本発明は、
可溶型hTRAILタンパク質配列の機能的断片であって、hTRAIL95より小さくない位置にあるアミノ酸で始まる断片、または70%以上の配列同一性を有する該機能的断片のホモログであるドメイン(a);および
免疫賦活性エフェクターペプチド配列であるをドメイン(b)を含み、
ドメイン(b)の配列が、ドメイン(a)のC末端および/またはN末端に付加している融合タンパク質に関する。
本発明の融合タンパク質に取り込まれたインターフェロン ガンマの配列を含むペプチドは、効果的にがん細胞を排除すると考えられる。
― メタロプロテアーゼMMPにより認識される配列、特に配列番号20により示されるPro Leu Gly Leu Ala Gly(一文字表示ではPLGLAG)、
― ウロキナーゼuPAにより認識される配列、特に配列番号21により示されるArg Val Val Arg(一文字表示ではRVVR)、および
それらの組み合わせ。
本発明の融合タンパク質の発現のために好ましい宿主細胞は大腸菌細胞である。
プラスミドの調製
標的融合タンパク質のアミノ酸配列を鋳型として使用して、それをコードしていおり、大腸菌における発現に最適化されたコドンを含むDNA配列を生成した。そのような手順により、大腸菌における標的タンパク質合成のその後の工程の効率を向上することができる。その後、得られたヌクレオチド配列を自動合成した。更に、制限酵素NdeI(リーディング鎖の5’末端)およびXhoI(リーディング鎖の3’末端)の切断部位を、標的タンパク質をコードしている得られた遺伝子に付加した。これらは、遺伝子をベクターpET28a(ノバジェン社)へクローン化するために使用した。それらは、タンパク質をコードしている遺伝子を他のベクターへクローン化するためにも使用できる。この構築物から発現された標的タンパク質は、N末端において、トロンビンにより認識される部位の後にポリヒスチジンタグ(6ヒスチジン)を有しており、これはその後のアフィニティークロマトグラフィーによる精製に利用される。
カナマイシン(30μg/mL)および100μM硫酸亜鉛を添加したLB培地に、一夜培養物を接種した。培養物を、600nmにおける光学濃度(OD)が0.60〜0.80になるまで37°Cで培養した。その後、IPTGを0.25〜1mMの範囲の最終濃度で添加した。25°Cで振盪しながら培養(3.5〜20時間)後、培養物を6、000gで25分間遠心分離した。細菌ペレットを、50mMKH2PO4、0.5MNaCl、10mMイミダゾールを含むpH7.4の緩衝液中に再懸濁した。懸濁液を、氷上で8分間超音波処理した(40%振幅、15秒パルス、10秒間隔)。得られた抽出物を、20000g、4°Cで40分間遠心分離して清澄化した。Ni−セルファロース(GEヘルスケア社)樹脂を、細菌細胞抽出物の調製に使用した緩衝液で平衡化して前処理した。

配列番号2のタンパク質は、347アミノ酸長および質量40kDaを有する融合タンパク質であり、TRAIL配列120−281のC末端でヒトインターフェロン アルファ(配列番号17)の165アミノ酸サブユニット2bがエフェクターペプチドとして付加している。エフェクターペプチドとTRAIL配列との間に、メタロプロテアーゼMMP(配列番号20)およびウロキナーゼuPA(配列番号21)により認識されるプロテアーゼ切断部位の配列が順次相互に隣接して組み込まれており、そのために、エフェクターペプチドが、融合タンパク質の内部移行の際に腫瘍環境で切断される。

配列番号45のタンパク質は、384アミノ酸長および質量44kDaを有する融合タンパク質であり、TRAIL配列95−281のN末端でヒトインターフェロン アルファのコンセンサス配列(配列番号47)がエフェクターペプチドとして付加している。エフェクターペプチド配列とTRAIL配列との間に、メタロプロテアーゼMMP(配列番号20)およびウロキナーゼuPA(配列番号21)により認識されるプロテアーゼ切断部位の配列の組み合わせが2つ組み込まれており、そのために、エフェクターペプチドが、融合タンパク質の内部移行の際に腫瘍環境で切断される。配列番号20および配列番号21の組み合わせの間で、融合タンパク質には、ペグ化用リンカー(配列番号22)が付加されている。タンパク質は、更にエフェクターペプチド配列とメタロプロテアーゼMMP切断部位との間にフレキシブルなグリシン−セリンリンカーGGSG(配列番号50)を含む。
融合タンパク質の抗腫瘍活性試験は、試験管テストとして腫瘍細胞株の細胞毒性試験を行い、生体内テストとしてマウスを使用して行った。比較の目的で、rhTRAIL114−281タンパク質およびプラセボを使用した。
例3、例5、例12および例14の融合タンパク質の調製物の構造品質を、円二色性(CD)により決定した。
MTT試験は、細胞の増殖、生存能力および細胞毒性を測定するために使用される比色定量試験である。ミトコンドリア酵素であるコハク酸塩テトラゾリウム還元酵素1による、黄色いテトラゾリウム塩MTT(4,5−ジメチル−2−チアゾリル)−2、5−ジフェニルテトラゾリウムブロミド)の非水溶性紫染料ホルマザンへの分解を利用する。MTTの還元は、生細胞においてのみ進行する。データ解析には、対照細胞と比較して処理された集団の細胞数の50%で還元が進行する濃度である、タンパク質のIC50濃度(単位ng/mL)の決定が含まれる。結果は、グラフパッド・プリズム5.0ソフトウェアを使用して解析した。テストは、文献の記載に基づいて行った(Celis JE, (1998), Cell Biology, a Laboratory Handbook, second edition, Academic Press, San Diego; Yang Y., Koh LW, Tsai JH., (2004) "Involvement of viral and chemical factors with oral cancer in Taiwan" Jpn J Clin Oncol, 34 (4), 176-183)。
EZ4U(バイオメディカ社)テストを使用して、非接着性細胞株におけるタンパク質の細胞障害活性をテストした。このテストは、テトラゾリウム塩の還元で生成されるホルマザンが水に可溶であるようにMTTを改変したものである。細胞生存能力の試験は、タンパク質(タンパク質を7つの濃度で各3ウェルずつ)と共に細胞を連続72時間培養した後に行った。これに基づいて、グラフパッド・プリズム5ソフトウェアによりIC50値(2回の独立した実験の平均値として)を決定した。
表5は、広範囲の最も一般的ながんに対応している様々な臓器由来の広範な腫瘍細胞パネルに対する、本発明の選択された融合タンパク質の試験管内での細胞障害活性テストの結果を示すものある。得られたIC50値により、融合タンパク質の高い細胞障害活性と、従ってがんの処置における潜在的な有用性が確認された。
タンパク質調製物の抗腫瘍活性を、ヒト結腸がんHCT116のマウスモデルにおいてテストした。
HCT116細胞は、10%ウシ胎児血清および2mMグルタミンを補充したOpti−MEM(インビトロジェン社、カタログ番号22600−134)を1:1の比率で添加したRPMI1640培地(ハイクローン社、ローガン、ユタ州、米国)中で維持した。マウスへの移植の日に、トリプシン(インビトロジェン社)で細胞を洗浄して、細胞をサポートから剥離し、その後、細胞を1300rpm,4°Cで8分間遠心分離し、HBSS緩衝液(Hanks培地)に懸濁し、カウントし、細胞数を25x106個/mLに希釈した。
本発明のタンパク質の抗腫瘍活性は、チャールス・リバー社(独国)から入手した7〜9週齢CDヌード(Crl:CD1−Foxn1nu1)または4〜5週齢Crl:SHO−PrkdcscidHrhrマウスを使用して試験した。マウスは、食糧および蒸留水は(適宜)自由摂取として、特定病原体感染防止条件下で保持した。動物を使用したすべての実験は、「Interdisciplinary Principles and Guidelines for the Use of Animals in Research, Marketing and Education」 issued by the New York Academy of Sciences' Ad Hoc Committee on Animal Research and were approved by the IV Local Ethics Committee on Animal Experimentation in Warsaw (No.71/2009) のガイドラインに従って行った。
ヒト結腸がんモデル
Crl:CD1−Foxn1 nu 1マウス
0日目に、0.2mLのHBSS緩衝液に懸濁した5x106個のHCT116細胞を0.5x25mm針付きシリンジ(BogMark社)を使用して、Crl:CD1−Foxn1nu1マウスの右側に皮下(sc)移植した。腫瘍の大きさが約 50〜140 mm3となったとき(14日目)、処置群の平均腫瘍サイズが約 70 mm3となるようにマウスをランダムに分別した。処置群には、例3、例12および例14の本発明の融合タンパク質調製物(10mg/kg)、比較参照としてrhTRAIL114−281(10mg/kg)、対照として製剤用緩衝液(5mMNaH2PO4、95mMNa2HPO4、200mMNaCl、5mMグルタチオン、0.1mMZnCl2、10%グリセリン、80mMショ糖、pH8.0)を投与した。調製物は8〜12日目および15〜19日目の10日間毎日静脈内投与した(i.v.)。治療群の平均腫瘍サイズが約1000mm3となったとき、マウスの脊髄を損壊して屠殺した。対照群には、rhTRAIL114−281を与えた。
0日目に、0.1mLのHBSS緩衝液に懸濁した5×106個のHCT116細胞を0.5×25mm針付きシリンジ(BogMark社)を使用して、Crl:SHO−PrkdcscidHrhrマウスの右側に皮下(sc)移植した。腫瘍の大きさが200〜700mm3となったとき(18日目)、処置群の平均腫瘍サイズが約400mm3となるようにマウスをランダムに分別した。処置群には、例14の本発明の融合タンパク質調製物(50mg/kg)、比較参照としてrhTRAIL114−281(20mg/kg)、対照として製剤用緩衝液(5mMNaH2PO4、95mMNa2HPO4、200mMNaCl、5mMグルタチオン、0.1mMZnCl2、10%グリセリン、80mMショ糖、pH8.0)を投与した。調製物は2日毎に6回静脈内投与した(i.v.)。実験の32日目に、マウスの脊髄を損壊して屠殺した。
PLC/PRF/5細胞
0日目に、HBSS:マトリゲル(3:1)混合物の緩衝液0.1mLに懸濁した7x106個のPLC/PRF/5細胞を0.5×25mm針付きシリンジ(BogMark社)を使用して、Crl:SHO−PrkdcscidHrhrマウスの右側に皮下(sc)移植した。腫瘍の大きさが140〜300mm3となったとき(31日目)、処置群の平均腫瘍サイズが約200mm3となるようにマウスをランダムに分別した。処置群には、例14の本発明の融合タンパク質調製物(20mg/kg)、比較参照としてrhTRAIL114−281(30mg/kg)、対照として製剤用緩衝液(5mMNaH2PO4、95mMNa2HPO4、200mMNaCl、5mMグルタチオン、0.1mMZnCl2、10%グリセリン、80mMショ糖、pH8.0)を投与した。調製物は3日毎に4回、その後2日毎に(q3dx4およびq2dx2)2回静脈内投与した(i.v.)。実験の32日目に、マウスの脊髄を損壊して屠殺した。
0日目に、HBSS:マトリゲル(3:1)混合物の緩衝液0.1mLに懸濁した7x106個のHepG2細胞を0.5×25mm針付きシリンジ(BogMark社)を使用して、Crl:SHO−PrkdcscidHrhrマウスの右側に皮下(sc)移植した。腫瘍の大きさが250〜390mm3となったとき(19日目)、処置群の平均腫瘍サイズが約330mm3となるようにマウスをランダムに分別した。処置群には、例14の本発明の融合タンパク質調製物(50mg/kg)、比較参照としてrhTRAIL114−281(30mg/kg)、対照として製剤用緩衝液(5mMNaH2PO4、95mMNa2HPO4、200mMNaCl、5mMグルタチオン、0.1mMZnCl2、10%グリセリン、80mMショ糖、pH8.0)を投与した。調製物は2日毎に6回静脈内投与した(i.v.)。実験の31日目に、マウスの脊髄を損壊して屠殺した。
TGI[%](腫瘍増殖阻害率)=(WT/WC)×100 −100%
実験結果は、平均値±標準偏差(SD)で示した。すべての計算およびグラフ作成は、グラフパッド・プリズム5.0ソフトウェアを使用して行った。
Claims (19)
- hTRAILタンパク質配列(配列番号16)の機能的断片であって、hTRAIL95からhTRAIL122までの範囲(両端を含む)のアミノ酸で始まり、hTRAIL281のアミノ酸で終わる断片、hTRAIL131、149、159、193、199、201、204、212、215、218および251からなる群もしくはhTRAIL195,269および214からなる群から選択された位置での1つ以上の変異を有する前記機能的断片のホモログ、またはD218H変異体もしくはY189N−R191K−Q193R−H264R−I266R−D269H変異体から選択される前記機能的断片のホモログであるドメイン(a);ならびに
― 配列番号19のインターフェロン ガンマの偽二量体;および
― 配列番号46のインターフェロン アルファ2bの偽二量体;
からなる群から選択される免疫賦活性エフェクターペプチド配列であるドメイン(b)を含み、
ドメイン(b)の配列が、ドメイン(a)のC末端またはN末端に付加している、融合タンパク質。 - ドメイン(a)が、95、116、120、121または122の位置のアミノ酸で始まる断片からなる群から選択される、請求項1に記載の融合タンパク質。
- 融合タンパク質が、ドメイン(a)とドメイン(b)との間に、メタロプロテアーゼMMPにより認識される配列、ウロキナーゼuPAにより認識される配列およびそれらの組み合わせからなる群から選択される、プロテアーゼ切断部位を含むドメイン(c)を少なくとも1つ包含する、請求項1または2に記載の融合タンパク質。
- メタロプロテアーゼMMPにより認識される配列が配列番号20であり、ウロキナーゼuPAにより認識される配列が配列番号21である、請求項3に記載の融合タンパク質。
- ドメイン(c)が、相互に隣接する、メタロプロテアーゼMMPにより認識される配列とウロキナーゼuPAにより認識される配列との組み合わせである、請求項3または4に記載の融合タンパク質。
