MDA−MB−231ヒト胸部癌細胞を、アメリカ培養細胞系統保存機関(ATCC、Manassas、VA)から得た。ピューロマイシンに耐性のヒト卵巣癌細胞をトランスフェクトしたMLS−pRSETB−mBananaは、Michal Neeman教授(Department of Biological Regulation、Weizmann Institute of Science、Rehovot、イスラエル)のご好意で提供された。
トランスフェクトには2つのプラスミド:ネオマイシンに対する耐性遺伝子を保有するpDsRed2−N1(Clontech、Palo Alto、CA)(図1A)、及びハイグロマイシンに対する耐性遺伝子を保有する、修飾型pDsRed−Monomer−Hyg−C1(Clontech、Palo Alto、CA)を用い、これらにおいて、図1Bにおいて示すプラスミドのDsRed−Monomer遺伝子をpDsRed2で置換した(pDsRed2−N1プラスミドから、図1C)。トランスフェクトにはLipofectamine(商標)2000(Invitrogen(商標))を、製造元のプロトコールに従って用いた。
MDA−MB−231−RFP細胞を、1mmol/Lピルビン酸ナトリウム、10%ウシ胎児血清(FCS)、250μg/mlハイグロマイシン、0.06mg/mlペニシリン、及び0.1mg/mlストレプトマイシンを補ったRPMI1640培地中に維持した。細胞を、加湿雰囲気中(5%CO2、95%空気)37℃で単層として増殖させた。MLS−mBanana細胞を、1mmol/Lピルビン酸ナトリウム、10%ウシ胎児血清(FCS)、10μg/mlピューロマイシン、0.06mg/mlペニシリン、及び0.1mg/mlストレプトマイシンを補ったMEM−α培地中に維持した。細胞を、加湿雰囲気中(5%CO2、95%空気)37℃で単層として増殖させた。
メスCD1ヌードマウス(7〜8週齢、約25g)を収容し、Institutional Animal Care and Use Committee of the Weizmann Institute of Science (Rehovot、イスラエル)のガイドライン(1996年)に従って動物施設中、餌及び水を自由に摂取させて飼い慣らした。
MDA−MB−231−RFP蛍光ヒト胸部癌細胞、及びMLS−mBanana蛍光ヒト卵巣癌細胞(食塩水100μl中5×106個)をトリプシン処理し、サブコフルーエンシー(subcofluency)で回収し、次いでメスマウスの背部又は乳房の脂肪パッド中に接種した。腫瘍を2つの望ましいサイズ−壊死性腫瘍(約1cm3、3〜4週間以内)及び非壊死性腫瘍(約0.5cm3、1〜2週間以内)に発達させた。
マウスに、85:15ケタミン:キシラジンの混合液30μlをi.p.注射することによって麻酔した。乳房の脂肪パッドにおける薬物の蓄積をモニターするために、15mg薬物/体重kgの化合物13、化合物25、又は化合物24をマウスの尾部静脈にi.v.注射した。赤色蛍光タンパク質(RFP)、pRSETB−mBanana、及び光増感剤の蛍光を、in vivoの光学画像化システム、IVIS(登録商標)100(Xenogen Corp.、Alameda、CA)によってモニターした。腫瘍画像化用のメインフィルターセットは、500〜550nmの励起フィルター及び575〜650nmの発光フィルターを含んでおり、組織の自己蛍光を差し引くためのバックグラウンドフィルターセットは460〜490nmの励起フィルター及び575〜650nmの発光フィルターを含んでいた。薬物画像化のメインフィルターセットは665〜695nmの励起フィルター及び810〜875nmの発光フィルターを含んでいた。同じ曝露時間の間に画像を得、定性的な比較を可能にするために同じ直線上のカラースケール上に図示する。
全身のin vivo画像からの関心領域(ROI)である腫瘍境界をマーキングし、円で囲んだ境界内の蛍光シグナルを光子/秒で表した。同じROIを用いて側副側における光子/秒を測定した。さらに、バックグラウンド測定用に、腫瘍及び側副側のROIを、3匹の非処置マウスで測定し、平均した。各ROIからの蛍光の測定値を面積で除して、光子/秒/cm2における標準化された蛍光シグナル強度を得た。化合物13注射15分後から216時間後まで、シグナル測定値を回収した。各時間点において、バックグラウンドを差し引き、腫瘍境界と側副側の間における蛍光の比として平均を計算した。
マウスに過剰(8.5μmol)の遊離c(RGDfK)ペプチドを注射し、1時間後に化合物13(140nmol)を注射した。対照群には化合物13(140nmol)だけを注射した。化合物13注射24時間後に蛍光画像を撮影した。蛍光画像化メインフィルターセットを、(vii)に記載した通りに用いた。
マウスの尾部静脈に化合物13 15mg/kgをi.v.注射した。マウスを屠殺し、腫瘍を切除し、半分に切断し、XenogenIVIS(登録商標)システムを用いて様々な時間間隔:MDA−MB−231−RFPに対して10分、1時間、4時間、及び24時間、3日、5日、及び7日、MLS−mBananaに対して7日画像化した。蛍光画像化に用いたフィルターセットは(vii)に記載されている。
切除実験後、腫瘍を3.7%ホルムアルデヒド中で固定し、パラフィンブロック中に包埋した。切片を、標準条件下、ヘマトキシリン−エオシン(H&E)で染色した。
麻酔したマウスに、化合物13 7.5mg/kg又は15mg/kgをi.v.注射した。腫瘍を10分間又は30分間照射した。薬物の光線の間隔は、薬物注射後8時間又は24時間であった。100mW/cm2の755nmダイオードレーザー(CeramOptec、ドイツ)での経皮照射を用いた。暗所の対照群では、マウスに薬物をi.v.注射し、暗所のケージに24時間配置した。明所の対照群では、マウスに薬物を注射せず、100mW/cm2で10分間照射した。PDT後最初の2日間、マウスに鎮痛薬を適宜投与した(毎日2.5mg/kgフルネキシン(Flunexin))。
蛍光タンパク質での腫瘍細胞のトランスフェクト
トランスフェクト方法を行って、安定してRFPを発現する細胞系を作り出した。このような細胞系は、in vivo及びin vitro(組織/細胞)で、蛍光顕微鏡及び他の蛍光画像化手段によって検出することができる。自発的な中央壊死を産生することが知られているヒト胸部癌MDA−MB−231細胞系が、この目的に選ばれた。2つのプラスミド(図1A、1C)を用い、安定なクローンを得、蛍光顕微鏡によって検出した(図2A〜2Bを参照されたい)。修飾したpDsRed−Monomer−Hyg−C1プラスミド(図1C)から産生したクローンは、強力な蛍光を表した。修飾したpDsRed−Monomer−Hyg−C1でトランスフェクトしたクローン3(図2B)が、さらなる使用に選ばれた。
壊死性腫瘍モデル−組織病理学的分析
MDA−MB−231−RFP細胞が適切な腫瘍モデルを産生することを立証するために、MDA−MB−231−RFP腫瘍を2つのサイズに発達させた。材料と方法、セクション(xi)に記載した通り、組織学的及び組織病理学的分析を行った。結果を図3A及び3Bに示す。約1cm3の大型の腫瘍は非常に著しい壊死領域を示し(図3A)、約0.5cm3の小型の腫瘍は壊死領域を示さなかった(図3B)。
原発性壊死性MDA−MB−231−RFP異種移植片腫瘍における化合物13の取込みのin vivoの蛍光画像化
in vivoの壊死性MDA−MB−231−RFP腫瘍(≧1cm3)における化合物13の蓄積パターンを試験した。図4A〜4B及び5A〜5Bは、XenogenIVIS(登録商標)システムを用いて、メスCD−1ヌードマウスの乳房パッドにおける、同所性ヒト胸部MDA−MB−231−RFP原発腫瘍における化合物13の蛍光シグナルの蓄積を例示するものである。上記材料と方法、セクション(vii)に記載したフィルターセットを用いて、全体の動物の画像を同時に記録した。1〜1.5時間ごとに9時間、及び化合物13注射24時間後(図4A〜4B)、次いで24時間ごとに次の7日間(図5A〜5B)にダイナミック蛍光画像を獲得した。化合物13の注射直後、動物全身からNIR蛍光を検出することができ、循環における薬物が高濃度であることを反映していた。肝臓における、及びある程度は腫瘍における蓄積に伴う循環からの急速な排除が、注射後最初の9時間に観察された(図4B)。その後日、化合物13は腫瘍に蓄積し続け、肝臓からは完全に排除され、≧3日から注射後7日のフォローアップ期間の終わりまで、選択的な腫瘍の画像化がもたらされ(図5B)、その後の排除は極めて遅かった。腫瘍のサイズ及び位置は、in vivoの全身画像の赤色によって見たところ、実験を通して変化しなかった(図4A及び5A)。同様の結果が、≧1cm3の腫瘍サイズの実験動物9匹に観察された。
原発性非壊死性MDA−MB−231−RFP異種移植片腫瘍における化合物13の取込みのin vivoの蛍光画像化
同様のパラメータを、次に、非壊死性である≦0.5cm3の腫瘍において試験した。図6A〜B及び7A〜7Bは、MDA−MB−231−RFPを移植したメスCD−1ヌードマウスにおける化合物13及びRFPからの蛍光シグナルを表す。薬物蓄積のパターンを、上記に記載したIVIS(登録商標)システムを用いて、及び例3におけるのと同様の時間間隔で画像化したが、その時間までに薬物が完全に排除されるので3日に限定した。これらの非壊死性腫瘍における蓄積パターンは、壊死性腫瘍に観察されたものと著しく異なっていた。化合物13のNIR蛍光は、注射後約2時間の腫瘍において、及び直後の肝臓において(薬物注射後3.5時間)ピーク値に到達した(図6B)。壊死性腫瘍からの化合物13の分解された蛍光とは対照的に、注射2日後には非壊死性腫瘍から実際に蛍光は観察され得なかった(図7A)。動物16匹からの蛍光の測定値の平均において、薬物注射1〜6.5時間後に腫瘍の蛍光のピークが検出された。
MDA−MB−231−RFP壊死性腫瘍における化合物13の取込み及び排除の動力学
壊死性腫瘍における化合物13の蓄積パターンを半定量的に評価するために、マウス9匹の壊死性腫瘍における化合物13に対する蛍光シグナルの平均を計算し、216時間にわたって、いくつかの時間点でプロットした(図8)。注射後12時間からその先、腫瘍からの蛍光シグナルは、側副側上の等しい対照領域の蛍光シグナルに比べて特徴的に強力になった。腫瘍と側副側の間の蛍光の比率はやがて増大し、192時間から約8のプラトーに到達した。
同所性MDA−MB−231−RFP原発性壊死性異種移植片腫瘍における化合物24の取込み
壊死性原発性MDA−MB−231−RFP腫瘍における、メタル化したコンジュゲート化合物24(c(RGDfK)−Pd−MLT)のin vivoの蓄積パターンを、例3に記載した同じセットの実験を用いて試験した。
同所性MDA−MB−231−RFP原発性非壊死性異種移植片腫瘍における化合物24の取込み
例6に示したのと同様の時間間隔を用いて、上記例4に記載したメスCD−1ヌードマウス上に移植したMDA−MB−231−RFP非壊死性腫瘍(約0.5cm3)において、化合物24及びRFPからの蛍光シグナルをさらに試験したが、その時間までに腫瘍から薬物が完全に排除されるので2日に限定した。図11A〜11B及び12A〜12Bに表した結果は、化合物13に関して、非壊死性腫瘍における蓄積パターンが、壊死性腫瘍に観察された蓄積パターンと著しく異なっていたことを実証している。化合物24のNIR蛍光は、注射後約2.5時間の腫瘍において、及び直後の肝臓において(薬物注射5.5時間後、図11B)ピーク値に到達した。壊死性腫瘍からの化合物24の蛍光の解像とは対照的に、非壊死性腫瘍から注射24時間後に実際に蛍光は観察できなかった(図12B)。ピーク腫瘍蛍光(動物5匹からの蛍光の平均測定値)は、薬物注射1〜4.5時間後に検出された。

上記の実験において得られた結果の普遍性を確立するために、壊死領域における化合物13の蓄積を、MLS−mBananaヒト卵巣癌細胞系から産生した様々な腫瘍タイプにおいてさらに試験した。図13A〜13B及び14A〜14Bに示す結果は、材料と方法、セクション(vii)に記載したXenogenIVIS(登録商標)システムを用いて、メスCD−1ヌードマウスの乳房パッド中に移植した皮下(s.c.)ヒト卵巣MLS−mBanana原発腫瘍における化合物13からの蛍光シグナルの蓄積を図示するものである。蓄積のパターンを、本明細書上記例3に記載したのと同様の時間間隔でモニターしたが、その時間までに薬物は完全に排除されるので4日に限定した。注射直後、化合物13のNIR蛍光は、肝臓及び腫瘍から大部分検出された。肝臓及び腫瘍における蓄積に伴う循環からの迅速な排除が、注射後最初の8時間に生じる(図13B)。2日後、化合物13は腫瘍中に蓄積し続け、一方、肝臓からは殆ど排除され、周囲の干渉のない、腫瘍の選択的な画像化がもたらされた(図14B)。MLS−mBanana壊死性腫瘍からの排除は、MDA−MB−231−RFP壊死性腫瘍からの排除よりも著しく速く、3日目には殆ど完了した。腫瘍のサイズ及び位置は、赤色のin vivoの全身の画像によって見られるように、実験を通して変化しなかった(図13A及び14A)。これらの結果は、≧1cm3の腫瘍サイズの動物3匹において観察された。
マウス乳房パッド中に埋め込まれたMLS−mBanana原発性非壊死性腫瘍における化合物13の取込み
長時間の薬物の蓄積を示さない、非壊死性MDA−MB−231−RFP腫瘍における結果に鑑みて、MLS−mBanana非壊死性腫瘍における長時間の蓄積を試験した。図15A〜15B及び16A〜16Bに示した結果は、MLS−mBanana非壊死性腫瘍における化合物13からの蛍光シグナルを表している。蓄積のパターンを、材料と方法、セクション(vi)に記載した通りに、例4に記載したのと同様の時間間隔を用いてモニターしたが、薬物はその時間までに殆ど完全に排除されるので4日に限定した。非壊死性腫瘍における蓄積パターンは、壊死性腫瘍における蓄積パターンといくらか類似していた。化合物13のNIR蛍光は、注射後約1時間の腫瘍においてピーク値に到達した(図15B)。肝臓における蓄積は、最初の1時間から検出された。
化合物13蓄積のc(RGDfK)部分に対する依存
化合物13の腫瘍の取込み、及び得られたNIR蛍光シグナルの蓄積はRGD部分によって駆り立てられるか否かを決定するために、2つの実験を行った。第1の実験において、化合物13注射後、様々な時間の蛍光シグナルの蓄積を、乳房パッドにMDA−MB−231−RFP非壊死性(≦0.5cm3)及び壊死性(約1cm3)腫瘍を移植したCD−1ヌードマウスに、遊離2H−MLT(化合物25)を注射した蛍光シグナルの蓄積と比べた。第2の実験は、化合物13と遊離c(RGDfK)の間の競合アッセイであった。
壊死性腫瘍及び非壊死性腫瘍の両方に対して、化合物25を最初に注射して1〜1.5時間ごとに8.5〜9時間、及び24時間(図18A〜18B、及び20A〜20B)、並びにその後3日間24時間ごとに(図19A〜19B及び21A〜21B)、ダイナミック蛍光画像を獲得した。化合物25のNIR蛍光シグナルは、任意の濃度を全て観察した場合、注射後5〜10分の腫瘍において最高濃度に到達した。蛍光シグナルの値は、(ずっと長い時間間隔で)化合物13に対して最大に観察された値よりも約2桁小さかった。蛍光シグナルは、注射後20分に、すでに有意でない値に低下した。一方、蛍光シグナルは、大部分肝臓中及び心臓中に蓄積した。同じ挙動が、壊死性腫瘍を有する動物3匹(図18B)、及び非壊死性腫瘍を有する動物2匹に(図20B)観察された。
特異的な結合を証明するために、遊離c(RGDfK)リガンドによって化合物13の蓄積を阻止しようという実験を行った。例3に記載したように、c(RGDfK)の事前の注射あり又はなしで、化合物13を投与した。結果を図22A〜22Dに実証する。化合物13を遊離c(RGDfK)投与後に投与した場合、腫瘍中の蓄積は24時間後に検出されなかったが(図22C)、対照群において、化合物13を単独投与した場合、腫瘍中の蓄積は検出可能であり(図22D)、c(RGDfK)部分による腫瘍に対する化合物13の特異的な結合を指摘していた。
MDA−MB−231−RFP壊死性腫瘍領域における化合物13の特異的な蓄積
乳房パッド中に埋め込まれた壊死性及び非壊死性の胸部腫瘍異種移植片の、著しく異なるダイナミック蛍光パターンは、腫瘍組織学と化合物13の蛍光シグナル蓄積の間の相関を調査する動機となった。そこで、化合物13を注射した後、示された時間に腫瘍を切除し、半分に切断した。材料と方法、セクション(vii)に記載したようにXenogenIVIS(登録商標)システムを用いて、いくつかの時間点に(注射後10分、1時間、4時間、及び24時間、3日、5日、及び7日)画像を撮影した。図23〜29に示した結果は、様々な時間点の化合物13の蛍光の蓄積を示している。各時間点に対して、動物3匹を試験した。薬物注射10分から4時間後まで、生存領域にのみ薬物の蛍光が観察された(図23〜25)。4時間では、壊死領域に向っていくらかの拡散があった。注射後3日から蓄積のパターンが復帰し、蛍光は壊死ゾーンに完全に移動し、周囲の生存細胞に向っていくらかの残りの蛍光の拡散があった(図27F)。これらの結果は、5日間及び7日間でも観察された(図28〜29)。重要なことに、注射後24時間に、壊死領域からの高い薬物の蛍光とともに特異的な腫瘍の蛍光がすでに明らかに観察されたが、腫瘍の境界は3日間隔と比べるとそれほど鋭く規定されていなかった(図26F)。このセットの実験において、時間の進行とともに化合物13は生存領域を通って壊死領域中に移動し、長時間そこに特異的に蓄積することが示された。
MDA−MB−231−RFP壊死性腫瘍領域における化合物24の特異的蓄積
化合物13に対して観察された蓄積のパターンに鑑みて、他のBchl誘導体を試験した。化合物24の蓄積のパターンをモニターし、得られた同様の結果が得られた。薬物注射9日後に切除した腫瘍に対する結果を図30A〜30Fに示す。画像を、上記に記載したとおりXenogenIVIS(登録商標)システムを用いて撮影した。図30Fに見られるように、薬物注射9日後、薬物は腫瘍の壊死領域中に明らかに存在した。これらの結果は、実験した動物全てに(N=3)観察された。薬物注射3日後及び5日後に腫瘍を切除した場合に、同様の結果が観察された(各時間点に対してN=3)。
MLS−mBanana壊死性腫瘍領域における化合物13の特異的な蓄積
MDA−MB−231−RFP腫瘍の壊死領域中に観察された化合物13及び化合物24両方の蓄積はこの現象の普遍性、並びに治療及び画像化におけるこの潜在的な適用を試験する動機付けをもたらした。そこで、様々なタイプの腫瘍−MLS−mBananaヒト卵巣癌を用いた。ここでは結果はいくらか異なっていた。ほとんどの場合、このタイプの腫瘍における壊死パターンは中央ではなく、即ち腫瘍の中央に特異的に位置せず、又は腫瘍において一箇所から産生されない、広範囲に広がる壊死が存在した。これらの場合、蓄積は長期間ではなかった。中央壊死が観察されたいくつかの場合において、蓄積パターンはMDA−MB−231−RFP腫瘍における蓄積パターンと同様であった。図31〜32は薬物注射7日後に得られた結果を示す。見られるように、中央壊死が観察された場合、薬物は腫瘍の壊死領域に明らかに存在し(図31F)、中央壊死が存在しない場合は存在しなかった(図32F)。
切除した腫瘍の画像及び壊死性腫瘍の組織学的切片
MDA−MB−231−RFP壊死性腫瘍を切除し、腫瘍の組織学的切片を染色した(ヘマトキシリン及びエオシン(H&E))。図33A〜33Dは、薬物注射4時間後に切除した腫瘍の、腫瘍生存領域及び壊死領域の組織学的切片を示す。これらの所見は、先に得られたRFP及び化合物13の組織分布を補完しなければならない。図33Aに見られるように、塊のバルク(balk)は、不透明で、褐色の壊死性組織からなる。図33Bから、肉眼と顕微鏡の特徴の間の相関に注目することができる。中央の壊死組織は好酸性から過好酸性であり、核溶解が広範で、核濃縮及び核崩壊は少なかった。壊死組織中への多核好中球の軽度の浸潤が、殆ど壊死領域の境界に存在した。生存領域は腫瘍の周囲に限られていた。これらは、高密度の細胞シートに整列された新生物細胞の無秩序な増殖からなっていた(図33D)。新生物細胞は、丸型から不規則であり、核:細胞質比が高く、小胞の核が不規則であった。注射4時間後の化合物13の蛍光は、生存領域及び壊死領域の両方に対して、組織学的結果に良好に相関した(図25F)。薬物を注射しなかった対照の腫瘍を含めて、試験した壊死性腫瘍全てに対して同様の相関が得られた。
化合物13を用いたin vivoのPDT試験
一般的に、in vivoのPDT試験に対して、上記に記載したようにメスマウスの背部又は乳房の脂肪パッド上に蛍光MDA−MB−231−RFP胸部癌細胞又はMLS−mBanana卵巣癌細胞を皮下的に接種することによって、マウス腫瘍モデルを産生し、壊死(≧1cm3)又は非壊死(約0.5cm3)のサイズに増殖させる。マウスを麻酔し、7.5〜15mg/kgの間の様々な濃度の薬物をi.v.注射する。腫瘍を10分間又は30分間照射し、薬物の光線の間隔は薬物注射後4〜24時間の間である。
ヌードマウス/ラットの乳房における局所性非浸潤性乳管癌(DCIS)モデルの確立
いくつかの細胞系を用いて、マウス及びラットにおけるDCISモデルを確立する。第1に、F−344ラットと同系であるMADB106細胞を、ラットの乳房パッドに上首尾に(同所性に)以前に埋め込んでおいた。このモデルは、これまでに、様々なRGD−Bchl誘導体を有するPDTの有効性、及びその後の抗腫瘍の長時間の免疫の発症をスクリーニングするために用いられている。ラット乳房癌腫瘍に対して同じプロトコールを用いて、2つのヒト細胞系及びさらなるマウス細胞系を同所性に埋め込み、肺及びリンパ節における転移を得る。hT47D及びHCC1395の最初の2つは、ATCC規定及び文献(Gazdarら、1998年)に従って増殖され、ヌード動物の乳房パッドに対する同種移植片として注射される管癌細胞系(HCC1395は、DCISにできるだけ近いステージ1の原発性乳管癌である)である。第3(4T1)は、細胞を尾静脈に注射して数週間後に肺の転移を産生するマウス乳房細胞系である。この様々な細胞系により、本発明者らは、胸部癌モデルとして、乳房癌の原発病変、局所的に再発したもの、及び遠隔性の転移に対する(Bchl誘導体/Bchl−RGD)−PDTの効果を研究できる。4T1細胞にルシフェラーゼ及び他の2つの細胞系をトランスフェクトする。pDsRed1−C1での4T1のトランスフェクトは現在進行中である。このような蛍光により、(1)Bchl−RGDベースの蛍光又はMRIを用いた検出の正確さの評価、(2)非常に高い感度での無処置の動物におけるBchl−PDT下の腫瘍の増殖及び退縮のオンラインのモニタリングが可能である。
一般概念としての壊死蓄積概念の確立
発生時に中央壊死を発生する腫瘍タイプが存在する。このような腫瘍の2つが、以下の細胞系から発生した腫瘍から試験するのに選択されている:ヒトDCIS MCF7胸部癌、ヒトDCIS MCF10DCIS(Taitら、2007年)、ヒトグリア芽細胞腫U87、ヒト炎症性胸部癌(IBC)WIBC−9(Shirakawaら、2001年)、及びRCC。
様々なRGD部分にコンジュゲートした化合物25の蓄積パターン
RGDターゲティングの関連性をさらに確立するために、様々なRGDペプチド及び陰性対照を用いて、MDA−MB−231−RFP腫瘍における蓄積のパターンを試験する。
壊死領域における他のBchl−RGD誘導体の蓄積
壊死における蓄積パターンが、治療及び/又は画像化の潜在性を有するBchl−RGD誘導体全てに対する一般的パラダイムであるか否かを調べるために、さらなるBchl−RGD誘導体を試験する。具体的には、化合物15(c(RGDfK)−Cu−MLT)及び化合物14(c(RGDfK)−Mn−MLT)誘導体を試験する。
体内分布アッセイ
この実験は、薬物の伝播及び身体の様々な器官における蓄積の経過を定量し、実際に薬物が最終的に腫瘍中に蓄積する普遍的な方法で実証することを目的とする。
画像化及び治療用の、放射性同位元素とのメタレートしたRGD−バクテリオクロロフィル誘導体
別の治療オプションは、薬物の中央の金属を放射性の金属で置き換えることである。Mの寿命が(治療に対して)比較的長いRGD−M−Bchl誘導体などの蓄積は、腫瘍の放射線治療に用いることができる。薬物が身体中に低濃度で留まるが、壊死領域中に次第に蓄積するような間隔で、薬物を投与することもできる。

- 腫瘍壊死領域の前記画像化が、
(a)壊死領域を有する腫瘍を有することが疑われる対象に、請求項1に記載のコンジュゲートを投与するステップであって、Mは2H、又は金属PdあるいはZnであるステップ、及び
(b)対象に照射し、コンジュゲート投与後少なくとも24〜48時間の間に1〜8時間の時間間隔で、疑われる領域の蛍光を測定するステップであって、24〜48時間後又はそれより長い時間蛍光を示す領域は前記腫瘍の壊死領域の存在を示すステップを含む、
ダイナミック蛍光画像化である、請求項1に記載の薬理組成物。 - 腫瘍壊死領域の診断のための前記画像化が、
(a)壊死領域を有する腫瘍を有することが疑われる対象に、請求項1に記載のコンジュゲートを投与するステップであって、Mは、64Cu、67Cu、99mTc、67Ga、201Tl、195Pt、60Co、111In、又は51Crからなる群から選択される放射性同位元素であるステップ、及び
(b)コンジュゲート投与後少なくとも24〜48時間の間に1〜8時間の時間間隔で、画像化スキャナーにおいて対象をスキャンし、疑われる領域の放射線レベルを測定するステップであって、24〜48時間後又はそれより長く放射線を示す領域は前記腫瘍の壊死領域の存在を示すステップを含む、
放射線診断技術である、請求項1に記載の薬理組成物。 - 腫瘍壊死領域の診断のための前記画像化が、
(a)壊死領域を有する腫瘍を有することが疑われる対象に、請求項1に記載のコンジュゲートを投与するステップであって、Mは、Mn3+、Cu2+、Fe3+、Eu3+、Gd3+、又はDy3+から選択される常磁性金属であるステップ、
(b)前記投与前(時間ゼロ)の患者体内の標的関心領域の少なくとも1つのMR画像、及び前記投与の少なくとも24〜48時間後、好ましくは96時間後の第2の時点又はそれを超える時間点の1つ又は複数のMR画像を産生することによって患者に磁気共鳴画像化を施すステップ、及び
(c)データを加工及び分析して前記腫瘍の壊死領域の存在又は非存在を診断するステップを含む、
分子磁気共鳴画像(MRI)化である、請求項1に記載の薬理組成物。 - (i)ダイナミック蛍光画像化に用いられるコンジュゲートが31−オキソ−15−メトキシカルボニルメチル−ロドバクテリオクロリン(Rhodobacteriochlorin)131−(2−スルホエチル)アミド−173−c(RGDfK)アミドカリウム塩、又はパラジウム31−オキソ−15−メトキシカルボニル−メチル−ロドバクテリオクロリン131−(2−スルホエチル)アミド−173−c(RGDfK)アミドカリウム塩であり、腫瘍が乳腺又は卵巣の腫瘍であり、壊死領域が薬物注射後3〜8日後に視覚化され、
(ii)前記放射線診断技術がポジトロン放出断層撮影法(PET)(ここで、Mが64Cu又は67Cuである)又は単一光子放射型コンピュータ断層撮影法(SPET)(ここでMが、99mTc、67Ga、195Pt、111In、51Cr、及び60Coからなる群から選択される放射性同位元素である)である、請求項10〜12のいずれか一項に記載の薬理組成物。 - 局在性の胸部癌、特に非浸潤性乳管癌(DCIS)のための低侵襲の処置、検出、及び予後のための請求項1に記載の薬理組成物であって、
(i)前記コンジュゲートが、腫瘍壊死領域に特異的に向い、蓄積する、請求項1に記載の、RGD含有ペプチド及びバクテリオクロロフィル誘導体のコンジュゲートであり;
(ii)MRI、蛍光、及びPET SCAN方法によって高い精度で患者の腫瘍を検出予知し、腫瘍マージンの画定し;そして
(iii)胸部温存及び再建を可能にする局在性の壊死領域が腫瘍標的光線力学的治療(PDT)により治療される;
薬理組成物。
