Description
個々のウシからの50μlの量の冷却しかつ保存された(BSM IIタブで)乳を、エッペンドルフチューブに移す。PBS中のアクリジンオレンジ(1mg/ml)50μl及びNa2H2EDTA(0.1g/ml,pH=7)10μlを1つの試薬として一度に添加し、混合する。更に、次いで100μlのPBSを上述の混合物に添加し、混合する。室温で1〜2分間反応させた後に、この混合物を標準フローサイトメーターで測定した。このフローサイトメーターでは、単に例示としてではあるが、サンプルを、14μl/分の速度で検出領域を通過して流す。識別マーカーがメタクロマティック染料であるために、細胞蛍光を用いて、細胞型が識別され得る。本実施例で使用される染料は、アクリジンオレンジであるので、本実施例では、監視される緑色(ここではFL1−Hと名づけられる)及び赤色(ここではFL2−Hと名づけられる)の強度で、蛍光が青色レーザー(488nm)によって励起される。400,000のFL1−H値が、トリガ信号として用いられる。本発明の構造において、光散乱信号もまた獲得され、側方散乱光(ここではSSC−Hと名づけられる)の強度が、獲得された信号からのノイズ(すなわち、計測される細胞に無関係の信号)を減らすために好都合に使用される。
個々のウシからの乳を保存し(BSM IIタブで)、分析前に40℃に加熱した。50μlの容量の乳をエッペンドルフチューブに移した。160μlのアクリジンオレンジ試薬(PBS中、0.3mg/mlのアクリジンオレンジ及び0.03mg/mlのNa2H2EDTA)を添加し混合した。室温で1分間反応させた後に、この混合物を、実施例1で使用された標準フローサイトメーターで測定した。このフローサイトメーターでは、サンプルが100μl/分の速度で測定され、青色レーザー(488nm)によって再励起させた。200,000のFL1−H値をトリガ信号として使用する。蛍光信号(FL1−H、FL2−H及び光散乱信号(側方散乱、SSC−H))が再度獲得される。
Claims (10)
- 感染の程度を判定するための方法において、
i)細胞型間の識別をもたらすのに十分な量で、識別マーカーを含む試薬を、1:200未満の希釈比で、哺乳類の乳に添加することによって未単離のサンプルを調製する工程と、
ii)前記サンプル中の異なる細胞型と別々に結合するようになる前記マーカーから得られる前記識別マーカーにおける差異に感受性がある検出システムを有するフローサイトメーターによって、前記サンプル上の前記マーカーによって識別された、白血球のリンパ球、単球、マクロファージ及び多形核細胞型からなる群から選択される体細胞の1つ以上の白血球型のそれぞれに関して細胞数を測定する工程と、
iii)総体細胞数を測定する工程と、
iv)1つ以上の感染の程度に関連して予め決められた1つ以上の基準値との前記測定された細胞数とを比較することによって、前記測定された細胞数に応じて、感染の程度の指標を判定する工程と、
を含むことを特徴とする方法。 - 請求項1に記載の方法において、希釈比が1:50未満であることを特徴とする方法。
- 請求項1に記載の方法において、希釈比が1:5未満であることを特徴とする方法。
- 請求項1に記載の方法において、前記指標を判定する工程が、前記測定された細胞数を、1つ以上の感染の程度に関連するようそれぞれ予め決められた1つ以上の基準値と比較する工程を更に含むこととを特徴とする方法。
- 請求項1〜4のいずれか一項に記載の方法において、前記哺乳類の乳は、牛乳であることと、前記感染の程度の指標を判定する工程が、乳腺炎の存在及び程度のうちの一方又は双方を判定する工程を含むこととを特徴とする方法。
- 請求項5に記載の方法において、前記乳腺炎の指標が、無症候性乳腺炎の指標を含むことを特徴とする方法。
- 請求項5又は請求項6に記載の方法において、臨床型乳腺炎に進行する可能性を示す感染の程度が判定されることを特徴とする方法。
- 請求項5の記載の方法において、メタクロマティック染料が、前記識別マーカーとして使用されることを特徴とする方法。
- 請求項8に記載の方法において、前記メタクロマティック染料が、0.2mg/mlを超える濃度でのアクリジンオレンジであることを特徴とする方法。
- 請求項5に記載の方法において、前記1つ以上の基準値が、基準からの前記測定された細胞計数の偏差の範囲を示す基準値を含むことを特徴とする方法。
