発明の分野
本発明は、精製されそして単離されている新規のヒト胸腺間質リンホポエチン(lymphopoietin)(TSLP)ポリペプチドおよびその断片、こうしたポリペプチドをコードする核酸、こうしたポリペプチドの組換え型を産生するための方法、これらのポリペプチドに対し生成された抗体、これらのポリペプチドに由来する断片化ペプチド、およびその使用に関する。
B細胞発生は詳細に研究されてきているが、造血幹細胞から成熟B細胞につながる経路には、いまだに空白がある。サイトカインがB細胞発生および増殖に影響を与え、そして重要な役割を果たすことが認識されている。B細胞発生に影響を与える既知のサイトカインには、IL−2、IL−4、IL−5、IL−6、IL−7、IFN−ガンマ、および顆粒球マクロファージコロニー刺激因子(GM−CSF)が含まれる。

好ましい配列

1つ以上のコドンが同一のアミノ酸をコードする可能性がある、遺伝暗号の既知の縮重のため、DNA配列は配列番号1に示されるものと異なり、そしてなお配列番号2のアミノ酸配列を有するポリペプチドをコードする可能性がある。こうした変異体DNA配列は、沈黙(silent)突然変異(例えば、PCR増幅中に発生する)から生じてもよいし、または天然配列の意図的な突然変異誘発の産物であってもよい。
本発明は、多様な型のポリペプチドおよびその断片を含み、天然発生である、または組換えDNA技術を伴う方法などの多様な技術を通じ産生されるものを含む。こうした型には、限定されるわけではないが、誘導体、変異体、およびオリゴマーと共に、融合タンパク質またはその断片が含まれる。
本発明のポリペプチドは、上述のような核酸配列にコードされる全長タンパク質を含む。特に好ましいポリペプチドは、配列番号2のアミノ酸配列を含み、特に好ましい断片は、配列番号2のアミノ酸29ないし159(成熟ポリペプチド配列)を含む。
一般的に、可溶性型の使用は特定の適用に有益である。可溶性ポリペプチドは宿主細胞から分泌されるため、組換え宿主細胞からのポリペプチドの精製が容易になる。さらに、可溶性ポリペプチドは、一般的に、静脈内投与に、より適している。
天然発生変異体と共に、ポリペプチドおよび断片由来変異体が本明細書に提供される。
変異体は、少なくとも80%同一であるアミノ酸配列を示してもよい。やはり意図されるのは、ポリペプチドまたは断片が、好ましいポリペプチドまたはその断片に、少なくとも90%同一、少なくとも95%同一、少なくとも98%同一、少なくとも99%同一、または少なくとも99.9%同一であるアミノ酸配列を含む、態様である。同一性パーセントは、視覚的検査および数学的計算により決定してもよい。あるいは、2つのタンパク質配列の同一性パーセントは、NeedlemanおよびWunsch(J. Mol. Bio. 48:443(1970))のアルゴリズムに基づき、そしてウィスコンシン大学遺伝学コンピューターグループ(UWGCG)より入手可能なGAPコンピュータープログラムを用い配列情報を比較することにより、決定してもよい。GAPプログラムの好ましいデフォルトパラメーターには:(1)HenikoffおよびHenikoff(Proc. Natl. Acad. Sci. USA, 89:10915(1992))に記載されるような、スコアリング・マトリックス、blosum62;(2)12のギャップ加重;(3)4のギャップ長加重;および(4)末端ギャップに対するペナルティなし、が含まれる。当業者に用いられる、配列比較の他のプログラムもまた、用いてもよい。
本発明に含まれるのは、ヒトTSLPポリペプチドを含むオリゴマーまたは融合タンパク質である。こうしたオリゴマーは、二量体、三量体、またはより高次のオリゴマーを含む、共有結合または非共有結合多量体型であってもよい。上述のように、好ましいポリペプチドは可溶性であり、そしてしたがってこれらのオリゴマーは可溶性ポリペプチドを含んでもよい。本発明の1つの側面において、オリゴマーはポリペプチド構成要素の結合能を維持し、そしてしたがって、二価、三価などの結合部位を提供する。
1つの代替物として、免疫グロブリンに由来するポリペプチドを用い、オリゴマーを調製する。抗体由来ポリペプチドの多様な部分(Fcドメインを含む)に融合している特定の異種性(heterologous)ポリペプチドを含む融合タンパク質の調製は、例えば、Ashkenaziら(PNAS USA 88:10535(1991));Byrnら(Nature 344:677(1990));並びにHollenbaughおよびAruffo(Current Protocols in Immunology中, “Construction of Immunoglobulin Fusion Proteins”, Suppl.4, 10.19.1−10.19.11(1992))に記載されている。
あるいは、オリゴマーはペプチドリンカー(スペーサーペプチド)を含むまたは含まない多数のポリペプチドを含む融合タンパク質である。適切なペプチドリンカーの中に、本明細書に援用される米国特許第4,751,180号および第4,935,233号に記載されるものがある。望ましいペプチドリンカーをコードするDNA配列は、いかなる適切な慣用的技術を用い、本発明のDNA配列の間に、そして該DNA配列と同じ読み枠で挿入してもよい。例えば、リンカーをコードする化学的に合成されたオリゴヌクレオチドを、配列間に連結してもよい。特定の態様において、融合タンパク質は、ペプチドリンカーにより分離された、2ないし4つの可溶性TSLPポリペプチドを含む。
本発明のオリゴマーを調製するための別の方法は、ロイシンジッパーの使用を伴う。ロイシンジッパードメインは、該ドメインが見られるタンパク質のオリゴマー化を促進するペプチドである。ロイシンジッパーは、元々、いくつかのDNA結合タンパク質で同定され(Landschulzら, Science 240:1759(1988))、そして以来、多様な異なるタンパク質で見出されてきている。既知のロイシンジッパーの中に、二量体化または三量体化する天然発生ペプチドおよびその誘導体がある。

発現系
本発明はまた、DNAを含む組換えクローニング用および発現ベクター、並びに、該組換えベクターを含む宿主細胞も提供する。DNAを含む発現ベクターを用い、該DNAにコードされる本発明のポリペプチドまたは断片を調製してもよい。ポリペプチドを産生するための方法は、ポリペプチドをコードする組換え発現ベクターで形質転換されている宿主細胞を、該ポリペプチドの発現を促進する条件下で培養し、その後培養から発現ポリペプチドを回収することを含む。当業者は、発現ポリペプチドを精製するための方法は使用する宿主細胞の種類、および該ポリペプチドが膜に結合しているかまたは宿主細胞から分泌される可溶性型かなどの要因にしたがい、多様であろうことを認識するであろう。

哺乳動物または昆虫宿主細胞培養系もまた、組換えポリペプチドを発現するのに使用してもよい。昆虫細胞において異種性タンパク質を産生するためのバキュロウイルス系がLuckowおよびSummers, Bio/Technology 6:47(1988)に概説されている。哺乳動物起源の樹立細胞株もまた、使用してもよい。適切な哺乳動物宿主細胞株の例には、サル腎臓細胞のCOS−7株(ATCC CRL 1651)(Gluzmanら, Cell 23:175(1981))、L細胞、C127細胞、3T3細胞(ATCC CCL 163)、チャイニーズハムスター卵巣(CHO)細胞、HeLa細胞、BHK(ATCC CRL 10)細胞株、およびMcMahanら(EMBO J. 10:2821(1991))に記載されるようなアフリカミドリザル(African green monkey)腎臓細胞株CV1(ATCC CCL 70)由来であるCV1/EBNA細胞株が含まれる。

単離および精製
本発明に含まれる「単離」ポリペプチドまたはその断片は、天然に見られる可能性がある環境と同一でない環境にあるポリペプチドまたは断片である。本発明に含まれる「精製」ポリペプチドまたはその断片は、例えば上述のような組換え発現系の精製産物のように、または天然発生細胞および/または組織などの非組換え供給源からの精製産物のように、他のタンパク質またはポリペプチドとの結合を本質的に含まない。
本発明の精製ポリペプチド(タンパク質、ポリペプチド、断片、変異体、オリゴマー、および他の型を含む)を、慣用的な結合アッセイなど、いかなる適切なアッセイにおいて、TSLP受容体に結合する能力に関し試験してもよい。例として、ポリペプチドを検出可能試薬(例えば、放射性核種、発色団、比色もしくは蛍光分光反応を触媒する酵素など)で標識してもよい。標識ポリペプチドを、TSLP受容体を発現している細胞と接触させる。細胞をその後、洗浄し、非結合標識ポリペプチドを除去し、そして細胞に結合している標識の存在を、標識の性質にしたがい選択される適切な技術により、決定してもよい。

以下に示すように、ネズミ遺伝子のシンテニー解析(syntenic analysis)により、配列番号1は染色体第5番のq21−q22領域にマッピングされている。したがって、当業者は、周知の技術を用い、ガードナー症候群、線腫様結腸ポリポーシス、遺伝性類線維腫疾患、ターコット症候群、および結腸直腸癌を含む、ヒト染色体第5番、および特に染色体第5番のq21−q22領域に関連する異常を解析するのに、配列番号1の核酸またはその断片を用いてもよい。これにより本マーカーが再配置または欠失されている状態を識別することが可能になる。さらに、配列番号1のヌクレオチドまたはその断片は、未知の位置の他の遺伝子をマッピングする、位置マーカーとして用いてもよい。

本発明のポリペプチドは、タンパク質精製試薬として使用を見出す。例えば、該ポリペプチドは、TSLP結合パートナー、例えばヒトTSLP受容体を精製するのに用いてもよい。特定の態様において、ポリペプチド(TSLP受容体に結合することが可能な、本明細書に記載されるいかなる型でもよい)を、慣用法により固体支持体に結合させる。1つの例として、タンパク質のアミノ酸側鎖上の官能基と反応するであろう官能基を含む、アフィニティークロマトグラフィーカラムが入手可能である(Pharmacia Biotech, Inc.、ニュージャージー州ピスカタウェイ)。あるいは、TSLPポリペプチド/Fcタンパク質(上述のようなもの)を、Fc部分との相互作用を通じ、プロテインAまたはプロテインG含有クロマトグラフィーカラムに結合させる。

研究用剤
本発明のポリペプチドの別の使用は、異なる細胞種上のTSLP/TSLP受容体相互作用を阻害することから生じる生物学的影響を研究するための研究ツールとしての使用である。ポリペプチドはまた、TSLPまたはTSLP受容体あるいはそれらの相互作用を検出するためのin vitroアッセイに使用してもよい。

損なわれていないタンパク質を用いる場合、これらのポリペプチド分子量マーカーの使用は、21,000ダルトンに近い見かけの分子量を有するタンパク質の見かけの分子量の決定の正確さの増加を可能にする。断片を用いる場合、断片の分子量の範囲にわたる分子量決定の正確さが増加する。
上述のように、ポリペプチドまたはペプチドフィンガープリントを、既知のタンパク質のデータベースに入力しまたは該データベースと比較し、質量分析を用いた未知のタンパク質の同定を補助してもよい(W.J. Henzelら, Proc. Natl. Acad. Sci. USA 90:5011−5015(1993);D. ヒトTSLP核酸配列は、EST IMAGEクローン1407260、寄託番号AA889581を配列決定することにより、得た。ネズミTSLP配列との比較により、本配列は、ESTクローンが部分的クローンであることを示唆した。内部プライマーを用い、いくつかのcDNAライブラリーをスクリーニングし、TSLP cDNAクローンの失われている3’端を得るために用いることが可能なcDNAの供給源を決定した。ヒトTSLP配列の2つの内部プライマーを用いた60サイクルのPCRの後、以下のcDNAライブラリーがTSLP配列に関し陽性であった:ヒト精巣、ヒト包皮線維芽細胞、および胎児脳(弱陽性);一方、全MoT、HS431、骨髄、HPT4、HBT3、W126、Hut102、PBT、Sk Hep、ヒト真皮線維芽細胞、Raji、ヒト胎盤、およびKBライブラリーはすべて陰性であった。
TSLP特異的ELISA:
TSLP含有試料の連続希釈(NaOHでpH 9にした50 mM NaHCO3中)で、Linbro/Titertek 96ウェル平底E.I.A.マイクロタイトレーションプレート(ICN Biomedicals Inc.、オハイオ州オーロラ)を、100:1/ウェルで被覆する。4℃で16時間インキュベーションした後、200:1の0.05% Tween−20を含むPBS(PBS−Tween)でウェルを6回洗浄する。その後、5%ウシ胎児血清(FCS)を含むPBS−Tween中、1μg/mlのFLAG(登録商標)−TSLP受容体と、ウェルを90分間インキュベーションし(ウェル当たり100:1)、その後、上述のように洗浄する。次に、各ウェルを、抗FLAG(登録商標)(5% FCSを含むPBS−Tween中、1μg/mlのモノクローナル抗体M2)と、ウェルを90分間インキュベーションし(ウェル当たり100:1)、その後、上述のように洗浄する。続いて、ポリクローナルヤギ抗mIgG1特異的西洋ワサビペルオキシダーゼ結合抗体(5% FCSを含むPBS−Tween中、商業的ストックの1:5000希釈)と、ウェルを90分間インキュベーションする(ウェル当たり100:1)。HRP結合抗体は、Southern Biotechnology Associates, Inc.、アラバマ州バーミンガムから得る。その後、ウェルを上述のように6回洗浄する。
ヒトTSLPのアミノ酸配列は、完全ヒトTSLPヌクレオチド配列の翻訳により、決定した。選択された読み枠は、ヒトTSLPとネズミTSLPの相同性に基づいた。
ヒトTSLP核酸配列は、EST IMAGEクローン1407260、寄託番号AA889581、およびクローン19F由来のさらなる3’配列の合成配列の標準的二本鎖配列決定により、決定した。
本実施例は、TSLPに結合するモノクローナル抗体を調製するための方法を例示する。こうした抗体を生成するのに使用してもよい適切な免疫原には、限定されるわけではないが、精製ヒトTSLPポリペプチドまたはその免疫原性断片、例えば細胞外ドメイン、またはヒトTSLPを含む融合タンパク質(例えば可溶性TSLP/Fc融合タンパク質)が含まれる。
ヒトTSLP mRNAの組織分布を、以下のように、ノーザンブロット解析により調べた。放射標識プローブのアリコットを、2つの異なるヒト多数組織ノーザンブロット(Clontech、カリフォルニア州パロアルト;Biochain、カリフォルニア州パロアルト)に添加した。ブロットを、10 X Denhardts、50 mM Tris pH 7.5、900 mM NaCl、0.1% ピロリン酸Na、1% SDS、200μg/ml サケ精子DNA中でハイブリダイズした。ハイブリダイゼーションは、先に記載されるように(Marchら, Nature 315:641−647(1985))、50%ホルムアミドで、63℃で一晩行った。その後、ブロットを、2 x SSC、0.1% SDSで、68℃で30分間、洗浄した。
全長ヒトTSLPを発現させ、そしてTSLP受容体に結合する能力に関し、試験してもよい。結合アッセイは、以下のように行うことが可能である。

- (a)配列番号2に示すアミノ酸配列からなるポリペプチド;および
(b)配列番号2のアミノ酸29ないし159、又は35ないし159のアミノ酸配列からなるポリペプチド;
からなる群より選択される、胸腺間質リンホポエチン(TSLP)のポリペプチドに特異的に結合する単離抗体又は抗体断片。 - 配列番号2に示すアミノ酸配列からなるポリペプチドに特異的に結合する、請求項1記載の単離抗体又は抗体断片。
