Description
神経系傷害又は疾患の部位への移植後に神経回復及び/又は神経再生を刺激することができる細胞が本明細書中に開示されている。ある態様において、上記細胞は骨髄接着幹細胞(MASCs)に由来しているが、in vitroでの処理及び培養後にある遺伝子のメチル化状態において改変している。したがって、本発明者は、1又は複数の遺伝子のメチル化状態の改変は前駆細胞を上記前駆細胞が有さなかった神経再生特性を有する子孫細胞に変換しうることを発見した。
1.細胞内の遺伝子のメチル化状態の改変方法であって:
(a)Notch細胞内ドメインをコードする配列を含むポリヌクレオチドで上記細胞をトランスフェクトすること;及び
(b)上記細胞又は上記細胞の1又は複数の子孫における遺伝子のメチル化状態がトランスフェクトされていない細胞における上記遺伝子と比較して改変されており、それにより上記遺伝子のメチル化状態が改変されている、上記トランスフェクトされた細胞を培養すること
を含む方法。
2.上記遺伝子がPITX2遺伝子である、態様1の方法。

37.in vitroでの培養をとおして前駆細胞に由来する細胞であって、ここで:
(a)上記細胞が神経組織の成長及び/又は再生を援助する;
(b)上記細胞における1又は複数の遺伝子のメチル化状態が上記前駆細胞に比較して改変されている;及び

50.神経再生細胞の同定方法であって、上記細胞における1又は複数の遺伝子のメチル化状態を分析することを含む方法であって、ここで上記分析された遺伝子のメチル化状態における変化は神経再生細胞を表している、方法。
51.上記分析が上記1又は複数の遺伝子の増加されたメチル化についてである、態様50の方法。

診断としてのメチル化状態
問題の1又は複数の遺伝子内の又は問題の1又は複数の遺伝子近辺の特定のCpG配列のDNAメチル化状態における変化の分析は細胞を同定するために及び異なるDNAメチル化パターンを有する他の細胞からそれを区別するために使用されうる。例えば、幹細胞又は他の型の前駆細胞において、特定のCpG配列がそのC残基上でメチル化され、そしてさらなる分化に際して、上記C残基が脱メチル化される場合、そのC残基の脱メチル化はその分化段階についてのマーカーとして使用されうる。逆に、C残基のメチル化は分化についてのマーカーとしてはたらきうる。メチル化状態における全体的な全か無かの変化は必要ではない;特定のCpG配列でのメチル化頻度における変化もまた診断でありうる。
前駆細胞
ある遺伝子のメチル化状態を改変することにより神経再生細胞に変換されうる前駆細胞はどんな型の非最終分化細胞でもありうる。例えば、例えば米国特許第5,843,780号;第6,200,806号及び第7,029,913号中に開示される全能幹細胞は前駆細胞として使用されうる。全能幹細胞は培養され(例えば、米国特許第6,602,711号及び第7,005,252号)、そして開示された方法の実施において前駆細胞としてまた使用されうるさまざまな型の多能性細胞(例えば、米国特許第6,280,718号;第6,613,568号及び第6,887,706号)に分化されうる。
Notch細胞内ドメイン
Notchタンパク質は細胞内シグナリングをとおして細胞分化に影響する、全ての後生動物中に見られる膜貫通型受容体である。Notch細胞外ドメインのNotchリガンド(例えば、Delta, Serrate, Jagged)との接触はNotchタンパク質の2のタンパク質分解切断をもたらし、そのうちの二番目のものはγ‐セクレターゼにより触媒され、そしてNotch細胞内ドメイン(NICD)を細胞質に放出する。マウスNotchタンパク質では、この切断はアミノ酸gly1743及びval1744の間で起こる。NICDは核に移動し、そこでそれは転写因子としてはたらき、追加の転写調節タンパク質(例えば、MAM、ヒストンアセチラーゼ)を補充し、さまざまな標的遺伝子(例えば、Hes1)の転写抑制を緩和する。
細胞培養及びトランスフェクション
細胞培養のための標準の方法は本分野において知られている。例えば、R. I. Freshney “Culture of Animal Cells: A Manual of Basic Technique,” Fifth Edition, Wiley, New York, 2005を参照のこと。
DNAメチル化の標的化改変の方法
前駆細胞の神経再生細胞への変換には、ある遺伝子のメチル化状態における変化が付随するので;メチル化状態の標的化改変は前駆細胞を神経再生細胞に変えるために使用されうる。
ヒトドナーから得られた骨髄吸引物を50ml管中に12.5ml等分に分け、そして12.5mlの生育培地(ペニシリン/ストレプトマイシン及び2mM L‐グルタミンで補充したαMEM中の10%FBS)を各管に添加した。上記管の内容物を反転により混合し、そして上記管を200×gで8分間遠心分離した。上部の透明な相を捨て、下部相の体積を新しい生育培地で25mlに合わせ、そして上記管を再び混合し、そして遠心分離した。上部層を再び除去した。各管中の下部相の体積を25mlに再び合わせ、そして全ての管の内容物を1つの250ml管中に集めた。Trypan Blue除外及び有核細胞カウントの決定による細胞濃度の決定後、細胞を1フラスコ当たり100×106総有核細胞の密度で1フラスコ当たり40mlの生育培地中のT225フラスコ中にまいた。上記フラスコをCO2インキュベーター内で37℃で3日間インキュベートし、その間MASCsは上記フラスコに接着した。
実施例2:神経再生細胞(NRCs)の調製
NRCs又はSB623細胞としても知られる、神経再生細胞を以下のように、M2代培養物から回収したMASCsから調製した。
A.トランスフェクション混合物の調製
神経再生細胞をNotch細胞内ドメインをコードするプラスミドでのM2代MASCsのトランスフェクションにより作出した。上記プラスミド(pN2)は、細胞内ドメインをコードする、ヒトNotch‐1タンパク質のアミノ酸1703‐2504をコードする配列をマルチクローニング部位に導入した、pCI‐ネオ骨格(Promega, Madison, WI)を含んだ。MASCsの各フラスコについて、40μgのプラスミド及び0.2mlのFugene6(商標)溶液を含む5mlのトランスフェクション混合物を使用した。上記トランスフェクション混合物を作出するために、(トランスフェクトされるべき細胞のフラスコの数に因り)適切な量のFugene(商標)溶液をガラスピペットを用いて滅菌250ml管中のαMEMに添加した。上記溶液を穏やかに混合し、そして室温で5分間インキュベートした。適切な量のプラスミドDNAをその後Fugene(商標)/αMEM混合物に一滴ずつ添加し、穏やかに混合し、そして室温で30分間インキュベートした。
B.トランスフェクション
トランスフェクションのために、M2代MASCsを(実施例1中に示されているような)トリプシン処理により回収し、そして1のT225フラスコ当たり40mlの生育培地中に2.5×106細胞の密度でまいた。上記細胞が50〜70%コンフルエンスに達したとき(通常18〜24時間以内)、それらを、それらの生育培地を1フラスコ当たり35mlのトランスフェクション培地(ペニシリン/ストレプトマイシンを含まないαMEM+10%FBS)で置き換えることにより、トランスフェクションのために準備した。
C.トランスフェクトされた細胞の選択
プラスミドDNAを組み込んだ細胞を、上記培地を1フラスコ当たり40mlの選択培地(100μg/ml G‐418を含む生育培地)で置き換えることによりトランスフェクション48時間後に選択した。新しい選択培地を、選択を開始した3日後、及び再び5日後に提供した。7日後、選択培地を除去し、そして上記細胞に40mlの生育培地を与えた。上記培養物をその後新しい生育培地を2〜3日毎に再び与えながら、約3週間(18〜21日間の範囲)生育させた。
D.トランスフェクトされた細胞の拡充
細胞数を形質転換され選択された細胞の懸濁物について決定し、そして上記細胞を1フラスコ当たり2×106細胞(生存細胞の約30%シーディングを提供する)でT‐225フラスコ中にまいた。この培養物をM2P1(#1代)と呼ぶ。M2P1培養物に2〜3日毎に新しい培地を与え、そして細胞が90〜95%コンフルエンスに達したとき(通常継代後4〜7日)、それらを回収し、そして1フラスコ当たり2×106細胞で再びまき、M2P2代を作出した。M2P2培養物が90〜95%コンフルエンスに達したとき、それらをさらなる分析のために回収した。
実施例3:MASCs及びNRCsのメチル化パターンの比較
MASCsを上記実施例1中に示されているように、3つの独立したヒトドナー(D33、D39及びD41と呼ばれる)各々から調製した。各MASCs調製物の一部を上記実施例2中に示されているように、神経再生細胞を調製するために使用した。ゲノムDNAをこれらの6つの細胞調製物の各々から単離し、そして上記3つのドナーの各々について、神経再生細胞からのDNAのメチル化状態をそれらのMASC前駆細胞からのDNAのそれと比較した。
1.MASCs及び間葉細胞系についての既知のDNAメチル化マーカー
2.ディファレンシャルメチル化ハイブリダイゼーションを用いたゲノムワイドなスクリーニングにおいてMASCsについてのメチル化マーカーとして同定された遺伝子;及び
3.胚幹細胞分化に対して影響を有することが文献中で報告されている遺伝子
にしたがって選択した。
[平均メチル化値(MASCs)−平均メチル化値(NRCs)]2/[[標準偏差(MASCs)]2+[標準偏差(NRCs)]2]
上記Fisher基準は特定のCpG部位でのメチル化値における変動性を示す。1超のFisherスコアは有意であると考えられる。
MASCsに比較して、NRCsにおいてROPN1L遺伝子中にメチル化状態の変化を含むアンプリコンのヌクレオチド配列をそのメチル化状態が改変されているヌクレオチドを正確に同定するために分析した。この分析の結果は表8中に示されている。
実施例3及び4中に示されているメチル化変化を有する、実施例2中に示されているように調製された神経再生細胞は中枢及び末梢神経系のさまざまな障害の治療において有用である。例えば、その開示を全ての目的のためにそれらを全体として本明細書中に援用する、共同所有のWO 2009/023251(Feb. 19, 2009)を参照のこと。
Claims (7)
- 骨髄接着間質細胞を、神経系傷害又は疾患の部位への移植後に神経回復及び/又は神経再生を刺激することができる細胞に変換する方法であって、
(i)前記骨髄接着間質細胞におけるPITX2、DNMT3b、IGF2R及びSDF4遺伝子のメチル化を増加させること、並びに
(ii)ROPN1L及びTMEM179遺伝子のメチル化を減少させること
を含み、
ここで前記骨髄接着間質細胞及びその子孫細胞のいずれも、Notch細胞内ドメイン(NICD)をコードする配列を含むポリヌクレオチドでトランスフェクトされていない、方法。 - in vitroでの培養をとおして骨髄接着間質細胞に由来する細胞であって、ここで:
(a)神経組織の成長及び/又は再生を援助し;
(b)PITX2、DNMT3b、IGF2R及びSDF4遺伝子のメチル化が前記骨髄接着間質細胞と比べて増加しており;
(c)ROPN1L及びTMEM179遺伝子のメチル化が前記骨髄接着間質細胞と比べて減少しており;並びに
(d)in vitroでの培養中、上記骨髄接着間質細胞及びその子孫のいずれも、Notch細胞内ドメイン(NICD)をコードする配列を含むポリヌクレオチドでトランスフェクトされていない、
細胞。 - 上記PITX2、DNMT3b、IGF2R及びSDF4遺伝子のメチル化状態が、前記骨髄接着間質細胞をメチル化ドメイン及びDNA結合ドメインを含む1又は複数の融合タンパク質と、又はメチル化ドメイン及びDNA結合ドメインを含む融合タンパク質をコードする1又は複数の核酸と接触させることにより増加されており、ここで上記DNA結合ドメインは、上記PITX2、DNMT3b、IGF2R及びSDF4遺伝子の各々の中の1又は複数の配列に結合するように改変されている、請求項1に記載の方法。
- 上記ROPN1L及びTMEM179遺伝子のメチル化状態が、前記骨髄接着間質細胞を脱メチル化ドメイン及びDNA結合ドメインを含む1又は複数の融合タンパク質と、又は脱メチル化ドメイン及びDNA結合ドメインを含む融合タンパク質をコードする1又は複数の核酸と接触させることにより減少されており、ここで上記DNA結合ドメインは、上記ROPN1L及びTMEM179遺伝子の各々の中の1又は複数の配列に結合するように改変されている、請求項1に記載の方法。
- 上記PITX2、DNMT3b、IGF2R及びSDF4遺伝子のメチル化状態が、前記骨髄接着間質細胞をメチル化ドメイン及びDNA結合ドメインを含む1又は複数の融合タンパク質と、又はメチル化ドメイン及びDNA結合ドメインを含む融合タンパク質をコードする1又は複数の核酸と接触させることにより増大されており、ここで上記DNA結合ドメインは、上記PITX2、DNMT3b、IGF2R及びSDF4遺伝子の各々の中の1又は複数の配列に結合するように改変されている、請求項2に記載の細胞。
- 上記ROPN1L及びTMEM179遺伝子のメチル化状態が、前記骨髄接着間質細胞を脱メチル化ドメイン及びDNA結合ドメインを含む1又は複数の融合タンパク質と、又は脱メチル化ドメイン及びDNA結合ドメインを含む融合タンパク質をコードする1又は複数の核酸と接触させることにより減少されており、ここで上記DNA結合ドメインは、上記ROPN1L及びTMEM179遺伝子の各々の中の1又は複数の配列に結合するように改変されている、請求項2に記載の細胞。
- 神経系傷害又は疾患の部位への移植後に神経回復及び/又は神経再生を刺激することができる、骨髄接着幹細胞に由来する子孫細胞の同定方法であって、細胞におけるPITX2、DNMT3b、IGF2R、SDF4、ROPN1L及びTMEM179遺伝子のメチル化状態を分析することを含み、ここで、
(i)PITX2、DNMT3b、IGF2R及びSDF4遺伝子のメチル化が増加し、並びに
(ii)ROPN1L及びTMEM179遺伝子のメチル化が減少している細胞を、上記子孫細胞として同定する方法。
