Description
本出願は、2015年7月29日に出願の米国特許出願第62/198,614号、2016年1月15日に出願の同第62/279,490号、および2016年2月8日に出願の同第62/292,755号の関連出願であり、米国特許法第119(e)の下でこれらの出願に基づく優先権を主張する。その全内容は、参照によりその全体が本明細書に組み込まれる。

(1)二倍体化は細胞分裂周期の失敗に起因して一定の確率で起こり、2つの一倍体娘細胞よりむしろ1つの二倍体娘細胞になる。
(2)正常な細胞分裂周期は2つの娘細胞を産生し、平均的に同時に起こる。
(3)一倍体細胞または二倍体細胞のいずれかを支持するいかなる選択優位性もごくわずかであり、細胞死および異数性の場合は考慮しない。
実施例1.方法
ヒト卵母細胞操作および単為発生ES細胞株誘導
ヒト卵母細胞供与手法およびpESとswaPS細胞株誘導手法は、これまでに記述されている15、35。手短に言うと、成熟したMII期卵母細胞をカルシウムイオノフォアおよび/または電気パルスを使用して活性化させ、続いてピューロマイシンとともに4時間培養した。極体放出と、一倍性を示す単一前核の存在とを確認し、卵母細胞を胚盤胞期まで発育させた。核ゲノムが別の卵母細胞の核ゲノムと交換されている卵母細胞を活性化させた後、swaPS細胞を誘導した15。栄養外胚葉38にレーザーアブレーションを施し、次いで誘導培地にROCK阻害剤Y−27632を10μM添加することによってES細胞株を誘導した35。プレーティングから2〜3日後に、残りの栄養外胚葉細胞にレーザーアブレーションを施し、次いで増殖物の手動採取が可能になるまで、ICM細胞を10〜14日間増殖させた。
特に明記しない限り、ヒトES細胞は、15%のKnockout Serum Replacement(KSR,Thermo Fisher Scientific)、2mMl−グルタミン、0.1mM非必須アミノ酸、50単位/mLペニシリン、50μg/mLストレプトマイシン、0.1mMβ−メルカプトエタノール、および8ng/mL塩基性線維芽細胞増殖因子(bFGF)を補充したノックアウトダルベッコ改変イーグル培地で構成された標準ヒトES細胞培地中で増殖停止マウス胚線維芽細胞のフィーダー層上で培養した。細胞はマイコプラズマがなく、加湿インキュベーター内に、37℃、5%のCO2で維持した。継代培養は、EDTAを含まないトリプシン溶液A(Biological Industries)を用いる穏やかなトリプシン処理による機械的な培養、または分離後TrypLE Express(Thermo Fisher Scietific)を用い、ROCK阻害剤Y−27632(Stemgent)10μMを加えて、1日もしくは最高2日間の酵素的な培養のいずれかで行った。一倍体ES細胞は、mTeSR1(STEMCELL Technologies)培地またはStemFitN.AK03(Ajinomoto)培地中でマトリゲル被覆プレート(Corning)上でフィーダーを含まない状態で増殖することも可能である。増殖物が急速に伸長することにより、3継代の早い段階で一倍体ES細胞の分離が可能になる。
ヒト単為発生ES細胞株において6〜8継代で、中期スプレッド解析またはサブ−2c細胞選別(表1および表2)のいずれかによって一倍体細胞を識別した後、一倍体ES細胞株を、二倍体細胞を基準として用いて、1c細胞集団を選別することによって確立した。一倍体ES細胞培養物は、3〜4継代ごとに1c細胞を選別する濃縮ラウンドによってさらに維持した。
核分裂停止を誘導するために、コルセミド(Biological Industries)100ng/mLを直接加えた培地中の増殖細胞を37℃、5%CO2の加湿インキュベーター内で、40分間インキュベートした。次いで、細胞をトリプシン処理し、室温、1000RPMで遠心分離して、20分で37℃に温めた低張液(塩化カリウム2.8mg/mLとクエン酸ナトリウム2.5mg/mL)に徐々に再懸濁した。細胞は、固定液(3:1のメタノール:酢酸)を加え、室温で5分間インキュベートして、固定した。遠心分離および固定液での再懸濁後、固定化を少なくとも3回繰り返した。中期スプレッドをスライド上で調製し、標準的なGバンド染色技術を用いて染色した。核型の完全性は、解析した中期の少なくとも80%に正常核型が観察された(一解析当たり最低20中期細胞)ことに基づいて、人類染色体国際命名規約(ISCN)に従って決定した。
細胞をリン酸緩衝生理的食塩水(PBS)で洗浄し、TrypLE SelectまたはTrypLE Express(Thermo Fisher Scientific) のいずれかを使用して解離させ、次いでヒトES細胞培地中で、Hoechst333422(Sigma−Aldrich)10μg/mLを用いて37℃で30分間染色した。遠心分離の後、細胞は、15%のKSRおよび10μMのROCK阻害剤Y−27632を含有するPBSに再懸濁し、70μmのセルストレーナー(Corning)を通して濾過し、BD FACSAria IIIまたはBD Influx(BD Biosciences)のいずれで405nmレーザーを使用して選別した。連続増殖のために、10μMのROCK阻害剤Y−27632を含有する新鮮培地で選別細胞を24時間平地培養した。比較解析のために、G1期細胞は、同質遺伝子一倍体濃縮培養物と、選別していない二倍体培養物から選別した。培養中で比較的最近(一倍体細胞濃縮から3継代以内)二倍体化した細胞を、一倍体濃縮培養物中のG2/M期ピークを選別することによって分離し、選別していない二倍体培養物からのG2/M期二倍体細胞と比較した。一倍体濃縮培養物もG2/M期一倍体とG1期二倍体の混合集団からなることに留意されたい。選別純度は、少量の選別試料を装置に再度流すことによって確認した。
すべてのDNA量プロファイルは、Hoechst33342染色とともに、フローサイトメトリーに基づいて作成した。一倍体細胞比率は、1c細胞のパーセンテージと、細胞周期の他の期に対するG1期細胞の相対的寄与とに基づいて推定した。メタノール固定細胞およびリボヌクレアーゼ処理細胞においてヨウ化プロピジウムとともにフローサイトメトリーを用いて各継代で2〜3反復のDNA量を解析することによる、連続濃縮ラウンド間の一倍体細胞の比率、ならびに7継代(30日間)にわたるh−pES10二倍体化動態の実験解析に基づいて、二倍体化率を推定した。二倍体化率は、データを指数関数的減衰曲線に適合させることによって推定した。DNA量および細胞表面分子の同時フローサイトメトリー解析については、細胞を洗浄し、解離させ、10μg/mLのHoechst 33342(Sigma−Aldrich)およびコンジュゲート抗体、または一次抗体とともに60分間インキュベートした後に1:200に希釈した二次抗体のいずれかの存在下で、氷上で30分間インキュベートした。DNA量および細胞内PDX1の同時フローサイトメトリー解析については、解離細胞を免疫蛍光手法に記載のように、DNA染色のためのHoechst33342を用いて処理した。一次抗体は、表3に詳述する。すべてのフローサイトメトリー手法において、試料は、70μmセルストレーナー(Corning Life Sciences)を通して濾過し、BD FACSAria IIIまたはBD Influx(BD Biosciences)で解析した。
他で記載されているように39、DNA FISHをヒト2番および4番染色体用のプローブ、および4’,6−ジアミジノ−2−フェニルインドール(DAPI)によるDNA染色を用いて行った。一倍性と二倍性は、それぞれ、単一ハイブリダイゼーションシグナルまたは二重ハイブリダイゼーションシグナルに基づいて核ごとに判定した。FISHの対象となるES細胞は、解析に先立ち数継代にわたりマトリゲル被覆MATEKガラスプレート上で増殖させた。
アルカリホスファターゼ染色は、Leukocyte Alkaline Phosphataseキット(Sigma−Aldrich)を用いて行った。免疫蛍光染色については、試料をPBSで洗浄し、4%パラホルムアルデヒドで10分間固定し、次いでブロッキング溶液(0.1%のTriton X−100および5%のロバ血清をPBSに溶解)中で透過処理し、ブロックした。一次抗体(表3)と、ブロッキング溶液で1:500に希釈した二次抗体とともに細胞をインキュベートし、DAPIをDNA染色で用いた。固定化および各インキュベーションステップに続いて、細胞をPBSで2回洗浄した。Zeiss LSM510 Meta共焦点顕微鏡を用いて画像を撮った。セントロメア定量化は、Z軸に沿って個々の平面にわたるセントロメアフォーカスを手動で計数することによって行なった。EdU染色は、Click−iT EdU Alexa Fluor 488 Imagingキット(Thermo Fisher Scientific)を使用して行なった。図1gおよび図5dのセントロメア染色の対象となるES細胞は、解析に先立ち、数継代にわたりマトリゲル被覆MATEKガラスプレート上で増殖させた。
遺伝子トラップ変異体ライブラリーを作成するために、4〜5×106一倍体pES10細胞の9反復(1c−細胞濃縮後1継代以内)を、Bio−Rad Gene Pulser(800μLのOpti−MEMに懸濁、4−mmキュベット、320V、250μF)を使用して20μgの5’−PTK−3’遺伝子トラップベクター52と、20μgのpCyL43 piggyBacトランスポサーゼプラスミド53とともに同時遺伝子導入し、100×20mmディッシュ上にDR3 MEFとROCK阻害剤Y−27632とともに再播種した。発現遺伝子座への挿入の選択は、0.3μg/mLピューロマイシンを用いて、遺伝子導入から48時間後に開始し、続いて約16,000耐性コロニーによって代表される単一ライブラリーにプールした。5’−PTK−3’だけによる遺伝子導入を陰性対照として用いた。6−TG−耐性変異体をスクリーニングするために、変異体ライブラリーをDR4 MEF上で6μMの6−TG(Sigma−Aldrich)の存在下で18日間増殖させ、この間6つの耐性コロニーを独立して分離し、特性を明らかにした。ゲノムDNAを抽出し(NucleoSpin 組織キット, MACHEREY−NAGEL)、前述のように54、splinkerettePCRを使用して、挿入部位を検出し、続いてPCR産物の精製とSanger配列決定(ABI PRISM 3730xl DNA Analyzer(Applied Biosystems))を行った。UCSC BLAT検索ツールを使用して、配列をヒトゲノム(GRCh38/hg38)にマップした。
全DNAは、NucleoSpin組織キット(MACHEREY−NAGEL)を使用して分離した。全RNAは、Qiagen RNeasyキットを使用し、メーカーのプロトコルに従って分離した。細胞当たりの全RNAレベルを判定するために、一倍体細胞および二倍体細胞を同じ培養物から、それぞれ1c(一倍体G1)集団と4c(二倍体G2/M)集団を選別することによって分離した。2継代にわたる増殖後、細胞を収取して、計数し、各細胞株および倍数性状態から400,000細胞を3つ組で分離した(pES10とpES12、一倍体と二倍体;合計12試料)。NanoDropを使用してRNA量を定量化した。
コピー数多型解析(CNV)をInfinium Omni2.5Exome−8 BeadChip一塩基多型(SNP)アレイ(Illumina)を使用し、メーカーのプロトコルに従って、G1期で選別した一倍体と二倍体のpES10細胞およびpES12細胞のDNA試料(表4)で行った。生データをGenome Studio Genotyping Module(Illumina)を使用して処理し、R統計プログラミング言語を使用する解析のためにlogR比率値を求めた。
全RNA試料(200ng−1μg、RNA完全性数(RIN)>9)をポリ(A)+RNAのプルダウンによってmRNAのために濃縮した。RNAーSeqライブラリーをTruSeq RNA Library Prepキット第2版(Illumina)を使用し、メーカーのプロトコルに従って調製し、次いでIllumina NextSeq 500を使用して 配列決定し、85bpの単一末端リードを作成した。
RNA−Seqリードを、TopHat(第2.0.8b版)を使用してヒト基準ゲノム(GRCh37/hg19)に整列させ、5つのミスマッチを認めた。マップした100万フラグメントごとのキロベース当たりのリード(RPKM)値を、Cuffquantを使用して定量化し、Cufflinks(第2.1.1版)のCuiffnormを使用して正規化し、相対的な遺伝子発現レベルを作成した。ピアソン相関および平均連結を使用してRPKM値を階層的クラスタリング解析した。一倍体と二倍体のヒトES細胞(各群につきn=4)中の全RNAと比較して差次的遺伝子発現の解析を以下のように2つの相補的戦略によって行った。第1に、有意差としてFDR<0.05を用い、2倍未満の差異を考慮して、デフォルトパラメータでCuffdiffを使用した。第2に、おそらくわずかだが一貫した転写の相違を識別するために、特定の群のすべての反復にわたる最小発現レベルが他の群のすべての反復にわたる最大発現レベルより高かった遺伝子に対して検査をした。次いで、統計的有意性を両側独立スチューデントt検定によって判定した。機能アノテーション濃縮解析をDAVID(Benjamini法を用いて統計的有意性を決定する)によって行った。インプリンティング解析には、表6に収載の75のヒトインプリント遺伝子(Geneimprintウェブサイトを参照されたい)が含まれていた。対照ES細胞株NYSCF1からのRNA−Seqデータは、他で37発表されている(GEO受入れ番号GSE61657)。最低基準を1にし、発現閾値を1より上に設定することによって平均化した基準二倍体試料中に発現しなかった遺伝子を除去した後、ゲノムワイド遺伝子発現の移動中央値プロットを、Rパッケージzoo(第1.7−12版)を使用して作成した。異なる組織からのRNA−Seqデータは、Genotype−Tissue Expression(GTEx)ポータル40から検索した。図4dの色分けスケールは、組織にわたる平均(左側スケール)およびES細胞二倍体とEB試料の各セットにわたる平均(右側スケール)に対する遺伝子発現レベルに対応する。発現マイクロアレイ解析をこれまでのように41、Affymetrix Human Gene1.0STアレイを用いて行った。
これまでに記載のように37、Infinium HD Methylationプロトコルに従ってInfinium HumanMethylation450 BeachChips(Illumina)を使用して、表4に詳述した試料からのゲノムDNAをDNAメチル化解析した。対照ES細胞株NYSCF1からのDNAメチル化データは、以前に公表されている(GEO受入れ番号GSE61657)37。データは、アレイ正規化(SWAN)内のサブセット四分位点を用いて、処理し、正規化し、次いでRパッケージChAMP(第1.4.0版)を使用してバッチ効果を調整した。多能性特異的遺伝子およびiDMRに関連したCpG部位でのDNAメチル化レベルは、前述のように解析した37。X染色体上のDNAメチル化レベルの解析のために、0.4未満の平均β値のプローブを除去した。DMR解析は、デフォルト設定を用いてChAMPのラッソ機能によって容易になった。次いで、DMRを近似によって遺伝子に割り当て、機能アノテーション濃縮をDAVID(Benjamini方法を用いて統計的有意性を決定する)によって解析した。
G1期の一倍体および2倍体の細胞集団の選別後、生存可能な単細胞の直径(2r)をCountess Automated Cell Counter(Invitrogen)で測定し、その表面積と体積をそれぞれ4πr2と4/3πr3として算出した。解析は、1n pES10、1n pES12、2n pES10および2n pES12に対してそれぞれ、7、4、8および4回の技術的反復を包んだ。
他に記載されているように42、mtDNAの相対的存在量を、ミトコンドリア遺伝子ND2のためのプライマー(順方向プライマー:5’−TGTTGGTTATACCCTTCCCGTACTA−3’(配列番号1);逆方向プライマー:5’−CCTGCAAAGATGGTAGAGTAGATGA−3’(配列番号2))を用いる定量的PCR(qPCR)によって解析し、核遺伝子BECN1のためのプライマー(順方向プライマー:5’−CCCTCATCACAGGGCTCTCTCCA−3’(配列番号3);逆方向プライマー:5’−GGGACTGTAGGCTGGGAACTATGC−3’E(配列番号4))を用いて核遺伝子に対して正規化した。解析は、Applied Biosystems 7300 Real−Time PCR SystemとともにPerfeCTa SYBR Green FastMix(Quanta Biosciencesを使用して行なった。解析は、表4に詳述のすべてのG1期選別試料を包含した(各群につきn=4、各細胞株対して生物学的反復2)。
EB分化は、EDTAを含まないトリプシン溶液A(Biological Industries)でES細胞コロニーを分離させ、続いて再懸濁させて、低接着プレート上のbFGFを含まないヒトES細胞培地中で細胞凝集体をさらに培養した。一倍体ES細胞の分化は、1c−細胞濃縮後2継代以内に開始した。21日後に、解離および10μg/mLHoechst33342(Sigma−Aldrich)を用いた37℃で30分間の染色後、G1期の選別していないおよび/または選別したEB細胞からEB RNAを抽出した。0.2%のゼラチン上に播種し、bFGFを含まないヒトES細胞培地中で伸長した、解離EB細胞で中期スプレッド解析を行った。
NCAM1陽性ES細胞由来のNPCを、わずかに改変した効率的な神経分化43のための10日間プロトコルを使用して得た44。分化は、1c−細胞濃縮後2継代以内に開始した。RNAは、G1期の一倍体NCAM1−陽性細胞を、Hoechst33342および1:200に希釈した抗ヒトNCAM−1/CD56一次抗体とCy3−コンジュゲート二次抗体(Jackson Immunoresearch Laboratories)とで同時染色することによって抽出した。
神経への分化は、修正したSMADシグナル伝達46の相乗阻害に基づいて、公表されているプロトコル45に従って以下のように行なった。分化は、マトリゲル被覆プレートでmTeSR1培地で培養された完全にコンフルエントなES細胞による1c細胞濃縮から2継代以内に、該培地を、bFGFを含まず、10μMのSB431542(Selleckchem)と2.5μMのLDN−193189(Stemgent)とを含有するヒトES細胞培地と交換して、開始し、4日間続けた。続いて、細胞を、10μMのSB431542と2.5μMのLDN−193189とを補充したN2培地45にさらに4日間保持し、続いてB−27(Thermo Fisher Scientific)と、10μEMのDAPT(Stemgent)とを補充したN2培地に2日間保持した。次いで、細胞を解離させ、0.01%のポリ−L−オルニチン(Sigma−Aldrich)およびラミニン被覆(4μg/mL、ThermoFisher Scientiic)プレート上に、10μMのROCK阻害剤Y−27632(Selleckchem)の存在下で再播種し、さらにY−27632を含まない同じ培地で続いて4日間培養した。神経培養物は、20日目の免疫染色とFISHによる解析まで、B−27と20ng−1 BDNF(R&D)を補充したN2培地で維持した。
マトリゲル被覆プレート(Corning)上のmTeSR1(STEMCELL Technologies)培地で増殖した80〜90%コンフルエントES細胞は、それぞれCHIR99021とIWP−2(Selleckchem)を用いるGSK3とWNTの連続阻害に基づく11日間のレジメン47を受けた。分化は、1c−細胞濃縮後、2継代以内に開始した。分化の11日目に、免疫染色のために1c−細胞を選別し、播種した。
ここで利用するプロトコルは、いくつかの最近の刊行物に基づいて開発した48〜50。フィーダーレス条件下で増殖したES細胞をSTEMdiff Definitive Endodermキット(Stemcell Technologies)を使用して3〜4日間胚体内胚葉に分化させた。組換え型ヒトKGF/FGF7(R&D Systems)、LDN−193189(Stemgent)、KAAD−シクロパミン(Stemgent)およびレチノイン酸(Stemgent)による処理を含む段階的プロトコルによって、以降の特異化を達成した。8〜11日目、EGF(R&D Sysem)を使用して、膵臓前駆細胞(PPC)を誘導した。分化は、1c−細胞濃縮後、2継代以内に開始した。
動物におけるすべての実験手順は、ヘブライ大学の倫理委員会の承認を得た。ES細胞をトリプシン処理し、約2×106細胞を100μLのヒトES細胞培地および100μLのマトリゲル(BD Biosciences)に再懸濁し、続いてNOD−SCIDII2rg−/−免疫不全マウス(Jackson Laboratory)に皮下注射した。注射から8〜12週間後、腫瘍を切開し、さらなる解析にかけた。組織学的スライドは、Leica CM1850クリオスタット(Leica Biosystems、10μm切片)を用いて、O.C.T.コンパウンド(Sakura Finetek)に凍結保存された腫瘍スライスから調製し、続いて免疫染色、ヘマトキシリンとエオシン染色、またはFISH解析を行なった。Hoechst33342染色によるフローサイトメトリーは、新たに切開した腫瘍からの解離細胞で行った。
本発明者らは、セントロメアタンパク質蛍光免疫染色に基づく一細胞解像度で倍数性を決定するための方法を考案した。各染色体は通常、セントロメアを1つ有するので、セントロメアを検出し、数え上げることができれば、個々の細胞での倍数性を可視化する手段がもたらされ、一方で識別のマーカーを共染色することによって細胞独自性を定義することが可能になると判断した。
一倍体細胞の持続のために、14の初期継代(継代≦9)ヒトpES細胞株の収集を作成し、解析した。すべての細胞株は、第二極体放出と単一前核を示している活性化卵母細胞に由来した。最初に、希少一倍体核の明白な定量的発見のための方法として中期スプッレッドおよびG−バンド法による染色体計数を利用した。個々の10pES細胞株の中で、低比率の一倍体中期を単一細胞株、pSE10だけに見いだした(1.3%、表1)。染色体コピー数が2以下(2c)に対応するDNA量を有する細胞を追加の4株から分離することを目標として、Hoechst33342染色により生存可能なFACSも利用し、第2の細胞株、pES12(表2)からの一倍体細胞の濃縮に成功した。
ヒトES細胞が一倍体および二倍体としての両方で存在する能力によって、本発明者らはこれらの2つの倍数性状態が遺伝子制御および細胞生物学のいくつかの側面において異なる可能性があるかどうかを調査することにした。細胞周期の同じ期で一倍体と二倍体のES細胞を解析するために、FACSを用いてG1期一倍体細胞(1c)を分離し、次いでこれらの細胞を未選別の二倍体培養物からの同質遺伝子G1期二倍体細胞(2c)と比較した(図3a、図9a)。観察される発現レベルの違いが絶対差を表わすよりもむしろ、各倍数性状態の全遺伝子発現に対して相対的であることを考慮して、RNA配列決定(RNA−Seq)を用いて、一倍体ES細胞と二倍体ES細胞のトランスクリプトームを比較することによって、推定上の倍数性の関連する差異を明らかにすることを目標とした。ゲノムスケールで、未分化一倍体ES細胞と二倍体ES細胞は互いに密接に、および分化胚様体(EB)とは別々にクラスター形成し、遺伝的背景の影響を超えて広がる類似点を示している(図3b)。それにもかかわらず、合計565の差次的に発現した遺伝子が識別され(>2倍の変化、偽陽性率(FDR)<0.05)、二倍体と比較して、一倍体において相対的に増加した遺伝子275および相対的に減少した遺伝子290に相当した(図9b)。
次に、単為発生由来の一倍体ヒトES細胞の分化能の評価を追求した。哺乳動物の単為発生が親ゲノムの非同等性のため制限されているが22、23、二倍体ヒト単為発生多能性幹細胞は、すべての胚系を生じさせるその能力によって明らかなように機能的に多能性である13、24、25。ヒト単為発生ES細胞が一倍体として多系列分化ができるかどうかに取り組むために、いくつかの分化アッセイを行い、続いて得られた細胞の倍数性と分化状態の特徴づけを行った、一倍体濃縮ES細胞と二倍体ES細胞の同時分化によって作製したEBは、21日目に外見から区別することができなく(図4a)、EB由来の解離一倍体細胞の形態は分化と一致していた(図10a)。特に、中期スプレッド解析により、一倍体核型が明らかになり(図4b、4/4中期)、および大部分のDNAプロファイル(約70%一倍体)は、h−pES10由来EB細胞とh−pES12由来EB細胞の両方がフローサイトメトリーによって確認された(図4c、図10b)。次いで、G1期選別一倍体ES細胞とEB細胞の遺伝子発現プロファイルを比較し、8種類の組織と多能性幹細胞にわたり明確な特異性を示した18の遺伝子に着目した。例えば、我々の遺伝子セットは、それぞれ脳、皮膚、筋肉、腎臓、肝臓、膵臓、肺および腸管で発現される、CHRM1(コリン受容体)、KRT17(ケラチン)、MYL1(ミオシン)、REN(レニン)、ALB(アルブミン)、CPA1(カルボキシペプチダーゼ)、SFTPD(界面活性剤)およびMALRD1(MAMおよびLDL受容体)を含んでいた(図4d)。これらの系統特異的遺伝子の発現が未分化ES細胞でごくわずかだったのに対して、すべてが一倍体と二倍体のEB細胞において発現された(図4d、図6c)。加えて、一倍体と二倍体のEB細胞は、3つの胚性胚葉すべての効率的な分化および体細胞運命の獲得に一致する、多能性特異的遺伝子のわずかな発現を示した。

Claims (31)
- 3つの胚性胚葉すべての一倍体体細胞へと分化可能な一倍体ヒトES細胞の濃縮集団を作製する方法であって、
a)ES細胞の集団からの試料中の一倍体中期細胞を識別することであって、前記ES細胞が人為的に活性化されたヒト卵母細胞に由来する、一倍体中期細胞を識別すること、および
b)一倍体ヒトES細胞の濃縮集団を作製するために細胞倍数性に基づいて前記ES細胞の集団からの前記一倍体中期細胞を選別すること
を含む、方法。 - 前記ES細胞の濃縮集団を培養中で少なくとも3継代の間維持することをさらに含む、請求項1に記載の方法。
- 前記試料中の前記一倍体中期細胞が中期スプレッド解析によって識別される、請求項1に記載の方法。
- 前記試料中の前記一倍体中期細胞がフローサイトメトリー、セントロメアタンパク質免疫蛍光染色またはDNA蛍光インサイチューハイブリダイゼーションによって識別される、請求項1に記載の方法。
- 前記選別ステップが蛍光活性化細胞選別(FACS)を少なくとも1サイクル含み、任意に前記選別ステップが2〜5サイクルのFACSを含み、かつ前記一倍体ヒトES細胞の濃縮集団が、実質的に純粋な一倍体ヒトES細胞集団である、請求項1に記載の方法。
- 前記一倍体体細胞は、神経細胞、心筋細胞、膵細胞、皮膚細胞、筋細胞、腎臓細胞、肝細胞、肺細胞および腸管細胞からなる群から選択される、請求項1に記載の方法。
- 前記一倍体体細胞は、成熟した神経である、請求項6に記載の方法。
- 3つの胚性胚葉すべての一倍体体細胞へと分化可能なin vitroでの一倍体ヒト胚性幹(ES)細胞の濃縮集団。
- 前記一倍体体細胞は、神経細胞、心筋細胞、膵細胞、皮膚細胞、筋細胞、腎臓細胞、肝細胞、肺細胞および腸管細胞からなる群から選択される、請求項8に記載の濃縮集団。
- 前記一倍体体細胞は、成熟した神経である、請求項9に記載の濃縮集団。
- 前記濃縮集団が少なくとも95%の一倍体ヒトES細胞を含む、請求項8〜10のいずれか1項に記載の濃縮集団。
- 請求項8〜11のいずれか1項に記載の一倍体ヒトES細胞の濃縮集団を含む、組成物。
- 前記濃縮集団が少なくとも95%の一倍体ヒトES細胞を含む、請求項1に記載の方法。
- 請求項8〜11のいずれか1項に記載の一倍体ヒトES細胞の濃縮集団を含む、分離された一倍体ヒトES細胞株。
- 請求項14に記載の一倍体ヒトES細胞株を作製する方法であって、
a)請求項1〜7のいずれか1項に記載の方法によって一倍体ヒトES細胞の濃縮集団を作製すること、
b)培養中で一倍体ヒトES細胞の濃縮集団を維持すること、および
c)培養中の前記ES細胞を3〜4継代ごとに選別することであって、前記選別することが細胞倍数性に基づく、選別すること、
それによって、一倍体ヒトES細胞株を作製することを含む、方法。 - 一倍体体細胞へと分化可能なin vitroでの一倍体多能性ヒト細胞の集団。
- 前記一倍体体細胞は、神経細胞、心筋細胞、膵細胞、皮膚細胞、筋細胞、腎臓細胞、肝細胞、肺細胞および腸管細胞からなる群から選択される、請求項16に記載の集団
- 前記一倍体体細胞は、成熟した神経である、請求項16または17に記載の集団。
- 前記一倍体多能性ヒト細胞の集団は、実質的に純粋な集団である、請求項16〜18のいずれか1項に記載の集団。
- 前記一倍体多能性ヒト細胞の集団は、内胚葉前駆細胞、中胚葉前駆細胞、外胚葉前駆細胞、または神経前駆細胞を含む、請求項16〜19のいずれか1項に記載の集団。
- 請求項16〜20のいずれか1項に記載の前記一倍体多能性ヒト細胞の集団を含む組成物。
- 請求項16〜20のいずれか1項に記載の前記一倍体多能性ヒト細胞の集団を作製する方法であって、
a)定方向分化の条件下で、3つの胚性胚葉すべての一倍体体細胞へと分化可能な一倍体ヒト胚性幹細胞を培養すること、
b)胚様体形成を誘導する条件下で、3つの胚性胚葉すべての一倍体体細胞へと分化可能な一倍体ヒトES細胞を培養すること、および胚様体を細胞に解離させること、または
c)テラトーマ形成を誘導する条件下で、3つの胚性胚葉すべての一倍体体細胞へと分化可能な一倍体ヒトES細胞を非ヒト哺乳動物に注入すること、および前記テラトーマを細胞に解離させること、
それによって、一倍体多能性ヒト細胞の集団を作製することを含む、方法。 - 細胞表面マーカーに基づいて解離細胞を選別することをさらに含む、請求項22に記載の方法。
- 培養中の分化した一倍体ヒト体細胞の集団。
- 前記分化した一倍体体細胞は、神経細胞、心筋細胞、膵細胞、皮膚細胞、筋細胞、腎臓細胞、肝細胞、肺細胞および腸管細胞からなる群から選択される、請求項24に記載の集団。
- 前記分化した一倍体体細胞は、成熟した神経である、請求項25に記載の集団。
- 請求項24〜26のいずれか1項に記載の前記分化した一倍体ヒト体細胞の集団を含む組成物。
- 請求項24〜26のいずれか1項に記載の前記分化した一倍体ヒト体細胞の集団を作製する方法であって、定方向分化の条件下で3つの胚性胚葉すべての一倍体体細胞へと分化可能な一倍体ヒトES細胞または一倍体体細胞へと分化可能な一倍体多能性ヒト細胞を培養すること、それによって、分化した一倍体ヒト体細胞の集団を作製することを含む、方法。
- 遺伝学的スクリーニングの方法であって、
a)細胞中で少なくとも1つの変異を誘導するために3つの胚性胚葉すべての一倍体体細胞へと分化可能なヒト一倍体ES細胞の濃縮集団を変異原に曝すこと、
b)前記変異を含有する前記濃縮集団においてヒト一倍体ES細胞を選択すること、および
c)前記ヒト一倍体ES細胞において前記変異の遺伝子型効果および/または表現型効果を識別すること
を含む、方法。 - 前記遺伝学的スクリーニングが順遺伝学的スクリーニングである、請求項29に記載の方法。
- 前記変異原が物理的変異原、化学的変異源および生物由来物質からなる群から選択される、請求項29に記載の方法。
