Description
したがって、この走化性運動のメカニズムを解明し、さらに走化性運動を調節する内在性因子や人工的化合物を得ることで、走化性運動が関与する様々な現象を人為的にコントロールできることが期待される。すなわち、白血球の運動を調節することによって免疫系をコントロールする薬剤や、炎症反応をコントロールする薬剤を得ることが期待できる。また、走化性運動の阻害剤によって、がん細胞の浸潤や転移を抑制することも期待できる。
しかしながら、現在、走化性運動メカニズムの詳細は不明であり、また、走化性運動を人為的にコントロールできる薬剤の開発は進んでいない。
わかったが、DIF-2は、DIF-1の分解産物ではないうえ、DIF-1とは別の経路で合成されることもわかった(文献2、3、5)。
したがって、DIFs発見以来20年あまり経過した現在でも、DIF-2特異的な生理的機能は、それがあるのかどうかも含め、まったくわかっていなかった。さらに、DIF-1に関しても、分化誘導以外の役割があるのではないかと推測されていたが、それは未解明であった。
これらの成果より、DIF誘導体が様々な薬理活性を有することが期待されるが、がん細胞の浸潤・転移に対する効果は検討されていなかった。
本発明はまた、前記走化性運動制御剤を含むがん転移阻害剤に関する。
本発明の走化性運動制御剤は、式(I)、(II)で表される化合物またはその薬学的に許容される塩を有効成分とする。
薬製剤の製造法で一般的に用いられている公知の手段に従って、該化合物またはその薬学的に許容される塩を、そのまま、あるいは薬理学的に許容される担体と混合して、例えば、錠剤(糖衣錠、フィルムコーティング錠を含む)、散剤、顆粒剤、カプセル剤、(ソフトカプセルを含む)、液剤、注射剤、坐剤、徐放剤等の医薬製剤として、経口的または非経口的(例、局所、直腸、静脈投与等)に安全に投与することができる。
式(I)、(II)の化合物またはその塩のがん細胞の浸潤・転移阻害剤中の含有量は、
製剤全体の約0.01ないし約100重量%である。
式(I)、(II)の化合物またはその塩の投与量は、投与対象、対象臓器、症状、投与
方法などにより異なり特に制限されないが、一般的に、患者(体重60kgとして)に対して、一日につき約0.1〜100mg、好ましくは約1.0〜50mg、より好ましく
は約1.0〜20mgである。
、D−マンニトール等が挙げられる。緩衝剤としては、例えばリン酸塩、酢酸塩、炭酸塩、クエン酸塩等の緩衝液等が挙げられる。無痛化剤としては、例えばベンジルアルコール等が挙げられる。防腐剤としては、例えばパラヒドロキシ安息香酸エステル類、クロロブタノール、ベンジルアルコール、フェネチルアルコール、デヒドロ酢酸、ソルビン酸等が挙げられる。抗酸化剤としては、例えば亜硫酸塩、アスコルビン酸、α−トコフェロール等が挙げられる。
なお、本発明の走化性運動制御剤はその他の薬剤と併用してもよい。
細胞性粘菌Ax2株(野生株)細胞を栄養存在下で十分な数にまで増殖させた後、飢餓処理(6時間)後、図2に示すアッセイ系を用いて、DIFsの存在下および非存在下において、様々な濃度のcAMP(粘菌の走化性誘導因子)に対する走化性運動の有無を検討した。
がん細胞の遊走運動(浸潤と転移)は、走化性運動によるものと考えられている。そこで、次に、哺乳類がん細胞の遊走に対するDIFsの効果を検討した。
図5に示すように、上段のウェルに膵臓がんYAPC細胞を入れ、下段ウェルに100 nMのリゾフォスファチジン酸(LPA:遊走因子)を添加し、DIFs(5-10 μM:薬理学的濃度を使用)の効果を比較検討した(図6)。なお、ControlとしてDIFsの溶剤 (0.1% EtOH)の効果も検討した。

- 下記式(I)で表される化合物またはその薬学的に許容される塩を有効成分とする、がん細胞の浸潤・転移阻害剤。
- 下記式(II)で表される化合物またはその薬学的に許容される塩を有効成分とする、がん細胞の浸潤・転移阻害剤。
- 式(I)で表される化合物が下記のいずれかの化合物である、請求項1に記載のがん細胞の浸潤・転移阻害剤。
