1−7、 CX3CR、 XCR1であって、総19個である(表1−1及び1−2)。増幅されたケモカイン受容体のDNAは、3100 Genetic Analyser (Applied BiosystemsInc., Foster City, CA, USA)を利用したDNA塩基配列分析技法を利用して、互いに重複する配列がないことを確認した。
IL−1βまたはTNF−αにより発現が増加するケモカイン受容体を選別するために、逆転写PCR法を利用して19個のケモカイン受容体のDNAを合成して試料DNAとして使用して、4、24または48時間IL−1βまたはTNF−αの刺激を受けた間葉系幹細胞のcDNAを、Omniscript RT Kit(Qiagen)を利用して合成してプローブcDNAとして使用した。ケモカイン受容体のDNAをナイロン膜(Amersham, GE healthcare, UK)にドッティング(dotting)して、紫外線(1200×100 μjules)を照射して固定し、前炎症性サイトカインの刺激を受けた間葉系幹細胞のcDNAをRediprime II random prime labeling system(GE Healthcare UK Limited. Amersham Place, Little Chalfont, Buckinghamshire, UK)とγ32P−dCTP(NEN Life Sciences, Boston, USA)を利用して標識した。膜に固定されたケモカイン受容体のDNAと標識された間葉系幹細胞のcDNAは、42℃で15時間ハイブリダイズした。膜を洗浄した後、蛍光イメージ分析器(FLA−7000, Fujifilm, Japan)でシグナルを検出し、密度計測器(TINA pixel analyzer version 2.10, Raytest Isotopen megerate GmbH, Straubenhardt, Germany)を利用して定量化した。定量化された結果値は、ペアt−検定(paired t−tests)を利用して、統計学的有意性を検証した(P−値<0.05)。
細胞増殖に対する影響を確認するために、MSCに50−500ng/mlのケモカインを処理して1日から4日間培養した。ケモカイン刺激後、各日付別にMTT法を利用して細胞増殖率を測定した。
軟骨芽細胞への分化誘導のために、10ng/ml TGF−β3(R&D system Inc, Minneapolis, MN, USA)及び30μg/ml アスコルビン酸(Sigma Co., St. Louis, MO, USA)が添加された基本培養液[10%牛胎児血清(GIBCO), 1%抗生剤(antibioticantimycotic)溶液(GIBCO)が含有されたDMEM−LG(GIBCO)]で14日間軟骨分化を誘導し、TGF−β3を処理しなかった間葉系幹細胞と比較し、軟骨分化状態を検査した。骨芽細胞への分化誘導のために骨芽細胞分化培養液[10mM β−glycerophosphate(Sigma), 100μM dexamethasone(Sigma), 50μg/ml ascorbic acid−2−phosphate(Sigma)が添加された基本培養液]で14日間分化誘導後、基本培養液で培養した間葉系幹細胞と比較した。
傷形成のためにシリコン培養挿入(silicon culture insert; IBIDI, Germany)を利用してMSCを培養した後、細胞に500ng/mLのケモカイン刺激を加えて、傷治癒程度を、移動した細胞数を数えて分析し、移動する細胞を実時間追跡して移動距離及び速度を測定した。
インビトロ細胞化学走性は、8μm孔大きさのトランスウェルインサート(Falcon, Germany)を利用して細胞を培養した後、マイクロポアを通過してケモカインに向いて移動した細胞をカウントして評価した。インビトロ細胞化学走性は、蛍光標識された1.5×106個のMSCが、尻尾血管に注入されてヌードマウス皮下に移植されたケモカイン(IL−8、 MIP−3a)を含有したPLGA支持体(韓国科学技術研究院バイオ素材センター キムスヒョン博士からの提供)に向いて移動することを実時間撮影装備(Optix, ART, USA)で観察した。
間葉系幹細胞で発現するケモカイン受容体
逆転写PCR法を行うために製作したヒトのケモカイン受容体に対するプライマー配列は表1−1及び1−2に示したとおりである。表1−1及び1−2のプライマーにより増幅された遺伝子の塩基配列は、DNA塩基配列分析法を通じて、他のケモカイン受容体の配列と重複しないことを確認した(結果は示していない)。RT−PCRを通じて、ヒトの骨髄由来の正常間葉系幹細胞から19個のケモカイン受容体の発現を分析した(図1)。3人以上の供与者(donor)から得た間葉系幹細胞からケモカイン受容体の発現を分析した結果、CCR2、 CCR6、 CCR7、 CCR8、 CCR9、 CCR10、 CXCR1、 CXCR5及びCXCR7が3人以上の間葉系幹細胞から共通的に発現した。
関節軟骨が損傷すると、前炎症性サイトカイン(IL−1βまたはTNF−α)が分泌されて、これに刺激された軟骨細胞などにより多様な化学走性因子の分泌が増加する[非特許文献23]。したがって、関節軟骨損傷モデルにおいて、間葉系幹細胞で発現するケモカイン受容体のうち、どのような受容体が増加するか調べるために、逆ドットブロット−ハイブリダイゼーションを利用して、3人の供与者から得た間葉系幹細胞において4、24及び48時間目にIL−1βまたはTNF−αの刺激により増加するケモカイン受容体を分析した。本研究では、代表供与者1人に対する結果だけを示しているが、反復実験を通じて、3人の供与者由来の間葉系幹細胞において、ケモカイン受容体の発現様相が全部類似していることを確認した(図2)。逆ドットブロット−ハイブリダイゼーションの結果を、各供与者の間葉系幹細胞に対する結果を密度計測器(densitometry)(TINA Pixel Analyzer version 2.10 (Raytest Isotopenmegerate GmbH, Straubenhardt, Germany)を利用して定量して、2倍以上(fold change≧2)増加した受容体を刺激時間によってまとめた結果、各供与者の間葉系幹細胞において、3人の供与者の間葉系幹細胞で共通的に前炎症性サイトカインにより2倍以上増加するケモカイン受容体をまとめてみると、CCR2、CCR4、CCR6、CXCR1、CXCR2、CXCR3などが3人の供与者の間葉系幹細胞で共通的に発現が増加して、その他の受容体は、発現増加率が統計的に有意ではなかった。
選別された候補物質の細胞移動に対する影響を評価するために、傷治癒技法を行った。既存報告によると、SDF1は、CXCR4とCXCR7を通じて間葉系幹細胞の化学走性を誘導すると知られている[非特許文献17]。したがって、本研究で選別された候補因子とSDF1とを比較してみた。まず、傷治癒効果が細胞増殖ではないことを確認するために、細胞増殖に対する影響を調べた結果、50、100、または500ng/mlの濃度で細胞毒性及び細胞増殖に対して何の影響を与えなかった(図2a)。4つの候補物質のうち、IL−8とMIP−3αが最も速い傷治癒結果を示し、多様な濃度のうち、500ng/mlの濃度で最もよい結果を確認した(図2b)。次に、動く細胞を実時間で追跡して、細胞の移動距離及び速度を測定した。MIP−3αが最も速い速度で最も長い距離を移動したことが確認された(図2c)。
14日間の間葉系幹細胞の骨、軟骨分化誘導の間、候補因子による影響を調べた結果、分化に対して影響を与えなかったことが確認された(図3)。したがって、本研究で発掘されたケモカインは、損傷した関節軟骨再生のために必要な自家間葉系幹細胞の流入を促進すると考えられる。
インビトロ及びインビボで選別された候補物質の間葉系幹細胞に対する化学走性を評価した。まず、トラスウェルインサートを利用したインビトロ実験で、TNFαに向いた細胞化学走性が観察された(図4a)。一方、選別された4つの候補物質のうち、IL−8とMIP−3αがインビトロで細胞化学走性を最も多く誘導して(図4b)、この2つの物質を併用して使用した時、化学走性は、さらに大きく増加することを確認した(図4c)。また、IL−8及びMIP−3αにより皮膚組織再生に必須的なケラチン細胞が流入できるかどうかをインビトロ実験で確認した結果、IL−8とMIP−3αがケラチン細胞の細胞化学走性を誘導した(図4d)。この結果に基づいて、IL−8とMIP−3αにより誘導された細胞化学走性をインビボでも確認した。PLGA支持体をPBSまたはIL−8及びMIP−3αにそれぞれ24時間浸した後、ヌードマウスなどの皮下に移植した後、尻尾血管を通じて注入された間葉系幹細胞の移動を、細胞に標識された蛍光を利用して確認した。約2週目に注入された間葉系幹細胞がIL−8及びMIP−3αから確認されて、6週目まで細胞の移動が増加して観察された(図5)。
支持体と共に移植された化学走性因子による炎症細胞の流入程度を調べて炎症反応を確認するために、6週目に移植された支持体を回収して、肉眼観察及びH&E染色法を行った。肉眼観察結果、PBS群に比べ、IL−8及びMIP−3α群の支持体は、移植された部位の周辺組織との融合がよくなっており、血管形成が多かった(図6a)。H&E染色の結果、PBS群は、移植された支持体の縁部位にのみ一部血管形成があって、その周辺にポア(pore)を通じてその細胞が浸透したが、IL−8またはMIP−3α群では、血管が支持体の中央部位まで多く形成されており、注入されたヒトの間葉系幹細胞が支持体中央部位まで浸透して細胞外基質を形成した。PBS、IL−8、MIP−3α群の3つとも大食細胞などを始めとした炎症性細胞が血管周辺で一部観察されたが、群間の差がなく、3つの群全部、炎症反応所見を確認することができなかった(図6b)。
Claims (9)
- 化学走性因子を含む生分解性支持体(biodegradable scaffolds)を有効成分として含み、
前記化学走性因子が、IL(interleukin)−8とMIP(Macrophage Inflammatory Protein)−3αとの組合せ、またはMIP−3αを含む、損傷した組織治療用移植組成物(implantable composition)。 - 前記生分解性支持体は、PLGA(poly(lactic−co−glycolic acid))、PLA(polylactic acid)、PGA(polyglycolic acid)、PCL(poly−ε−caprolactone)、PAA(poly(amino acid))、ポリアンハイドライド、ポリオルトエステル、コラーゲンゲル、ハイドロゲル、ポリビニルアルコールスポンジ、ゼラチン、ポリサッカライド、ポリフォスファゼン、ポリアクリレート、ポリエチレンオキシド−ポリプロピレングリコールブロック共重合体及びこれらの混合物から構成された群から選択されることを特徴とする、請求項1に記載の組成物。
- 前記損傷した組織は、骨組織、関節組織または皮膚組織であることを特徴とする、請求項1に記載の組成物。
- 前記関節組織は、股関節、肩、膝、足首、手首、肘、足根骨、尺骨、脊髄骨、足首骨、腸骨または顎関節であることを特徴とする、請求項3に記載の組成物。
- 前記皮膚組織は、表皮、真皮または皮下組織であることを特徴とする、請求項3に記載の組成物。
- 前記組成物は、損傷した組織部位に幹細胞またはケラチン細胞(keratinocyte)の細胞移動能を増大させることを特徴とする、請求項1に記載の組成物。
- 前記幹細胞は、自家(autologous)幹細胞であることを特徴とする、請求項6に記載の組成物。
- 前記幹細胞は、間葉系幹細胞または外胚葉幹細胞であることを特徴とする、請求項6に記載の組成物。
- 前記組成物は、哺乳動物に適用できることを特徴とする、請求項1に記載の組成物。
